Designing of inhibitors against drug tolerant Mycobacterium tuberculosis (H37Rv).

BACKGROUND: Mycobacterium tuberculosis (M.tb) is the causative agent of tuberculosis, killing ~1.7 million people annually. The remarkable capacity of this pathogen to escape the host immune system for decades and then to cause active tuberculosis disease, makes M.tb a successful pathogen. Currently available anti-mycobacterial therapy has poor compliance due to requirement of prolonged treatment resulting in accelerated emergence of drug resistant strains. Hence, there is an urgent need to identify new chemical entities with novel mechanism of action and potent activity against the drug resis

Skin tolerability associated with transdermal drug delivery systems: an overview.

As transdermal patches become more widely prescribed, it is important that clinicians understand: (a) the common causes of skin reactions with these medications; (b) how to minimize these reactions; and (c) how to manage the signs and symptoms. Here we review published data for skin reactions with patch medications approved within the past decade. Overall, the most common application site signs and symptoms appear to be localized redness (erythema) or itching, sometimes accompanied by swelling (edema). Typically, these are mild to moderate in severity, transient in nature, and occur in 20% to

Comparative tolerability of drug therapies used to treat incontinence and enuresis.

Drug therapy for incontinence and enuresis has met with varying degrees of success. Currently, there is no medication available that specifically targets the lower urinary tract without having untoward effects elsewhere in the body. Patients with urge incontinence are the most difficult group to treat. The agents most commonly used to treat urge incontinence, i.e. anticholinergics, musculotropics and tricyclic antidepressants, are limited in their effectiveness and have anticholinergic adverse effects. Other medications with theoretical treatment potential such as nonsteroidal anti-inflammator

The ocular tolerability of a new ophthalmic drug delivery system (NODS).

In an open label, two period cross-over study we compared the ocular tolerability of NODS vs. Isopto-NaturaleR eye drops. Thirty healthy volunteers were asked to rate the application system they were using for ocular discomfort, utilizing a visual analog scale in which 100 = no ocular discomfort and 0 = severe ocular discomfort. In addition each subject had to answer a questionnaire and underwent a full ophthalmic control before, at day 7, at day 14 and at day 21 of the study. Each application form had to be applied to one eye only once a day for 7 days, followed by a washout period of 1 week.

Mitochondrial metabolic-transcriptional axis drives adaptive drug tolerance across major lung cancer subtypes.

Drug resistance remains the first and most significant barrier to long-term curative benefits in lung cancer patients. Traditional resistance mechanisms focused on oncogenic driver mutations are inadequate to explain reversible tumor phenotypic plasticity under therapeutic stress. Increasing evidence has placed mitochondrial metabolic rewiring as the core signaling hub bridging energy homeostasis to nuclear epigenetic reprogramming; however, a comparative review of this metabolic-transcriptional axis in lung adenocarcinoma (LUAD), lung squamous cell carcinoma (LUSC), and small cell lung cancer

Cannabis and tolerance: acute drug impairment as a function of cannabis use history.

Cannabis use history as predictor of neurocognitive response to cannabis intoxication remains subject to scientific and policy debates. The present study assessed the influence of cannabis on neurocognition in cannabis users whose cannabis use history ranged from infrequent to daily use. Drug users (N = 122) received acute doses of cannabis (300 μg/kg THC), cocaine HCl (300 mg) and placebo. Cocaine served as active control for demonstrating neurocognitive test sensitivity. Executive function, impulse control, attention, psychomotor function and subjective intoxication were significantly worse

Bowman-birk proteinase inhibitor confers heavy metal and multiple drug tolerance in yeast.

Cultured Coptis japonica cells show tolerance to various toxic compounds. By yeast functional screening of cadmium (Cd) plates with its cDNA library, we isolated a gene encoding Bowman-Birk proteinase inhibitor (CjBBI). The yeast transformant of CjBBI showed multiple tolerance to various drugs adding to Cd, and revealed reduced Cd accumulation in cells. Preferential organs for Cjbbi expression were aerial parts of intact plants, and the subcellular localization of CjBBI was shown, using its green fluorescent protein fusion, to be the apoplast. Induction of Cjbbi expression by Cd treatment sugg

Albendazole treatment of echinococcosis in humans: effects on microsomal metabolism and drug tolerance.

We prospectively studied the effect of albendazole on microsomal reserve and on first-pass activation to albendazole sulfoxide in patients with hydatid disease. An aminopyrine breath test was performed in 12 patients while they were receiving albendazole treatment and while they were not. Excretion of 14CO2 in breath averaged 0.70%.kg.mmol-1 +/- 0.20%.kg.mmol-1 without treatment and 0.54%.kg.mmol-1 +/- 0.14%.kg.mmol-1 with treatment (p less than 0.005). Plasma levels of albendazole sulfoxide were measured 4 hours after the morning dose during the first and second half of the 4-week treatment c

Targeting drug-tolerant persister cells: Let's bring on the TROPs!

In this issue of Cancer Cell, Liao et al. identify TROP2 as an EGFR tyrosine kinase inhibitor-induced vulnerability in drug-tolerant persister lung cancer cells. They show that anti-TROP2 antibody-drug conjugates target this residual population, supporting an "induce-target-kill" strategy to delay resistance and extend the benefit of EGFR tyrosine kinase inhibition.

Pharmacological agents targeting drug-tolerant persister cells in cancer.

Current cancer therapy can be effective, but the development of drug resistant disease is the usual outcome. These drugs can eliminate most of the tumor burden but often fail to eliminate the rare, "Drug Tolerant Persister" (DTP) cell subpopulations in residual tumors, which can be referred to as "Persister" cells. Therefore, novel therapeutic agents specifically targeting or preventing the development of drug-resistant tumors mediated by the remaining persister cells subpopulations are needed. Since approximately ninety percent of cancer-related deaths occur because of the eventual developmen

Safety and tolerability of available urate-lowering drugs: a critical review.

INTRODUCTION: Urate-lowering therapy (ULT) is the cornerstone of gout management, which is a widespread chronic illness characterized by hyperuricemia, arthropathy, tophus development, and urolithiasis. Since asymptomatic increased serum urate levels are associated with a higher risk of cardiovascular, renal and metabolic disorders, a larger use of ULTs in the general population is expected in the near future. AREAS COVERED: This review will focus on the safety and tolerability profile of the available urate-lowering drugs: xanthine oxidase inhibitors (XOIs), uricosuric agents and injectable

Effectiveness and tolerability of antiepileptic drugs in 104 girls with Rett syndrome.

UNLABELLED: Approximately 60-80% of girls with Rett Syndrome (RTT) have epilepsy, which represents one of the most severe problems clinicians have to deal with, especially when patients are 7-12years old. The aim of this study was to analyze the antiepileptic drugs (AEDs) prescribed in RTT, and to assess their effectiveness and tolerability in different age groups from early infancy to adulthood. We included in this study 104 girls, aged 2-42years (mean age 13.9years): 89 had a mutation in MECP2, 5 in CDKL5, 2 in FOXG1, and the mutational status was unknown in the remaining 8. Epilepsy was pre

House of cellulose - a new hideout for drug tolerant Mycobacterium tuberculosis.

Mycobacterium tuberculosis (Mtb) causes tuberculosis (TB). The treatment of TB requires administration of multiple drugs for long durations because of the unusual drug tolerance of Mtb. The phenotypic drug tolerance of genetically drug-susceptible Mtb in humans can be explained by its ability to form biofilms. Recent studies from different laboratories suggest that Mtb forms biofilms that harbour drug-tolerant bacteria. These findings have created a new area of research in the field of mycobacterial physiology. Recently, my laboratory has reported that Mtb cells organise themselves into biofil

Tolerance of nonsteroidal antiinflammatory drugs in patients with inflammatory bowel disease.

OBJECTIVE: We sought to examine whether use of nonsteroidal antiinflammatory drugs (NSAIDs) in an outpatient inflammatory bowel disease (IBD) population is associated with an increased likelihood of active disease. METHODS: We reviewed records of initial outpatient visits of IBD patients to the principal author from June 1995 to December 1997, with regard to use of aspirin and other NSAIDs and disease activity. RESULTS: Of 40 Crohn's patients seen with active disease, three (7.5%) were using NSAIDs; 14 of 72 (19.4%) Crohn's patients seen with inactive disease were using NSAIDs. Fifty-eight u

[Influence of co-administered antiepileptic drugs on nitrazepam tolerance in mice].

In the treatment of epilepsy, benzodiazepines are often administered in combination with other antiepileptic drug (s) because of the development of tolerance. In this study, the influence of concurrently administered antiepileptic drugs on tolerance to the anticonvulsant action of the nitrazepam (NZP) was studied using an animal tolerance model. Mice were given vehicle, NZP alone or NZP concurrently with one of six antiepileptic drugs (carbamazepine CBZ, phenytoin PHT, zonisamide ZNS, vigabatrin VGB, lamotrigine LTG, or flunarizine FNR) twice daily for 5 days. Tolerance was assessed by the abi

GSTA1 Conferred Tolerance to Osimertinib and Provided Strategies to Overcome Drug-Tolerant Persister in EGFR-Mutant Lung Adenocarcinoma.

INTRODUCTION: The generation of drug-tolerant persister (DTP) cancer cells remains a major challenge in treating lung adenocarcinoma (LUAD) patients with EGFR tyrosine kinase inhibitors (TKIs), as these cells eventually drive drug resistance and disease progression. However, the mechanisms underlying DTP formation are poorly understood, limiting therapeutic options upon the emergence of DTP state or resistance after TKI therapy. METHODS: In this study, we analyzed samples from LUAD patients receiving frontline osimertinib therapy (including baseline, DTP, and stable resistance states) to diss

Drug-Tolerant, Chemiluminescent Lateral Flow Immunoassay Platform for the Determination of Neutralizing Anti-Drug Antibodies.

Administration of mAb therapeutics often elicits the production of neutralizing anti-drug antibodies (nADA) in patients. Current nADA assays are drug-sensitive and detect only free nADA, thereby overlooking drug-bound antibodies and reducing sensitivity. An extra preparatory step of acid dissociation is often applied to resolve the immunocomplexes and subsequently detect the resultant free nADA separately. This tedious sample preparation limits them to lab settings and exposes the antibodies to harsh conditions. Here, a modified lateral flow assay platform, namely sequential binding flow immun
