[Duck hepatitis B virus DNA in multiple primary cancers and metastatic tumors in duck liver].

Duck hepatitis B virus (DHBV) DNA was detected in both multiple primary cancers and intrahepatic metastatic tumors in duck liver by Southern blot technique. Among 7 ducks with multiple primary tumors in the liver, the banding pattern of integrated DHBV DNA in different tumors was identical in 3 animals but different in 2. One duck showed a similar hybridization pattern in different tumor nodules and the remaining one was negative for DHBV DNA. Integrated DHBV DNA was also identified in one duck with metastatic tumor in the lung, in which the hybridization pattern was the same as that of the pr

[Isolation from ducks of a hypervirulent strain of duck plague virus and an avian type 6 paramyxovirus].

This paper describes the isolation and identification of a duck plague virus (DP) and a paramyxovirus (PMV6), from the livers and intestines collected in 4-month old mule ducks, under fattening, exhibiting 75% mortality and necrotic-haemorrhagic gross lesions. These viruses were isolated in specific pathogen free (SPF) muscovy duck eggs and SPF chicken eggs respectively. Then the DP virus was adapted to duck and chicken fibroblasts. The disease was reproduced in 2-week old SPF muscovy ducklings, intramuscularly inoculated with the previous organs, as well as in contact ducks. From them, only t

Characteristics of the febrile response in Pekin ducks.

We measured body temperature in Pekin ducks for 22 h after intravenous injection of the lipopolysaccharide (LPS) of gram negative bacteria at doses of 0, 1, 10, and 100 micrograms.kg body mass (-1). The ducks developed monophasic fevers showing increases in peak temperature reached and duration of fever with increases in dose of LPS. Body temperatures of unrestrained telemetered ducks without access to food and water were similar to those of saline-injected controls in the fever experiments, but were lower in the morning than when the same birds had access to food and water. This nocturnal hyp

Intrapulmonary CO2-rise time and ventilation in ducks.

Many investigators have reported finding intrapulmonary chemoreceptors (IPCs) in several species of birds and reptiles; however, the role of IPCs in ventilatory control in birds has not been identified. This study was undertaken to assess the role of intrapulmonary CO2 dynamics on the control of breathing. Ducks were unidirectionally ventilated, and their breathing was monitored with a pneumotachograph connected to a personal computer. A gas-mixing system controlled by the computer adjusted the rate of rise of airway CO2 concentration ([CO2]). On inspiration, the computer removed CO2 from the

A Comparative Analysis of Transcriptome-Wide Differential Gene Expression and Alternative Polyadenylation in the Ovaries of Meat Ducks and Laying Ducks.

Significant differences in reproductive performance exist between meat-type ducks (e.g., Qiangying Duck, QD) and laying-type ducks (e.g., Shaoxing Duck, SD). The molecular mechanisms underlying these differences, particularly concerning ovarian development and function, remain incompletely understood. This study aimed to comprehensively characterize the ovarian transcriptomes of these two duck types, focusing on differential gene expression and post-transcriptional regulatory events. We performed an integrated full-length transcriptome analysis of ovarian tissues from these two breeds using Pa

Simultaneous Protective Immune Responses of Ducks against Duck Plague and Fowl Cholera by Recombinant Duck Enteritis Virus Vector Expressing Pasteurella multocida OmpH Gene.

Duck enteritis virus and Pasteurella multocida are major duck pathogens that induce duck plague and fowl cholera, respectively, in ducks and other waterfowl populations, leading to high levels of morbidity and mortality. Immunization with live attenuated DEV vaccine containing P. multocida outer membrane protein H (OmpH) can provide the most effective protection against these two infectious diseases in ducks. We have recently reported the construction of recombinant DEV expressing P. multocida ompH gene using the CRISPR/Cas9 gene editing strategy with the goal of using it as a bivalent vaccine

Transcriptional analysis of duck liver infected with duck hepatitis A virus type 1 isolate HA5.

Duck Hepatitis A Virus Type 1 (DHAV-1) is a major pathogen in ducklings, characterized by severe hepatomegaly and punctate hepatic hemorrhage. In this study, we investigated host gene expression dynamics in specific-pathogen-free (SPF) ducklings infected with the DHAV-1 isolate HA5 using high-throughput RNA sequencing (RNA-seq). We performed comprehensive transcriptomic analyses, integrating Clusters of Orthologous Groups (COG) classification, Gene Ontology (GO) annotation, and Kyoto Encyclopedia of Genes and Genomes (KEGG) pathway enrichment. By combining these data with viral replication kin

Pathogenicity difference analysis of novel duck reovirus NY01 between in semi-muscovy duck and shelduck.

Recently, the waterfowl industry of China has suffered significant economic losses due to the novel duck reovirus (NDRV), an emerging pathogen. Its characteristic pathological manifestations include marked splenomegaly with diffuse hemorrhagic necrosis, multi-organ congestion, and parenchymal cell degeneration. Clinically recovered ducks frequently exhibit persistent growth retardation syndrome. In this study, a wild-type strain of NDRV, designated NY01, was isolated and identified from clinical spleen samples using viral isolation and cultivation techniques. To evaluate its pathogenic potenti

Effect of duck interferon-α and an anti-cap protein polyclonal antibody against duck circovirus.

Duck circovirus (DuCV) is one of the most prevalent viruses in the duck breeding industry, and causes persistent infection and severe immunosuppression. Currently, there is a serious lack of prevention and control measures and no commercial vaccine against DuCV. Therefore, effective antiviral drugs are important for treating DuCV infection. Interferon (IFN) is an important component of antiviral innate immunity, but it remains unclear whether duck IFN-α has a clinical effect against DuCV. Antibody therapy is an important way to treat viral infections. The DuCV structural protein (cap) is immun

Pathogenicity of duck circovirus and fowl adenovirus serotype 4 co-infection in Cherry Valley ducks.

Duck circovirus (DuCV) is one of the most prevalent infectious viruses in the duck industry in China. Although the clinical symptoms vary, it often causes immunosuppression in the host and leads to secondary infection with other pathogens. Fowl adenovirus serotype 4 (FAdV-4) mainly infects chickens and causes hydropericardium hepatitis syndrome. However, the incidence of infection in ducks has increased in recent years, and the phenomenon of mixed infection with DuCV is very common, resulting in more severe clinical morbidity. However, there is no systematic study evaluating the presence of mi

Duck plague virus infection alter the microbiota composition and intestinal functional activity in Muscovy ducks.

Intestinal damage from the duck plague virus (DPV) infection affects intestinal inflammation factors expression and barrier dysfunction. Here we report findings from the pathogenicity of the intestinal tract, intestinal morphological, intestinal permeability, inflammatory cytokines, and tight junction gene expression in 72 two-wk-old Muscovy ducks exposed to DPV. The characterization of intestinal metabolites and their classification were examined using 16-sequencing technology. The primary outcomes of the study evaluated the correlation between intestinal microbiota characteristics and the de

Construction of the recombinant duck enteritis virus delivering capsid protein VP0 of the duck hepatitis A virus.

Duck hepatitis A virus type 1 (DHAV-1) disease causes significant economic losses to the duck industry. Duck enteritis virus (DEV) is frequently used as a viral vector for aquatic poultry vaccination, but no recombinan DEV expressing DHAV-1 VP0 has been developed. In this study, we established a system for rescuing the DEV C-KCE vaccine strain by transfecting cells with six fosmid DNAs. We generated a recombinant virus (rDEV-ul41VP0) by inserting the VP0 gene of DHAV-1 into the ul41 region in the DEV C-KCE genome. DHAV-1 VP0 was stably expressed in the rDEV-ul41VP0 infected cells, but did not

A recombinant adenovirus expressing the E protein of duck Tembusu virus induces protective immunity in duck.

Duck Tembusu virus disease, caused by the duck Tembusu virus (DTMUV), can lead to a severe reduction in egg production and growth retardation in laying ducks and ducklings, respectively. In this study, we engineered a novel recombinant adenovirus expressing the E protein of DTMUV (rAd-E) in AAV-293 cells (analyzed by western blot and indirect immunofluorescence assays). Intramuscular immunization of Cherry Valley ducks with rAd-E was performed to evaluate host cellular and humoral immune responses. Compared to the phosphate-buffered saline administered group and the negative control wild-type

Duck interferon regulatory factor 1 acts as a positive regulator in duck innate antiviral response.

In mammals, IFN regulatory factor (IRF) 1 can activates cellular genes and promotes viral resistance against some DNA and RNA viruses. Most IRFs have been identified as critical regulators in the IFN responses in both mammals and fish. In ducks, however, the functional role of IRF1 remains unknown. Here, we identified duck IRF1 (duIRF1) is essential to counteract viral invasion. duIRF1 is most abundant in duck spleen, and virus infection or poly(I:C) stimulation significantly induced duIRF1 expression in vivo and in vitro. Overexpression of duRF1 induces the expression of type I IFN-β, type II

Induction of immune responses in ducks with a DNA vaccine encoding duck plague virus glycoprotein C.

BACKGROUND: A DNA vaccine expressing glycoprotein C (gC) of duck plague virus (DPV) was evaluated for inducing immunity in ducks. The plasmid encoding gC of DPV was administered via intramuscular (IM) injection and gene gun bombardment. RESULTS: After immunization by both routes virus-specific serum antibody and T-cell responses developed. Vaccination of ducks by IM injection induced a stronger humoral, but weaker cell-mediated immune response. In contrast, a better cell-mediated immune response was achieved by using a gene gun to deliver DNA-coated gold beads to the epidermis with as little

Duck hepatitis B virus infection, aflatoxin B1 and liver cancer in domestic Chinese ducks.

The oncogenicity of Duck hepatitis B virus (DHBV) is unclear since hepatocellular carcinomas (HCCs) have been reported only in domestic ducks in Qidong, an area of China where hepatitis B virus (HBV) and aflatoxin B1 (AFB1) are risk factors for liver cancer in man. In order to better define the association between DHBV infection, AFB1 and HCC we analysed a series of 16 duck liver samples collected from local farms in Qidong. HCC was found in eight and cirrhosis in one of these samples. Furthermore bile duct proliferation, characteristic of AFB1 exposure in ducks and other animal species, was f
