Brassinosteroids (BRs) are plant steroid hormones with structural similarity to mammalian sex steroids and ecdysteroids from insects. The BRs are synthesized from sterols and are essential regulators of cell division, cell elongation and cell differentiation. In this work we show that voriconazole, an antifungal therapeutic drug used in human and veterinary medicine, severely impairs plant growth by inhibiting sterol-14α-demethylation and thereby interfering with BR production. The plant growth regulatory properties of voriconazole and related triazoles were identified in a screen for compound

In contrast to previously described transformations of insect epidermal cells, morphological changes in the larval epidermis of Manduca sexta involve large changes in cell volume: a threefold increase at the beginning and a twofold decrease at the end of the last feeding phase. The volume changes are determined, in part, or entirely, by changes in cell height (cell elongation and cell shortening). Initial cell elongation occurs in a region-specific manner, whereas subsequent cell shortening affects all of the epidermis equally. As shown by ligation experiments and hormone treatments in tissue

Whitefly (Bemisia tabaci Gennadius) is a hemipteran phyto polyphagous sucking insect pest which is an important pest of cotton that causes economic losses to the crop by reducing its yield and quality. Ecdysteroids such as 20-hydroxy ecdysone (20-E), play a significant role in larval moulting, development, and reproduction in pterygota insects. Receptor of 20-E, that is Ecdysone Receptor (BtEcR) of Bemisia tabaci has been targeted to prevent fundamental developmental processes. To identify potent inhibitors of BtEcr, 98,072 natural compounds were retrieved from ZINC database. A structure-based

Sida tuberculata R.E.Fr. (Malvaceae) is a medicinal plant widely found in Southern Brazil, and popularly used for inflammatory disorders and to pain relief. A phytochemical analysis followed by an investigation about antinociceptive potential and mechanism of action were performed with leaves and roots extracts. Methanolic extracts, designated as S. tuberculata leaves extract (STLE) and S. tuberculata roots extract, were analyzed both by UHPLC–MS. The in vivo antinociceptive potential of STLE (10–300 mg kg−1) was assessed in mice subjected to the acetic acid‐induced abdominal writhes and forma

Parasitism by a parasitoid wasp alters the physiology and behavior of its host insects. This study investigated the effects of successful and unsuccessful parasitization on individual host development and testis growth in several moth species. Although the solitary koinobiont endoparasitoid wasp, Cotesia vestalis (Hymenoptera: Braconidae), primarily parasitizes the diamondback moth (Plutella xylostella, Lepidoptera: Plutellidae), its host range is relatively broad. Previous research confirmed successful parasitization of P. xylostella, Mythimna separata (Noctuidae), and Helicoverpa armigera ar

Ingestion of vertebrate blood is essential for egg maturation and transmission of disease-causing parasites by female mosquitoes. Prior studies with the yellow fever mosquito, Aedes aegypti, indicated blood feeding stimulates egg production by triggering the release of hormones from medial neurosecretory cells in the mosquito brain. The ability of bovine insulin to stimulate a similar response further suggested this trigger is an endogenous insulin-like peptide (ILP). A. aegypti encodes eight predicted ILPs. Here, we report that synthetic ILP3 dose-dependently stimulated yolk uptake by oocytes

Forkhead box O (FoxO) transcription factors act downstream of insulin signaling and play conserved roles in development and metabolic homeostasis in insects. However, whether FoxO participates in 20-hydroxyecdysone (20E)-mediated pupation and fat body remodeling in honeybee larvae remains unclear. Here, we show that FoxO is highly expressed during the prepupal and pupal stages of honeybee development. RNA interference (RNAi)-mediated silencing of FoxO delayed pupation, inhibited ecdysteroid biosynthesis and 20E signaling, and ultimately led to pupal lethality. Knockdown of FoxO also suppressed

In some insect species, two sites of juvenile hormone (JH) synthesis have been reported: the very well documented corpora allata that secrete JH for "general use", and the reproductive system, in particular the male accessory glands, in which the function of the sometimes huge amounts of JH (e.g. in Hyalophora cecropia) remains to be clarified. A recent finding in Schistocerca gregaria, namely that suppression of the ecdysteroid peak preceding a molt by RNAi of the Halloween genes spook, phantom and shade does not impede normal molting, challenges the (never experimentally proven) classical co

At the end of each developmental stage, the yellow fever mosquito Aedes aegypti performs the ecdysis behavioral sequence, a precisely timed series of behaviors that culminates in shedding of the old exoskeleton. Here we describe ecdysis triggering hormone-immunoreactive Inka cells located at branch points of major tracheal trunks and loss of staining coincident with ecdysis. Peptides (AeaETH1, AeaETH2) purified from extracts of pharate 4th instar larvae have--PRXamide C-terminal amino acid sequence motifs similar to ETHs previously identified in moths and flies. Injection of synthetic AeaETHs

The presence of specific binding sites for radiolabelled vertebrate-type and arthropod-type steroid hormones was investigated in several organs including salivary gland, and central nervous system of third instar Calliphora vicina larvae by thaw-mount autoradiography. Ponasterone A, a 20-hydroxyecdysone agonist and 20-hydroxyecdysone are the only steroids which bind to nuclear high affinity binding sites. These binding sites are DNA associated while nucleoli show no tracer binding. Ecdysone, an endogenous 20-hydroxyecdysone precursor, is taken up by target cells but no significant nuclear bind

Although the spatial distribution of plant metabolites is critical for understanding their biosynthesis, transformation, transport, and accumulation, the tissue-specific organization of saponins in Paris polyphylla Smith var. yunnanensis (PPY) remains poorly understood. In this study, saponins, flavonoids, and ecdysteroids in the rhizomes, fibrous roots, stems, leaves, and calyces of PPY were analyzed using UPLC-IM-MS. Based on these results, desorption electrospray ionization-ion mobility-mass spectrometry imaging (DESI-IM-MSI) was employed to map their spatial distributions, with collision c

Portunus trituberculatus is an economically important marine crab in China, and the mature ovaries of females possess high nutritional and market value. The Y-organ (YO) is an important endocrine gland in crustaceans that primarily synthesizes ecdysteroids and other steroid hormones. Although its functional status influences ovarian development, the underlying regulatory mechanisms remain poorly understood. This study elucidates how the TGF-β signaling pathway regulates YO function and ovarian development in this species. Unmated and mated females were treated with FK506 (pathway activator) an

Insect development and metamorphosis are regulated by two major hormones, juvenile hormone and ecdysteroids. Despite being the key regulator of insect developmental transitions, the metabolic pathway of the primary steroid hormone, 20-hydroxyecdysone (20E), especially its inactivation pathway, is still not completely elucidated. A cytochrome P450 enzyme, CYP18A1, has been shown to play key roles in insect steroid hormone inactivation through 26-hydroxylation. Here, we identified two CYP18 (BmCYP18A1 and BmCYP18B1) orthologs in the lepidopteran model insect, Bombyx mori. Interestingly, BmCYP18A

Aedes albopictus is one of the most important vectors of Dengue, which poses a serious threat to public health. The bacterial microbiota has an effect on the parameters of mosquitos, such as larval development and fecundity, and it has emerged as a promising field to be explored for novel environmentally friendly control strategies. Rahnella sp. are present in many insects, including Ae. Albopictus, and play a role in bacterial-insect interactions; however, the role of the bacteria in mosquito biology has not yet been characterized. In this study, we characterized the Rahnella isolate RAeA1 ob

Two new phytoecdysteroids, (24Z)-20,26-dihydroxy-24(28)-dehydro-28-methyl-ecdysone (1), (24Z)-22,25-epoxy-2,3,14,20-tetrahydroxyergosta-7,24(28)-dien-28-methyl-6-one (2), together with eleven known compounds (3 - 13) were isolated from the whole plant of Tournefortia sibirica L. The chemical structures of the compounds were elucidated by 1D and 2D NMR and HR-ESI-MS and compared with data from the literature. The isolated compounds were evaluated for their inhibitory effects on NO, TNF-α and IL-6 production in LPS-induced RAW 264.7 cells. Among them, compounds 6, 7, 9, and 10 showed anti-inflam

In Drosophila melanogaster, ecdysis triggering hormone (ETH) is the key factor triggering ecdysis behaviour and promoting trachea clearance. However, whether ETH plays the dual roles in non-dipteran insects is unknown. In this survey, we found that Ldeth mRNA levels were positively correlated with circulating 20-hydroxyecdysone (20E) titers in Leptinotarsa decemlineata. Ingestion of an ecdysteroid agonist halofenozide or 20E stimulated the transcription of Ldeth, whereas RNA interference (RNAi) of ecdysteroidogenesis (LdPTTH or LdSHD) or 20E signalling (LdEcR, LdUSP or LdFTZ-F1) genes inhibite

The ventromedial cells (VM cells) of the moth Manduca sexta belong to a peptide hormone signaling hierarchy that directs an episodic and stereotyped behavior pattern, ecdysis. The VM cells respond to declining ecdysteroid titers at the end of the final larval molt with a transcription-dependent decrease in spike threshold and the abrupt release of the previously stockpiled neuropeptide, eclosion hormone (EH). This report describes whole-cell patch-clamp recordings of acutely isolated VM cell somata made to identify membrane currents that may underlie the increase in VM cell excitability during

The steroid hormone ecdysone controls genetic regulatory hierarchies underlying insect molting, metamorphosis and, in some insects, reproduction. Cytogenetic and molecular analysis of ecdysone response in Drosophila larval salivary glands has revealed regulatory hierarchies including early genes which encode transcription factors controlling late ecdysone response. In order to determine whether similar hierarchies control reproductive ecdysone response, we have investigated ecdysone-regulated gene expression in vitellogenic mosquito ovaries and fat bodies. Here, we identify the homologue of th

LIM homeodomain transcription factor 1 beta (Lmx1b), a member of the LIM-homeodomain family, plays essential roles in tissue differentiation and organ development in diverse eukaryotes, yet its biological functions in insects remain largely unknown. Here, we identify Lmx1b as a highly effective RNA interference (RNAi) target in the 28-spotted ladybeetle, Henosepilachna vigintioctopunctata, a serious solanaceous pest. RNAi-mediated silencing of HvLmx1b prevented larval molting and pupation, resulting in complete mortality. HvLmx1b knockdown markedly reduced endogenous 20-hydroxyecdysone (20E) t

As a tropomyosin-binding component, troponin T (TnT) is essential for the Ca2+ regulation of striated muscles' contraction and locomotion activity, but its impacts on the growth and development of insects have rarely been reported. In this study, TnT was identified and functionally characterized in Tribolium castaneum by RNA interference (RNAi) and transcriptome analysis. The TnT of T. castaneum contained a 1152 bp open reading frame encoding 383 amino acids. It displayed the highest expression in late pupae and was highly expressed in the integument and CNS. Both the larval and early pupal in
