Edaravone protects against cobalt chloride-induced dysfunctions in apoptosis and invasion in trophoblast cells.

This study was performed to investigate the effects of edaravone on hypoxia-induced trophoblast cell proliferation, apoptosis, and invasion. The trophoblast cell line HTR-8 (H8) was treated with cobalt chloride (CoCl2 ) with or without a 1-hr edaravone pretreatment, followed by assessment of intracellular reactive oxygen species (ROS) levels, cell proliferation, apoptosis, migration, and invasion. Metrics of apoptosis included measurement of cysteine-aspartic acid protease 3 (CASP3) activity as well as BAX and BCL2 expression. Migration and invasion phenotypes were complemented with expression

Edaravone-based precision antioxidative therapy alleviates intervertebral disc degeneration via the Nrf2/Aldh3a1 axis.

Oxidative stress-induced dysfunction of nucleus pulposus (NP) cells is a key driver of intervertebral disc degeneration (IDD), promoting accelerated cellular senescence and extracellular matrix (ECM) degradation. Moreover, effective regulation of redox homeostasis within the avascular intervertebral disc remains challenging due to rapid drug clearance and poor targeting. To address these issues, we developed a precision antioxidative therapeutic strategy by integrating edaravone (ED), an FDA-approved free radical scavenger, into a novel reactive oxygen species (ROS)-responsive hydrogel (ED@HFA

Nephroprotective role of edaravone against polystyrene nanoplastic-induced endoplasmic reticulum stress in Wistar rats.

BACKGROUND: Plastics in particular nanoplastics pose the major environmental threat worldwide. Nanoplastics refers to smaller plastic components with size less than 100 nm. Previous studies indicated that plastics could accumulate in blood and various other tissues, leading to hepatotoxicity, nephrotoxicity, neurotoxicity, and reproductive toxicity. Endoplasmic Reticulum (ER) stress disrupts the functionality of ER due to various factors, leading to the accumulation of unfolded and misfolded proteins inside the ER. The current study aimed to investigate the protective effect of edaravone again

Edaravone attenuates acrylamide-induced nephrotoxicity by modulating the endoplasmic reticulum stress pathway in rats.

Acrylamide (ACR) is an environmental toxicant reported to have nephrotoxic effects. Edaravone is a free radical scavenger proven to have antioxidant properties. However, the role of edaravone in regulating endoplasmic reticulum stress (ER stress) in ACR-induced nephrotoxicity remains unclear. Thus, the present study aims to explore the role of ER stress and edaravone in an ACR-induced nephrotoxicity rat model. In this study, 24 male albino Wistar rats were randomly divided into four groups. The control group received normal saline orally for 28 days. The acrylamide group received acrylamide (1

Edaravone attenuates ACSL4-dependent ferroptosis in spinal motor neurons following cardiac arrest in rats.

The contribution of acute spinal motor neuron injury following cardiac arrest (CA) remains poorly understood. This study aimed to investigate the role of ferroptosis in CA-induced spinal cord injury and to evaluate the neuroprotective effects of edaravone. Asphyxial CA was induced in rats for 5 min, followed by resuscitation. Edaravone was administered immediately after the return of spontaneous circulation (ROSC). At 24 h post-ROSC, The CA group exhibited significant hindlimb motor deficits and reduced survival rates. Histological analysis revealed selective injury of choline acetyltransferas

Edaravone provides neuroprotection against chronic heat stress-induced behavioral dysfunction through adaptive proteomic remodeling.

Heat stress is an increasing global health concern associated with high mortality and multi-organ dysfunction. The brain is particularly vulnerable to heat stress, which can lead to behavioral impairments. At the molecular level, heat stress disrupts proteins involved in synaptic communication, mitochondrial function, and cellular homeostasis. Current treatments are mainly symptomatic, highlighting the need for targeted neuroprotective strategies. Edaravone, a potent free-radical scavenger with anti-inflammatory properties, has demonstrated neuroprotective effects in various neurological condi

Edaravone dextranol alleviates ferroptosis, Cuproptosis, and blood-brain barrier damage after acute cerebral infarction.

Edaravone dextrose (EDB) is a commonly used clinical treatment for cerebral infarction due to its anti-inflammatory and free radical scavenging properties. However, its potential additional neuroprotective mechanisms need to be further investigated. In this study, we evaluated the effects of EDB on ferroptosis, cuproptosis, and blood-brain barrier (BBB) disruption after cerebral infarction in vivo and in vitro by constructing a mouse middle cerebral artery occlusion (MCAO) model, and an oxygen-glucose deprivation/reperfusion (OGD/R) model of neurons and brain microvascular endothelium. Edaravone-loaded poly(amino acid) nanogel inhibits ferroptosis for neuroprotection in cerebral ischemia injury.

Neurological injury caused by ischemic stroke is a major cause of permanent disability and death. The currently available neuroprotective drugs fail to achieve desired therapeutic efficacy mainly due to short circulation half-life and poor blood-brain barrier (BBB) permeability. For that, an edaravone-loaded pH/glutathione (pH/GSH) dual-responsive poly(amino acid) nanogel (NG/EDA) was developed to improve the neuroprotection of EDA. The nanogel was triggered by acidic and EDA-induced high-level GSH microenvironments, which enabled the selective and sustained release of EDA at the site of ische

Repurposed Edaravone, Metformin, and Perampanel as a Potential Treatment for Hypoxia-Ischemia Encephalopathy: An In Vitro Study.

Hypoxia-ischemia encephalopathy results from the interruption of oxygen delivery and blood flow to the brain. In the developing brain, it can lead to a brain injury, which is associated with high mortality rates and comorbidities. The hippocampus is one of the brain regions that may be affected by hypoxia-ischemia with consequences on cognition. Unfortunately, clinically approved therapeutics are still scarce and limited. Therefore, in this study, we aimed to test three repurposed drugs with good pharmacological properties to evaluate if they can revert, or at least attenuate, the deleterious
