Current Methods to Investigate Nociception and Pain in Zebrafish.

Pain is an unpleasant, negative emotion and its debilitating effects are complex to manage. Mammalian models have long dominated research on nociception and pain, but there is increasing evidence for comparable processes in fish. The need to improve existing pain models for drug research and the obligation for 3R refinement of fish procedures facilitated the development of numerous new assays of nociception and pain in fish. The zebrafish is already a well-established animal model in many other research areas like toxicity testing, as model for diseases or regeneration and has great potential

Is there a role for long-chain omega3 or oil-rich fish in the treatment of atrial fibrillation?

Atrial fibrillation is the most common cardiac arrhythmia in Europe and north America, and recently it was described as an epidemic. Treatment and management of this arrhythmia consists of using drugs, external electrical cardioversion and in extreme cases, internal electrical pacing. Despite treatment, this arrhythmia continues to impact on morbidity and mortality. The possible benefit from dietary interventions in relation to the primary and secondary prevention of atrial fibrillation have largely been overlooked. Our hypothesis is that increasing the intake of long-chain polyunsaturated ome

Effects of local anesthesia of the cerebellum on classical fear conditioning in goldfish.

BACKGROUND: Besides the amygdala, of which emotion roles have been intensively studied, the cerebellum has also been demonstrated to play a critical role in simple classical fear conditioning in both mammals and fishes. In the present study, we examined the effect of local administration of the anesthetic agent lidocaine into the cerebellum on fear-related, classical heart-rate conditioning in goldfish. METHODS: The effects of microinjection of the anesthetic agent lidocaine into the cerebellum on fear conditioning were investigated in goldfish. The fear conditioning paradigm was delayed clas

Efficiency of four secondary enrichment protocols in differentiation and isolation of Listeria spp. and Listeria monocytogenes from smoked fish processing chains.

Four secondary enrichment protocols (conventional methods: UVM II, Fraser 24 h and Fraser 48 h: Impedimetric method: Listeria electrical detection medium) were studied for their ability to isolate Listeria spp. and Listeria monocytogenes from fish and environmental samples collected along the processing chain of cold-smoked fish. From all methods, Listeria spp. and L. monocytogenes were respectively present in 56 and 34 of 315 samples analysed. Fraser broth incubated for 48 h gave the fewest false negative Listeria spp. results [4/56; (7.1%)], but concurrently only 15/34 (44.1%) samples were c

[The effect of temperature changes on evoked potentials of the fish olfactory bulb].

Perches were kept at 10 degrees C and fixed in a chamber through which water was perfused. During the experiment the water temperature was changed. The potentials induced by electric stimulation of the olfactory nerve were recorded from the dorsal surface of the olfactory bulb. The oscillations of the evoked potentials separated into several components when the temperature was lowered. Six components were found in the evoked response of the olfactory bulb at 8 degrees C.

The dose-response relation for the antinociceptive effect of morphine in a fish, rainbow trout.

There have been suggestions that analgesics be used by fish researchers. But in the absence of dose-response data for morphine, this suggestion seems imprudent. The purpose of the present study was to develop a dose-response relationship in fish using six doses of morphine. The response (movement of the fins or tail) to a noxious stimulus (electrical shock to the face region) was monitored before and after a dose of morphine intraperitoneally (i.p.). The i.p. dose of morphine ED(50) in rainbow trout was 6.7 ± 0.8 mg/kg (n = 12 at each dose). The plasma morphine concentration EC(50) was 4.1 ± 1

Infestation of Rhinogobius flumineus (Family Gobiidae) by the Myxozoan Myxobolus nagaraensis in the Nagara River System, Central Japan.

Myxobolus nagaraensis is a species of myxozoa, a parasitic cnidarian that moves between fish and annelids, and was described in 2007 as a parasite of the genus Rhinogobius. The purpose of the present study was to clarify the actual status of parasitism of Rhinogobius flumineus by M. nagaraensis. Collection surveys conducted in the fall/winter season confirmed infestation at 18 of 38 locations of tributaries of the Nagara river. At all eight sites with high infection rates (> 9%) during fall/winter, the number of parasitized individuals decreased dramatically in spring/summer. The body length o

A sustainable strategy for generating highly stable human skin equivalents based on fish collagen.

Tissue engineered skin equivalents are increasingly recognized as potential alternatives to traditional skin models such as human ex vivo skin or animal skin models. However, most of the currently investigated human skin equivalents (HSEs) are constructed using mammalian collagen which can be expensive and difficult to extract. Fish skin is a waste product produced by fish processing industries and identified as a cost-efficient and sustainable source of type I collagen. In this work, we describe a method for generating highly stable HSEs based on fibrin fortified tilapia fish collagen. The fo

Central control of cardiorespiratory interactions in fish.

Fish control the relative flow rates of water and blood over the gills in order to optimise respiratory gas exchange. As both flows are markedly pulsatile, close beat-to-beat relationships can be predicted. Cardiorespiratory interactions in fish are controlled primarily by activity in the parasympathetic nervous system that has its origin in cardiac vagal preganglionic neurons. Recordings of efferent activity in the cardiac vagus include units firing in respiration-related bursts. Bursts of electrical stimuli delivered peripherally to the cardiac vagus or centrally to respiratory branches of c

Long-term editing of brain circuits using an engineered electrical synapse.

Electrical signalling across distinct populations of brain cells underpins cognitive and emotional function. However, approaches that selectively regulate electrical signalling between two cellular components of a mammalian neural circuit remain sparse. Here we engineered an electrical synapse composed of two connexin proteins1 found in Morone americana (white perch fish)-connexin 34.7 and connexin 35-to accomplish mammalian circuit modulation. By exploiting protein mutagenesis, devising a new in vitro system for assaying connexin hemichannel docking, and performing computational modelling of

Deltamethrin is toxic to the fish (crucian carp, Carassius carassius) heart.

Pyrethroids are extensively used for the control of insect pests and disease vectors. Pyrethroids are regarded safe due to their selective toxicity: they are effective against insects but relatively harmless to mammals and birds. Unfortunately, pyrethroids are very toxic to fishes. The high toxicity of pyrethroids to fishes is only partly explained by slow metabolic elimination of pyrethroids, suggesting that some molecular targets in vital organs of the fish body are sensitive to pyrethroids. To this end we tested the effect of deltamethrin (DM) on fish (crucian carp, Carassius carassius) hea

Bio-Inspired Electrolocation Based on Electrostatic Interfacial Enhancement.

As one of the remote sensing abilities, electrolocation allows organisms to hunt, navigate, and interact with the environment by sensing electric fields. Mimicking this capability in artificial systems will provide a powerful strategy for remote object detection and inspection. However, current artificial approaches are restricted to conductive medium or charged targets, while electrolocation of uncharged targets in the non-conductive medium has not been achieved yet. In this work, an electrostatic interfacial enhancement-based electrolocation system (EIEEs) that fundamentally expands the scop

Switchable Adhesion of Hydrogels to Plant and Animal Tissues.

The ability to "switch on" adhesion between a thin hydrogel and a biological tissue can be useful in biomedical applications such as surgery. One way to accomplish this is with an electric field, a phenomenon termed electroadhesion (EA). Here, it is shown that cationic gels can be adhered by EA to tissues across all of biology. This includes tissues from animals, including humans and other mammals; birds; fish; reptiles (e.g., lizards); amphibians (e.g., frogs), and invertebrates (e.g., shrimp, worms). Gels can also be adhered to soft tissues from plants, including fruit (e.g., plums) and vege

Consequences of electroshock-induced narcosis in fish muscle: from mitochondria to swim performance.

Adult zebrafish Danio rerio were exposed to an electric shock of 3 V and 1A for 5 s delivered by field backpack electrofishing gear, to induce a taxis followed by a narcosis. The effect of such electric shock was investigated on both the individual performances (swimming capacities and costs of transport) and at cellular and mitochondrial levels (oxygen consumption and oxidative balance). The observed survival rate was very high (96·8%) independent of swimming speed (up to 10 body length s-1 ). The results showed no effect of the treatment on the metabolism and cost of transport of the fish. N

Sustainable feed processing methods to enhance growth and digestibility in freshwater aquaculture: A review.

Freshwater fish farming is one of the fastest-growing food sectors, contributing significantly to global food security. Despite the impressive global growth of aquaculture, high operational costs remain a major challenge, with feed production costs constituting the highest proportion of expenses. To reduce reliance on expensive materials, alternative ingredients have been explored, but issues such as reduced digestibility may arise. To address these challenges, various processing methods have been developed, ranging from traditional to advanced technologies. This review provides comprehensive

Electricity in the treatment of nervous system disease.

Electricity has been used in medicine for almost two millenniums beginning with electrical chocks from the torpedo fish and ending with the implantation of neuromodulators and neuroprostheses. These implantable stimulators aim to improve functional independence and quality of life in various groups of disabled people. New indications for neuromodulation are still evolving and the field is rapidly advancing. Thanks to modern science and computer technology, electrotherapy has reached a degree of sophistication where it can be applied relatively safely and effectively in a variety of nervous sys

Reduced ventricular excitability causes atrioventricular block and depression of heart rate in fish at critically high temperatures.

At critically high temperature, cardiac output in fish collapses as a result of depression of heart rate (bradycardia). However, the cause of bradycardia remains unresolved. To investigate this, rainbow trout (Oncorhynchus mykiss; acclimated at 12°C) were exposed to acute warming while electrocardiograms were recorded. From 12°C to 25.3°C, electrical excitation between different parts of the heart was coordinated, but above 25.3°C, atrial and ventricular beating rates became partly dissociated because of 2:1 atrioventricular (AV) block. With further warming, atrial rate increased to a peak val
