Gut microbial signatures of advanced hepatocellular carcinoma and their potential diagnostic value.

BACKGROUND: Hepatocellular carcinoma (HCC) is a prevalent and lethal malignancy worldwide. Gut microbiota play crucial roles in liver disease progression and may offer noninvasive diagnostic value, yet microbial signatures specific to advanced HCC remain unclear. METHODS: Seventy-six participants, including early-stage HCC (HCC12), advanced HCC (HCC34), liver cirrhosis (LC), and healthy controls (CG), were prospectively enrolled. Fecal samples underwent 16S rRNA sequencing to characterize microbial diversity and community composition. Differential taxa were identified using Kruskal-Wallis tes

Two low-toxic Klebsiella pneumoniae strains from gut of black soldier fly Hermetia illucens are multi-resistance to sulfonamides and cadmium.

In recent years, pollution of antibiotics and heavy metal has often been reported in organic wastes. Saprophytic insects have been recorded as biological control agents in organic waste management. During organic waste conversion, the intestinal bacteria of the saprophytic insects play an important role in digestion, physiology, immunity and prevention of pathogen colonization. Black soldier fly (BSF) Hermetia illucens has been widely used as saprophytic insects and showed tolerance to sulfonamides (SAs) and cadmium (Cd). Diversity and changes in gut microbiota of black soldier fly larvae (BSF

Improvement of the quality and safety of low-salt fish sauce by reconstruction of microbial community through cooperative fermentation of starters.

Low-salt fermentation can speed up the fish sauce production, but it will lead to bad flavor and biogenic amine increase due to the metabolisms of spoilage microorganisms. In this work, cooperative fermentation of Tetragenococcus muriaticus, Bacillus subtilis, and Staphylococcus edaphicus that were isolated from traditional fish sauce were used to improve the quality and safety of low-salt fish sauce. The high-throughput sequencing found that these starters could significantly inhibited the spoilage microorganisms and reconstructed the microbial community with the total abundance over 99 % dur

Age-Related Microbial Differences in Newly Diagnosed Crohn's Disease Reveal the Distinct Enrichment of Oral-Associated Taxa in Pediatric Patients.

BACKGROUND/AIMS: Crohn's disease (CD) presents differently by age of onset, but the microbial distinctions between pediatric and adult CD are currently unclear. METHODS: In this study, the gut microbiota of fecal samples collected from newly diagnosed patients with CD (n=88; pediatric 44, adult 44) and their age- and sex-matched healthy controls (HCs, n=112) was analyzed using 16S rRNA gene amplicon sequencing. Differential abundance analysis was performed using MaAsLin2 to identify age-specific microbial signatures, and enterotypes were classified using Dirichlet multinomial mixture modeling

Lineage-dependent Microbial Shifts in the Reproductive Tract of Black Soldier Flies (Hermetia illucens) Reveal Novel Mating-linked Associations.

Reproductive microbiota is known to modulate host immunity, reproductive physiology, and longevity, yet their interactions with host genetics, sex, and mating status remain relatively underexplored in most insects. Some studies suggest that species (and even populations) can vary in the microbial communities associated with their reproductive tissues. Sexually transmitted microbes can also directly influence population growth and reproductive fitness, making them relevant even for applied insect management strategies. The economically relevant, Black Soldier Fly (BSF; Hermetia illucens), is kn

Exclusive dependence of IL-10Rα signalling on intestinal microbiota homeostasis and control of whipworm infection.

The whipworm Trichuris trichiura is a soil-transmitted helminth that dwells in the epithelium of the caecum and proximal colon of their hosts causing the human disease, trichuriasis. Trichuriasis is characterized by colitis attributed to the inflammatory response elicited by the parasite while tunnelling through intestinal epithelial cells (IECs). The IL-10 family of receptors, comprising combinations of subunits IL-10Rα, IL-10Rβ, IL-22Rα and IL-28Rα, modulates intestinal inflammatory responses. Here we carefully dissected the role of these subunits in the resistance of mice to infection with

Metagenomic and ribosomal transcript profiles of diabetic foot osteomyelitis in Hispanic patients: underestimated bacteria in biofilm persistence.

BACKGROUND: Diabetic foot osteomyelitis (DFO) is a serious complication of diabetes and a leading cause of lower-limb amputations. Conventional culture-based diagnostics often underestimate the microbial diversity of infected bone tissue. This study represents the first characterization of both total and ribosomally active bone microbiota in Hispanic patients with DFO using high-throughput 16S rRNA gene sequencing. The work aims to contribute to the inclusion of underrepresented populations in microbiome research and informing molecular-based antimicrobial strategies. METHODS: Bone specimens

Improvement of gut microbiome and intestinal permeability following splenectomy plus pericardial devascularization in hepatitis B virus-related cirrhotic portal hypertension.

The gut microbiome is an essential component of the intestinal mucosal barrier, critical in regulating intestinal permeability. Microbiome dysbiosis and intestinal permeability changes are commonly encountered conditions in patients with cirrhosis and are closely related to its development and further complications. However, alterations in the gut microbiome and intestinal permeability in chronic hepatitis B virus (HBV) patients with cirrhotic portal hypertension after undergoing a splenectomy plus pericardial devascularization (SPD) have not been investigated. This study recruited 22 patients

Modeling pathogen-driven neonatal late-onset sepsis: a modification to the murine cecal slurry.

INTRODUCTION: Neonatal sepsis is a major cause of neonatal morbidity and mortality. Reliable animal models are essential to our understanding of late-onset sepsis, but notable limitations exist in the current standard murine cecal slurry model. We sought to refine the existing model by using an injection of known stock slurry ("NEC'teria") cultured from an infant who died of necrotizing enterocolitis to better mimic sepsis following the translocation of neonatal specific bacterial pathogens from the intestine into the peritoneum. METHODS: To induce sepsis, neonatal mice (P7 and P14 - P16) wer

Lactobacillus rhamnosus GG treatment improves intestinal permeability and modulates microbiota dysbiosis in an experimental model of sepsis.

Decrease of 'health‑benefiting' microbes and increase of pathogenic bacteria (a condition termed dysbiosis) in intensive care unit patients is considered to induce or aggravate sepsis (gut‑origin sepsis). Orally administered probiotics have been effective in the prevention of nosocomial infections. However, the mechanisms of probiotic‑induced anti‑infection and anti‑sepsis remain to be explored. In the present study, 4‑week‑old C57BL6 mice were orally administrated with Lactobacillus rhamnosus GG (LGG) or normal saline (control) 4 weeks prior to cecal ligation and puncture (CLP). A subset of t

Characterization of the probiotic properties of Lacticaseibacillus rhamnosus LR6 isolated from the vaginas of healthy Korean women against vaginal pathogens.

The human microbiome exhibits intricate populations across the body, with the vaginal tract serving as an ecosystem characterized by the prevalence of the genus Lactobacillus. Disruptions in the vaginal microbiota, which are frequently linked to variables such as sexual activity, hormonal fluctuations, and excessive use of antibiotics, can result in vaginal dysbiosis and the development of diseases such as bacterial vaginosis (BV) and candidiasis. Lactobacillus species, owing to their capacity to create an acidic environment through the production of lactic acid, have a key function within thi
