Stabilization of membrane bound enzyme profiles by sodium selenite in N-nitrosodiethylamine induced and phenobarbital promoted hepatocarcinogenesis in rats.

As part of a substantial effort to curtail the adverse health effects posed by hepatoma, studies have been conducted to elucidate the possible mechanism for the anticarcinogenic action of sodium selenite against N-nitrosodiethylamine induced hepatocarcinogenesis. Sodium selenite administered through drinking water at a dose of 4 ppm before initiation, or during initiation and/or during the promotion period of carcinogenesis exerted an in vivo stabilizing effect on cell membranes in rat hepatoma. This was demonstrated in normal rats and in animals whose biomembranes were rendered fragile by ind

Gelatin-Stabilized Water-in-Water Emulsions for Biocatalysis: Interfacial Enzyme Confinement, In Situ Product Extraction, and Cryogenic Recyclability.

Water-in-water (W/W) emulsions offer high biocompatibility for enzymatic processes but are difficult to stabilize with conventional oil-water surfactants due to their ultralow interfacial tension. Here, we design a gelatin-stabilized PEG/dextran (PEG/Dex) emulsion platform enabling integrated "reaction-separation-recovery" biocatalysis. By tuning the pH and polymer concentration, programmable microarchitectures such as core-shell, Janus, or triple-phase droplets were achieved. In cascade oxidation of guaiacol, the compartmentalized system reached an 86% yield of tetraguaiacol─50% higher than i

Reciprocal regulation of eNOS, H2S and CO-synthesizing enzymes in human atheroma: Correlation with plaque stability and effects of simvastatin.

The molecular and cellular mechanisms underlying plaque destabilization remain obscure. We sought to elucidate the correlation between NO, H2S and CO-generating enzymes, nitro-oxidative stress and plaque stability in carotid arteries. Carotid atherosclerotic plaques were collected from 62 patients who had undergone endarterectomy due to internal artery stenosis. Following histological evaluation the plaques were divided into stable and unstable ones. To investigate the impact of simvastatin we divided patients with stable plaques, into those receiving and to those not receiving simvastatin. Ex

Atmospheric steam outperforms other thermal treatments in elevating the storage stability of oat flour: Revealed via enzyme inactivation, volatile profiles, and lipidomics.

Oats, as a representative nutritious grain, are rich in various bioactive compounds, but the high endogenous lipid-related enzymes make their flour highly susceptible to hydrolytic and oxidative rancidity during storage, thus affecting nutrition and sensory quality. In this study, four thermal stabilization technologies, including microwave (MW), stir-frying (SF), far-infrared (FIR), and atmospheric steam (AS), were used to improve the storage stability of oat flour. The results showed that all treatments reduced initial lipase (LA) activity by more than 91% and delayed lipid deterioration to

Novel Treatment Strategy for Patients With Urea Cycle Disorders: Pharmacological Chaperones Enhance Enzyme Stability and Activity in Patient-Derived Liver Disease Models.

Urea cycle disorders (UCDs) are inherited diseases causing recurrent life-threatening metabolic decompensations due to impaired hepatic ammonia detoxification and decreased ureagenesis. Ornithine transcarbamylase (OTC) deficiency (OTCD) is X-linked and the most common and often fatal UCD. In male hemizygous patients, disease severity primarily depends on the pathogenic sequence variant, while in heterozygous females, disease severity also depends on the X-chromosomal inactivation (XCI) pattern. Females with unfavorable XCI predominantly expressing the mutant OTC protein may be severely affecte

Assessing the compatibility of choline-based deep eutectic solvents for the structural stability and activity of cellulase: Enzyme sustain at high temperature.

As a new generation of 'green solvents' deep eutectic solvents (DESs) represents a promising alternative to the conventional solvents. Their environmental-benign nature and designer properties promote their utility in biocatalysis. Enzymes are marginally stable when exposed to physical/chemical disturbances. One such enzyme is cellulase which is a propitious catalyst for the depolymerization of cellulose under mild conditions. Therefore, their stability is a prerequisite condition to match demands of biorefineries. To address this issue of low stability, activity and thermal denaturation of ce

Two novel angiotensin I-converting enzyme inhibitory peptides from garlic protein: In silico screening, stability, antihypertensive effects in vivo and underlying mechanisms.

This study aimed to screen novel angiotensin I-converting enzyme (ACE) inhibitory peptides from garlic proteins and to explore their underlying antihypertensive mechanisms in vivo. After simulated hydrolysis and in silico screening, two novel peptides (MGR and HDCF) were obtained with the highest ACE inhibitory activity (IC50 of 4.50 μM and 26.38 μM) and acted as competitive inhibitors. They interacted with key residues in the ACE receptor mainly through hydrogen bonding and exhibited excellent stability against high temperature, extreme pH, and gastrointestinal digestion. A Functional Variant in Ubiquitin Conjugating Enzyme E2 L3 Contributes to Hepatitis B Virus Infection and Maintains Covalently Closed Circular DNA Stability by Inducing Degradation of Apolipoprotein B mRNA Editing Enzyme Catalytic Subunit 3A.

Hepatitis B virus (HBV) infection is a common infectious disease, in which nuclear covalently closed circular DNA (cccDNA) plays a key role in viral persistence, viral reactivation after treatment withdrawal, and drug resistance. A recent genome-wide association study has identified that the ubiquitin conjugating enzyme E2 L3 (UBE2L3) gene is associated with increased susceptibility to chronic HBV (CHB) infection in adults. However, the association between UBE2L3 and children with CHB and the underlying mechanism remain unclear. In this study, we performed two-stage case-control studies includ

Microbial Necromass and Extracellular Enzyme Activities Are Associated with Depth-Dependent Soil Carbon Stabilization Along a Wildfire-Severity Gradient.

Wildfire can alter soil organic carbon (SOC) pools and microbial pathways of carbon stabilization, but depth-dependent links between microbial necromass and stable carbon pools remain unclear. We investigated a wildfire-severity gradient in a subtropical coniferous forest in Guizhou, China, including four severity classes (unburned, light, moderate, and severe) and two soil layers (0-20 and 20-40 cm). We measured easily oxidizable organic carbon (EOC), recalcitrant organic carbon (ROC), SOC, amino sugars, microbial necromass carbon (MNC), extracellular enzyme activities, and carbohydrate-activ

Evaluation of stabilizing additives to protect activities of cytochrome P450 enzymes for in vitro drug testing and pharmacogenetic studies: Focus on CYP2D6.

In vitro and ex vivo studies on drug metabolism and stability are vital for drug development and pre-clinical safety assessment. Traditional in vitro models, such as liver enzyme (S9) fractions and microsomes, often fail to account for individual variability. Personalized models, including 3D cell models and organoids, offer promising alternatives but may not fully replicate physiological processes, especially for Cytochrome P450 (CYP) families involved in extrahepatic metabolism. A major challenge in these studies is the low stability and expression of CYP enzymes. This study aimed to stabili

pH and CO2/N2 dual responsive Pickering emulsion stabilized by shellac nanoparticle-enzyme conjugates for synthesis of phytosterol esters.

The development of green and efficient catalytic systems is crucial for sustainable chemical processes. This study engineered shellac nanoparticle-lipase conjugates for biphasic biocatalysis. Taking advantage of the inherent amphiphilicity and pH-triggered interfacial behavior of the shellac, the conjugates exhibited dual functionality as intelligent Pickering emulsifiers and interfacial biocatalysts. The covalent conjugation strategy achieved a protein loading capacity of 107.97 ± 5.24 mg/g under optimized conditions while preserving interfacial responsiveness. Reversible emulsification/break

Harnessing polyhydroxylated pyrrolidines as a stabilizer of acid alpha-glucosidase (GAA) to enhance the efficacy of enzyme replacement therapy in Pompe disease.

To discover small molecules as acid alpha-glucosidase (GAA) stabilizers for potential benefits of the exogenous enzyme treatment toward Pompe disease cells, we started from the initial screening of the unique chemical space, consisting of sixteen stereoisomers of 2-aminomethyl polyhydroxylated pyrrolidines (ADMDPs) to find out two primary stabilizers 17 and 18. Further external or internal structural modifications of 17 and 18 were performed to increase structural diversity, followed by the protein thermal shift study to evaluate the GAA stabilizing ability. Fortunately, pyrrolidine 21, posses

Vinyl sulfone-amino-alkyl supports: heterofunctional matrixes to prevent enzyme release and stabilize lipases via covalent immobilization.

New trifunctional supports were prepared (amino-octyl-vinyl sulfone (VS)- and amino-hexyl-VS-agarose) and compared to octyl-VS-agarose. They were utilized to immobilize the lipases A and B from Candida antarctica (CALA and CALB). After incubation to generate some enzyme-support bonds and blocking with different nucleophiles, SDS-PAGE analyses showed that all enzyme molecules become covalently immobilized on the support. In all VS biocatalysts, the blocking reagent presented a great effect in the properties of enzymes. The best blocking agents promoted a significant enzyme stabilization compare

A Universal Structural Grammar in Enzyme Fold for Predicting Drug Target Stability: Deciphering Directional Scaffolding via Multi-Stage Pearson Correlation of Asymmetric Contact Matrices.

Background/Objectives: Traditional protein contact analysis often fails to distinguish between local, sequence-driven motifs and global, tertiary scaffolding, which ensures structural determinism. While deep-learning models do not fully elucidate the 'why', they do reveal the underlying directional rules of the stability landscape. In this study, we analyzed 475 non-redundant Protein Data Bank (PDB) structures categorized into SCOP classes (all-α, all-β, α/β, α+β) of the hydrolase superfamily. Methods: To isolate the structural anchors of the global fold, we applied a sequence separation filte
