Ependymal Vps35 Promotes Ependymal Cell Differentiation and Survival, Suppresses Microglial Activation, and Prevents Neonatal Hydrocephalus.

Hydrocephalus is a pathologic condition associated with various brain diseases, including Alzheimer's disease (AD). Dysfunctional ependymal cells (EpCs) are believed to contribute to the development of hydrocephalus. It is thus of interest to investigate EpCs' development and function. Here, we report that vacuolar protein sorting-associated protein 35 (VPS35) is critical for EpC differentiation, ciliogenesis, and survival, and thus preventing neonatal hydrocephalus. VPS35 is abundantly expressed in EpCs. Mice with conditional knock-out (cKO) of Vps35 in embryonic (Vps35GFAP-Cre and Vps35Emx1-

Sox8 and Sox9 regulate differentiation and nuclear positioning of retinal Müller glia.

Temporal patterning of retinal progenitor cells governs the sequential generation of retinal cell types, with gliogenesis occurring late in development. Sox8 and Sox9, members of the SoxE transcription factor family, are highly expressed in late-stage retinal progenitor cells and mature Müller glia, yet their functional roles remain incompletely defined. Here, we have employed gain- and loss-of-function approaches, single-cell multiomic profiling, and injury models to investigate Sox8 and Sox9 function. Overexpression of human SOX8 and SOX9 in early-stage mouse retinal progenitor cells both su

Müller Glia Subtypes Define Neuro-Glial Associations and Spatial Morphogen Axes in the Zebrafish Retina.

Müller glia are instrumental macroglia of the vertebrate retina, once thought to be a homogeneous population. Now Müller glia are generally accepted as transcriptionally heterogeneous, and new evidence suggests functional diversity may exist in the way these cells respond to retinal injury. It remains unclear, however, whether this functional heterogeneity is limited to a transient phenotype that stems from injury or a fundamental feature of the healthy retina. Here, we investigate Müller glia heterogeneity in the uninjured zebrafish retina across development and adulthood using a comprehensiv

Light and Electron Microscopic Characterization of Vascular and Mitochondrial Structures in the Camel Retina.

Camels have unique morphological traits that enable them to adapt well to harsh conditions. This work aims to describe the vascular architecture of the camel retina and investigate its cellular components with a focus on the distribution of mitochondria in Muller cells and photoreceptors, using light and electron microscopy. The camel retina is euangiotic in which blood vessels extend in the inner retina from the nerve fiber layer to the outer plexiform layer. The pericytes are embedded in the basement membrane of the retinal capillaries, and overlapping of pericytes could be observed. Glial c

IL-33 regulates Müller cell-mediated retinal inflammation and neurodegeneration in diabetic retinopathy.

Diabetic retinopathy (DR) is characterised by dysfunction of the retinal neurovascular unit, leading to visual impairment and blindness. Müller cells are key components of the retinal neurovascular unit and diabetes has a detrimental impact on these glial cells, triggering progressive neurovascular pathology of DR. Amongst many factors expressed by Müller cells, interleukin-33 (IL-33) has an established immunomodulatory role, and we investigated the role of endogenous IL-33 in DR. The expression of IL-33 in Müller cells increased during diabetes. Wild-type and Il33-/- mice developed equivalent

Two different populations of Müller cells stabilize the structure of the fovea: an optical coherence tomography study.

PURPOSE: To document with spectral-domain optical coherence tomography the structural stabilization of the fovea and the sealing of outer macular defects by Müller cells. METHODS: A retrospective case series of 45 eyes of 34 patients is described. RESULTS: In cases of a cystic disruption of the foveola as in macular telangiectasia type 2 and vitreomacular traction, the Müller cell cone provides the structural stability of the fovea. In cases of a detachment or disruption of the Müller cell cone, e.g., in foveal pseudocysts, outer lamellar holes, and degenerative and tractional lamellar holes

Attenuation of Retinal Vascular Development in Neonatal Mice Subjected to Hypoxic-Ischemic Encephalopathy.

A significant proportion of children that survive hypoxic-ischemic encephalopathy (HIE) develop visual impairment. These visual deficits are generally attributed to injuries that occur in the primary visual cortex and other visual processing systems. Recent studies suggested that neuronal damage might also occur in the retina. An important structure affecting the viability of retinal neurons is the vasculature. However, the effects of HIE on the retinal neurovasculature have not been systemically evaluated. Here we investigated whether exposure of postnatal day 9 (P9) neonatal mice to HIE is s

Brain organoid models of SZT2-related disease reveal an overproduction of outer radial glial cells through mTORC1 activation.

Biallelic loss-of-function variants of Seizure Threshold 2 (SZT2) cause neurodevelopmental diseases with developmental delay, epilepsy, and macrocephaly. SZT2 forms the KICSTOR complex, which represses the mechanistic target of rapamycin complex 1 (mTORC1) amino acid-sensitive pathway. SZT2 dysfunction is thought to cause abnormal activation of the mTOR pathway, underlying the pathogenesis of SZT2-related diseases. We previously reported constitutive activation of mTORC1 in lymphoblastoid cell lines derived from patients with SZT2-related disease. However, the impact of SZT2 dysfunction on hum

Insights into CD154-mediated pathways in ocular hypertensive glaucoma: The role of Müller cells and P2X7 in retinal neuroprotection and therapeutic potential.

An elevation of pathologic intraocular pressure (IOP) is the greatest risk factor for glaucoma. CD154 has been reported to bind to CD40 expressed by orbital fibroblasts and be involved in immune and inflammatory responses. However, the function and mechanism of CD154 in ocular hypertensive glaucoma (OHG) are not fully understood. We isolated and characterized Müller cells and subsequently examined the effect of CD154 on ATP release from those cells. After being cocultured with CD154-pretreated Müller cells, retinal ganglion cells (RGCs) were treated with P2X7 siRNAs or a P2X7 inhibitor. Furthe

Purinergic Receptor P2Y2 Regulates Neurogenesis and Functional Recovery Following Spinal Cord Injury in Zebrafish.

In contrast to mammals, adult zebrafish (Danio rerio) undergo neuronal regeneration following spinal cord injury (SCI), thereby allowing successful functional recovery. The adaptive regenerative response to SCI in zebrafish includes injury-induced proliferation of ependymal radial glial cells (ERGs) and neurogenesis. The molecular mechanisms underlying injury-induced neurogenesis in "regenerative" species, such as zebrafish, have not been fully elucidated and are an area of great interest. In mammals, the purinergic receptor P2Y2 has been shown to mediate NSC proliferation and neurogenesis, bu

N6-methyladenosine (m6A) modification regulates HSPA1A and HSPA1B expression in Müller cells under high glucose stress.

Müller cells (MCs) represent the major glial cells that are responsible for maintaining retinal homeostasis. In diabetic retinopathy, Müller cell activation occurs in the initial stages, playing a role in many pathological processes, such as neovascularization, neuronal dysfunction, and inflammatory retinal environment. As the most common RNA modification in eukaryotes, N6-methyladenosine (m6A) exerts dynamic and reversible control over cellular functions in the context of high glucose (HG) stress. Here, we performed combined m6A and RNA sequencing to elucidate the landscape of m6A modificatio

The lack of Irs2 induces changes in the immunocytochemical expression of aromatase in the mouse retina.

Insulin receptor substrate (Irs) belongs to a family of proteins that mediate the intracellular signaling of insulin and IGF-1. Insulin receptor substrate 2 (Irs2) is necessary for retinal function, since its failure in Irs2-deficient mice in hyperglycemic situation promotes photoreceptor degeneration and visual dysfunction, like in diabetic retinopathy. The expression of P450 aromatase, which catalyzes androgen aromatization to form 17ß-estradiol, increases in some neurodegenerative diseases thus promoting the local synthesis of neuroestrogens that exert relevant neuroprotective functions. Dexamethasone protects retinal ganglion cells but not Müller glia against hyperglycemia in vitro.

Diabetic retinopathy (DR) is a common complication of diabetes, for which hyperglycemia is a major etiological factor. It is known that retinal glia (Müller cells) and retinal ganglion cells (RGCs) are affected by diabetes, and there is evidence that DR is associated with neural degeneration. Dexamethasone is a glucocorticoid used to treat many inflammatory and autoimmune conditions, including several eye diseases like DR. Thus, our goal was to study the effect of dexamethasone on the survival of RGCs and Müller glial cells isolated from rat retinas and maintained in vitro under hyperglycemic

Loss of ABCA4 from photoreceptor discs is associated with glial transcriptomic changes in retinal organoids.

BACKGROUND: Loss-of-function mutations in the ABCA4 gene cause Stargardt disease (STGD1), the most common inherited macular dystrophy leading to progressive central vision loss. METHODS: Here, we generated human induced pluripotent stem cell-derived retinal organoids harboring a premature stop codon in exon-24 of ABCA4 to evaluate the impact of this mutation on mRNA and protein levels in a human model. RESULTS: Immunofluorescence analysis revealed the absence of ABCA4 protein in the mutant photoreceptor outer segment discs, while single-cell RNA sequencing detected no major transcriptional a

Nonsense Mutation in USH2A Exon-13 Activates the Innate Immune Response in Müller Glial Cells.

Pathological USH2A mutations cause Usher syndrome type II, characterized by progressive retinitis pigmentosa and hearing and balance impairment. This study aims to investigate the cellular mechanisms underlying USH2A-related retinal degeneration using human induced pluripotent stem cell (hiPSC)-derived retinal organoids. The introduction of a homozygous nonsense mutation in the USH2A hotspot exon-13 resulted in normal photoreceptor development but loss of ciliary localization of usherin long form B and its interacting proteins, ADGRV1 and whirlin. Notably, single-cell RNA sequencing revealed u

An ancient cholinergic cell triad of a light-sensitive, a pigmented and a glial cell could regulate stemness.

A close spatiotemporal relation between end of mitosis, of butyryl- (BChE) and then acetylcholinesterase (AChE) expressions in early neural tissues has been established, yet the underlying mechanisms are not understood. With this observation in mind, cell culture work analyzing cholinergic functions in the developing vertebrate retina was revealing. For proper cell stratification in avian and rodent retinal 3D-reaggregates (organoids) and explants, Müller glial cells (MCs) and the retinal pigmented epithelium (RPE) are essential actors. Network formation depends on a ChAT+ amacrine cell and BC

PMS. Lineage trajectories and fate determinants of postnatal neural stem cells and ependymal cells in the developing ventricular zone.

The ventricular zone (VZ) harbors the largest neurogenic niche in the adult mammalian brain and is consisted of neural stem cells (NSCs) and multiciliated ependymal cells (EPCs). Previous lineage tracing studies showed that both NSCs and EPCs were derived from radial glial cells (RGCs). However, the transcriptomic dynamics and the molecular mechanisms guiding the cell fate commitment during the differentiation remain poorly understood. In this study, we analyzed the developing VZ of mice at single-cell resolution and identified three distinct cellular states of RGCs: bipotent glial progenitor

Circulating ghrelin crosses the blood-cerebrospinal fluid barrier via growth hormone secretagogue receptor dependent and independent mechanisms.

Ghrelin is a peptide hormone mainly secreted from gastrointestinal tract that acts via the growth hormone secretagogue receptor (GHSR), which is highly expressed in the brain. Strikingly, the accessibility of ghrelin to the brain seems to be limited and restricted to few brain areas. Previous studies in mice have shown that ghrelin can access the brain via the blood-cerebrospinal fluid (CSF) barrier, an interface constituted by the choroid plexus and the hypothalamic tanycytes. Here, we performed a variety of in vivo and in vitro studies to test the hypothesis that the transport of ghrelin acr
