Epigenetic memory and allostatic integrity: a theoretical integration of the hierarchical-circular model using a longitudinal methylation signature of dementia.

INTRODUCTION: The Hierarchical-Circular Model of Biological Memory (HCM) proposes that dementia results from disrupted circular information flow across five levels: genetic, epigenetic, allostatic, PNEI network, and interoceptive. Allostatic integrity-the adaptive capacity for circular flow-determines whether pathology translates into dementia. OBJECTIVE: To evaluate whether a recently published blood DNA methylation signature for incident dementia is consistent with the predictions of the HCM. METHODS: We systematically mapped Zhang et al.'s findings (44 CpGs, 44 DMRs, pathway enrichments,

Epigenetic Clocks of Biological Aging and Risk of Incident Mild Cognitive Impairment and Dementia: The Women's Health Initiative Memory Study.

Aging is the strongest risk factor for dementia; however, few studies have examined the association of biological aging with incident dementia. We analyzed 6069 cognitively unimpaired women (mean age = 70.0 ± 3.8 years) in the Women's Health Initiative Memory Study to examine the association of accelerated biological aging, measured with second and third-generation epigenetic clocks (AgeAccelPheno and AgeAccelGrim2, and DunedinPACE, respectively) with incident mild cognitive impairment (MCI) and probable dementia. Multivariable Cox proportional hazards models adjusted for age, education, race,

Hepatitis C Virus: An Overview of Its Chronic Impact on Liver Function, Metabolic Dysregulation, Inflammatory-Oxidative Pathogenesis and Epigenetic Memory.

Hepatitis C virus (HCV) infection is a global health concern, chronically affecting over 71 million people. It primarily targets the liver but also causes systemic complications through inflammation, oxidative stress, and metabolic dysregulation. HCV is a highly variable RNA virus with six major genotypes that are mainly transmitted via blood. Often asymptomatic, the infection progresses silently to chronic hepatitis C (CHC), which can lead to fibrosis, cirrhosis, and hepatocellular carcinoma (HCC). Direct-acting antivirals (DAAs) have revolutionized treatment, achieving cure rates above 95%,

Epigenetic alterations and memory: key players in the development/progression of chronic kidney disease promoted by acute kidney injury and diabetes.

Chronic kidney disease (CKD) is a highly prevalent global public health issue and can progress to kidney failure. Survivors of acute kidney injury (AKI) have an increased risk of progressing to CKD by 8.8-fold and kidney failure by 3.1-fold. Further, 20% to 40% of individuals with diabetes will develop CKD, also known as diabetic kidney disease (DKD). Thus, preventing these kidney diseases can positively impact quality-of-life and life-expectancy outcomes for affected individuals. Frequent episodes of hyperglycemia and renal hypoxia are implicated in the pathophysiology of CKD. Prior periods o

Perinatal S-Adenosylmethionine Supplementation Represses PSEN1 Expression by the Cellular Epigenetic Memory of CpG and Non-CpG Methylation in Adult TgCRD8 Mice.

DNA methylation, the main epigenetic modification regulating gene expression, plays a role in the pathophysiology of neurodegeneration. Previous evidence indicates that 5'-flanking hypomethylation of PSEN1, a gene involved in the amyloidogenic pathway in Alzheimer's disease (AD), boosts the AD-like phenotype in transgenic TgCRND8 mice. Supplementation with S-adenosylmethionine (SAM), the methyl donor in the DNA methylation reactions, reverts the pathological phenotype. Several studies indicate that epigenetic signatures, driving the shift between normal and diseased aging, can be acquired duri

Shared and distinct biological circuits in effector, memory and exhausted CD8+ T cells revealed by temporal single-cell transcriptomics and epigenetics.

Naïve CD8+ T cells can differentiate into effector (Teff), memory (Tmem) or exhausted (Tex) T cells. These developmental pathways are associated with distinct transcriptional and epigenetic changes that endow cells with different functional capacities and therefore therapeutic potential. The molecular circuitry underlying these developmental trajectories and the extent of heterogeneity within Teff, Tmem and Tex populations remain poorly understood. Here, we used the lymphocytic choriomeningitis virus model of acute-resolving and chronic infection to address these gaps by applying longitudinal

Early-life blockade of NMDA receptors induces epigenetic abnormalities in the adult medial prefrontal cortex: possible involvement in memory impairment in trace fear conditioning.

RATIONALE: Several findings indicate that early-life dysfunction of N-methyl-D-aspartate (NMDA) receptors might cause schizophrenia-like abnormalities in adulthood that might be induced by impairments in epigenetic regulation. OBJECTIVES: In the present study, we investigated whether postnatal blockade of NMDA receptors (within the first 3 weeks of life) by the competitive antagonist CGP 37849 (CGP) might affect some epigenetic markers in the adult medial prefrontal cortex (mPFC). METHODS: Histone H3 phosphorylation at serine 10 (H3S10ph), histone H3 acetylation at lysine 9 or 14 (H3K9ac or

Sensory cortical and subcortical auditory neurophysiological changes predict cue-specific extinction behavior enabled by the pharmacological inhibition of an epigenetic regulator during memory formation.

Extinction learning and memory have been broadly investigated at both behavioral and neural levels, but sensory system contributions to extinction processes have been less explored. Using a sound-reward extinction paradigm in male rats, we reveal both cortical and subcortical forms of plasticity associated with the cue-specificity of behavioral extinction memory. In the auditory cortex, frequency tuning narrowed by up to two-thirds of an octave around the remembered extinguished sound cue. Subcortical signals revealed in the auditory brainstem response (ABR) in the same animals developed small

Interleukin 1β triggers synaptic and memory deficits in Herpes simplex virus type-1-infected mice by downregulating the expression of synaptic plasticity-related genes via the epigenetic MeCP2/HDAC4 complex.

Extensive research provides evidence that neuroinflammation underlies numerous brain disorders. However, the molecular mechanisms by which inflammatory mediators determine synaptic and cognitive dysfunction occurring in neurodegenerative diseases (e.g., Alzheimer's disease) are far from being fully understood. Here we investigated the role of interleukin 1β (IL-1β), and the molecular cascade downstream the activation of its receptor, to the synaptic dysfunction occurring in the mouse model of multiple Herpes simplex virus type-1 (HSV-1) reactivations within the brain. These mice are characteri

Developmental toxicity of metals: Three decades of insights into mechanisms, epigenetics, and transgenerational inheritance.

Since the early 1990s, considerable progress has been made in understanding the teratogenic and embryotoxic effects of metals in mammalian systems. The present updated review synthesizes over three decades of research findings, examining the developmental toxicity of metals across four categories: (a) metals of greatest toxicological significance (arsenic, cadmium, lead, mercury, and uranium), (b) essential trace metals (chromium, cobalt, manganese, selenium, and zinc), (c) other metals with evident biological interest (nickel and vanadium), and (d) metals of pharmacological interest (aluminum

Metabolic-epigenetic imprinting of macrophages in endometriosis: a working model for lesion persistence and treatment.

Endometriosis is an estrogen-dependent inflammatory disorder that often persists or returns even after visible lesions have been removed. The behavior of peritoneal macrophages may help explain this clinical durability. These cells participate in lesion implantation, vascularization, fibrosis, defective immune clearance, and pain, but it is uncertain why disease-supporting phenotypes are maintained. We argue that repeated exposure to the endometriotic niche may leave macrophages with a metabolically conditioned epigenetic memory rather than only a short-lived polarization state. Lactate, succi

Reproductive toxicity and transgenerational effects of microplastics: Evidence from experimental models and underlying epigenetic mechanisms.

Micro- and nanoplastics (MNPs) are emerging environmental contaminants with ubiquitous distribution and growing concern for reproductive health. Recent detection of MNPs in human follicular fluid and seminal plasma suggests that these particles can enter the reproductive microenvironment and interfere with gametogenesis and reproductive function in both females and males. On the basis of summarizing the toxic effects of MNPs on the male and female reproductive systems, this review focuses on their potential impacts on offspring and subsequent generations. Current evidence suggests that MNPs co

Paternal preconception exercise and sperm epigenetic mechanisms: a systematic review on offspring health.

INTRODUCTION: Paternal biology before conception can influence offspring health beyond DNA sequence inheritance. Paternal exercise is a modifiable exposure that can remodel the male germline epigenome through DNA methylation, histone modifications, and small non-coding RNAs, potentially affecting offspring development and metabolism. METHODS: This systematic review searched PubMed/MEDLINE, Scopus, Web of Science, LILACS, and EMBASE without year restrictions. Experimental animal studies were eligible if sires exercised before conception and reported sperm or offspring epigenetic outcomes along

Multiple generation distinct toxicant exposures and epigenetic transgenerational inheritance of sex‑specific outcross impacts on sperm epigenetics and obesity pathology.

Environmental factors can influence the epigenetic inheritance of pathology across generations. Prior studies found various toxicants increased pathology in their descendants through epigenetic transgenerational inheritance. Previously, three generations of rats were exposed separately to vinclozolin (F0 generation), a jet fuel hydrocarbon mixture (F1 generation), and dichlorodiphenyltrichloroethane (DDT) (F2 generation), producing a transgenerational F5 generation. The current study extends this study by breeding this transgenerational F5 generation with wild-type rats to create maternal and

Bull Fertility and Epigenetics: Exploring Influences Beyond Genetics.

Infertility has been a concern throughout the ages and is still a significant problem in several species, including human beings and farm animals. Many research projects have focused on exploring the genetic basis of male infertility, but thus far, they have been able to explain not more than 15% of infertile cases. Therefore, much remains to be understood, and we need to involve several other approaches to fully understand the aetiology of male infertility. One such promising approach is epigenetics, which has the potential to explain the aetiology of numerous disorders partially. Epigenetics

Spermatogenic timing, dose, and route of administration as determinants of paternal transmission of developmental neurotoxicity.

Growing evidence challenges the long-standing assumption that the paternal germline is insulated from environmental perturbation and identifies preconception paternal exposure as an underrecognized pathway of developmental neurotoxicity. Across mammalian models, paternal exposure to substances of abuse, including cocaine, nicotine, ethanol, morphine, and cannabinoids, induces reproducible, often sex-specific alterations in offspring brain function and behavior, including changes in cognition, stress responsivity, reward processing, and affective behaviors, accompanied by transcriptional and ep

Transgenerational Effect of a Mixture of Pyrethroids in the Testis of Rats: A Transcriptome and Epigenome Analyses.

Pyrethroids exert toxic effects on multiple organ systems, including the male reproductive tract. We previously demonstrated the toxicity of a mixture of pyrethroids in rats when administered at doses equivalent to human exposure. It is possible that the toxic effects observed in the exposed generations may be transmitted to the next generations. In this study, the transgenerational toxicity on the transcriptome and epigenome was evaluated in the testes of rats exposed to a mixture of pyrethroids and in the testes of rats obtained from subsequent generations (up to F3 generation). Exposure to

The bull sperm epigenome: role in male fertility and transmission to the offspring.

Extensive DNA methylation and small non-coding RNA modifications occur during male germline differentiation, which are essential for normal spermatogenesis and sperm maturation. Recent evidence has underscored the role of these key regulators in male fertility. However, due to significant environmental and technical variations across the studies, the identification of epigenetic biomarkers relevant to fertility remains challenging. Nevertheless, recent studies using large cohorts and integrative approaches have highlighted sperm epigenetic signatures that improve fertility prediction. In addit

Sperm tRNA-derived fragments: molecular mechanisms and clinical implications for paternal epigenetic inheritance.

Sperm tRNA-derived fragments (tRFs) have emerged as novel regulators of paternal epigenetic inheritance. By modulating embryonic epigenetic reprogramming, transposon silencing, and intergenerational genetic programming, tRFs mediate the transgenerational transmission of paternal environmental information, extending conventional notions of genetic inheritance. This review systematically elaborates the molecular mechanisms, clinical applications, and potential interventional strategies of tRFs. Through an in-depth analysis of how tRFs regulate the epigenetic landscape during embryonic developmen
