Methanogenesis from acetate: a nonmethanogenic bacterium from an anaerobic acetate enrichment.

A methanogenic acetate enrichment was initiated by inoculation of an acetate-mineral salts medium with domestic anaerobic digestor sludge and maintained by weekly transfer for 2 years. The enrichment culture contained a Methanosarcina and several obligately anaerobic nonmethanogenic bacteria. These latter organisms formed varying degrees of association with the Methanosarcina, ranging from the nutritionally fastidious gram-negative rod called the satellite bacterium to the nutritionally nonfastidious Eubacterium limosum. The satellite bacterium had growth requirements for amino acids, a peptid

The role of lactate metabolism in shaping intestinal microbiota and health.

Lactate, particularly the D-isomer, which is produced exclusively by microbial fermentation, is usually low or undetectable in the healthy colon but can accumulate when there is a microbial imbalance, contributing to low pH and inflammation. Intestinal lactate-utilizing bacteria, such as Anaerobutyricum soehngenii, Anaerostipes species, and Coprococcus catus, convert lactate to short-chain fatty acids, including butyrate, potentially important for health. Here, we mined genomes of lactate utilizers for genes involved in propionate formation and conducted metabolic network modeling to investiga

Effects of in vitro fermentation of Astragalus polysaccharide on gut microbiota and neurotransmitter metabolites in patients with major depressive disorder.

BACKGROUND: Major depressive disorder (MDD) is a common and heterogeneous mental illness with an unclear pathogenesis and often suboptimal treatment outcomes. Astragalus polysaccharide (APS) has shown potential antidepressant effects and may serve as a functional food; however, its impact on the gut microbiota and metabolome in MDD remains unexplored. METHODS: Using an in vitro fermentation model coupled with 16S rRNA sequencing and targeted metabolomics, we characterized the gut microbial composition and neurotransmitter metabolites in fecal samples from 15 MDD patients and 15 healthy subjec

Identifying and ranking causal microbial biomarkers for colorectal cancer at different cancer subsites and stages: a Mendelian randomization study.

INTRODUCTION: The gut microbiome is directly involved in colorectal carcinogenesis, but much of the epidemiological evidence for the effect of the gut microbiome on colorectal cancer (CRC) risk comes from observational studies, and it is unclear whether identified microbial alterations are the cause or consequence of CRC development. METHODS: Univariate Mendelian randomization (MR) analysis and multivariate MR analysis based on Bayesian model averaging were performed to comprehensively explore the microbial risk factors associated with CRC. The Network Module Structure Shift method was used t

Alterations in gut microbiota and metabolite profiles in patients with infantile cholestasis.

BACKGROUND: Infantile cholestasis (IC) is the most common hepatobiliary disease in infants, resulting in elevated direct bilirubin levels. Indeed, hepatointestinal circulation impacts bile acid and bilirubin metabolism. This study evaluates changes in the gut microbiota composition in children with IC and identifies abnormal metabolite profiles associated with microbial alterations. RESULTS: The gut microbiota in the IC group exhibits the higher abundance of Veillonella, Streptococcus and Clostridium spp. (P < 0.05), compared to healthy infants (CON) group. Moreover, the abundance of Ruminoco

Consumption of partially hydrolysed guar gum stimulates Bifidobacteria and butyrate-producing bacteria in the human large intestine.

Partially hydrolysed guar gum (PHGG) is a water-soluble dietary fibre that is non-digestible in the upper gastrointestinal tract. It is believed that PHGG benefits the health of hosts by altering the colonic microbiota and stimulating short-chain fatty acid (SCFA) production. However, it remains unclear which bacteria ferment PHGG in the human large intestine. In this study, the effect of PHGG on faecal bacteria was analysed to specify the bacteria that contribute to the fermentation of PHGG in the human large intestine. Ten healthy volunteers consumed PHGG (6 g/day) for 2 weeks. Faeces were c

[Pathogen spectrum and resistance status of exclusively anaerobic odontogenic infections].

Out of 440 dentogenic pyogenic infections, 171 exclusively caused by anaerobes were investigated to understand the importance of anaerobic bacteria in dental pyogenic processes better. Grampositive anaerobic bacteria dominated. The predominant grampositive isolates in monoinfections were Peptostreptococci and in the case of mixed infections, strains of the genus Eubacterium. Strains of Prevotella and Porphyromonas dominated the gramnegative anaerobic spectra. The resistance to penicillin was very low. Altogether, only one strain of Prevotella oris and one strain of Prevotella oralis showed res

Paecilomyces cicadae-fermented Radix astragali modulates short-chain fatty acids metabolism in the intestine through gut microbiota and ameliorates hyperuricaemia.

BACKGROUND AND PURPOSE: Hyperuricaemia, a severe metabolic disorder linked to gout, is increasingly prevalent worldwide. Radix astragali, as a drug-food homologous material, has significantly expanded the market for novel functional foods. The solid-state fermentation products of R. astragali and Paecilomyces cicadae (RPF) promise to be candidates for lowering uric acid. Here, we have investigated the effects of RPF, using a model of hyperuricaemia in rats. EXPERIMENTAL APPROACH: Hyperuricaemia, and damage to liver and kidney, was induced in male Sprague-Dawley rats receiving a high purine di

Microbiome Responses of Beef Steers to Rotational Grazing of Toxic Endophyte-Infected Tall Fescue Under Fall Conditions.

Fescue toxicosis results from ingestion of tall fescue infected with the ergot alkaloid (EA)-producing endophyte Epichloë coenophiala. The gastrointestinal microbiota is the first point of interaction with EAs and may be modulated by them. To investigate this, rumen and fecal samples were collected from steers grazing toxic (E+) or non-toxic fescue for 14 days, followed by a pasture switch. Microbial communities were characterized using 16S rRNA amplicon sequencing (bacterial V4, archaeal V6-V8 regions). Short-term E+ grazing did not affect alpha diversity in either matrix, but beta diversity

Peri-implantitis biofilm from explanted implants in Korean patients: microbial and functional profiling.

Peri-implantitis is an inflammatory disease affecting tissues surrounding dental implants, with microbial biofilms recognized as the primary etiological factor. However, most previous studies analyzed samples from peri-implant pockets, and research on biofilms directly attached to explanted implant surfaces remains limited. This study compared the microbial composition and functional characteristics of biofilms from explanted implant surfaces in peri-implantitis cases with subgingival plaque from healthy controls. A total of 41 samples (peri-implantitis n=19, healthy controls n=22) were obtain
