Lipidome profiling of neutrophil-derived extracellular vesicles unveils their contribution to the ensemble of synovial fluid-derived extracellular vesicles during joint inflammation.

The molecular signature of cell-derived extracellular vesicles (EVs) from synovial fluid (SF) offers insights into the cells and molecular processes associated with joint disorders and can be exploited to define biomarkers. The EV-signature is determined by cargo molecules and the lesser-studied lipid bilayer. We here investigated the lipidome of SF-EVs in inflamed joints derived from Rheumatoid Arthritis (RA) and Spondyloarthritis (SpA) patients, two autoimmune-driven joint diseases, and compared these signatures to the lipid profile of equine SF-EVs obtained during induced acute synovitis. Extracellular Fluid/Intracellular Fluid Volume Ratio as a Novel Risk Indicator for All-Cause Mortality and Cardiovascular Disease in Hemodialysis Patients.

BACKGROUND: In hemodialysis patients, fluid overload and malnutrition are accompanied by extracellular fluid (ECF) expansion and intracellular fluid (ICF) depletion, respectively. We investigated the relationship between ECF/ICF ratio (as an integrated marker reflecting both fluid overload and malnutrition) and survival and cardiovascular disease (CVD) in the context of malnutrition-inflammation-arteriosclerosis (MIA) complex. METHODS: Seventy-seven patients from a single hemodialysis unit were prospectively enrolled. The ECF/ICF volume was measured by segmental multi-frequency bioimpedance a

Non-canonical cleavage of GPR125/ADGRA3 releases extracellular receptor fragments detectable in cerebrospinal fluid.

Adhesion G protein-coupled receptors (aGPCRs) are crucial for barrier tissues and typically autoproteolytically cleaved into N- and C-terminal fragments (NTF and CTF) that remain non-covalently associated until separation is triggered. Here, we show that human GPR125 (ADGRA3), expressed in choroid plexus epithelium, is not cleaved at the GPCR autoproteolysis site (GPS), consistent with its divergent SLS motif. Instead, it undergoes non-canonical proteolytic processing that generates and releases atypical, non-canonical N-terminal fragments (ncNTFs), a process further enhanced by insulin. Using

Follicular fluid-derived small extracellular vesicles during individual in vitro maturation improve blastocyst development.

INTRODUCTION: We evaluated the impact of follicular fluid-derived small extracellular vesicles (FF-sEVs) supplementation during oocyte maturation in vitro on bovine embryo outcomes, comparing group and individual culture systems. METHODS: Follicular fluid was aspirated from dominant follicles of four nulliparous Holstein heifers at 4.5 days post-ovulation. Small extracellular vesicles were isolated, characterized, and pooled to ensure balanced donor contribution. To confirm uptake, FF-sEVs were fluorescently labelled and co-cultured with cumulus-oocyte complexes (COCs) during in vitro maturat

CD9+ and CD82+ extracellular vesicles in synovial fluid differentiate aseptic from septic endoprosthesis loosening.

Aim: Extracellular vesicles (EVs) hold great promise as emerging biomarkers for a variety of diseases. However, their clinical application is still hindered by complex and time-consuming isolation procedures. A clinically relevant scenario where improved biomarker-based diagnostics are urgently needed is prosthetic joint loosening, which may result from either aseptic inflammation or periprosthetic joint infection (PJI) - two conditions requiring fundamentally different therapeutic approaches. This study investigated whether imaging flow cytometry (IFCM) enables the discrimination between asep

Extracellular Vesicles From Follicular Fluid in Infertile Women: Size, Morphology and miRNA Content Analysis.

The declining birth rates and fertility challenges in Europe have intensified global concerns over rising infertility, particularly among women. This study decisively investigates follicular fluid-related extracellular vesicles (FF-EVs) from infertile patients with polycystic ovary syndrome (PCOS) or diminished ovarian reserve (DOR) undergoing in vitro fertilization (IVF), comparing them to a healthy control group. We have identified significant variations in protein content and polydispersity in crude follicular fluid using UV-Vis absorption and dynamic light scattering (DLS) techniques. Furt

Cerebrospinal fluid-driven extracellular vesicle as a potential diagnostic biomarker for multiple system atrophy.

Given that Parkinsonian disorders, including Parkinson's disease (PD) and multiple system atrophy-parkinsonism (MSA-P), often have similar clinical presentations, making a clinical diagnosis can be challenging. In the present study, we extracted and analyzed extracellular vesicles (EVs) from the cerebrospinal fluid of individuals diagnosed with PD (N = 16), MSA-P (N = 16), and amyotrophic lateral sclerosis (ALS) (N = 16), with the latter serving as the control group. Transcriptomic analysis identified 7426 and 8003 differentially expressed RNAs between MSA and ALS, and MSA and PD, respectively

Inflammatory proteins and neutrophil extracellular traps increase in burn blister fluid 24h after burn.

Burn wound blister fluid is a valuable matrix for understanding the biological pathways associated with burn injury. In this study, 152 blister fluid samples collected from paediatric burn wounds at three different hospitals were analysed using mass spectrometry proteomic techniques. The protein abundance profile at different days after burn indicated more proteins were associated with cellular damage/repair in the first 24 h, whereas after this point more proteins were associated with antimicrobial defence. The inflammatory proteins persisted at a high level in the blister fluid for more than

Oviduct Fluid Extracellular Vesicles Change the Phospholipid Composition of Bovine Embryos Developed In Vitro.

Oviduct fluid extracellular vesicles (oEVs) have been proposed as bringing key molecules to the early developing embryo. In order to evaluate the changes induced by oEVs on embryo phospholipids, fresh bovine blastocysts developed in vitro in the presence or absence of oEVs were analyzed by intact cell MALDI-TOF (Matrix assisted laser desorption ionization-Time of flight) mass spectrometry (ICM-MS). The development rates, cryotolerance, and total cell number of blastocysts were also evaluated. The exposure to oEVs did not affect blastocyst yield or cryotolerance but modified the phospholipid co

Surface Profiling of Extracellular Vesicles from Plasma or Ascites Fluid Using DotScan Antibody Microarrays.

DotScan antibody microarrays were initially developed for the extensive surface profiling of live leukemia and lymphoma cells. DotScan's diagnostic capability was validated with an extensive clinical trial using mononuclear cells from the blood or bone marrow of leukemia or lymphoma patients. DotScan has also been used for the profiling of surface proteins on peripheral blood mononuclear cells (PBMC) from patients with HIV, liver disease, and stable and progressive B-cell chronic lymphocytic leukemia (CLL). Fluorescence multiplexing allowed the simultaneous profiling of cancer cells and leukoc

Effects of extracellular matrix protein functionalized fluid membrane on cell adhesion and matrix remodeling.

In order to study cellular responses and extracellular matrix protein remodeling mediated by biomaterials coating, we proposed a biomimetic construct containing protein-conjugated supported lipid bilayers (SLBs) as a cell culture platform. Single or multi-component proteins-bound SLBs were fabricated by conjugating type I collagen and/or fibronectin on the N-hydroxysulfosuccinimide-functionalized SLBs. The proposed protein-conjugated systems were quantitatively characterized by the quartz crystal microbalance with dissipation. NIH 3T3 fibroblasts were cultured on the model constructs and on ox

Acute extracellular fluid volume changes increase ileocolonic resistance to saline flow in anesthetized dogs.

We determined the effect of acute extracellular fluid volume changes on saline flow through 4 gut segments (ileocolonic, ileal, ileocolonic sphincter and proximal colon), perfused at constant pressure in anesthetized dogs. Two different experimental protocols were used: hypervolemia (iv saline infusion, 0.9% NaCl, 20 ml/min, volume up to 5% body weight) and controlled hemorrhage (up to a 50% drop in mean arterial pressure). Mean ileocolonic flow (N = 6) was gradually and significantly decreased during the expansion (17.1%, P < 0.05) and expanded (44.9%, P < 0.05) periods while mean ileal flow

GW4869 Depletes Macrophages and Increases Number of Extracellular Vesicles in Murine Peritoneal Cavity Fluid.

Pharmacological strategies to modulate extracellular vesicle (EV) release in vivo are gaining traction, yet their broader effects on local immune microenvironments remain unclear. In this study, we assessed how repeated intraperitoneal (i.p.) dosing of the neutral sphingomyelinase inhibitor GW4869 reshapes the peritoneal cavity. GW4869 administration triggered a strong local inflammatory reaction, was accompanied by a selective depletion of resident peritoneal macrophages and was associated with higher EV counts 24 h after the final injection. Targeted macrophage depletion in the absence of GW

Isolation of extracellular vesicles from minimal volume ascites fluid using strong anion exchange beads.

Ovarian cancer (OC) remains a leading cause of gynecologic cancer mortality due to late-stage diagnosis and limited early detection strategies. Ascites fluid, a pathological hallmark of OC, is a rich source of tumor-derived extracellular vesicles (EVs) that reflect the tumor microenvironment and hold promise for biomarker discovery. However, isolating EVs from minimal ascites volumes (<100 µl) poses technical challenges using conventional methods like ultracentrifugation or size-exclusion chromatography (SEC). This study explores the application of strong anion exchange (SAX) magnetic beads (M

Distinctive features of blood- and ascitic fluid-derived extracellular vesicles in ovarian cancer patients.

BACKGROUND: Ovarian cancer (OC) is a highly aggressive malignancy characterized by early dissemination of cancer cells from the surface of the ovary to the peritoneum. To gain a deeper understanding of the mechanisms associated with this intraperitoneal spread, we aimed to characterize the role of extracellular vesicles (EVs) in metastatic colonization in OC. METHODS: To this purpose, a total of 150 samples of ascitic fluids, blood serum, tumor and normal tissues from 60 OC patients, were extensively analyzed to characterize the EVs released in blood and ascitic fluids of OC patients, in term

Phosphorylated tau in cerebrospinal fluid-derived extracellular vesicles in Alzheimer's disease: a pilot study.

Alzheimer's disease (AD) is a debilitating neurodegenerative disorder characterized by brain aggregation of β-amyloid (Aβ) peptides and phosphorylated tau (P-tau) proteins. Extracellular vesicles (EVs) can be isolated and studied for potential roles in disease. While several studies have tested plasma-derived EVs in AD, few have analyzed EVs from cerebrospinal fluid (CSF), which are potentially more closely related to brain changes. This study included 20 AD patients and 20 cognitively unimpaired (CU) participants. Using a novel EV isolation method based on acoustic trapping, we isolated and p

RNA Profiles of Tear Fluid Extracellular Vesicles in Patients with Dry Eye-Related Symptoms.

Currently, diagnosing and stratifying dry eye disease (DED) require multiple tests, motivating interest in a single definitive test. The purpose of this study was to investigate the potential for using tear fluid extracellular vesicle (EV)-RNA in DED diagnostics. With a role in intercellular communication, nanosized EVs facilitate the protected transport of diverse bioactive molecules in biofluids, including tears. Schirmer strips were used to collect tears from 10 patients presenting with dry eye-related symptoms at the Norwegian Dry Eye Clinic. The samples comprised two groups, five from pat
