The responses of inhibitory neurons/synapses to motoneuron injury in the cranial nervous system remain to be elucidated. In this study, we analyzed GABAA receptor (GABAAR) and GABAergic neurons at the protein level in the transected rat facial nucleus. Immunoblotting revealed that the GABAARα1 protein levels in the axotomized facial nucleus decreased significantly 5-14 days post-insult, and these levels remained low for 5 weeks. Immunohistochemical analysis indicated that the GABAARα1-expressing cells were motoneurons. We next examined the specific components of GABAergic neurons, including gl

Descending brainstem projections from the pedunculopontine tegmental nucleus (PPN) were studied in the rat by use of the anterograde tracer Phaseolus vulgaris-leucoagglutinin (PHA-L) and the retrograde tracer lectin-conjugated horseradish peroxidase (HRP-WGA). Results of these experiments demonstrated prominent bilateral projections to the pontomedullary reticular nuclei, but direct connections to the motor and sensory nuclei of the cranial nerves could not be ascertained. The PPN fibers terminated mainly in the pontine reticular nuclei oralis and caudalis and in ventromedial portions (pars al

The number of neuroglia was estimated in the retrofacial, facial and trigeminal motor nuclei of the mouse brain at 6, 25, 28 and 31 months of age. There was no statistically significant variation in neuroglial number in either the facial or trigeminal motor nuclei despite changes in neuron number. There was a decrease in the number of neuroglia in the retrofacial nucleus between 25 and 28 months which was slightly greater than the decrease in neuronal number, followed by a small increase in neuroglial number between 28 and 31 months of age. The glia-to-neuron ratio in the facial and trigeminal

8-L-Arginine vasopressin ([Arg8]VP) receptors are expressed transiently in the rat facial nucleus during the perinatal period. Electrophysiological studies suggest that at least part of these receptors is located on facial motoneurones. In the present study we report that, in the adult rat, unilateral section of a facial nerve results in a massive and transient reexpression of [Arg8]VP receptors in the deeferented facial nucleus. Data were obtained by quantitative film autoradiography. During the first 2 postoperative weeks, binding of an iodinated ligand selective for V1a-type receptors incre

Horseradish peroxidase was applied to a ligation of the chorda tympani nerves running alongside the submandibular duct in order to label the preganglionic parasympathetic salivatory neurons in the brain stem. Labelled neurons were found in the ipsilateral motor trigeminal nucleus, lateral reticular formation and in the facial motor nuclei of both sides. However, the labelling of neurons in the motor nuclei of the trigeminal and facial nucleus was caused by the uptake of horseradish peroxidase by axons in the musculature surrounding the injection site which were probably damaged by the dissecti

OBJECTIVE: Weakness and synkinesis (involuntary cocontraction of different muscle groups) are common sequelae after facial nerve injury. We describe a rabbit model of facial nerve axotomy and repair, which can be used to study such sequelae and propose a grading tool to assess the facial movement outcomes. Using this rabbit model, we assess the effect of delaying facial nerve repair on the quality of the clinical result.
METHODS: A total of 15 rabbits (30 facial halves) were divided into 4 groups: control, facial nerve main trunk axotomy and immediate repair, axotomy and repair at 2.5 weeks a

Facial motoneurons respond to peripheral transection of the facial nerve with a number of molecular changes. In order to obtain insight into the transcriptional mechanisms underlying the changes induced by axotomy, the expression of a number of immediate early genes was investigated after facial nerve lesion in the rat. Some immediate early genes (such as c-fos, c-jun or jun B) are known to encode transcription factors that bind to DNA at sites that regulate gene expression and they could therefore contribute to long-term changes in motoneurons. Northern blot analysis of RNA extracted from the

OBJECT: Hemifacial spasm (HFS) is thought to be due to a hyperactive facial motor nucleus consequent to chronic neurovascular contact. The lateral spread (LS) response is presumed to reflect changes in facial motor neuron excitability. Facial muscle motor evoked potentials (MEPs) use the same efferent pathway as LS, therefore the authors speculated that these potentials should reflect differences consistent with changes at the facial motor nucleus level.
METHODS: Monitoring of LS and bilateral facial MEP was performed in 10 consecutive patients undergoing MVD for HFS. Ipsilateral facial MEPs

Cytofluorometric quantification of the accumulation of somatopetally transported fluoroisothiocyanate (FITC)-labelled wheat germ agglutinin (WGA) and Concanavalin A (Con A) were performed on sections of the mouse facial nucleus. The lectins were injected bilaterally in the snout region of six-week-old mice eleven days after one of the facial nerves had been crushed. Analysis of variance revealed a significantly higher uptake of both WGA and Con A in the regenerating than in the control nerve. WGA appeared to be taken up to a higher degree than Con A, both in normal and regenerating nerve. Afte

The development of facial nuclei in animal models of disease is poorly understood, but autism is sometimes associated with facial palsy. In the present study, to investigate migration of facial neurons and initial facial nucleus formation in an animal model of autism, rat embryos were treated with valproic acid (VPA) in utero at embryonic day (E) 9.5 and their facial nuclei were analyzed by in situ hybridization at E13.5, E14.5 and E15.5. Signals for Tbx20, which is expressed in early motor neurons, appeared near the floor plate at the level of the vestibular ganglion and extended caudolateral

Motoneuron number and expression of cytoplasmic RNA and ribosomal RNA (rRNA) gene transcription activity in the facial nucleus were examined quantitatively and chronologically for up to 4 weeks in rats after facial nerve axotomy and avulsion in order to elucidate interrelationships in axonal changes. The right facial nerves of adult Fischer rats were avulsed at a portion of the outlet or axotomized at a portion of the foramen stylomastoideus. The number of large motoneurons in the facial nucleus was reduced by 40% 2 weeks after avulsion and by 70% 4 weeks after avulsion but displayed a 19% los

Deafferentation of motoneurons after facial nerve injury is a well-documented phenomenon but whether synaptic inputs to facial motoneurons are completely restored after reinnervation is unknown. Here, we tested the hypothesis that deficits in motor performance after transection/suture of the facial nerve (facial-facial anastomosis, FFA) in adult rats are associated with incomplete recovery of synaptic inputs. At 2 months after FFA, we found, in congruence with previous results, that the amplitude of whisking had recovered to only 31 % of control (sham operation). In the same FFA-treated rats,

The sensory modalities of taste and touch, for the anterior tongue, are relegated to separate cranial nerves. The lingual branch of the trigeminal nerve mediates touch: the chorda tympani branch of the facial nerve mediates taste. The chorda tympani also contains efferent axons which originate in the superior salivatory nucleus. The central projections of these two nerves have been visualized in the hamster by anterograde labelling with horseradish peroxidase (HRP). Afferent fibers of the chorda tympani distribute to all rostral-caudal levels of the solitary nucleus. They synapse heavily in th

The effects of stimulation of the vestibular nerve, spinal trigeminal nucleus, facial and hypoglossal nuclei of the cranial nerves on the neuronal activity in the lateral vestibular nucleus of Deiters were studied in cats anaesthetized with pentobarbitone. Stimulation of these nuclei was found to produce antidromic and synaptic activation of Deiters' neurons. Descending axon collaterals of the vestibular neurons to these brainstem structures were revealed. Stimulation of the VIIIth nerve, spinal trigeminal and facial nuclei evoked mono- and polysynaptic excitatory postsynaptic potentials in De

Central pathways converging upon the trigeminofacial motor nuclei of the mallard were studied in order to elucidate neuroanatomically the presumed influence of primary sensory trigeminal afferents upon jaw muscle activity. The techniques used included the Fink-Heimer I method after lesions, and axonal transport labeling following injections of 3H-leucine or of HRP for retrograde identification of the neurons of origin. A general description is given of the trigeminofacial motor complex. Jaw closer muscles are innervated by trigeminal motor neurons, and facial motor neurons innervate the jaw de

We report a case of left-sided horizontal gaze palsy, ipsilateral adduction weakness, and left peripheral facial weakness, all of which indicate the lesion in the left median pontine tegmentum. The enhanced MRIs revealed a discrete left median pontine tegmental lesion, involving the abducens nucleus, MLF, and facial nerve knee. This lesion spared the area of the left PPRF. Among these structures, the area of the abducens nucleus seems to be responsible for the unilateral horizontal gaze palsy. We are not aware of any previous precise neuroradiological documentation of unilateral paralysis of c
