During pregnancy, maternal plasma concentrations of the peroxidation-susceptible polyunsaturated fatty acids (polyenes) increase. In addition, the proportion of polyenes is higher in neonatal plasma than in maternal plasma. To study whether these increased amounts of polyenes affect antioxidant levels, we measured lipid-soluble antioxidants in maternal and neonatal plasmas obtained during thirty-five normal pregnancies. These values were then related to the degree of phospholipid-fatty acid unsaturation. Maternal plasma levels of tocopherols and lutein increased during pregnancy, as assessed a

Epstein-Barr virus (EBV) drives over 200,000 cancer cases annually, including diffuse large B-cell lymphoma, Burkitt lymphoma, and classic Hodgkin lymphoma-malignancies that frequently originate from germinal centers (GCs), which are physiologically hypoxic (O2 < 1%). However, conventional transformation models are typically conducted under 21% O2-an artificial condition that fails to replicate the hypoxic GC microenvironment and may obscure critical metabolic vulnerabilities that are therapeutically targetable. Therefore, therapeutic targets identified under 21% O2 conditions may not fully tr

Gut microbiota plays an essential role in the progression of nonalcoholic fatty liver disease (NAFLD), making the gut-liver axis a potential therapeutic strategy. Bacteroides genus, the enriched gut symbionts, has shown promise in treating fatty liver. However, further investigation is needed to identify specific beneficial Bacteroides strains for metabolic disorders in NAFLD and elucidate their underlying mechanisms. In this study, we observed a positive correlation between the abundance of Bacteroides thetaiotaomicron (B. theta) and the alleviation of metabolic syndrome in the early and end

We previously reported that glycation induces insulin resistance in the hearts of newborn pups from a gestational diabetes mellitus (GDM) rat model. Administration of n-3 unsaturated fatty acids suppressed glycation and improved signaling in GDM rat pups. In this study, we investigated their effects on cranial neurons using the GDM rat model and PC12 cells derived from rat adrenal pheochromocytomas. Additionally, we examined whether n-3 and n-7 unsaturated fatty acids (cis-palmitoleic acid [CPA] and trans-palmitoleic acid [TPA]) ameliorate the detrimental effects of high glucose exposure on ra

The ability to detect spoilage and the nutrient content of salmon is critical for ensuring food safety and determining market value. In this paper, we document the development of a hydrophilic-lipophilic balanced solid-phase microextraction arrow that offers better extraction performance than two other commercial devices. The devices were also compared using two operational models: headspace microextraction and direct immersion. The multidimensional statistical analysis and heatmap analysis for the headspace microextractions showed that the content levels of volatile metabolites including hydr

BACKGROUND: Postmenopausal osteoporosis (PMOP) is a serious problem for the women over 50 years old. Natural product puerarin (PUE) has been proven to improve PMOP with high safety. PMOP is a metabolic disorder affecting bone metabolism, indicating that endogenous metabolites amelioration may be a novel strategy for PMOP therapy. However, what the metabolic profile of POMP will be after PUE treatment is still obscure.
PURPOSE: We purpose to figure out the metabolic characteristics of PMOP and to explore the intrinsic mechanism on the anti-osteoporosis efficacy after PUE treatment based on the

Aromatic amine drugs like aminoglutethimide (AG) and related congeners have been shown to produce phenyl radicals through metabolism by myeloperoxidase (MPO)/H(2)O(2), which has been proposed to play a role in drug-induced agranulocytosis. AG has also been shown to induce MPO protein radical formation, but the ultimate fate of these metabolically generated phenyl radicals is still unknown. We tested the reactivity of linoleic acid (LA) and GSH with aniline-based compounds in the presence of horseradish peroxidase (HRP)/H(2)O(2) by measuring oxygen consumption. We found a qualitative correlatio

Diabetic nephropathy (DN) is the leading cause of end-stage renal disease, and the protein glycation pathway has a major role in DN pathogenesis. Accumulation of advanced glycation end products (AGEs) upregulates the Receptor for Advanced Glycation End products (RAGE)-NF-κB pathway, leading to elevated oxidative stress and inflammation. The present study aims to shed light on the anti-glycation mechanism of unsaturated fatty acids (UFAs), such as oleic acid (OA), linoleic acid (LA), and docosahexaenoic acid (DHA), on glycated human serum albumin (G. HSA) in an in vitro setup using multispectro

BACKGROUND: In dentistry, gingival hyperpigmentation is a common aesthetic issue caused mainly by excessive melanin deposits. Traditional surgical methods for depigmentation, like surgical techniques, can be effective, but they often associated with pain after the procedure and longer healing times. Because of their anti-inflammatory and antioxidant qualities, omega-3 polyunsaturated fatty acids (PUFAs) have demonstrated potential benefits in tissue regeneration and dermatological depigmentation. OBJECTIVE: To investigate the effectiveness of locally and systemically administered Omega-3 (PUFA

Specific locations of carbon-carbon double bonds (C═C) in lipids often play an essential role in biological processes, and there has been a booming development in C═C composition analysis by mass spectrometry. However, a universal derivatization and fragmentation pattern for the annotation of C═C positions in lipids is still challenging and attractive. To expand this field in lipidomics, a flexible and convenient N-tosylaziridination method was developed, with high derivatization efficiency, sensitivity, and specificity. The derivatization was very fast (15 s), and C═C numbers as well as locat

Staphylococcus aureus is a major human pathogen that secretes several toxins associated with the pathogenesis of sepsis and pneumonia. Its antibiotic resistance is notorious, and its biofilms play a critical role in antibiotic tolerance. We hypothesized fatty acids might inhibit S. aureus biofilm formation and the expressions of its virulence factors. Initially, the antibiofilm activities of 27 fatty acids against a methicillin-sensitive S. aureus strain were investigated. Of the fatty acids tested, three C18 unsaturated fatty acids, that is, petroselinic, vaccenic, and oleic acids at 100 μg/m

Free fatty acid (FFA) intake regulates blood pressure and vascular reactivity but its direct effect on contractility of systemic arteries is not well understood. We investigated the effects of saturated fatty acid (SFA, palmitic acid), polyunsaturated fatty acid (PUFA, linoleic acid), and monounsaturated fatty acid (MUFA, oleic acid) on the contractility of isolated mesenteric (MA) and deep femoral arteries (DFA) of Sprague-Dawley rats. Isolated MA and DFA were mounted on a dual wire myograph and phenylephrine (PhE, 1-10 µM) concentration-dependent contraction was obtained with or without FFAs

Primary angle-closure glaucoma (PACG) is a severe chronic neurodegenerative disease in Asia. Identification of important metabolites associated with PACG is crucial for early intervention and advancing knowledge of the disease mechanism. We applied gas chromatography-mass spectrometry (GC-MS) for targeted metabolomic analysis on serum samples from 38 newly diagnosed PACG patients and 48 controls. Palmitoleic acid (PA), linoleic acid (LA), γ-linolenic acid (GLA) and arachidonic acid (ARA) were identified as important metabolites associated with PACG. PA and GLA were significantly elevated (p <

Incubation of Schistosoma mansoni lung-stage larvae in 90% corn oil for 6 hr was shown to elicit exposure of their, otherwise masked, apical membrane antigens to binding of anti-schistosome antibodies in the indirect membrane immunofluorescence test (IF). The possibility that unsaturated fatty acids (FA) are responsible for this effect was herein supported by IF data on ex vivo lung-stage larvae of S. mansoni and S. haematobium incubated for 1/2-2 hr with 80% corn oil, 50% olive oil, or 10-20 microM arachidonic acid. Treatment with unsaturated FA followed by filipin staining for cholesterol vi

Unsaturated fatty acids, such as oleic acid, increase both the affinity for Ca2+ and the maximum effect of the Ca(2+)-ATPase of red blood cells [Niggli et al. (1981) J. Biol. Chem. 256, 8588-8592]. With the aim of examining the structural and kinetic details of the interaction between unsaturated fatty acids and the enzyme, we designed and synthesized 8-(5'-azido-O-hexanoylsalicylamido)octanoic acid (AS86), a photoactivatable analogue of unsaturated fatty acids. AS86, interacting noncovalently with the enzyme, shares with oleic acid the following properties: (i) it binds reversibly to the plas

This study is undertaken to reveal the molecular origins of non-ideal aggregation in ionic liquid (IL)-surfactant systems by investigating the micellization behaviour of a double-tailed imidazolium-based 1,3-didecyl-2-methylimidazolium chloride in binary mixtures with sodium oleate and sodium linoleate. Surface tension measurements indicate significant non-ideality, with much lower critical micelle concentrations (CMCs) and very negative interaction parameters, owing to strong synergistic interactions. Electrostatic ion pairs form upon mixing oppositely charged surfactants, providing enhanced

Acne vulgaris is a disease of pilosebaceous units with multifactorial pathogenesis, including hyperkeratinization, increased sebum secretion, and inflammation. Recently, it was suggested that acne subjects may have also impaired skin barrier. We hypothesized that excess unsaturated free fatty acids (UFFA) present in the sebum may cause barrier impairment associated with increased follicular stratum corneum (SC) thickening and inflammation seen in acne. Therefore, epidermal and sebaceous lipid profiles from acne and healthy subjects were analyzed and an in vitro epidermal tissue model was devel

AIM: To investigate the effect of ectogenesis unsaturated fatty acid on carbachol-induced muscarinic current (ICCh) and its mechanism.
METHODS: Using the whole-cell patch-clamp technique, ICCh was recorded in single smooth muscle cell isolated from the antral circular smooth muscles of guinea-pig stomach.
RESULTS: Arachidonic acid (AA) was added in external perfusing solution and AA inhibited ICCh to 46 %+/-8 %, 23 %+/-5 %, and 3.8 %+/-0.9 % at 1, 3, and 5 micromol/L. Another unsaturated fatty acid, linoleic acid (LA) also inhibited ICCh in a dose-dependant manner. LA inhibited ICCh to 69 %+

The influence of unsaturated fatty acid molecular geometry on the assembly and digestive properties of starch-lipid complexes remains unclear. Here, five UFAs with distinct chain lengths and configurations were incorporated into gelatinized wheat starch via ultrasound-assisted dynamic high-pressure microfluidization to construct hierarchical complexes. Multiscale analyses showed that UFA geometry governed amylose helix inclusion, V-type crystallinity, and lamellar packing. Monounsaturated UFAs promoted compact ordered assemblies, whereas highly unsaturated UFAs generated looser interfacial str
