Eco-friendly spectrofluorimetric determination of fenbendazole in pharmaceutical, plasma, and milk samples using erythrosine B as a fluorescent probe.

A simple, sensitive, and sustainable spectrofluorimetric method was developed to determine fenbendazole (FEN) based on its fluorescence-quenching interaction with erythrosine B (EB). Under the optimized conditions, FEN efficiently quenched the native fluorescence of EB (λ ex = 527 nm, λ em = 544 nm). The decrease in fluorescence intensity was proportional to FEN concentration over 0.050-3.0 µg mL-1 (r 2 = 0.9997), with limits of detection and quantification of 0.014 and 0.043 µg mL-1, respectively. Stern-Volmer analysis, thermodynamic studies, Job's method, spectral analysis, and quantum-chemi

Drug-Induced Liver Injury Following Co-ingestion of Veterinary Fenbendazole and Ivermectin for Prostate Cancer: A Case Report.

The self-administration of veterinary-grade anthelmintics, such as fenbendazole and ivermectin, has gained visibility in online communities due to unverified claims regarding potential anticancer effects. Despite these claims, these agents lack human regulatory approval due to unestablished safety profiles. We report a rare case of severe drug-induced liver injury (DILI) in a 65-year-old male with prostate cancer resulting from the concurrent ingestion of veterinary-grade fenbendazole and ivermectin. The patient presented to his primary care provider with a 2-week history of fatigue, jaundice,

Anticancer Evaluation of Methoxy Poly(Ethylene Glycol)-b-Poly(Caprolactone) Polymeric Micelles Encapsulating Fenbendazole and Rapamycin in Ovarian Cancer.

PURPOSE: We aimed to inhibit ovarian cancer (OC) development by interfering with microtubule polymerization and inhibiting mTOR signaling. To achieve this, previously developed micelles containing fenbendazole and rapamycin were applied. METHODS: Herein, we prepared micelles for drug delivery using fenbendazole and rapamycin at a 1:2 molar ratio and methoxy poly(ethylene glycol)-b-poly(caprolactone)(mPEG-b-PCL) via freeze-drying. We revealed their long-term storage capacity of up to 120 days. Furthermore, a cytotoxicity test was performed on the OC cell line HeyA8, and an orthotopic model was

A small-scale survey of fenbendazole resistance in Ascaridia galli and Heterakis gallinarum, two common ascarid parasites of poultry.

Resistance to benzimidazole (BZ) anthelmintics is a widespread problem in parasitic nematodes that infect production animals such as sheep and goats, and an emerging issue in ascarid parasites of poultry. Ascarid parasites are highly prevalent across commercial poultry species and are associated with significant production losses. The BZ drug fenbendazole (FBZ) is the only approved treatment for ascarid infections in poultry, and previously, FBZ resistance has been identified in Ascaridia dissimilis, the large ascarid of turkeys, and Heterakis gallinarum, the cecal ascarid. Here, we have condu

Gastrointestinal parasites of captive European bison Bison bonasus (L.) with a sign of reduced efficacy of Haemonchus contortus to fenbendazole.

The history of European bison Bison bonasus Linnaeus, 1758 has been stormy since its extinction in the wild after the First World War. Due to the fact that the species was restored from just 12 founders, further expansion has suffered from low genetic variability, rendering the bison vulnerable to various pathogens due to inbreeding depression. Parasites are recognised as a key biological threat to bison population. Thus, parasitological examination including monitoring of the level of anthelmintic resistance in a herd should be a routine procedure involved in management and protection of Euro

Efficacy of concomitant early summer treatment with fenbendazole and clorsulon against Fasciola hepatica and gastrointestinal nematodes in calves in Louisiana.

The efficacy, safety, and compatibility of fenbendazole (FBZ) and clorsulon (CLN) were tested after oral administration of label recommended and of higher (5x) dosage rates to calves naturally infected with gastrointestinal nematodes and Fasciola hepatica. Results for 42 calves allotted to 4 treatment groups indicated a similar efficacy against mature F hepatica by FBZ (5 mg/kg of body weight) and CLN (7 mg/kg) in a combined oral suspension, compared with CLN (7 mg/kg) alone (100 vs 99% reduction). A lesser efficacy was observed against immature flukes (88.6 and 84.9% reduction, respectively).

[Morphofunctional changes in the digestive system of the nematode Passalurus ambiguus (Rudolphi, 1819) after use of the anthelmintics albendazole, fenbendazole, and ivermectin].

The paper presents the results of histological and electron microscopic studies of the tissues of the digestive system of the nematode Passalurus ambiguus (Rudolphi, 1819) after use of the anthelminthics albendazole, fenbendazole, and ivermectine. They demonstrate that albendazole and fenbendazole cause irreversible structural changes. Less pronounced destruction of different parts of the digestive system occurs after the use of ivermectine. All the drugs affect the mid-gut of Passalurus.

Synergistic anti-tumor effect of fenbendazole and diisopropylamine dichloroacetate in immunodeficient BALB/c nude mice transplanted with A549 lung cancer cells.

BACKGROUND: Lung cancer remains one of the leading causes of cancer-related deaths worldwide. Recent studies suggest that fenbendazole (FZ), even at micromolar doses, shows promising anticancer potential but can cause liver toxicity in some patients. Diisopropylamine dichloroacetate or vitamin B15 (DADA), known for its hepatoprotective properties, has also demonstrated antitumor properties and may reduce FZ-induced liver injury. Our research aimed to evaluate the synergistic anticancer effects of FZ and DADA in vivo lung cancer models. METHODS: Immunodeficient BALB/c nude mice (Foxn1nu) were

Optimization and Pharmacokinetic Evaluation of Synergistic Fenbendazole and Rapamycin Co-Encapsulated in Methoxy Poly(Ethylene Glycol)-b-Poly(Caprolactone) Polymeric Micelles.

PURPOSE: We aimed to develop a nanocarrier formulation incorporating fenbendazole (FEN) and rapamycin (RAPA) with strong efficacy against A549 cancer cells. As FEN and RAPA are poorly soluble in water, it is difficult to apply them clinically in vivo. Therefore, we attempted to resolve this problem by encapsulating these drugs in polymeric micelles. METHODS: We evaluated drug synergy using the combination index (CI) values of various molar ratios of FEN and RAPA. We formed and tested micelles composed of different polymers. Moreover, we conducted cytotoxicity, stability, release, pharmacokine

Efficacy of fenbendazole against gastrointestinal nematodes in naturally infected goats in Maputo Province, Mozambique using in vivo, in vitro and molecular assessment.

Anthelmintic resistance occurs worldwide in strongyles of ruminants but data from low-income countries are sparse and rarely apply most up to date methods, while effects of management practices in these countries are poorly documented. In Mozambique, benzimidazole resistance has been previously reported; the present study followed this up in detail, applying in vivo faecal egg count (FEC) reduction test (FECRT), in vitro egg hatch test (EHT) and molecular deep amplicon sequencing approaches targeting the internal transcribed spacer 2 (ITS-2, nemabiome) and the isotype 1 β-tubulin gene to deter

In vivo porcine multi-omics integration identifies microbiome-driven histamine elevation and lasting gut perturbations following Ascaris suum infection and fenbendazole treatment.

Ascaris roundworms impair human and swine health. While treatments using anthelmintic drugs are generally effective in eliminating worms, their effects on the gut microenvironment remain poorly understood. Here we applied integrated multi-omics to characterize infection- and treatment-associated alterations in the pig-Ascaris system. In vitro anaerobic cultures were conducted as supportive validation of selected observations. Ascaris suum infection altered microbial composition and dysregulated 182 serum and fecal metabolites, including histamine and p-cresol sulfate. Compared with time-matche

Efficacy of fenbendazole formulated in a commercial primate diet for treating specific pathogen-free baboons (Papio cynocephalus anubis) infected with Trichuris trichiura.

Trichuris trichiura is a common intestinal nematode parasite of captive baboons. We evaluated the efficacy of fenbendazole formulated in a commercial primate diet (FBZ-PD) for treating specific pathogen-free (SPF) baboons (Papio cynocephalus anubis) naturally infected with Trichuris trichiura. Twenty-nine baboons, housed indoors in 3 separate rooms, were fed FBZ-PD for 5 d, whereas 4 baboons housed in another isolated area served as untreated controls. The efficacy of FBZ-PD was measured as reduction in the number of T. trichiura eggs in host feces after treatment as determined by quantitative

Treatment of fish parasites. 3. Effects of levamisole HCl, metrifonate, fenbendazole, mebendazole, and ivermectin on Anguillicola crassus (nematodes) pathogenic in the air bladder of eels.

The effects of five nematocidal drugs (levamisole HCl, metrifonate, fenbendazole, mebendazole, and ivermectin) on the nematode Anguillicola crassus, pathogenic in eels and recently introduced in Europe, were tested under in vivo conditions. The resulting tissue alterations were studied by means of light and electron microscopy. It was found that levamisole and metrifonate were most effective in a freshwater bath (1 mg/l) for 24 h. The 50% lethal dose of levamisole was 250 mg/l per 24 hours, whereas that of metrifonate was only 10 mg/l per 2 hours. Morphological studies of the nematodes showed

Computational insights into fenbendazole encapsulation by Cucurbit[n]urils (n = 7, 8): Implications for drug delivery and nanocarrier design through integrated structural, electronic, thermodynamic, and spectroscopic investigations.

Fenbendazole (fenben), a benzimidazole anthelmintic with emerging anticancer potential, faces significant challenges in clinical applications due to its poor aqueous solubility and potential toxicity at high doses. To address these limitations, this study explores the encapsulation of fenben within cucurbit[7]uril (CB[7]) and cucurbit[8]uril (CB[8]) using computational methods to evaluate host-guest interactions, stability, and electronic properties. Density functional theory (DFT) calculations at the APFD/6-31+G(d) level revealed that CB[7] forms more stable complexes with fenben than CB[8],
