Maternal Nutritional Environment and the Development of the Melanocortin System.

The maternal nutritional and/or metabolic environment is crucial for future offspring health outcomes, and impairments during critical periods of development can alter the development of brain circuits that regulate energy balance, predisposing individuals to metabolic disorders throughout life. Epigenetic changes, changes in cell number and/or organ structure, and cellular metabolic differentiation could be some of the fetal adaptations leading to the development of metabolic disorders later in life. Here, we review animal models showing that the nutritional environment to which the offspring

Fetal programming of polycystic ovary syndrome.

Polycystic ovary syndrome (PCOS) is a common endocrine disorder that affects up to 6.8% of reproductive age women. Experimental research and clinical observations suggest that PCOS may originate in the very early stages of development, possibly even during intrauterine life. This suggests that PCOS is either genetically-transmitted or is due to epigenetic alterations that develop in the intrauterine microenvironment. Although familial cases support the role of genetic factors, no specific genetic pattern has been defined in PCOS. Several candidate genes have been implicated in its pathogenesis

Fetal and neonatal levels of omega-3: effects on neurodevelopment, nutrition, and growth.

Nutrition in pregnancy, during lactation, childhood, and later stages has a fundamental influence on overall development. There is a growing research interest on the role of key dietary nutrients in fetal health. Omega-3 polyunsaturated fatty acids (n-3 LCPUFAs) play an important role in brain development and function. Evidence from animal models of dietary n-3 LCPUFAs deficiency suggests that these fatty acids promote early brain development and regulate behavioral and neurochemical aspects related to mood disorders (stress responses, depression, and aggression and growth, memory, and cogniti

Hypoxia: From Placental Development to Fetal Programming.

Hypoxia may influence normal and different pathological processes. Low oxygenation activates a variety of responses, many of them regulated by hypoxia-inducible factor 1 complex, which is mostly involved in cellular control of O2 consumption and delivery, inhibition of growth and development, and promotion of anaerobic metabolism. Hypoxia plays a significant physiological role in fetal development; it is involved in different embryonic processes, for example, placentation, angiogenesis, and hematopoiesis. More recently, fetal hypoxia has been associated directly or indirectly with fetal progra

Maternal and Placental Risk Factors for Small Gestational Age and Fetal Malnutrition.

AIMS: This case-control study was conducted to identify maternal and placental risk factors of small-for-gestational-age (SGA) and fetal malnutrition. METHODS: Cases comprised 104 consecutively delivered SGA neonates (determined as per INTERGROWTH- 21st standard). An equal number of next-born gestation and gender-matched appropriatefor- gestational age (AGA) neonates served as controls. Maternal risk factors were enquired, and placentae were evaluated by clinical and histopathological examination. Nutrition of the neonates was assessed by the clinical assessment of nutrition (CAN) score. Univ

Management of pregnancy and childbirth in a patient with glutaric aciduria type II.

Glutaric aciduria type II (GA II) is a genetic disorder that interferes with the body's ability to break down proteins and fats to produce energy. Signs and symptoms vary greatly depending on the age of onset and severity of the condition. Pregnancy may be a high-risk period in women affected by metabolic disorders, because the mother must guarantee wellness for both her and fetus. A 37-year-old primigravida woman affected by a mild form of GA II presented to our high-risk department. During pregnancy a dietician strictly controlled her diet and nutritional intake. The fetal growth was regular

Neuroinflammation in Autism: Plausible Role of Maternal Inflammation, Dietary Omega 3, and Microbiota.

Several genetic causes of autism spectrum disorder (ASD) have been identified. However, more recent work has highlighted that certain environmental exposures early in life may also account for some cases of autism. Environmental insults during pregnancy, such as infection or malnutrition, seem to dramatically impact brain development. Maternal viral or bacterial infections have been characterized as disruptors of brain shaping, even if their underlying mechanisms are not yet fully understood. Poor nutritional diversity, as well as nutrient deficiency, is strongly associated with neurodevelopme

The role of vitamin A and its pro-vitamin carotenoids in fetal and neonatal programming: gaps in knowledge and metabolic pathways.

Vitamin A (VA) and its pro-vitamin carotenoids are naturally occurring lipophilic compounds involved in several cellular processes and metabolic pathways. Despite their broad spectrum of activities in the general population, dietary deficiencies of these compounds can potentially affect pregnancy outcomes. Since maternal nutritional status and diet composition during pregnancy and lactation can have long-lasting effects in offspring until adulthood, this study presents an overview of VA and the role of pro-VA carotenoids during pregnancy and lactation - the nutrition, metabolism, and biologica

Maternal microbiota and fetal brain development: a multidisciplinary review of cognitive, neurological, and metabolic outcomes.

OBJECTIVE: The prenatal period represents a critical window for the programming of the central nervous system. This multidisciplinary narrative review aims to synthesize current evidence regarding the maternal microbiota-gut-fetal brain axis, examining how maternal gut composition may influence fetal neurogenesis, synaptogenesis, and long-term neurobehavioural outcomes. MECHANISM: We analyse three primary pathways of influence: the systemic translocation of neuroactive bacterial metabolites (specifically short-chain fatty acids), the modulation of the maternal immune system leading to materna

Hypothyroidism impairs development of the gastrointestinal tract in the ovine fetus.

Growth and maturation of the fetal gastrointestinal tract near term prepares the offspring for the onset of enteral nutrition at birth. Structural and functional changes are regulated by the prepartum rise in cortisol in the fetal circulation, although the role of the coincident rise in plasma tri-iodothyronine (T3) is unknown. This study examined the effect of hypothyroidism on the structural development of the gastrointestinal tract and the activity of brush-border digestive enzymes in the ovine fetus near term. In intact fetuses studied between 100 and 144 days of gestation (dGA; term ∼145

Thiamin deficiency on fetal brain development with and without prenatal alcohol exposure.

Adequate thiamin levels are crucial for optimal health through maintenance of homeostasis and viability of metabolic enzymes, which require thiamine as a co-factor. Thiamin deficiency occurs during pregnancy when the dietary intake is inadequate or excessive alcohol is consumed. Thiamin deficiency leads to brain dysfunction because thiamin is involved in the synthesis of myelin and neurotransmitters (e.g., acetylcholine, γ-aminobutyric acid, glutamate), and its deficiency increases oxidative stress by decreasing the production of reducing agents. Thiamin deficiency also leads to neural membran

Maternal Nutrition Status Associated with Pregnancy-Related Adverse Outcomes.

Although maternal nutrition has an impact on fetal development and gestational outcome, tracking maternal nutrition in outpatient practice is still complex and involves proper technical capacitation in this area. Nevertheless, the association between nutritional variables may broaden the ability to predict the occurrence of gestational disorders and prevention management. We aimed to identify factors that could indicate the probability of adverse outcomes in mid-pregnancy. From a cohort of 1165 nulliparous pregnant women without any previous disease, the nutritional status was assessed by body

Zinc nutrition in fetal alcohol syndrome.

Because alcohol has an adverse effect on zinc homeostasis, this study was designed to study if zinc content of the diet modifies the severity of fetal alcohol syndrome in a mouse model. The effect of varying zinc intake on the progeny of pregnant mice fed a liquid diet containing 15% of the calories as ethanol was studied. Prenatal mortality was higher when the mothers consumed alcohol with inadequate zinc intake. Because of the adverse effect of alcohol on zinc homeostasis and because zinc deficiency has been shown to potentiate alcohol embryopathy, one group was given zinc supplementation to

Behind the Curtain of Abnormal Placentation in Pre-Eclampsia: From Molecular Mechanisms to Histological Hallmarks.

Successful human pregnancy needs several highly controlled steps to guarantee an oocyte's fertilization, the embryo's pre-implantation development, and its subsequent implantation into the uterine wall. The subsequent placenta development ensures adequate fetal nutrition and oxygenation, with the trophoblast being the first cell lineage to differentiate during this process. The placenta sustains the growth of the fetus by providing it with oxygen and nutrients and removing waste products. It is not surprising that issues with the early development of the placenta can lead to common pregnancy d

Melatonin treatment in fetal and neonatal diseases.

This literature review aims to address the main scientific findings on oxidative stress activity in different gestational disorders, as well as the function and application of melatonin in the treatment of fetal and neonatal changes. Oxidative stress has been associated with the etiopathogenesis of recurrent miscarriages, preeclampsia, intrauterine growth restriction, and stillbirth. Both, the exacerbated consumption of the antioxidant enzymes superoxide dismutase, catalase and glutathione peroxidase, and the increased synthesis of reactive oxygen species, such as superoxide, peroxynitrite, an

Prenatal stress, hormones, and fetal brain development: gender differences.

Prenatal stress is increasingly recognized as a critical factor influencing fetal brain development, with potential long-term risks for neurodevelopmental and psychiatric disorders. This review explores the effects of prenatal stress on the fetal brain, with particular emphasis on sex differences and the interplay between genetic, epigenetic, and environmental factors. The activation of the hypothalamic-pituitary-adrenal (HPA) axis in response to maternal stress leads to increased levels of glucocorticoids, which can cross the placenta and alter fetal brain structure and function. Key regions

Protein turnover in tissues of the fetal rat after prolonged maternal malnutrition.

Protein turnover in fetal diaphragm, heart, liver, and brain was determined at 21 days gestation in fetal rats whose mothers had received a protein-restricted diet (6% protein) throughout gestation. This diet resulted in severe combined protein-calorie malnutrition. Fetal body weight was significantly decreased at days 19-22 gestation versus controls (27% protein diet) when pregnant animals were protein-restricted (e.g. 40% decrease in body weight at day 22 gestation versus controls). Protein synthesis was determined by intravenous injection of "massive" amounts of [3H]phenylalanine to pregnan
