Influence of self-assembly regenerated silk fibroin nanofibers on the properties of electrospun materials.

In this study, self-assembly regenerated silk fibroin (RSF) nanofibers were prepared and observed by Atomic Force Microscope (AFM). Then RSF films containing nanospheres and nanofibers were prepared and dissolved with poly (L-lactide-co-ε-caprolactone) (PLCL) with a blending ratio of 30/70 in hexafluoro-2-propanol (HFIP). In order to determine whether different nanostructures in the solution influence the morphological, structural, and mechanical properties of the final electrospun materials, flat membranes were prepared and characterized by Scanning Electron Microscope (SEM), Fourier Transfor

Silk Fibroin Counteracts Fibroblast Senescence to Restore ECM Homeostasis in Aged Skin.

Skin aging is characterized by a progressive decline in regenerative capacity, primarily driven by fibroblast senescence, oxidative stress, chronic inflammation, and the degradation of type I/III collagen, culminating in an extracellular matrix (ECM) imbalance. Current injectable fillers-such as hyaluronic acid, collagen, and PLLA-provide temporary structural support but fail to address the underlying cellular senescence or restore ECM homeostasis, highlighting the need for regenerative biomaterials. Silk fibroin (SF), a natural protein, self-assembles into a β-sheet-rich scaffold that structu

Silk fibroin scaffolds loaded with growth factors can reverse liver fibrosis.

Currently, there are no effective pharmacological interventions in clinical practice to reverse liver fibrosis. This study explores the therapeutic potential of regenerated silk fibroin (RSF) scaffolds loaded with dual growth factors for reversing hepatic fibrosis. A chronic liver fibrosis mouse model was induced using carbon tetrachloride (CCl4) combined with a high-fat diet. RSF scaffolds, with or without hepatocyte growth factor (HGF) and fibroblast growth factor-4 (FGF-4), were implanted onto the liver surface to assess antifibrotic efficacy. Liver function was evaluated using biochemical

Design Strategies and Emerging Applications of Electrospun Silk Fibroin Nanofibers for Tissue Engineering.

Electrospun silk fibroin nanofibers (SFNFs) combine exceptional biocompatibility, tunable mechanical properties, and a native extracellular matrix (ECM)-mimetic architecture, making them compelling scaffolds for tissue engineering. Despite rapid progress, current research often pursues isolated material enhancements, lacking a cohesive strategy that aligns scaffold design with the complex biophysical and biochemical microenvironments of targeted tissues. To bridge this gap, this review presents a "design-application coupling" framework that systematically integrates SFNF composition, processin

Bio-enhanced silk fibroin-based scaffolds for chronic diabetic foot ulcers.

Diabetic foot ulcers (DFUs) represent a growing clinical challenge, driven by an aging global population and the increasing prevalence of diabetes. Affecting millions worldwide, DFUs remain one of the most serious complications of diabetes, frequently progressing to infection, amputation, and elevated mortality. Standard treatments typically include pressure offloading, circulation improvement, infection control, and topical wound care; however, many chronic DFUs fail to respond to these interventions due to persistent inflammation, impaired vascularization, and microbial burden. These challen

Evaluation of Bi-layer Silk Fibroin Grafts for Inlay Vaginoplasty in a Rat Model.

BACKGROUND: Autologous tissues derived from bowel, buccal mucosa and skin are primarily used to repair or replace diseased vaginal segments as well as create neovaginas for male-to-female transgenders. These grafts are often limited by scarce tissue supply, donor site morbidity and post-operative complications. Bi-layer silk fibroin (BLSF) biomaterials represent potential alternatives for vaginoplasty given their structural strength and elasticity, low immunogenicity, and processing flexibility. The goals of the current study were to assess the potential of acellular BLSF scaffolds for vaginal

Precisely Printable Silk Fibroin/Carboxymethyl Cellulose/Alginate Bioink for 3D Printing.

Three-dimensional (3D) bioprinting opens up many possibilities for tissue engineering, thanks to its ability to create a three-dimensional environment for cells like an extracellular matrix. However, the use of natural polymers such as silk fibroin in 3D bioprinting faces obstacles such as having a limited printability due to the low viscosity of such solutions. This study addresses these gaps by developing highly viscous, stable, and biocompatible silk fibroin-based inks. The addition of 2% carboxymethyl cellulose sodium and 1% sodium alginate to an aqueous solution containing 2.5 to 5% silk

Development of a bioactive silk fibroin bilayer scaffold for wound healing and scar inhibition.

In cases of deep skin defects, spontaneous tissue regeneration and excessive collagen deposition lead to hyperplastic scars. Conventional remedial action after scar formation is limited with a high recurrence rate. In this study, we designed a new artificial skin bilayer using silk fibroin nanofibers films (SNF) as the epidermis, and silk fibroin (SF) / hyaluronic acid (HA) scaffold as the dermal layer. The regenerated SF film was used as a binder to form a functional SNF-SF-HA bilayer scaffold. The bilayer scaffold showed high porosity, hydrophilicity, and strength, and retained its shape ove

Unveiling the Rational Development of Stimuli-Responsive Silk Fibroin-Based Ionogel Formulations.

We present an approach for the rational development of stimuli-responsive ionogels which can be formulated for precise control of multiple unique ionogel features and fill niche pharmaceutical applications. Ionogels are captivating materials, exhibiting self-healing characteristics, tunable mechanical and structural properties, high thermal stability, and electroconductivity. However, the majority of ionogels developed require complex chemistry, exhibit high viscosity, poor biocompatibility, and low biodegradability. In our work, we overcome these limitations. We employ a facile production pro

Robust Silk Fibroin/Graphene Oxide Aerogel Fiber for Radiative Heating Textiles.

Aerogel fibers with ultrahigh porosity and ultralow density are promising candidates for personal thermal management to reduce the energy waste of heating an entire room, and play important roles in reducing energy waste in general. However, aerogel fibers generally suffer from poor mechanical properties and complicated preparation processes. Herein, we demonstrate hierarchically porous and continuous silk fibroin/graphene oxide aerogel fibers (SF/GO) with high strength, excellent radiative heating performance, and thermal insulation performance through coaxial wet spinning and freeze-drying.

Evaluation of Neurosecretome from Mesenchymal Stem Cells Encapsulated in Silk Fibroin Hydrogels.

Physical and cognitive disabilities are hallmarks of a variety of neurological diseases. Stem cell-based therapies are promising solutions to neuroprotect and repair the injured brain and overcome the limited capacity of the central nervous system to recover from damage. It is widely accepted that most benefits of different exogenously transplanted stem cells rely on the secretion of different factors and biomolecules that modulate inflammation, cell death and repair processes in the damaged host tissue. However, few cells survive in cerebral tissue after transplantation, diminishing the thera

Enzymatically Cross-Linked Silk Fibroin-Based Hierarchical Scaffolds for Osteochondral Regeneration.

Osteochondral (OC) regeneration faces several limitations in orthopedic surgery, owing to the complexity of the OC tissue that simultaneously entails the restoration of articular cartilage and subchondral bone diseases. In this study, novel biofunctional hierarchical scaffolds composed of a horseradish peroxidase (HRP)-cross-linked silk fibroin (SF) cartilage-like layer (HRP-SF layer) fully integrated into a HRP-SF/ZnSr-doped β-tricalcium phosphate (β-TCP) subchondral bone-like layer (HRP-SF/dTCP layer) were proposed as a promising strategy for OC tissue regeneration. For comparative purposes,

Silk fibroin scaffolds loaded with angiogenic genes in adenovirus vectors for tissue regeneration.

Vascularization remains a critical challenge in dermal tissue regeneration. In this study, a vascular endothelial growth factor (VEGF165) and angiopoietin-1 (Ang-1) dual gene coexpression vector that encoded green fluorescent protein (GFP) was constructed from an arginine-glycine-aspartic acid-modified adenovirus. Silk fibroin (SF) scaffolds loaded with adenovirus vectors were fabricated by freeze-drying method. In vitro, the human endothelial-derived cell line EA.hy926 was infected with adenovirus vectors and then expressed GFP, secreted VEGF165 and Ang-1, and promoted cell proliferation effe

Electrospun silk fibroin-hydroxybutyl chitosan nanofibrous scaffolds to biomimic extracellular matrix.

Silk fibroin (SF)-hydroxybutyl chitosan (HBC) blend nanofibrous scaffolds were fabricated using 1,1,1,3,3,3-hexafluoro-2-propanol (HFIP) and trifluoroacetic acid (TFA) as solvents to biomimic the native ECM by electrospinning. SEM results showed that the average nanofibrous diameter increased when the content of HBC was raised from 20% to 100%. Whereas water contact angle measurements confirmed that SF/HBC nanofibrous scaffolds with different weight ratios were of good hydrophilicity. Both the tensile strength and the elongation at break were improved obviously when the weight ratio of SF to H

Biomimetic nanofibrous scaffolds: preparation and characterization of chitin/silk fibroin blend nanofibers.

Electrospinning of chitin/silk fibroin (SF) blend solutions in 1,1,1,3,3,3-hexafluoro-2-propanol (HFIP) was investigated to fabricate a biomimetic nanostructured scaffolds for tissue engineering. The morphology of the electrospun chitin/SF blend nanofibers was investigated with a field emission scanning electron microscope (FE-SEM). The average diameters of chitin/SF blend fibers decreased from 920 to 340nm, with the increase of chitin content in blend compositions. The miscibility of chitin/SF blend fibers was examined by solution viscosity measurement. The chitin and SF were immiscible in th

Robust Silk Fibroin Aerogel Fibers Designed by Dual Cross-Linking for Thermal Insulation.

Silk fibroin (SF) is ideal for aerogel fibers owing to its abundance, biodegradability, biocompatibility, and structural tunability, yet pristine SF aerogel fibers have insufficient performance for practical use. Here, a dual cross-linking strategy combining enzymatic cross-linking and Zn2+ coordination was used, leveraging their noncompetitive inhibition to produce SF aerogel fibers featuring low density, high strength, and outstanding thermal insulation. SF concentration governs the pore architecture formed by enzymatic cross-linking. Zn2+ ions complex with SF molecular chains, creating a mo
