Abstract A challenge to selectively breeding fish populations for improved disease resistance in aquaculture is an incomplete understanding of how artificial selection affects innate immunity at the host-pathogen level. The objective of this study was to determine whether Rainbow Trout Oncorhynchus mykiss bred for differential susceptibility to bacterial cold-water disease exhibited altered tissue damage and cellular inflammatory response following experimental challenge with Flavobacterium psychrophilum. Fish from disease-resistant (ARS-Fp-R) and disease-susceptible (ARS-Fp-S) lines were expe

In order to identify genes encoding the outer membrane proteins (OMPs) of the myxobacter Flavobacterium columnare G(4), the expression library of the bacterium was screened by using rabbit antisera developed against its OMPs. Positive colonies of Escherichia coli M15 containing fragments encoding the bacterial OMPs were selected for cloning the relevant genes by genomic walking methods. Two genes encoding a membrane-associated zinc metalloprotease and prolyl oligopeptidase are reported in this paper. The membrane-associated zinc metalloprotease gene (map) is 1800 bp in length, coding for 449 a

Withholding feed has been suggested as a strategy to manage infectious disease of channel catfish, Ictalurus punctatus (Rafinesque). In a previous study, we demonstrated that deprivation of feed for as little as 7 days reduced innate resistance of catfish to Flavobacterium columnare. This study was conducted to evaluate the effect of feeding regimens [no feeding (NF), fed once every other day to satiation (FEOD) and fed once daily to satiation (FD)] on organosomatic indices, physiological changes and susceptibility of channel catfish to F. columnare. Fish that were not fed for 2 and 4 weeks ha

β-Galactosidases are widely used to produce lactose-free dairy products, but efficient lactose hydrolysis under refrigerated conditions remains limited by the low activity of many commercial enzymes. In this study, a cold-adapted β-galactosidase OUC-JST from Flavobacterium algicola was characterized and engineered. OUC-JST exhibited maximal activity at 20 °C and pH 7.0. Structure-guided mutagenesis identified R110I, which shifted the optimal temperature to 15 °C, while K1023D increased activity approximately 3-fold. The combined variant R110I/K1023D (OUC-JST-N) retained the optimal temperature

RATIONALE: Acquired hemophilia A (AHA) is a rare bleeding disorder characterized by the development of autoantibodies directed against coagulation factor VIII (FVIII). Approximately 50% of cases are idiopathic, while the remaining are associated with autoimmune conditions, malignancies, or infections. To date, there have been no reported cases of AHA secondary to Flavobacterium odoratum (Frullania odoratum) infection.
PATIENT CONCERNS: A male patient underwent ureteroscopic lithotripsy with double-J stent placement. Postoperatively, he developed hematuria, a right medial thigh hematoma, fever

Flavobacterium psychrophilum, the causative agent of bacterial cold-water disease (BCWD) and rainbow trout fry syndrome (RTFS), remains one of the most important bacterial pathogens affecting salmonid aquaculture. In this study, 24 F. psychrophilum isolates recovered during a 12-month surveillance programme from two rainbow trout farms in Türkiye were characterized using phenotypic, serological, antigenic, phylogenomic and experimental infection approaches. Phylogenomic analyses showed that most isolates belonged to CC-ST10, whereas four isolates were assigned to the CC-ST90/ST108 lineage, rep

BACKGROUND: The study investigated the transcriptional level expression dynamics of nonspecific (iNOS and TLR22) and specific (IgM, IgD, and IgZ) immune genes in rohu (Labeo rohita) following challenge with formalin-inactivated and virulent Flavobacterium columnare in the specific tissues, such as gill, kidney, liver, gut, and skin. METHODS AND RESULTS: Fish were immunized on days 0 and 14 with 7.98 × 107 CFU/mL F. columnare and challenged with live bacteria on day 35 (7.98 × 108 CFU/mL). Immune gene expression was analyzed at multiple time points. Samples for long-term monitoring were taken a

Flavobacterium psychrophilum is a pathogenic bacterium affecting Chilean salmonid farms. High antigenic and genetic diversity exists among Chilean F. psychrophilum isolates, but the distribution thereof among farms is poorly understood. These epidemiological data are key for developing isolate-specific vaccines. The present study isolated F. psychrophilum in diseased Atlantic salmon (Salmo salar) and rainbow trout (Oncorhynchus mykiss) from five freshwater farms between 2018 and 2019. Each farm only raised one salmonid species and was geographically separated from and did not share culturing w

Fish are covered by a watery gel-mucus, mainly secreted by the goblet cells, serving as the physical and biochemical barrier between the external environment and the interior milieu, playing more important roles in fish that without scale. Despite the important roles of mucus in fish immunity, the knowledge of detailed molecular events happened during infection process is still limited. While most studies were focused on characterizing the protein and enzyme activities in the mucus following challenge, no studies have examined the gene expression profiles in fish mucus. In this regard, herein

Legionella pneumophila, the agent of Legionnaires' disease pneumonia, is transmitted to humans following the inhalation of contaminated water droplets. In aquatic systems, L. pneumophila survives much of time within multi-organismal biofilms. Therefore, we examined the ability of L. pneumophila (clinical isolate 130 b) to persist within biofilms formed by various types of aquatic bacteria, using a bioreactor with flow, steel surfaces, and low-nutrient conditions. L. pneumophila was able to intercalate into and persist within a biofilm formed by Klebsiella pneumoniae, Flavobacterium sp. or Pseu

We analyzed here the complete genome sequences of a highly virulent Flavobacterium columnare Pf1 strain isolated in our laboratory. The complete genome consists of a 3,171,081 bp circular DNA with 2784 predicted protein-coding genes. Among these, 286 genes were predicted as antibiotic resistance genes, including 32 RND-type efflux pump related genes which were associated with the export of aminoglycosides, indicating inducible aminoglycosides resistances in F. columnare. On the other hand, 328 genes were predicted as pathogenicity related genes which could be classified as virulence factors, g

Columnaris is an important bacterial disease of freshwater fish and is increasingly recognised as being caused by a species complex historically referred to as Flavobacterium columnare. A two-step molecular workflow was applied to ornamental fish with clinical signs of columnaris disease from a multispecies commercial distributor operating a shared recirculating system in São Paulo, Brazil. Skin and fin lesion scrapings were Gram stained, and lesion-derived tissue DNA was screened by ISR-PCR for the columnaris disease complex. ISR-PCR yielded the expected amplicon in 61/123 samples (49.6%) fro

Flavobacterium columnare is an important pathogen causing columnaris disease, which can result in high mortality in freshwater fish worldwide. Understanding the immune response in infection status of fish may be essential for developing effective prevention and treatment strategies. In this study, transcriptomes of liver and head kidney tissues in grass carp (Ctenopharyngodon idella) were compared under symptomatic and asymptomatic statuses following the immersion infection of F. columnare. Significant differences in expression of genes were observed between fish showing disease symptoms and t

OBJECTIVE: Columnaris disease is a leading cause of disease-related losses in the catfish industry of the southeastern United States. The term "columnaris-causing bacteria" (CCB) has been coined in reference to the four described species that cause columnaris disease: Flavobacterium columnare, F. covae, F. davisii, and F. oreochromis. Historically, F. columnare, F. covae, and F. davisii have been isolated from columnaris disease cases in the catfish industry; however, there is a lack of knowledge of which CCB species are most prevalent in farm-raised catfish. The current research objectives we

OBJECTIVES: The aim of this study was to determine mutations associated with a quinolone-resistant (QR) phenotype of Flavobacterium columnare isolates.
METHODS: The susceptibility of 53 F. columnare isolates to 11 antimicrobials, including 2 quinolones, was investigated by the disk diffusion method. Oxolinic acid (OXO) was subsequently chosen for minimum inhibitory concentration (MIC) assay. Sequence analysis of four genes within the quinolone resistance-determining regions (QRDRs) of OXO-resistant F. columnare compared with susceptible isolates was subsequently performed.
RESULTS: The disk

The hemolytic activity of cells of smooth and rough phenotypic variants of the Gram-negative fish pathogen Flavobacterium psychrophilum was investigated in two different assays, a microplate and an agarose hemolysis assay, using rainbow trout erythrocytes. The smooth cells showed a high and the rough cells a negligible, concentration dependent, hemolytic activity in the microplate assay. Both smooth and rough cells showed a rather weak hemolytic activity, with two distinct hemolytic patterns, in the agarose assay. The hemolytic activity of the cells was not regulated by iron availability and c

TonB-dependent siderophore receptors play a critical transport role for Flavobacterium columnare virulence formation and growth, and have become valuable targets for the development of novel antimicrobial agents. Traditional Chinese medicine has demonstrated notable efficacy in the treatment of fish diseases and includes potential antibacterial agents. Herein, we performed molecular docking-based virtual screening to discover novel TonB-dependent siderophore receptor inhibitors from traditional Chinese medicine and provide information for developing novel antibacterial agents. Firstly, we effi

Structural colors (SC), generated by light interacting with nano-structured materials, are responsible for the brightest and most vivid coloration in nature. Despite being widespread within the tree of life, there is little knowledge of the genes involved. Partial exceptions are some Flavobacteriia in which genes involved in a number of pathways, including gliding motility and polysaccharide metabolism, have been linked to SC. A previous genomic analysis of SC and non-SC bacteria suggested that the pterin pathway is involved in the organization of bacteria to form SC. Here, we focus on moeA, a

BACKGROUND: Columnaris disease, a prevalent disease among farmed and wild freshwater fish, is caused by the Flavobacterium columnare group, which includes four distinct species: F. columnare, F. oreochromis, F. covae, and F. davisii. Among these, F. oreochromis, F. covae, and F. davisii are particularly prevalent in farmed freshwater fish in Thailand. In this study, a comparative genomic analysis of 22 isolates was conducted to elucidate virulence factors, antibiotic resistance genes (ARGs), genomic islands (GIs), phages, insertion elements (ISs), and clustered regularly interspaced short pali

NOD-like receptors are significant contributors to the immune response of fish against different types of pathogen invasion. NOD1 and NOD2 genes of yellow catfish (Tachysurus fulvidraco) were identified and characterized in this study. Yellow catfish NOD1 and NOD2 have open reading frames (ORFs) of 2841 and 2949 bp, encoding 946 and 982 amino acids, respectively. Both NOD1 and NOD2 are intracellular proteins lacking transmembrane regions and signal peptides. Sequence homology analysis revealed that the protein sequences of NOD1 and NOD2 of yellow catfish are highly similar to those of channel
