[Oxidative stress and antioxidant defense in alcoholic liver disease and chronic hepatitis C].

In the past decade it became accepted that free radicals, lipid peroxidation and antioxidant defense play a role in various tissues damages, thus in certain liver diseases as well. Since only limited data have been reported concerning the oxidative stress in viral hepatitis, a comparative study was performed in patients (pts) with chronic hepatitis C and alcoholic liver disease. In addition, the effects of a flavonolignan drug silymarin were assessed. 10 pts with chronic hepatitis C, 5 pts with alcoholic hepatitis and 13 pts with alcoholic cirrhosis have been investigated. Biochemical liver te

Silibinin is a natural molecular glue that inhibits PD-L1 glycomaturation.

BACKGROUND: The adaptive upregulation of the immune-checkpoint PD-L1 in response to interferon-γ (IFNγ) protects solid tumors from cytotoxic lymphocytes and undermines adoptive cell immunotherapies. Currently, pharmacologically interrupting the trafficking of nascent PD-L1 to the surface of cancer cells is not feasible in a clinical setting. HYPOTHESIS/PURPOSE: We investigated whether silibinin (SBN), the primary bioactive flavonolignan found in Silybum marianum (milk thistle) seeds, could impede the post-translational glycomaturation of PD-L1 and enhance T-cell-mediated antitumor cytotoxicit

Silymarin: a review of its clinical properties in the management of hepatic disorders.

UNLABELLED: The mechanisms of action of silymarin involve different biochemical events, such as the stimulation of the synthetic rate of ribosomal RNA (rRNA) species through stimulation of polymerase I and rRNA transcription, protecting the cell membrane from radical-induced damage and blockage of the uptake of toxins such as alpha-amanitin. Studies in patients with liver disease have shown that silymarin increases superoxide dismutase (SOD) activity of lymphocytes and erythrocytes, as well as the expression of SOD in lymphocytes. Silymarin has also been shown to increase patient serum levels

Silymarin sensitizes human colorectal cancer cells to 5-ALA-mediated photodynamic therapy by enhancing cytotoxicity and apoptotic signaling in vitro.

BACKGROUND: Photodynamic therapy (PDT) using 5-aminolevulinic acid (5-ALA) generates reactive oxygen species (ROS) that can kill tumor cells, but treatment efficacy may be limited by cellular antioxidant defenses. Silymarin, a flavonolignan complex from Silybum marianum, exhibits context-dependent antioxidant/pro-oxidant activity and has been suggested to modulate cancer redox homeostasis. This study evaluated whether silymarin pre-treatment enhances PDT-induced cytotoxicity in human colorectal cancer cells in vitro. METHODS: HCT-116 human colorectal carcinoma cells were cultured under standar

Novel Investigations of Flavonoids as Chemopreventive Agents for Hepatocellular Carcinoma.

We would like to highlight the application of natural products to hepatocellular carcinoma (HCC). We will focus on the natural products known as flavonoids, which target this disease at different stages of hepatocarcinogenesis. In spite of the use of chemotherapy and radiotherapy in treating HCC, patients with HCC still face poor prognosis because of the nature of multidrug resistance and toxicity derived from chemotherapy and radiotherapy. Flavonoids can be found in many vegetables, fruits, and herbal medicines that exert their different anticancer effects via different intracellular signalin

Study of Liposomes Containing Extract from the Leaves of Protium heptaphyllum (Aubl.) March in Animals Submitted to a Mutagenic Model Induced by Cyclophosphamide.

Cyclophosphamide (CPA) is an alkylating agent used as a chemotherapy agent in the treatment of cancer, but it has immunosuppressive effects. Protium heptaphyllum (P. heptaphyllum) is a plant rich in triterpenes and flavonoids, with some bioactive and therapeutic properties presented in the literature. Thus, the present study aimed to investigate the chemoprotective potential of P. heptaphyllum extract inserted into liposomes against oxidative damage chemically induced by CPA. Male Swiss mice received 1.5 mg/kg of P. heptaphyllum liposomes as a pre-treatment for 14 consecutive days (via gavage)

Phytochemical-Based Nanomedicine for Advanced Cancer Theranostics: Perspectives on Clinical Trials to Clinical Use.

In the current chapter, a new strategic compilation of phytochemicals with potent antitumor properties has been addressed, most importantly focusing on cell cycle arrest and apoptotic signaling mechanism. A promising approach in tumor prevention is to eliminate cancer cells preferably via cell cycle arrest and programmed cell death with lesser harm to neighboring normal cells. Cancer cells have a survival advantage to escape apoptosis and relentlessly divide to proliferate, gearing up the cell cycle process. Recently, the use of phytochemical-derived conjugated chemotherapeutic agents has incr

Emerging agents that target signaling pathways to eradicate colorectal cancer stem cells.

Colorectal cancer (CRC) represents the third most commonly diagnosed cancer and the second leading cause of cancer death worldwide. The modern concept of cancer biology indicates that cancer is formed of a small population of cells called cancer stem cells (CSCs), which present both pluripotency and self-renewal properties. These cells are considered responsible for the progression of the disease, recurrence and tumor resistance. Interestingly, some cell signaling pathways participate in CRC survival, proliferation, and self-renewal properties, and most of them are dysregulated in CSCs, includ
