Thrombocytopenia: diagnosis with flow cytometry and antiplatelet antibodies.

OBJECTIVE: To identify antiplatelet antibodies by flow cytometry (direct method) in patients with thrombocytopenia. METHODS: Between January 1997 and March 2004 a total of 15100 patients were referred to the Centro de Hematologia de São Paulo for hematological investigation of several diagnoses (anemia, leukopenia, thrombocytopenia, coagulation abnormalities, adenomegaly, leukemia and others). Of those, 1057 were referred because of thrombocytopenia and were divided into two groups: Group Idiopathic thrombocytopenic purpura, with no identifiable cause; and Group Other thrombocytopenia, which

Diagnosing PNH with FLAER and multiparameter flow cytometry.

BACKGROUND: PNH is an acquired hematopoietic stem cell disorder leading to a partial or absolute deficiency of all glycophosphatidyl-inositol (GPI)-linked proteins. The classical approach to diagnosis of PNH by cytometry involves the loss of at least two GPI-linked antigens on RBCs and neutrophils. While flow assays are more sensitive and specific than complement-mediated lysis or the Hams test, they suffer from several drawbacks. Bacterial aerolysin binds to the GPI moiety of cell surface GPI-linked molecules and causes lysis of normal but not GPI-deficient PNH cells. FLAER is an Alexa488-lab

DNA flow cytometry of follicular non-Hodgkin's lymphoma.

S-phase fraction, an index of cellular proliferation, and DNA ploidy were measured by DNA flow cytometry in a retrospective study of lymph node biopsy specimens from 83 cases (before treatment) of follicular non-Hodgkin's lymphoma, Working Formulation categories B and C. The correlations between these measures and survival, clinical stage, symptoms and histopathological factors were investigated. Aneuploidy was rare (n = 16) and had no effect on length of survival or transformation to high grade lymphoma. The overall mean S-phase fraction was 3.6%; for the whole series increasing S-phase fract

Nucleic acid flow cytometry in large cell lymphoma.

Between 1978 and 1985, 140 patients with large cell lymphoma (27 follicular, 92 diffuse, 5 immunoblastic, and 16 transformed) had DNA-RNA cytometry performed on involved tissue. DNA-RNA features were correlated with treatment outcome and compared to other established prognostic factors in 63 newly diagnosed patients who received uniformly intensive therapy. Significantly better outcome was noted for previously untreated patients with intermediate RNA content (RNA index, 1.0-1.8), diploid DNA content, and (during the initial 12-month follow-up) low proliferative activity. Of patients followed b

Isolation of Mononucleated and Binucleated Hepatocytes by Flow Cytometry.

Polyploid hepatocytes are one of the unique features of the liver. Some polyploid hepatocytes have chromosomes in a single nucleus (e.g., 1x4n, 1x8n), while others separate their chromosomes into two nuclei (e.g., 2x2n, 2x4n). In ploidy research, hepatocytes are typically sorted according to their cellular ploidy, revealing their contribution to tumorigenesis and cellular senescence. However, the conventional sorting method fails to distinguish 1x4n from 2x2n, or 1x8n from 2x4n cells, leaving it unclear whether hepatocytes with the same cellular ploidy but different nuclear configurations are

Detection and Characterization of Senescent Cells with Flow Cytometry.

Cellular senescence, a stable cell cycle arrest induced by various stressors or DNA damage, plays a critical role in various aspects of human physiology, including embryonic development, wound healing, and age-related diseases like cancer. Despite its discovery nearly 50 years ago, a universal marker for senescent cells remains elusive. Current identification methods, such as the detection of senescence-associated β-galactosidase activity (SA-β-Gal), immunohistochemistry, and qPCR, have limitations, including subjective assessment of the results and complex prerequisites. Flow cytometry (FC) h

Flow cytometry in the diagnosis and prognosis of multiple myeloma.

Multiparameter flow cytometry is an effective tool in the diagnosis and monitoring of plasma cell neoplasms such as multiple myeloma. This review focuses on how flow cytometry can be used to identify and characterize plasma cells. It discusses the immunophenotype of normal plasma cells and how this can be used to distinguish them from neoplastic plasma cells. The prognostic impact of particular immunophenotypic aberrancies is reviewed. Finally, it describes approaches to post-treatment monitoring myeloma by flow cytometry, including measurable residual disease detection and its impact on disea

Advances in Monitoring and Prognostication for Lymphoma by Flow Cytometry.

Flow cytometry (FC) is a well-established method important in the diagnosis and subclassification of lymphoma. In this article, the role of FC in lymphoma prognostication will be explored, and the clinical role for FC minimal/measurable residual disease testing as a monitoring tool for mature lymphoma will be introduced. Potential pitfalls of monitoring for residual/recurrent disease following immunotherapy will be presented.

Imaging flow cytometry for the characterization of extracellular vesicles.

Extracellular Vesicles (EVs) are potent bio-activators and inter-cellular communicators that play an important role in both health and disease. It is for this reason there is a strong interest in understanding their composition and origin, with the hope of using them as important biomarkers or therapeutics. Due to their very small size, heterogeneity, and large numbers there has been a need for better tools to measure them in an accurate and high throughput manner. While traditional flow cytometry has been widely used for this purpose, there are inherent problems with this approach, as these i

Usefulness of flow cytometry in the diagnosis of mycosis fungoides.

BACKGROUND: The pathologic evaluation of mycosis fungoides (MF) is a challenging area in dermatopathology. OBJECTIVE: We sought to determine the usefulness of flow cytometry for the diagnosis of MF from skin biopsy specimens. METHODS: Skin biopsy specimens from 22 patients with a clinical suggestion for MF were evaluated by 4-color flow cytometry. The results were correlated with the International Society for Cutaneous Lymphoma (ISCL) MF diagnostic score and molecular studies for T-cell receptor gene rearrangement. RESULTS: A T-cell abnormality by flow cytometry was identified in all 11 pat

Diagnosis of mediastinal masses in dogs by flow cytometry.

BACKGROUND: Biopsy of mediastinal masses can be invasive, but the procedure may be necessary if cytology of a mass aspirate is inconclusive. The 2 most common mediastinal masses, lymphoma and thymoma, may both be comprised of small lymphocytes. We investigated the ability of flow cytometry to distinguish between these 2 neoplasms. HYPOTHESIS: Flow cytometry of mediastinal mass aspirates may provide a definitive diagnosis of thymoma or lymphoma, reducing the need for biopsy. ANIMALS: Dogs with mediastinal masses presenting to the Veterinary Teaching Hospital/Animal Cancer Center were included

Detection of myeloperoxidase by flow cytometry in acute leukemia.

The value of flow cytometric detection of myeloperoxidase (MPO) in the differential diagnosis of acute leukemia was evaluated in 57 cases of acute leukemia and in 9 leukemia cell lines. Cells were fixed and permeabilized with Fix & Perm cell permeabilization kit at room temperature for 15 min each, and stained with anti-MPO monoclonal antibody (MPO-7) by direct immunofluorescence. One myeloid cell line, HL-60, was MPO-positive, while the other myeloid cell lines (KG-1, K-562, and MEG-01) as well as lymphoid cell lines (KM-3, NALM-6, Raji, REH, and T-ALL-1) were MPO-negative as previously descr

Imaging flow cytometry reveals a subset of TdT negative T-ALL blasts with very low forward scatter on conventional flow cytometry.

BACKGROUND: Studies in T-cell acute lymphoblastic leukemia (T-ALL) have shown that leukemic blast populations may display immunophenotypic heterogeneity. In the clinical setting, evaluation of measurable residual disease during treatment and follow-up is highly dependent on knowledge of the diversity of blast subsets. Here, we set out to evaluate whether variation in expression of the blast marker, TdT, in T-ALL blasts could correspond to differences in morphometric features. METHODS: We investigated diagnostic bone marrow samples from six individual T-ALL patients run in parallel on imaging

Guidelines for Gating Flow Cytometry Data for Immunological Assays.

"Gating" refers to the selection of successive sub-populations of cells for analysis in flow cytometry. While automated analysis algorithms have seen a greatly increased use, there is still heavy reliance on manual gating, based on expert knowledge of cell characteristics. This can lead to variability in how gates are applied, even between individuals experienced in the field. For clinical assays, guidelines often exist, and software may automate the gating process. But there are no comprehensive guidelines across the various types of immunological assays performed in research settings using f

Flow cytometry method to identify arginase-1 expressing MDSCs.

Myeloid-derived suppressor cells (MDSCs) contribute to immune suppression in cancer through arginase-1 (ARG1), which limits T-cell activation. Conventional assays such as ELISA or qPCR cannot determine the cellular source of ARG1 in blood. Recognizing the importance of ARG1 in immunosuppression, we developed and validated a standardized flow cytometry method using a commercially available ARG1 antibody to measure intracellular ARG1 in MDSC subsets. This method includes detailed instructions, including experimental optimization using positive and negative controls, to help other researchers per

Platelet flow cytometry: An evolving platform for clinical hematology.

Flow cytometry has transformed platelet biology and diagnostics through high-resolution, single-cell analysis of surface markers, activation states, and intracellular signaling. This review traces its evolution from early applications in glycoprotein profiling and immune thrombocytopenia to clinical roles in HIT, VITT, and FNAIT. Advances in spectral cytometry, phosphoflow, and fluorescent barcoding have increased analytical depth and supported development of whole blood-compatible platelet function assays. Flow cytometry now enables characterization of platelet subpopulations, assessment of c

Identifying ovarian cortex cell subpopulations using multicolor flow cytometry.

CONTEXT: Ovarian tissue autotransplantation is currently the only proven technique for reusing ovarian tissue after fertility preservation by ovarian tissue cryopreservation. However, one of its limitations relates to the quality of the grafts, both in terms of the number of surviving follicles and the quality of the stromal environment, which is essential for follicular development and graft revascularization. The aim of this study was to validate a technique for characterizing and functionally qualifying ovarian tissue in order to identify cell sub-populations of interest. MATERIALS: Ovaria

Deep Immunophenotyping in ME/CFS Using Spectral Flow Cytometry.

Immune dysfunction is reported to play a significant role in the etiology of myalgic encephalomyelitis/chronic fatigue syndrome (ME/CFS). To gain an understanding of the underlying immune abnormalities associated with this complex condition, a comprehensive approach for characterizing immune cell subsets and their inferred functional states is essential. We developed a high-dimensional flow cytometry method that enables detailed immunophenotyping of peripheral blood mononuclear cells (PBMCs) from ME/CFS patients. By simultaneously measuring over 40 markers on individual cells within one sample

Detection of mulberry bodies using molecular imaging flow cytometry.

BACKGROUND: Fabry disease is a hereditary disorder that leads to the accumulation of glycolipids, such as globotriaosylceramide, because of the absence or decreased activity of the enzyme alpha-galactosidase A, causing various organ dysfunctions. Urinary mulberry bodies (MBs) are specific markers of Fabry disease and can be identified in urinary sediments through microscopic visual evaluation. However, MBs are present in small quantities in urine, and some are difficult to distinguish morphologically from other urinary components, necessitating a highly accurate and objective automated detecti
