The gut microbiota regulates acute foreign body reaction and tissue repair after biomaterial implantation.

We hypothesized that the host microbiome may influence foreign body responses following biomaterial implantation. To test this, we implanted a variety of clinically relevant biomaterials into germ-free or antibiotic-treated mice. Surprisingly, these mice displayed less fibrous tissue deposition, reduced host cell recruitment to the implant site, and differential expression of angiogenic and inflammatory markers. These observations were reversed upon fecal microbiome reconstitution, confirming a causal role of the host microbiome. In a clinically relevant disease model, microbiome-depleted mice

Sabra dermatitis: combined features of delayed hypersensitivity and foreign body reaction to implanted glochidia.

A striking dermatitis referred to by its colloquial designation of sabra dermatitis is associated with glochidia inoculation from the Opuntia cactus commonly known as the prickly pear. We report a 45-year-old woman who had an unexpected encounter with a cactus plant during a trip to Texas. She brushed up against the plant and was aware that she had been inoculated with several spines of the plant. Five days later she developed erythematous papules on the digits accompanied by swelling. The biopsy showed a very striking granulomatous reaction pattern within the dermis. There was a linear patter

Foreign-Body Reaction and Osteolysis in Dorsal Lunate Dislocation Repair With Bioabsorbable Suture Anchor.

Background: In recent years the use of biodegradable suture anchors for treating tendon and ligament pathology in hand surgery became popular. These materials are biocompatible, radiolucent, and load sharing, as they incrementally transfer load to surrounding bone during the resorption process. Despite these numerous advantages, polyglycolic (PGA) and poly-L-lactic acid (PLLA) have become a problem because of the potential risk for foreign body reactions. Methods: This article presents a case of an intraosseous foreign body reaction and massive osteolysis of the proximal carpal after dorsal lu

Deletion of the chemokine receptor CCR2 attenuates foreign body reaction to implants in mice.

Subcutaneous implantation of synthetic materials and biomedical devices often induces abnormal tissue healing - the foreign body reaction - which impairs their function. Here we investigated the role of the chemokine receptor CCR2 in this reaction to subcutaneous implants in mice. We measured angiogenesis, inflammation and fibrogenesis induced by implantation, for 1, 4, 7 and 14days, of polyether-polyurethane sponges in mice with genetic deletion of CCR2 (KO) and WT mice. Blood flow was determined by dye diffusion and laser Doppler perfusion techniques. Cytokines (VEGF, TNF-α, CCL2, TGF-β1) we

Foreign body reaction to hemostatic materials mimicking recurrent brain tumor. Report of three cases.

Chemical agents routinely used in neurosurgery to achieve intraoperative hemostasis can cause a foreign body reaction, which appears on magnetic resonance (MR) images to be indistinguishable from recurrent tumor. Clinical and/or imaging evidence of progression of disease early after surgical resection or during aggressive treatment may actually be distinct features of granuloma in these circumstances. A series of three cases was retrospectively analyzed for clinical, imaging, surgical, and pathological findings, and the consequences they held for further disease management. All patients were b

Foreign Body Granulomatous Reaction of the Breast Mimicking Carcinoma: A Radiologic-Pathologic Diagnostic Pitfall.

Foreign body granulomatous reactions of the breast are uncommon benign entities that may closely mimic carcinoma on imaging. We report the case of a woman in her seventies who presented with persistent breast pain and an enlarging lesion detected on imaging. Mammography demonstrated architectural distortion and focal asymmetry, while ultrasound revealed an irregular hypoechoic mass with posterior acoustic shadowing, leading to classification as a Breast Imaging-Reporting and Data System (BI-RADS) 4 suspicious lesion. Ultrasound-guided core needle biopsy showed fibrous breast tissue with foreig

Foreign body-type giant cell reaction post intracranial endovascular clot retrieval: a postmortem case.

BACKGROUND AND AIMS: Endovascular clot retrieval (ECR) has revolutionised acute ischaemic stroke management. While procedural safety is well established, little attention has been paid to vascular responses to hydrophilic polymer coatings used on retrieval devices. This report presents postmortem histopathological findings implicating a foreign body giant cell reaction secondary to hydrophilic polymer exposure as a cause of recurrent stroke following ECR. METHODS: A detailed clinical and neuropathological review was conducted on a 72-year-old patient who experienced three sequential ischaemic

Mechanically Compliant, Precision-Porous Brain Implants Reduce the Foreign Body Reaction and Guide Regeneration.

Central nervous system (CNS) diseases and injuries might be treated by implanted devices, tissue regenerative scaffolds, or drug delivery platforms. However, inflammatory CNS responses limit these interventions and may worsen outcomes following damage to the CNS. Via the foreign body reaction, macrophages and glial cells trigger a "glial scar" around implants, reducing device performance, scaffold regenerative ability, or drug delivery potential. Previous studies have shown that stiffness of CNS implants significantly affects glial encapsulation, but few studies have investigated materials tha

Immunomodulatory Biomaterials: Tailoring Surface Properties to Mitigate Foreign Body Reaction and Enhance Tissue Regeneration.

The immune cells have demonstrated the ability to promote tissue repair by removing debris, breaking down the extracellular matrix, and regulating cytokine secretion profile. If the behavior of immune cells is not well directed, chronic inflammation and foreign body reaction (FBR) will lead to scar formation and loss of biomaterial functionality. The immunologic response toward tissue repair or chronic inflammation after injury and implantation can be modulated by manipulating the surface properties of biomaterials. Tailoring surface properties of biomaterials enables the regulation of immune

Outflow graft foreign body reaction and thrombosis in HeartMate 3 left ventricular assist device.

Outflow cannula occlusion is an infrequent complication occurring among recipients of continuous flow left ventricular assist devices. Hereby, we present a case of a 66-year-old man with a HeartMate 3 left ventricular assist device that presented 18 months after implantation and during his admission, evidence of poor left ventricular unloading and forward flow uncovered intrinsic and extrinsic outflow cannula obstruction. The patient's course was complicated by cardiogenic shock, diffuse alveolar hemorrhage and multiorgan failure. Post-mortem examination suggested foreign body reaction as a po

Polysialic Acid Hydrogel for Reducing Foreign Body Reactions in Implanted Polyurethane Materials.

Long-term retention of polyurethane (PU) materials as vascular channels, due to foreign body reactions causing fiber encapsulation, will eventually lead to material failure. In this research, an interfacial interpenetrating grafting approach is adopted to graft PSA onto the surface of the PU material (PU-sg-PSA), which exhibits excellent resistance to fibrous encapsulation. The experimental results indicated that, compared with the unmodified PU material, the PU-sg-PSA material exhibited a reduced level of non-specific protein adhesion. In terms of macrophage behavior, macrophages on the PU-sg

Foreign body reactions causing spinal cord tethering: a case-based update.

BACKGROUND: Patients operated on for myelomeningocele (MMC) and lipomeningocele (LMC) can suffer from late functional worsening that, in many cases, is due to spinal cord tethering by the post-repair scarring process. ILLUSTRATIVE CASES: In this case-based update, we report two patients operated on for MMC and LMC, respectively, who presented the clinical manifestations of spinal cord tethering, which we attributed to severe foreign body reactions to the materials used at their primary back surgery. In the first case, the cause of the tethering was an intense fibrotic scar around the silk sut

A Combinatory Therapy of Metformin and Dexamethasone Reduces the Foreign Body Reaction to Intraneural Electrodes.

Neural electrodes used for bidirectional communication between the nervous system and external devices like prosthetic limbs have advanced in neuroprosthetic applications. However, their effectiveness is hindered by the foreign body reaction, a natural immune response causing inflammation and fibrosis around the implanted device. This process involves protein adsorption, immune cell recruitment, cytokine release, and fibroblast activation, leading to a fibrous capsule formation and a decrease in electrode functionality. Anti-inflammatory and antifibrotic strategies have the potential to dimini
