Induction of DNA-DNA cross-link formation in human cells by various psoralen derivatives.

Furocoumarin-induced DNA damage, monoadducts, and cross-links were measured in normal human, xeroderma pigmentosum, and Fanconi's anemia cells after exposure to near-UV (356 nm). At similar concentrations and near-UV doses, photoaddition by 8-methoxypsoralen was twice that by angelicin and the substitution of bromodeoxyuridine for thymidine in one strand of DNA did not alter the binding. The rate of cross-linking by 8-methoxypsoralen was twice that of 5-methoxypsoralen. Low frequencies of cross-links were detected from angelicin and 3-carbethoxypsoralen but none were detected from 5-geranoxyps

The many faces of phytophotodermatitis.

Phytophotodermatitis is a common cutaneous phototoxic reaction produced by contact with a variety of plant substances, followed by sunlight exposure. Phytophotodermatits can simulate child abuse, impetigo, superficial lymphangitis, and larva migrans. The most frequent presentation is hyperpigmentation of the dorsum of the hands, but there are other less common findings such as blisters or lesions mimicking solar lentigines and melasma.

Colloid-amyloid bodies in PUVA-treated human psoriatic patients.

In 4 cases of PUVA-treated psoriatic patients a number of colloid bodies and amyloid-like deposits were seen at the dermo-epidermal junction, in a very similar fashion the colloid bodies and the amyloid substances are deposited in lichen planus and in primary cutaneous anyloidoses respectively. In some instances such a deposition was found within the epidermis and contained remnants of cellular debris. Serial sections revealed that a part of the intraepidermal deposit is located in the dermis. Some deposition is partially composed of typical amyloid filaments and partially of nonamyloidal fila

Diversity of Secondary Metabolites in Roots from Conium maculatum L.

Conium maculatum is known as highly toxic plant, due to piperidine alkaloids present in the aerial parts. In a first attempt, in various tap root samples, however, alkaloids could not be detected. The present study describes active compounds in the tap roots from 16 populations harvested at maturity. The compounds were extracted with dichloromethane from root pieces of single plants and analyzed by gas chromatography-mass spectrometry. Ten bioactive compounds were evaluated: five furocoumarins, two prenylated coumarins, two aliphatic C17-polyacetylenes and the phenylpropanoid elemicin. A high

Phototoxic effect of Thamnosma texana (Dutchman's breeches) in sheep.

Oral exposure of sheep to the airdried, aerial portions of Thamnosma texana (Dutchman's breeches) resulted in severe photosensitization. Sheep fed the plant at 9 or 12 g/kg of body weight/day and held in direct sunlight exhibited signs of phototoxicosis within 24 to 48 hours. The clinical signs manifested were increased body temperature; photophobia; edema of the muzzle, ears, and vulva; keratoconjunctivitis with edema of the cornea; and exudative dermatitis of the skin of the ears, muzzle, and vulva. Lesser dosages of the plant produced similar effects after several days, except that corneal

Thio and Seleno Derivatives of Angelicin as Efficient Triplet Harvesting Photosensitizers: Implications in Photodynamic Therapy.

Photodynamic therapy (PDT) is widely accepted in medical practice for its targeted induction of apoptosis in cancerous cells. Angelicin (Ang) has traditionally been known for its efficacy in cancer treatment and its capability to enter a photoexcited triplet state. This study has comprehensively assessed the effects of substituting individual chalcogen atoms at three specific positions in Angelicin, with the objective of facilitating access to this elusive triplet state to enhance its role as a photosensitizer in PDT. The study scrutinizes various enhancements and factors that are crucial for

[Sosnowsky's hogweed - toxicology and threat to health].

Sosnowsky's hogweed (Heracleum sosnowskyi Manden.) is a genus of plants in the family Apiaceae which also includes Giant Hogweed (Heracleum mantegazzianum Sommier and Levier). They are both found in Central Europe, mainly in neglected green areas or riversides. Sosnowsky's hogweed was brought to Poland from the Soviet Union in the 1950s to be used in animal feed production. Intended goals couldn't be achieved and the plant spread throughout grounds distant to the primarily cultivated lands. Sosnowsky's hogweed is especially hazardous in direct contact with human skin. It results from the conte

Severe Phytophotodermatitis Caused by Mulberry Tree: A Case Report and Literature Review.

Phytophotodermatitis (PPD) is an uncommon dermatologic condition that occurs after exposure to furocoumarins found in certain plant saps, which become activated upon ultraviolet A radiation exposure. This condition is frequently misdiagnosed as cellulitis, allergic dermatitis, or other common skin disorders. Here, we present a case of severe PPD in a 52-year-old male who initially developed a rash and blistering on his forearm following outdoor activity. He was initially misdiagnosed with cellulitis; however, his symptoms persisted despite antibiotic treatment, prompting further evaluation. A

Coumarins from Magydaris pastinacea as inhibitors of the tumour-associated carbonic anhydrases IX and XII: isolation, biological studies and in silico evaluation.

In an in vitro screening for human carbonic anhydrase (hCA) inhibiting agents from higher plants, the petroleum ether and ethyl acetate extracts of Magydaris pastinacea seeds selectively inhibited hCA IX and hCA XII isoforms. The phytochemical investigation of the extracts led to the isolation of ten linear furocoumarins (1-10), four simple coumarins (12-15) and a new angular dihydrofurocoumarin (11). The structures of the isolated compounds were elucidated based on 1 D and 2 D NMR, MS, and ECD data analysis. All isolated compounds were inactive towards the ubiquitous cytosolic isoform hCA I a

Psoralen Derivatives: Recent Advances of Synthetic Strategy and Pharmacological Properties.

Psoralen or furocoumarin is a linear three ring heterocyclic compound. Psoralens are planar, tricyclic compounds, consisting of a furan ring fused to a coumarin moiety. Psoralen has been known for a wide spectrum of biological activities, spanning from cytotoxic, photosensitizing, insecticidal, antibacterial to antifungal effect. Thus, several structural changes were introduced to explore the role of specific positions with respect to the biological activity. Convenient approaches utilized for the synthesis of psoralen skeleton can be categorized into two parts: (i) the preparation of the tric

Mojito-Induced Phytophotodermatitis: A Case of Lime-Triggered Skin Reaction.

INTRODUCTION: Phytophotodermatitis is a cutaneous reaction resulting from skin contact with certain plant-derived phototoxic compounds, followed by ultraviolet exposure. Lime-induced phytophotodermatitis, sometimes called "lime disease" or "margarita photodermatitis," is one of the most common forms, caused by furocoumarins in lime juice or peel. Despite being well described, the condition remains underdiagnosed and is frequently mistaken for allergic dermatitis, burns, cellulitis, or even abuse, particularly when occurring in atypical linear or patterned distributions. 4-Hydroxymethyl- and 4-methoxymethylfuro[2,3-h]quinolin-2(1H)-ones: synthesis and biological properties.

4-Hydroxymethyl-1,6,8-trimethylfuro[2,3-h]quinolin-2(1H)-one (HOFQ) was prepared by a new profitable way, which allowed to synthesize also 4-methoxymethyl-1,6,8-trimethylfuro[2,3-h]quinolin-2(1H)-one (MOFQ), and 4-hydroxymethyl-6,8-dimethylfuro[2,3-h]quinolin-2(1H)-one (HOHFQ). Some biological activities of the three compounds were studied in comparison with 8-MOP. In the dark, they inhibited topoisomerase II, leading to a moderate antiproliferative activity in mammalian cells. The antiproliferative activity was also tested upon UVA irradiation in mammalian cells: all compounds showed higher a

Effects of herbicides on the concentration of poisonous compounds in plants: a review.

Herbicides may raise, lower, or not affect the concentration of poisonous compounds in plants. Herbicides, 2,4,5-T and silvex controlled Wasatch milkvetch (Astragalus miser var oblongifolius) and markedly reduced the concentration of miserotoxin, a poisonous aliphatic nitro compound. The concentration of phototoxic furocoumarins in the leaves of spring parsley (Cymopterus watsonii) decreased to nontoxic concentrations 4 to 5 weeks after treatment with 2,4-D. The hydrocyanic acid content of wild cherry (Prunus spp) decreased after treatment with 2,4-D and 2,4,5-T, but increased in Sudangrass (S

New enantiomers from Ficus variolosa Lindl. ex Benth. and their anti-inflammatory and neuroprotective activities.

Chemical investigation of the whole plant of Ficus variolosa Lindl. ex Benth. led to the discovery of two pairs of previously unreported enantiomers, 1a/1b and 2a/2b, which were separated by chiral HPLC. Their structures were characterised through detailed spectroscopic interpretation, and the stereochemical assignments were achieved by matching the experimental ECD spectra with those calculated at the TDDFT level. Notably, to the best of our knowledge, 2a/2b represents the first enantiomerically resolved example of this type of furocoumarin. The isolates were evaluated for their inhibitory ef
