Fluorescence and Electron Microscopy of Fusobacterium nucleatum-Induced Autophagy.

Fusobacterium nucleatum, an opportunistic oral anaerobe, has emerged as a key microbial contributor of colorectal cancer (CRC) progression and chemoresistance through its modulation of host cellular pathways. Autophagy-a conserved degradation process-has been identified as a central mechanism by which F. nucleatum enhances tumor cell survival. Here, we describe a standardized platform for detecting and quantifying F. nucleatum-induced autophagy in CRC cells using immunofluorescence microscopy with an mRFP-GFP-LC3 reporter to measure autophagic flux, complemented by ultrastructural analysis of

Host-Microbe Mechanisms of Fusobacterium nucleatum in Colorectal Cancer.

The oral-derived pathobiont Fusobacterium nucleatum has increasingly been implicated in colorectal cancer (CRC) progression through its ability to modulate inflammatory, immunological, and molecular pathways. F. nucleatum has emerged as a key oral-derived pathobiont frequently associated with tumor aggressiveness and alterations in the colorectal tumor microenvironment. This review critically examines recent mechanistic evidence from in vitro, in vivo, and advanced experimental models to elucidate how F. nucleatum interacts with host pathways involved in colorectal carcinogenesis. Fusobacterium, quiescent niches, and therapy response in colorectal cancer.

In this issue of Cancer Cell, Galeano Niño et al. report that tumor-infiltrating bacteria like Fusobacterium species induce a quiescent, chemoresistant state in cancer cells in the colon by disrupting epithelial contacts and suppressing proliferation and transcription. These studies highlight microbial-tumor interactions as potential modulators of therapeutic response in colon cancer.

Fusobacterium nucleatum: a transboundary pathogen in host-microbiota networks.

Fusobacterium nucleatum (Fn), a gram-negative anaerobic bacterium,, transitions from oral commensal to systemic pathogen in colorectal cancer (CRC), inflammatory bowel disease (IBD), arthritis, and preterm birth. This review synthesizes Fn's subspecies-specific pathogenic mechanisms. Its virulence factors enable immune evasion, inflammation modulation, and tumor progression. In the gastrointestinal tract, Fn disrupts intestinal barrier integrity through paracellular and apoptotic pathways, activates Th17/ T regulatory (Treg) immune balance alterations, and induces macrophage polarization. Type IV pili facilitated natural competence in Fusobacterium nucleatum.

OBJECTIVES: Many bacterial species naturally take up DNA from their surroundings and recombine it into their chromosome through homologous gene transfer (HGT) to aid in survival and gain advantageous functions. Herein we present the first characterization of Type IV pili facilitated natural competence in Fusobacterium nucleatum, which is a Gram-negative, anaerobic bacterium that participates in a range of infections and diseases including periodontitis, preterm birth, and cancer. METHODS: Here we used bioinformatics on multiple Fusobacterium species, as well as molecular genetics to character

Fusobacterium nucleatum endocarditis: a case report and literature review.

Fusobacteria is anaerobic gram-negative rods, which frequently colonize the oral cavity and can rarely cause deadly diseases in humans. The two most commonly found in this group of bacteria are fusobacterium necrophorum and fusobacterium nucleatum. Only a handful of cases of endocarditis due to fusobacterium have been reported. We describe an 86-year-old male who had a recent tooth extraction presenting to the emergency department with weakness lightheadedness, and pain in his right elbow. He had a low-grade fever of 100.8°. The patient was discharged home but came back less than 24 hours with

Pathogenic Mechanisms of Fusobacterium nucleatum on Oral Epithelial Cells.

Periodontitis is an oral chronic inflammatory disease and may cause tooth loss in adults. Oral epithelial cells provide a barrier for bacteria and participate in the immune response. Fusobacterium nucleatum (F. nucleatum) is one of the common inhabitants of the oral cavity and has been identified as a potential etiologic bacterial agent of oral diseases, such as periodontitis and oral carcinomas. F. nucleatum has been shown to be of importance in the development of diverse human cancers. In the dental biofilm, it exhibits a structural role as a bridging organism, connecting primary colonizers

Characterization of Fusobacterium necrophorum subsp. necrophorum outer membrane proteins.

Liver abscesses are of major economic importance to the cattle industry. These are mainly associated with the presence of Fusobacterium necrophorum, a non-spore forming and Gram-negative anaerobe. There are two main subspecies, F. necrophorum subspecies necrophorum and subsp. funduliforme, and they differ molecularly, morphologically, biochemically and in virulence. Previous studies have shown that the outer membrane proteins (OMP) of F. necrophorum subsp. necrophorum are important for its successful binding to immobilized bovine adrenal gland capillary endothelial (EJG) cells. In this study,

Fusobacterium necrophorum septicemia following Epstein-Barr virus infectious mononucleosis.

We report herein a case of a 20-year-old previously healthy man who presented, 25 days after the onset of clinically and serologically confirmed Epstein-Barr virus (EBV) infectious mononucleosis, Fusobacterium necrophorum septicemia. The diagnosis of postanginal septicemia was confirmed by repeated demonstration of fusiform, obligate anaerobic, Gram-negative bacilli in anaerobic blood cultures, identified as F. necrophorum 15 days after admission. This case report aims at underlining the need of taking into consideration the possibility of severe Fusobacterium septicemia in previously healthy

Fusobacterium necrophorum infections in England and Wales 1990-2000.

In response to a marked increase in both the number of Fusobacterium necrophorum bacteraemia reports to the PHLS Communicable Disease Surveillance Centre and the number of F. necrophorum isolates referred to the PHLS Anaerobe Reference Unit in 1999, the data from both sources on F. necrophorum infections were reviewed for the decade 1990-2000. There were 208 reports of F. necrophorum bacteraemia (average 19/year; range 14-34/year) with a peak in incidence in the late winter months; 68% were from male patients and the peak age range was 16-23 years. Of 205 referred isolates of F. necrophorum, 1

Effect of thymoquinone on Fusobacterium nucleatum‑associated biofilm and inflammation.

Periodontitis affects oral tissues and induces systemic inflammation, which increases the risk of cardiovascular disease and metabolic syndrome. Subgingival plaque accumulation is a trigger of periodontitis. Fusobacterium nucleatum (FN) contributes to subgingival biofilm complexity by intercalating with early and late bacterial colonizers on tooth surfaces. In addition, inflammatory responses to FN are associated with the progression of periodontitis. Nigella sativa Lin. seed, which is known as black cumin (BC), has been used as a herbal medicine to treat ailments such as asthma and infectious

Effect of IgY on Periodontitis and Halitosis Induced by Fusobacterium nucleatum.

Fusobacterium nucleatum is a morbific agent in periodontitis and halitosis. Egg yolk antibody (IgY) was obtained from egg yolks from chickens stimulated with F. nucleatum. This study was to assess the effectiveness of IgY on periodontitis and halitosis caused by F. nucleatum in vitro and in vivo. The growth of F. nucleatum was inhibited (p <0. 05) by different concentrations of IgY in vitro and the results of a Halimeter show volatile sulfur compounds (VSCs) were reduced to 904 ± 57 ppb at a concentration 40 mg/ml of IgY. The changes of fatty acids of F. nucleatum were determined using GC-MS.

Atopobium and Fusobacterium as novel candidates for sarcoidosis-associated microbiota.

Sarcoidosis is a granulomatous disease that mainly affects the lung. A role of microbial factors in disease pathogenesis is assumed, but has not been investigated systematically in a large cohort. This cross-sectional study compared the lung microbiota of 71 patients with sarcoidosis, 15 patients with idiopathic pulmonary fibrosis (non-infectious controls) and 10 healthy controls (HCs). Next-generation sequencing of 16S DNA was used on bronchoalveolar lavage samples to characterise the microbial composition, which was analysed for diversity and indicator species. Host genotypes for 13 known sar

Antibody development to Fusobacterium necrophorum in patients with peritonsillar abscess.

A polymicrobial mixture of aerobic and anaerobic bacteria is commonly recovered from peritonsillar abscess (PTA) aspirates. Previous studies have suggested a role for Fusobacterium necrophorum (FN) in the development of PTA. The purpose of the current study was to explore whether anti-FN antibodies were produced in patients with PTA. We developed a novel immunofluorescence-based method to measure anti-FN antibody levels in acute and convalescent sera from 15 patients with PTA and 47 patients with chronic tonsillar conditions (controls) undergoing acute or elective tonsillectomy, respectively.

Fusobacterium necrophorum endocarditis Case report and review of the literature.

Fusobacterium necrophorum is a gram-negative anaerobic bacillus that can cause serious systemic infections typically in previously healthy young adults. Lemierre's syndrome, also known as post-anginal sepsis or necrobacillosis, is the infection most usually associated with F. necrophorum. However, F. necrophorum is also a very rare cause of anaerobic endocarditis. Mortality and rates of thromboembolism are high with F. necrophorum endocarditis. In this article, we describe a case of F. necrophorum native valve infective endocarditis. The patient was treated with penicillin plus clindamycin fol

[Bacterial meningitis as a complication of Fusobacterium necroforum infection in adults].

Fusobacterium necrophorum, an anaerobic, gram-negative rod, belongs to the physiological flora of the oropharynx. It causes Lemierre's syndrome characterized by oropharyngeal infection, septic thrombophlebitis of the neck, in particular of the internal jugular vein, and metastatic abscesses, predominantly in the lungs. Rarely, and mainly in children, it causes meningitis. Here we report the clinical course of a 25-year-old woman with F. necrophorum meningitis. She presented with incomplete, right third nerve palsy. Within a few days, she developed fever, meningism and progressive reduction of
