Modulation of Host Immunity by Human Respiratory Syncytial Virus Virulence Factors: A Synergic Inhibition of Both Innate and Adaptive Immunity.

The Human Respiratory Syncytial Virus (hRSV) is a major cause of acute lower respiratory tract infections (ARTIs) and high rates of hospitalizations in children and in the elderly worldwide. Symptoms of hRSV infection include bronchiolitis and pneumonia. The lung pathology observed during hRSV infection is due in part to an exacerbated host immune response, characterized by immune cell infiltration to the lungs. HRSV is an enveloped virus, a member of the Pneumoviridae family, with a non-segmented genome and negative polarity-single RNA that contains 10 genes encoding for 11 proteins. These in

TGR5/Cathepsin E signaling regulates macrophage innate immune activation in liver ischemia and reperfusion injury.

The role and underlying mechanism of plasma membrane-bound G protein-coupled bile acid receptor (TGR5) in regulating macrophage innate immune activation during liver ischemia and reperfusion (IR) injury remains largely unclear. Here, we demonstrated that TGR5 depletion in myeloid cells aggravated liver injury with increased macrophage infiltration and enhanced inflammation in livers post-IR. While TGR5 deficiency enhanced mobility and proinflammatory M1 polarization of macrophages, TGR5 agonist enhanced the anti-inflammatory effect of TGR5 both in vivo and in vitro. Microarray profiling reveal

Pilot-scale duckweed-CO2 wastewater bioremediation system: a synergistic approach for nutrient removal, carbon capture, and biomass production.

This study presents a pilot-scale duckweed-based wastewater treatment system enhanced with CO2 fertilization to improve nutrient removal, CO2 capture, and biomass productivity. Three strains of Landoltia punctata were cultivated continuously for five months. In spring, CO2 supplementation significantly increased the biomass yield and starch and protein productivity (by more than 12.2, 2.4 and 3.5 g/m2/d, respectively), representing increases of 189-259 %, 1,108-2,330 % and 143-166 %, respectively, compared with those of the control group. The removal rates of total nitrogen (TN) and total phos

From inflammation to healing: the crucial role of GPR91 activation and SDH inhibition in chronic diabetic wound recovery.

BACKGROUND: Diabetic foot ulcers (DFU) typically exhibit impaired healing due to dysregulated re-epithelialization and excessive inflammation. Succinate, a key metabolic intermediate, is now understood to regulate inflammation through G Protein-Coupled Receptor 91 (GPR91) and succinate dehydrogenase (SDH), although its role in DFU remains unclear. METHODS: Co-cultures of M2 macrophages and epithelial cells, along with clinical samples, were used to analyze the expression of GPR91 and SDH. Functional assays were performed using high glucose (HG)-treated M2 macrophages (HG-M2) and an in vivo mo

Optimal Protein Intake in Pre-Dialysis Chronic Kidney Disease Patients with Sarcopenia: An Overview.

Multi-factors, such as anorexia, activation of renin-angiotensin system, inflammation, and metabolic acidosis, contribute to malnutrition in chronic kidney disease (CKD) patients. Most of these factors, contributing to the progression of malnutrition, worsen as CKD progresses. Protein restriction, used as a treatment for CKD, can reduce the risk of CKD progression, but may worsen the sarcopenia, a syndrome characterized by a progressive and systemic loss of muscle mass and strength. The concomitant rate of sarcopenia is higher in CKD patients than in the general population. Sarcopenia is also

Characteristics of Radish Seed Protein and Its In Vitro Digested Products After Ultrasound-Heat Co-Treatment.

Radish seed, a traditional Chinese herbal medicine rich in oil, generates a significant byproduct called radish seed meal after oil extraction, which contains high levels of protein. To develop novel plant-based proteins, this study investigated the effects of heat treatment (37-95°C, 30 min) with or without ultrasonication (20 kHz, 40% amplitude, 10 min) on the characteristics of radish seed protein (RSP) extracted from radish seed meal. Results showed that RSP particle size decreased after heating at 37-65°C (with or without sonication) but increased at 75-95°C. The combination of heat treat

An O/W antibacterial emulsion stabilized by protein-polyphenol complexes to control food spoilage: A study on whey protein and caffeic acid.

This study used a non-covalent complex of whey protein (WP) and caffeic acid (CA) as an interface-stabilizing agent. The interaction between WP and CA was analyzed through spectroscopy and molecular dynamics simulations. A Pickering emulsion (WC-PE) with oregano essential oil (OEO) was created using the WP-CA complex and characterized. This complex was then combined with sodium alginate (SA) to produce calcium alginate gel beads (WC-PE-CAB), which were tested for their ability to preserve shrimp freshness. FT-IR spectroscopy and molecular dynamics showed that CA mainly binds to β-lactoglobulin

alpha-Thrombin upregulates G alpha i3 in human vascular endothelial cells.

BACKGROUND AND PURPOSE: During thrombosis, alpha-thrombin becomes sequestered by fibrin and the subendothelial basement membrane, and it is available to interact with the vasculature over prolonged periods. In this study, we investigated the long-term effect of alpha-thrombin on G alpha i3 and G alpha s levels in human vascular endothelial cells (EC). Because obesity is associated with changes in receptor signaling in many animal models, we also explored the influence of this clinical risk factor. METHODS: Primary cultures of human EC were exposed to alpha-thrombin for 16 hours, and immunolog

Metabolites of Synthetic Cannabinoid 5F-MDMB-PINACA Retain Affinity, Act as High Efficacy Agonists and Exhibit Atypical Pharmacodynamic Properties at CB1 Receptors.

Synthetic cannabinoid receptor agonists (SCRAs) are a large group of abused psychoactive compounds that elicit numerous toxic effects not observed with cannabis, including death. Abuse of third-generation SCRA 5F-MDMB-PINACA (also known as 5F-ADB) has been associated with over 40 fatalities. This SCRA is metabolized to several active phase I metabolites, including excessively high post-mortem serum concentrations of an ester hydrolysis metabolite, 5F-MDMB-PINACA-M7 (M7). Although high serum concentrations of M7 (and other active metabolites) have been suggested to contribute to 5F-MDMB-PINACA

Early applications of granulocyte colony-stimulating factor (G-CSF) can stabilize the blood-optic-nerve barrier and ameliorate inflammation in a rat model of anterior ischemic optic neuropathy (rAION).

Granulocyte colony-stimulating factor (G-CSF) was reported to have a neuroprotective effect in a rat model of anterior ischemic optic neuropathy (rAION model). However, the therapeutic window and anti-inflammatory effects of G-CSF in a rAION model have yet to be elucidated. Thus, this study aimed to determine the therapeutic window of G-CSF and investigate the mechanisms of G-CSF via regulation of optic nerve (ON) inflammation in a rAION model. Rats were treated with G-CSF on day 0, 1, 2 or 7 post-rAION induction for 5 consecutive days, and a control group were treated with phosphate-buffered

Nutritional and Functional Potential of Rapeseed and Canola Proteins: Advances in Protein Extraction, Phenolic Removal, and Food Applications.

Oilseeds, such as rapeseed and canola, are promising plant protein sources, containing approximately 35%-50% protein on a dry weight basis, comparable to soy (30%-55%) and pea (23%-30%). However, their application in foods is limited by anti-nutritional factors (ANFs), residual oils, and phenolic compounds, which impair solubility, digestibility, and functional performance. This review critically evaluates protein extraction and refining strategies from rapeseed and canola meals, emphasizing phenolic removal to enhance functionality. Conventional alkaline extraction followed by isoelectric pre

Effects of Heat Treatment on the Structural and Functional Properties of Phaseolus vulgaris L. Protein.

The paper presents the effect of heat treatment at 80 °C at different times (3, 5, 7, and 9 min) on the structural and functional properties of Phaseolus vulgaris L. protein (PVP, bean protein powder). Surface and structure properties of PVP after heat treatment were analyzed using a Fourier transform infrared spectrometer (FTIR), a fluorescence spectrophotometer, a visible light spectrophotometer, a laser particle size analyzer, and other equipment. The secondary structure and surface hydrophobicity (H0) of PVP changed significantly after heat treatment: the β-sheet content decreased from 25.

Meeting physical activity guidelines in conjunction with higher protein intake: associations with appendicular lean soft tissue index in middle aged adults with cancer.

BACKGROUND: Loss of muscle mass is a common concern among patients with cancer. The aim of this study was to examine whether meeting the World Health Organization physical activity guidelines in combination with a higher vs. lower than the recommended daily allowance (RDA) protein intake is associated with greater appendicular lean soft tissue index (ALSTI) in adults aged 40-59 years with cancer from the National Health and Nutrition Examination Survey. METHODS: Participants were categorized by physical activity levels (moderate ≥ 150 min/week or vigorous ≥ 75 min/week) and protein intake (>

Cannabinoid Receptor 1 Participates in Liver Inflammation by Promoting M1 Macrophage Polarization via RhoA/NF-κB p65 and ERK1/2 Pathways, Respectively, in Mouse Liver Fibrogenesis.

UNLABELLED: Macrophage M1/M2 polarization mediates tissue damage and inflammatory responses. Cannabinoid receptor (CB) 1 participated in liver fibrogenesis by affecting bone marrow (BM)-derived monocytes/macrophages (BMMs) activation. However, the knowledge of whether CB1 is involved in the polarization of BMMs remains limited. Here, we found M1 gene signatures (including CD86, MIP-1β, tumor necrosis factor, IL-6, and inducible nitric oxide synthase) and the amount of M1 macrophages (CD86+ cells, gated by F4/80) were significantly elevated in carbon tetrachloride (CCl4)-induced mouse injured l

Opsin-G11-mediated signaling pathway for photic entrainment of the chicken pineal circadian clock.

Light is a major environmental signal for entrainment of the circadian clock, but little is known about the intracellular phototransduction pathway triggered by light activation of the photoreceptive molecule(s) responsible for the phase shift of the clock in vertebrates. The chicken pineal gland and retina contain the autonomous circadian oscillators together with the photic entrainment pathway, and hence they represent useful experimental models for the clock system. Here we show the expression of G11alpha, an alpha subunit of heterotrimeric G-protein, in both tissues by cDNA cloning, Northe

Additive renoprotective effects of oral calcitriol on patients with IgA nephropathy receiving RAS blockers.

BACKGROUND: Patients with immunoglobulin A nephropathy (IgAN) and moderate proteinuria are at risk of progressive renal failure despite the maximal use of renin-angiotensin system blockers (RASBs). This study evaluated the efficacy of active vitamin D in reducing proteinuria in patients with IgAN receiving RASBs. METHODS: In this double-blind, placebo-controlled trial, we enrolled biopsy-confirmed patients with IgAN and persistent proteinuria (0.5-3.5 g/day) after maximal use of RASBs. Patients were randomized to receive either 0.25 μg/day of calcitriol or placebo for 12 months. The primary o

RGS Proteins in Heart: Brakes on the Vagus.

It has been nearly a century since Otto Loewi discovered that acetylcholine (ACh) release from the vagus produces bradycardia and reduced cardiac contractility. It is now known that parasympathetic control of the heart is mediated by ACh stimulation of G(i/o)-coupled muscarinic M2 receptors, which directly activate G protein-coupled inwardly rectifying potassium (GIRK) channels via Gβγ resulting in membrane hyperpolarization and inhibition of action potential (AP) firing. However, expression of M2R-GIRK signaling components in heterologous systems failed to recapitulate native channel gating k

Two opposite signal transducing mechanisms regulate a G-protein-coupled guanylyl cyclase.

Membrane-bound guanylyl cyclase (GC) is regulated by muscarinic receptors (mAChRs). Carbamylcholine (CC) induces a "dual" biological response on GC activity. Thus, an activation is observed at 0.1 nM and a maximal response at 1 nM CC. However, at higher agonist concentration (> 100 nM), there is an agonist-dependent inhibition of GC. This CC dual response is affected by 4-DAMP and HDD (M3 antagonists), which produce a right-shift of the CC curve; the maximal CC dose response with 4-DAMP is more potent than that with HDD. Moreover, AFDX-DS (an M2 antagonist) increases basal activity and decreas
