Unexpected variability of gametogenic pathways and interspecific chromosomal leakage in Pelophylax esculentus hybrids.

Hybridization is common among animals and plants, but often leads to sterility due to improper chromosome pairing. Some hybrids overcome sterility through modified gametogenesis, enabling clonal or hemiclonal reproduction. In water frog hybrids from the Pelophylax esculentus complex, hybridogenesis typically ensures hemiclonal reproduction. These hybrids inherit haploid chromosomal sets (n = 13) from both parental species, Pelophylax lessonae and Pelophylax ridibundus. During hybrid gametogenesis, one genome is eliminated while the other is clonally transmitted. We analyzed hybrid males and fe

Fancm has dual roles in the limiting of meiotic crossovers and germ cell maintenance in mammals.

Meiotic crossovers are required for accurate chromosome segregation and producing new allelic combinations. Meiotic crossover numbers are tightly regulated within a narrow range, despite an excess of initiating DNA double-strand breaks. Here, we reveal the tumor suppressor FANCM as a meiotic anti-crossover factor in mammals. We use unique large-scale crossover analyses with both single-gamete sequencing and pedigree-based bulk-sequencing datasets to identify a genome-wide increase in crossover frequencies in Fancm-deficient mice. Gametogenesis is heavily perturbed in Fancm loss-of-function mic

An atypical Arp2/3 complex is required for Plasmodium DNA segregation and malaria transmission.

Plasmodium parasites, the causative agents of malaria, undergo crucial developments within the mosquito vector, initiated by the formation of male and female gametes. Male gametogenesis involves three rapid rounds of mitosis without nuclear or cell division, followed by a single round of DNA segregation and nuclear division during gamete budding. How the cell organizes the segregation of eight genomes from a single octoploid nucleus into eight haploid gametes is currently unknown. Here we discovered an atypical Arp2/3 complex in Plasmodium important for DNA segregation during male gametogenesi

Heterozygosity analysis of spontaneous 2n female gametes and centromere mapping of the diploid Hevea brasiliensis based on full-sib triploid populations.

Unreduced megagametophytes via second-division restitution were confirmed through heterozygosity analysis, and four candidate physical centromeres of rubber were located for the first time. The evaluation of maternal heterozygosity restitution (MHR) is vital in identifying the mechanism of 2n gametogenesis and assessing the utilization value of 2n gametes. In this study, three full-sib triploid populations were employed to evaluate the MHR of 2n female gametes of rubber tree clone GT1 and to confirm their genetic derivation. The 2n female gametes of GT1 were derived from second-division restit

Optional Endoreplication and Selective Elimination of Parental Genomes during Oogenesis in Diploid and Triploid Hybrid European Water Frogs.

Incompatibilities between parental genomes decrease viability of interspecific hybrids; however, deviations from canonical gametogenesis such as genome endoreplication and elimination can rescue hybrid organisms. To evaluate frequency and regularity of genome elimination and endoreplication during gametogenesis in hybrid animals with different ploidy, we examined genome composition in oocytes of di- and triploid hybrid frogs of the Pelophylax esculentus complex. Obtained results allowed us to suggest that during oogenesis the endoreplication involves all genomes occurring before the selective

Germ cell-intrinsic effects of sex chromosomes on early oocyte differentiation in mice.

A set of sex chromosomes is required for gametogenesis in both males and females, as represented by sex chromosome disorders causing agametic phenotypes. Although studies using model animals have investigated the functional requirement of sex chromosomes, involvement of these chromosomes in gametogenesis remains elusive. Here, we elicit a germ cell-intrinsic effect of sex chromosomes on oogenesis, using a novel culture system in which oocytes were induced from embryonic stem cells (ESCs) harboring XX, XO or XY. In the culture system, oogenesis using XO and XY ESCs was severely disturbed, with

An essential role of the basal body protein SAS-6 in Plasmodium male gamete development and malaria transmission.

Gametocytes are the sole Plasmodium parasite stages that infect mosquitoes; therefore development of functional gametes is required for malaria transmission. Flagellum assembly of the Plasmodium male gamete differs from that of most other eukaryotes in that it is intracytoplasmic but retains a key conserved feature: axonemes assemble from basal bodies. The centriole/basal body protein SAS-6 normally regulates assembly and duplication of these organelles and its depletion causes severe flagellar/ciliary abnormalities in a diverse array of eukaryotes. Since basal body and flagellum assembly are

The role of LLPS of biomacromolecules in the gonads and related diseases.

Liquid-liquid phase separation (LLPS), an emerging mechanism of intracellular organization and regulation, has attracted increasing attention in reproductive biology in recent years. By promoting the spatiotemporal compartmentalization of biomolecules, LLPS is involved in a wide range of essential biological processes, including transcriptional regulation, chromatin remodeling, and mRNA stabilization and translation. Increasing evidence has demonstrated that LLPS plays indispensable roles in the development, maturation, and differentiation of both sperm and oocytes, while also contributing to

Reproductive characteristics and gametogenic cycle of the scleractinian coral Dendrophyllia ramea.

The present study marks a pioneering investigation into the reproductive cycle of the scleractinian coral Dendrophyllia ramea. This is one of the first reproduction studies conducted in the Mediterranean Sea for a colonial azooxanthellate coral. Coral samples were collected in 2017 (May and October) and 2018 (February and July) in the Alborán Sea (SW Mediterranean). This location was selected due to its rarity as one of the few sites where this species thrives at depths shallower than 40 m. These samples were used to study the sexual patterns, fertilization mechanisms and gametogenic cycles by

Loss of serine/threonine protein phosphatase 6 severely impairs sexual stage development in malaria parasite Plasmodium berghei.

Protein phosphorylation plays a critical role during the development of malaria parasites. Here, we performed a functional analysis of the Plasmodium berghei Ser/Thr protein phosphatase 6 (PbPP6), which is associated with the plasma membrane of macrogametes and ookinetes. Compared to wild-type P. berghei, the genetic disruption of pbpp6 (∆pbpp6) resulted in reduced asexual growth of the parasites and prolonged survival of infected mice. The ∆pbpp6 parasites showed impaired gametogenesis, particularly affecting male gametogenesis, which substantially decreased both ookinete formation and mosqui

Epigenetics of oogenesis.

Epigenetic changes include all modifications affecting the expression of genes without changing the nucleotide sequence of the genome. Most studied epigenetic changes include DNA methylation, histone alterations and non-coding RNAs. DNA methylation is an important epigenetic mark, protecting the genome during gametogenesis and early embryo development. Demethylation process is a genome-wide event, taking place in two distinct waves during gametogenesis. The first event helps restore naïve pluripotency of the zygote, while the second event aids in the loss of parental epigenetic memory and faci

Epidemiology of gestational trophoblastic disease.

The epidemiology of gestational trophoblastic disease is not well understood. Methodologic problems with published reports limit the interpretation of incidence data, although the frequency of hydatidiform mole appears to be about one per 1000 pregnancies. No consistent temporal trends in rates of either hydatidiform mole or choriocarcinoma are evident. Hydatidiform mole appears to be caused by abnormal gametogenesis and fertilization. Age, ethnicity, and a history of hydatidiform mole appear to be important risk factors for hydatidiform mole. Age, ethnicity, a history of hydatidiform mole or

A Microfluidics-Based Assay for the Discovery of Gametogenic Rejuvenation Factors in Saccharomyces cerevisiae.

Saccharomyces cerevisiae progressively ages through asymmetrical cell divisions, retaining age-associated damage in the mother cell while producing young daughter cells. Gametogenesis completely resets the replicative lifespan of aged cells, thus rewinding the aging clock for the next generation. Although gametogenic rejuvenation in yeast has been described before, techniques to quantify the resetting of replicative lifespan at high throughput have been lacking. Previous methods for quantifying rejuvenation following gametogenesis were limited to low-throughput, labor-intensive approaches that

Analysis and gonadal localization of Speedy A mRNA transcript, a novel gene associated with early germline cells in the scallop, Argopecten purpuratus.

The Speedy A (spdya) gene is a member of the Speedy/RINGO family, encoding a spdya protein associated with cellular cycle and meiosis in vertebrates. Results from genetic analyses indicated spdya conditional knockout mice are sterile, suggesting that this protein has essential functions in mammalian reproduction. There, however, are no published reports on the localization of spdya mRNA in the germline or in somatic cell lineages within the gonads from mollusks or other invertebrate species. Using a previously obtained transcriptome assembly from the scallop Argopecten purpuratus, an economica

Identification and preliminary characterization of Plasmodium falciparum proteins secreted upon gamete formation.

Malaria long-term elimination depends on parasite transmission control. Plasmodium sexual stage maturation in the mosquito, including egress from the host erythrocyte, is one of the prime targets for transmission-blocking interventions. This work aims to identify candidate molecules potentially involved in gamete emergence from the host erythrocyte, as novel transmission blocking targets. We analyzed by quantitative mass spectrometry the proteins released/secreted by purified Plasmodium falciparum gametocytes upon induction of gametogenesis. The proteome obtained showed a good overlap (74%) wi

Epitranscriptomic modifications in embryonic development: insights into natural and ART-induced mechanisms and implications.

BACKGROUND: Mammalian embryo development involves a complex process governed by multiple layers of cellular and molecular regulation mechanisms. ART is widely used around the world to assist fertility in humans, with ∼12 million babies being born by ART in the last 40 years. These technologies are also used extensively for reproductive purposes in other mammalian species that have many analogies with human reproductive biology. Epitranscriptomic marks, including RNA modifications such as N6-methyladenosine (m6A) and N1-methyladenosine (m1A), modulate gene expression during gametogenesis and em

Cyprinid Juji (Gobiocypris rarus) as a model fish to study germ cell development and gonadal differentiation.

Although the cyprinid zebrafish has been widely used to study gametogenesis and gonadal differentiation, it has remarkable limitations, such as the loss of sex determinants in laboratory strains, a juvenile ovary stage in both sexes, and oocyte defects generally leading to sex reversal. Here, we develop a novel small cyprinid model for studying gonadal differentiation and gametogenesis, Juji (Gobiocypris rarus), which possesses a genetic sex determination system. We generate a transgenic line of Juji, Tg(ddx4:EGFP-UTRddx4), enabling lifetime visualization of germline development, and find that

Sex-Specific Transcriptome Signatures in Pacific Oyster Hemolymph.

BACKGROUND/OBJECTIVES: Sex determination and differentiation exhibit remarkable molecular diversity across taxa, driven by genetic, epigenetic, and environmental factors. Invertebrates with sequential hermaphroditism, such as the Pacific oyster (Magallana gigas), represent a poorly understood system despite their role as keystone species and contribution to a substantial aquaculture industry. METHODS: To identify sex-related molecular markers during gametogenesis, we repeatedly sampled hemolymph from artificially conditioned oysters over two months, and sex phenotypes were assigned at the end

How annual course of photoperiod shapes seasonal behavior of diploid and triploid oysters, Crassostrea gigas.

In this work, we study if ploidy (i.e. number of copies of chromosomes) in the oyster Crassostrea gigas may introduce differences in behavior and in its synchronization by the annual photoperiod. To answer to the question about the effect of the seasonal course of the photoperiod on the behavior of C. gigas according to its ploidy, we quantified valve activity by HFNI valvometry in situ for 1 year in both diploid and triploid oysters. Chronobiological analyses of daily, tidal and lunar rhythms were performed according the annual change of the photoperiod. In parallel, growth and gametogenesis
