Gardnerella vaginalis is a facultative anaerobic bacterium that inhabits the genitourinary tract of both healthy women and those with bacterial vaginosis. We report a case of G. vaginalis bacteremia associated with severe toxic encephalopathy in a young woman. Anaerobic blood cultures yielded pure growth of small gram-variable rods later identified as G. vaginalis by both rapid biochemical tests and 16S rRNA gene sequencing. The patient recovered after treatment with amoxicillin-clavulanate according to the in vitro susceptibility testing. The complete genome of G. vaginalis isolate from blood

A commercial system (Affirm VP Microbial Identification Test; MicroProbe Corp.) for detection of vaginal pathogens was evaluated with 176 consecutive women attending a sexually transmitted disease clinic for genital complaints. Vaginal swab specimens were used for culture of Gardnerella vaginalis and Trichomonas vaginalis, preparation of a vaginal smear for Gram stain interpretation, and wet mount evaluation. An additional swab was used to evaluate the 30-min nonisotopic oligonucleotide probe test. The automated probe system detected G. vaginalis in 69 (95%) of 73 women having > 5 x 10(5) CFU

Since it was first reported in the 1950s, bacterial vaginosis (BV) has become a globally vital concern among women of childbearing age. Gardnerella vaginalis is widely recognized as the primary causative agent responsible for BV development. G. vaginalis has a strong tendency to form biofilms which have been linked to widespread antimicrobial tolerance and recurrent or persistent BV episodes. Our study demonstrated that proteins constitute more than 50% of the G. vaginalis biofilm matrix, which significantly protects the bacterium from degradation by lysozyme, and that LasA, an elastase derive

BACKGROUND: Bacterial vaginosis (BV) affects 20-50% of reproductive-age female patients annually, arising when opportunistic pathogens outcompete healthy vaginal flora. Many patients fail to resolve symptoms with a course of metronidazole, the current first-line treatment for BV. Our study was designed to identify genomic variation associated with metronidazole resistance among strains of Gardnerella vaginalis spp. (GV), a genus of biogenic-amine-producing bacteria closely associated with BV pathogenesis, for the development of a companion molecular diagnostic.
METHODS: Whole-genome sequencin

A 20 year old female, 14 days post partum, presented to the Emergency Department in severe respiratory distress. Imaging of her chest revealed a left tension hydropneumothorax with significant mediastinal displacement. A chest drain was inserted and over 4L of cloudy-brown malodourous fluid was drained. Gardnerella Vaginalis was isolated on culture of the fluid. The pyohydropneumothorax and associated sepsis, was refractory to management with a chest drain and antimicrobial therapy. She required a video-assisted thoracoscopic surgery and decortication of her unexpanded lung. She ultimately mad

The vaginal epithelial barrier, which integrates mechanical, immune, chemical, and microbial defenses, is pivotal in safeguarding against external pathogens and upholding the vaginal microecological equilibrium. Although the widely used metronidazole effectively curtails Gardnerella vaginalis, a key pathogen in bacterial vaginosis, it falls short in restoring the vaginal barrier or reducing recurrence rates. Our prior research highlighted Lactobacillus crispatus CCFM1339, a vaginally derived Lactobacillus strain, for its capacity to modulate the vaginal epithelial barrier. In cellular models,

Bacterial vaginosis (BV) is the most common vaginal infection in reproductive women, which is characterized by depleted level of lactic acid bacteria and overgrowth of anaerobes such as Gardnerella vaginalis spp. Lactic acid bacteria have been known to be beneficial for amelioration of BV, since they produce antimicrobial substances against G. vaginalis spp. The objectives of this study were to characterize different fractions of cell-free supernatant of Lactobacillus paracasei CH88 (LCFS) and investigate antibacterial activity of the LCFS fractions against G. vaginalis in-vitro and in-vivo. We report the first documented case of Gardnerella vaginalis peritonitis in a 55-year-old man undergoing continuous ambulatory peritoneal dialysis (CAPD) for renal failure of unknown etiology. He was started on CAPD three months after catheter insertion and simultaneous umbilical hernia repair with mesh placement. His follow-up was notable for poor compliance and geographical isolation. He presented to the emergency department for abdominal pain associated with an abscess at the catheter exit site. Microbiological culture identified G. vaginalis, an organism typically found in the female vagin

We present a case of a male patient with a history of renal allograft transplant four years prior to admission who presented with acute kidney injury. There were initial concerns for acute transplant rejection due to elevated creatinine. Urine cultures were positive for Gardnerella vaginalis. Renal biopsy results were concerning for chronic pyelonephritis. The patient developed hypertension, fever, and hematuria during the hospital course. He was initially treated with metronidazole, vancomycin, and cefepime. Broad-spectrum antibiotics were quickly discontinued, and the patient improved with m

Gardnerella vaginalis is the main pathogen that causes bacterial vaginosis. In the healthy vaginal microecological environment of a woman, the lactobacilli produce lactate and hydrogen peroxide to inhibit the growth of pathogens such as G. vaginalis. The lack of lactobacilli results in a high pH and low hydrogen peroxide in the vagina which facilitate G. vaginalis growth, leading to the imbalance of the vaginal microecology. In this study, lactate and hydrogen peroxide were added to a G. vaginalis culture medium to simulate the co-culture of the lactobacilli and G. vaginalis, and then the gene

Gardnerella spp. are considered a hallmark of bacterial vaginosis, a dysbiosis of the vaginal microbiome. There are four cpn60 sequence-based subgroups within the genus (A, B, C and D), and thirteen genome species have been defined recently. Gardnerella spp. co-occur in the vaginal microbiome with varying abundance, and these patterns are shaped by a resource-dependent, exploitative competition, which affects the growth rate of subgroups A, B and C negatively. The growth rate of rarely abundant subgroup D, however, increases with the increasing number of competitors, negatively affecting the g

In a study of 2224 adult women from the cytology clinic of the University College Hospital, Ibadan, Nigeria, the prevalence of 'specific vaginal infection' (i.e. Gardnerella vaginalis, Trichomonas vaginalis and Candida albicans) was 14.5%. For individual organisms, the rates were 9.76% for G. vaginalis, 2.52% for T. vaginalis and 2.20% for C. albicans. About half of the patients were asymptomatic while others were referred from other clinics with vaginal discharge, cervical erosion, post-coital bleeding, intermenstrual bleeding and various other symptoms and signs. The infections were almost u

A double-blind, randomized, therapeutic study was carried out in 289 patients with vaginal discharge and growth of Gardnerella vaginalis (GV) and no growth of Neisseria gonorrhoeae, Trichomonas vaginalis or Candida. Treatment consisted of either pivampicillin (Pondocillin), 700 mg twice daily for seven days, or metronidazole, 500 mg twice daily for seven days. The effect was evaluated on the basis of the patient's statement and on the result of culture for GV immediately after end of treatment. Evaluated from the culture results metronidazole was significantly more effective than pivampicillin

INTRODUCTION: Peritonsillar abscess (PTA) is usually caused by common oropharyngeal microorganisms. Gardnerella vaginalis, which is primarily associated with bacterial vaginosis, is rarely implicated in oropharyngeal or deep neck infections.
CASE PRESENTATION: An otherwise healthy 18-year-old man developed a left PTA that resolved after incision and drainage, 5 days of intravenous ampicillin/sulbactam, and subsequent oral antimicrobial therapy. Initial abscess culture yielded only normal oral flora. Eight days after discharge, he engaged in oral sex with a female partner. Four days later, PTA

Current treatments for bacterial vaginosis (BV) often result in recurrent disease. Gardnerella, a key player in BV pathogenesis, forms biofilms on vaginal epithelial cells. Recombinant endolysins have shown to specifically kill Gardnerella, but not commensal lactobacilli, in vitro. This study evaluated the pharmacodynamics of BNT331-endolysin (BNT331-EL) on vaginal samples from 49 women with BV (Nugent score ≥7, Amsel criteria, and clue cells). Whole genome sequencing confirmed BV-associated community state types IV-B and III, with Gardnerella dominating in 53% of samples and present in 86%. E

Gardnerella species are associated with bacterial vaginosis (BV) and have been investigated as etiological agents of the condition. Nonetheless, the isolation of this taxon from healthy individuals has raised important questions regarding its etiological role. Recently, using advanced molecular approaches, the Gardnerella genus was expanded to include several different species that exhibit differences in virulence potential. Understanding the significance of these different species with respect to mucosal immunity and the pathogenesis and complications of BV could be crucial to solving the BV
