BACKGROUND/OBJECTIVES: Ulp1 is a vital regulator of the cell cycle, with its absence leading to G2/M phase arrest in Saccharomyces cerevisiae. This study aims to investigate the role of Ulp1 in cell cycle regulation in Pichia pastoris and to elucidate its mechanisms of action, particularly through the modulation of the gene INO1.
METHODS: We generated Ulp1 knockout strains in Pichia pastoris using the FLP-FRT system and performed RNA sequencing (RNA-seq) to analyze gene expression changes. We assessed cell proliferation in Ulp1 knockout and INO1 overexpressing strains, as well as the effects

Engineered conditional gene expression is used in appraisal of gene function and pathway relationships. For pathogens like the fungus Candida albicans, conditional expression systems are most useful if they are active in the infection environment and if they can be utilized in multiple clinical isolates. Here, we describe such a system. It employs the RBT5 promoter and can be implemented with a few PCRs. We validated the system with RBT5 promoter fusions to two genes that promote filamentation and polarized growth, UME6 and HGC1, and with efg1Δ/Δ mutants, which are defective in an activator of

In the last decades, Candida albicans has served as the leading causal agent of life-threatening invasive infections with mortality rates approaching 40% despite treatment. Candida albicans (C. albicans) exists in three biological phases: yeast, pseudohyphae, and hyphae. Hyphae, which represent an important phase in the disease process, can cause tissue damage by invading mucosal epithelial cells then leading to blood infection. In this review, we summarized recent results from different fields of fungal cell biology that are instrumental in understanding hyphal growth. This includes research o

Cryptococcus neoformans is the top-ranked W.H.O. fungal priority pathogen, but tools for generating conditional mutations are limited. Auxin-inducible degron systems permit rapid and effective cellular depletion of a tagged protein of interest upon adding a small molecule. These tools are invaluable, particularly for studying essential genes, which may play important roles in pathogen biology. AID2 is one such system that improves on previous strategies. This system achieves greater sensitivity and specificity through an auxin derivative, 5-Ph-IAA, alongside an OsTIR1F74G mutant. We adapted th

The yeast-to-filament transition is important for dimorphic fungi to adapt to different environmental conditions. Transcriptional activators that activate downstream target genes in response to environmental stimulations are particularly important for the control of this morphological transition. However, only a limited number of key activators of filamentation have been identified. Here, we report YlAaf1 and YlAaf2, a novel pair of SANT domain-containing transcription factors that promote filamentation in the dimorphic yeast Yarrowia lipolytica. We show that both deletion and co-overexpressio

In eukaryotes, the glycosylphosphatidylinositol (GPI) modification of many glycoproteins on the cell surface is highly conserved. The lipid moieties of GPI-anchored proteins undergo remodelling processes during their maturation. To date, the products of the PER1, GUP1 and CWH43 genes of the yeast Saccharomyces cerevisiae have been shown to be involved in the lipid remodelling. Here, we focus on the putative GPI remodelling pathway in the methylotrophic yeast Ogataea minuta. We found that the O. minuta homologues of PER1, GUP1 and CWH43 are functionally compatible with those of S. cerevisiae. D

Virulence of Candida is linked with its ability to form biofilms. Once established, biofilm infections are nearly impossible to eradicate. Biofilm cells live immersed in a self-produced matrix, a blend of extracellular biopolymers, many of which are uncharacterized. In this study, we provide a comprehensive analysis of the matrix manufactured by Candida albicans both in vitro and in a clinical niche animal model. We further explore the function of matrix components, including the impact on drug resistance. We uncovered components from each of the macromolecular classes (55% protein, 25% carboh

Pneumocystis pneumonia is the most common serious opportunistic infection in patients with HIV/AIDS. Furthermore, Pneumocystis pneumonia is a feared complication of the immunosuppressive drug regimens used to treat autoimmunity, malignancy, and posttransplantation rejection. With an increasing at-risk population, there is a strong need for novel approaches to discover diagnostic and vaccine targets. There are multiple challenges to finding these targets, however. First, Pneumocystis has a largely unannotated genome. To address this, we evaluated each protein encoded within the Pneumocystis gen

INTRODUCTION: The ability to acquire iron and maintain iron homeostasis is crucial for the virulence of the human pathogenic fungus Cryptococcus neoformans. This study investigates the role of Bud32, a core virulence kinase and component of the KEOPS complex, within the iron regulatory network of C. neoformans.
METHODS: We used gene deletion techniques to study the phenotypic effects of BUD32 gene knockout and conducted proteomic and metabolomic analyses to assess changes in protein expression and metabolite levels in the mutant. Additionally, we performed in vivo phosphoproteomics analysis t

The tripeptide glutathione is involved in cellular defense mechanisms for xenobiotics and reactive oxygen species. This study investigated glutathione-dependent mechanisms in the model organism Aspergillus nidulans. A recombinant dimeric protein of A. nidulans glutathione reductase (GR) contained FAD and reduced oxidized glutathione (GSSG) using NADPH as an electron donor. A deletion strain of the GR gene (glrA) accumulated less intracellular reduced glutathione (GSH), indicating that the fungal GR contributes to GSSG reduction in vivo. Growth of the deletion strain of glrA was temperature-sen

Spray-induced gene silencing (SIGS) uses exogenous double-stranded RNA (dsRNA) to suppress pathogen virulence or growth-related genes, offering a promising strategy for controlling grey mould caused by Botrytis cinerea at the gene level. In this study, we investigated the molecular mechanisms underlying the efficient uptake of dsRNAs by B. cinerea, making significant progress towards addressing key gaps in our understanding of SIGS. We identified four B. cinerea genes, BcBMP1, BcSOD1, BcGAR2, and BcVELB, as effective targets for grey mould control by employing an Arabidopsis thaliana SIGS scre

Cell fate decisions in eukaryotes are regulated by interconnected networks of transcription factors (TFs) that drive heritable changes in identity. However, much is unknown about how TFs act together to control cell fate, despite links to cellular dysfunction and disease when TF function is aberrant. Here, we addressed the interplay between TFs that control heritable switching in the diploid fungal pathogen Candida albicans. This species can propagate in two distinct cell states, white and opaque, with epigenetic transitions between states regulated by a core network of eight TFs plus >100 aux

Cryptococcus neoformans is a significant human fungal pathogen, particularly concerning for immunocompromised individuals. Key kinases, Hsl101 and Urk1, are critical for crossing the blood-brain barrier, although the specific mechanisms by which they do so remain undefined. In this study, we systematically investigate the impact of HSL101 or URK1 deletion on the proteomic and metabolomic profiles of C. neoformans. By generating a deletion mutant for HSL101, we observed profound alterations in the expression levels of 366 proteins and 421 metabolites, highlighting significant disruptions in key

Coccidioides is a dimorphic fungal pathogen responsible for the mammalian disease commonly known as Valley fever. While previous studies have characterized transcriptomic changes associated with its life stage transitions, the contribution of small RNAs (sRNAs)-key regulators of virulence in other pathogenic fungi-remains unexplored. Here, we profile sRNA expression across three Coccidioides posadasii morphologies-arthroconidia, mycelia, and spherules-analyzing both intracellular and extracellular RNA fractions. Utilizing RiboMarker sRNA and RNA fragment library preparation, we achieved enhanc

To gain a better understanding of the transcriptional response of Aspergillus fumigatus during invasive pulmonary infection, we used a NanoString nCounter to assess the transcript levels of 467 A. fumigatus genes during growth in the lungs of immunosuppressed mice. These genes included ones known to respond to diverse environmental conditions and those encoding most transcription factors in the A. fumigatus genome. We found that invasive growth in vivo induces a unique transcriptional profile as the organism responds to nutrient limitation and attack by host phagocytes. This in vivo transcript

AmyR, a Zn(II)2Cys6 transcription factor, is a key regulator of carbohydrate-active enzyme production and oligosaccharide and polysaccharide utilization in filamentous fungi, but its roles in asexual development, environmental adaptation, and fruit spoilage remain unknown. This study aimed to characterize its functions using multi-phenotypic and transcriptional analyses in Aspergillus niger, a filamentous fungus widely used in fermentation and food processing, as well as a food spoilage and human pathogenic fungus. Deletion of amyR decreased amylolytic activity by repressing the expression of

Sporocarp formation is part of the reproductive stage in the life cycle of many mycorrhizal macrofungi. Sporocarp formation is accompanied by a transcriptomic switch and profound changes in regulation of the gene families that play crucial roles in the sporocarp initiation and maturation. Since sporocarp growth requires efficient water delivery, in the present study, we investigated changes in transcript abundance of six fungal aquaporin genes that could be cloned from the ectomycorrhizal fungus Laccaria bicolor strain UAMH8232, during the initiation and development of its basidiocarp. Candida albicans is a common human fungal pathogen that is also a commensal of the oral cavity and gastrointestinal tract. C. albicans pathogenesis is linked to its transition from budding yeast to filamentous morphologies including hyphae and pseudohyphae. The centrality of this virulence trait to C. albicans pathobiology has resulted in extensive characterization of a wide range of factors associated with filamentation with a strong focus on transcriptional regulation. The vast majority of these experiments have used in vitro conditions to induce the yeast-to-filament transition. Taking adva

Sex-specific homeodomain (HD) proteins are key regulators of cell identity and sexual development in fungi, typically functioning as heterodimers to govern transcription. In the human fungal pathogens Cryptococcus neoformans and Cryptococcus deneoformans, the HD proteins Sxi1α and Sxi2a (Sex-inducer 1α and 2a) have been characterized as interacting components that play critical roles in sexual development during α x a sexual reproduction. α cells are the predominant mating type in natural populations of Cryptococcus, and unisexual (same-sex) mating can also occur in certain genetic backgrounds

Fusarium wilt of banana, caused by Fusarium oxysporum f. sp. cubense (Foc), is one of the most damaging plant diseases known. Foc race 1 (R1) decimated the Gros Michel-based banana (Musa acuminata) trade, and now Foc tropical race 4 (TR4) threatens global production of its replacement, the Cavendish banana. Here population genomics revealed that all Cavendish banana-infecting Foc race 4 strains share an evolutionary origin distinct from that of R1 strains. Although TR4 lacks accessory chromosomes, it contains accessory genes at the ends of some core chromosomes that are enriched for virulence
