Laboratory colonisation and genetic bottlenecks in the tsetse fly Glossina pallidipes.

BACKGROUND: The IAEA colony is the only one available for mass rearing of Glossina pallidipes, a vector of human and animal African trypanosomiasis in eastern Africa. This colony is the source for Sterile Insect Technique (SIT) programs in East Africa. The source population of this colony is unclear and its genetic diversity has not previously been evaluated and compared to field populations. METHODOLOGY/PRINCIPAL FINDINGS: We examined the genetic variation within and between the IAEA colony and its potential source populations in north Zimbabwe and the Kenya/Uganda border at 9 microsatellite

[Genetic analysis of Yersinia pestis strains isolated in China].

OBJECTIVE: The strains of Yersinia pestis isolated in different period and different natural foci in China were analyzed. METHODS: Traditional and molecular biological methods were used. Rhamnose fermentation, rRNA gene copy number, nitrite reduction, and the glycerol fermentation were important characters for typing, and pulse field gel electrophoresis (PFGE) and random amplified polymorphic DNA (RAPD) profile could reflect the genetic distance between the strains. RESULTS: The strains could be divided into 15 genetic types by those 6 characters with each of them covered an isolated geograp

Beyond two alleles: multiallelic genotypic selection and its estimation from time-series data.

Genetic diversity is central to evolutionary change, with both natural selection and random genetic drift depending on variation within a population. An individual in a diploid population carries two alleles per locus, yet the population as a whole can harbour many alleles, giving rise to a rich spectrum of homozygous and heterozygous genotypes. Such multiallelic variation is common at biologically and medically important loci such as the major histocompatibility complex, the ABO blood group system, and genes underlying monogenic diseases. However, much of population genetic theory and data an

Biosurfactants direct the evolutionary dynamics of surface-associated microbial systems.

Surface-associated microbial systems experience spatial constraints that can intensify stochastic lineage loss through genetic drift and alter how natural selection acts on competing lineages. A key feature of these systems is that steep resource gradients typically confine growth to a narrow band of cells located at the biomass periphery. This restricts the effective population size and constrains lineage interactions, thereby accelerating stochastic lineage exclusion. Here, we demonstrate that biosurfactants, which are secreted by many bacteria, can modulate the evolutionary dynamics of thes

Vaccines and viral antigenic diversity.

Antigenic diversity among ribonucleic acid (RNA) viruses occurs as a result of rapid mutation during replication and recombination/reassortment between genetic material of related strains during co-infections. Variants which have a selective advantage in terms of ability to spread or to avoid host immunity become established within populations. Examples of antigenically diverse viruses include influenza, foot and mouth disease (FMD) and bluetongue (BT). Effective vaccination against such viruses requires surveillance programmes to monitor circulating serotypes and their evolution to ensure tha

Evolution and Antigenic Advancement of N2 Neuraminidase of Swine Influenza A Viruses Circulating in the United States following Two Separate Introductions from Human Seasonal Viruses.

Two separate introductions of human seasonal N2 neuraminidase genes were sustained in U.S. swine since 1998 (N2-98) and 2002 (N2-02). Herein, we characterized the antigenic evolution of the N2 of swine influenza A virus (IAV) across 2 decades following each introduction. The N2-98 and N2-02 expanded in genetic diversity, with two statistically supported monophyletic clades within each lineage. To assess antigenic drift in swine N2 following the human-to-swine spillover events, we generated a panel of swine N2 antisera against representative N2 and quantified the antigenic distance between wild

Why are some genetic diseases common? Distinguishing selection from other processes by molecular analysis of globin gene variants.

Various processes (selection, mutation, migration and genetic drift) are known to determine the frequency of genetic disease in human populations, but so far it has proved almost impossible to decide to what extent each is responsible for the presence of a particular genetic disease. The techniques of gene and haplotype analysis offer new hope in addressing this issue, and we review relevant studies of three haemoglobinopathies: sickle cell anaemia, and alpha and beta thalassaemia. We show how for each disease it is possible to recognize a pattern of regionally specific mutations, found in ass

The key role of patrilineal inheritance in shaping the genetic variation of Dagestan highlanders.

The Caucasus region is a complex cultural and ethnic mosaic, comprising populations that speak Caucasian, Indo-European and Altaic languages. Isolated mountain villages (auls) in Dagestan still preserve high level of genetic and cultural diversity and have patriarchal societies with a long history of isolation. The aim of this study was to understand the genetic history of five Dagestan highland auls with distinct ethnic affiliation (Avars, Chechens-Akkins, Kubachians, Laks, Tabasarans) using markers on the male-specific region of the Y chromosome. The groups analyzed here are all Muslims but

Microevolutionary divergence and plasticity of laryngeal shape in the house mouse (Mus musculus).

The morphology of the larynx is essential for vocalization, respiration, and airway protection, yet the sources of phenotypic variation within species are not well understood. This study investigated whether genetic background exerts a stronger influence than environmental factors on laryngeal morphology in two house mouse strains. Using geometric morphometrics, we analyzed the size and shape of 79 laryngeal specimens and examined the effects of body size, genetics, obesity, exercise, and social housing. Our results demonstrate that genetic background significantly shapes laryngeal structure.

The evolutionary dynamics of transposable elements in eukaryote genomes.

Transposable elements (TEs) are ubiquitous components of eukaryotic genomes. They have considerably affected their size, structure and function. The sequencing of a multitude of eukaryote genomes has revealed some striking differences in the abundance and diversity of TEs among eukaryotes. Protists, plants, insects and vertebrates contain species with large numbers of TEs and species with small numbers, as well as species with diverse repertoires of TEs and species with a limited diversity of TEs. There is no apparent relationship between the complexity of organisms and their TE profile. The p

[Prevalence and distribution of genetic diseases in Quebec: impact of the past on the present].

The prevalence and distribution of genetic diseases in the province of Quebec has been influenced by its population history. The current French Canadian population stems from 8,500 pioneers who left France for Nouvelle-France between 1608 and 1759. After the English conquest of Nouvelle-France in 1759, the French Canadian population remained mostly genetically isolated, for linguistic, cultural, and religious reasons. The migration of a small number of French individuals to Nouvelle-France created a founder effect. Subsequent migrations inland have created smaller regional founder effects. The

Parapatric genetic divergence among deep evolutionary lineages in the Mediterranean green crab, Carcinus aestuarii (Brachyura, Portunoidea, Carcinidae), accounts for a sharp phylogeographic break in the Eastern Mediterranean.

BACKGROUND: Recently, population genetic studies of Mediterranean marine species highlighted patterns of genetic divergence and phylogeographic breaks, due to the interplay between impacts of Pleistocene climate shifts and contemporary hydrographical barriers. These factors markedly shaped the distribution of marine organisms and their genetic makeup. The present study is part of an ongoing effort to understand the phylogeography and evolutionary history of the highly dispersive Mediterranean green crab, Carcinus aestuarii (Nardo, 1847), across the Mediterranean Sea. Recently, marked divergenc

Loss of genetic diversity in Culex quinquefasciatus targeted by a lymphatic filariasis vector control program in Recife, Brazil.

Recife is one of the largest cities in north-eastern Brazil and is endemic for lymphatic filariasis transmitted by Culex quinquefasciatus. Since 2003 a control program has targeted mosquito larvae by elimination of breeding sites and bimonthly application of Bacillus sphaericus. To assess the impact of this program on the local vector population we monitored the genetic diversity and differentiation of Cx. quinquefasciatus using microsatellites and a B. sphaericus-resistance associated mutation (cqm1(REC)) over a 3-year period. We detected a significant but gradual decline in allelic diversity

Ashkenazi Jewish genetic disorders.

The frequency of several genes responsible for 'single-gene' disorders and disease predispositions is higher among Ashkenazi Jews than among Sephardi Jews and non-Jews. The disparity is most likely the result of founder effect and genetic drift, rather than heterozygote advantage. The more common Mendelian Ashkenazi Jewish genetic disorders are summarized, and examples of variable expressivity and penetrance, inconsistent genotype-phenotype correlation, and potential modifiers are presented. The importance of genetic counseling in both the pre- and post-test phases of population screening is e

Neandertal roots: Cranial and chronological evidence from Sima de los Huesos.

Seventeen Middle Pleistocene crania from the Sima de los Huesos site (Atapuerca, Spain) are analyzed, including seven new specimens. This sample makes it possible to thoroughly characterize a Middle Pleistocene hominin paleodeme and to address hypotheses about the origin and evolution of the Neandertals. Using a variety of techniques, the hominin-bearing layer could be reassigned to a period around 430,000 years ago. The sample shows a consistent morphological pattern with derived Neandertal features present in the face and anterior vault, many of which are related to the masticatory apparatus

Comparing raccoon major histocompatibility complex diversity in native and introduced ranges: Evidence for the importance of functional immune diversity for adaptation and survival in novel environments.

The adaptive potential of invasive species is related to the genetic diversity of the invader, which is influenced by genetic drift and natural selection. Typically, the genetic diversity of invaders is studied with neutral genetic markers; however, the expectation of reduced diversity has not been consistently supported by empirical studies. Here, we describe and interpret genetic diversity at both neutral microsatellite loci and the immune-related MHC-DRB locus of native and invasive populations of raccoon to better understand of how drift and selection impact patterns of genetic diversity d

Endogenous aldehydes: A driver of clonal hematopoiesis from within?

Detoxification of endogenous aldehydes is critical for preserving genomic integrity in hematopoietic stem cells. In this issue, Kamimae-Lanning et al.1 show that excess formaldehyde can drive clonal hematopoiesis through attrition of blood-forming progenitors, accelerating neutral drift in the absence of known genetic drivers of positive selection.

Age-driven developmental drift in the pathogenesis of idiopathic pulmonary fibrosis.

Idiopathic pulmonary fibrosis (IPF) is a progressive and usually lethal disease of unknown aetiology. A growing body of evidence supports that IPF represents an epithelial-driven process characterised by aberrant epithelial cell behaviour, fibroblast/myofibroblast activation and excessive accumulation of extracellular matrix with the subsequent destruction of the lung architecture. The mechanisms involved in the abnormal hyper-activation of the epithelium are unclear, but we propose that recapitulation of pathways and processes critical to embryological development associated with a tissue spe

Regional differences in craniofacial diversity and the population history of Jomon Japan.

The people associated with the Jomon culture, the Neolithic inhabitants of Japan, are one of the key groups in the population history of East Asia, because they retain many archaic characters that may be traced back to Eurasian Upper Palaeolithic hunter-gatherers. In this study, the regional diversity of the Jomon skeletal series was estimated by applying the R-matrix method to 34 craniofacial measurements. The patterns of intraregional variation indicate little effect on the genetic structure of the Jomon from long-term gene flow stemming from an outside source. The regional diversities were

Distribution and medical impact of loss-of-function variants in the Finnish founder population.

Exome sequencing studies in complex diseases are challenged by the allelic heterogeneity, large number and modest effect sizes of associated variants on disease risk and the presence of large numbers of neutral variants, even in phenotypically relevant genes. Isolated populations with recent bottlenecks offer advantages for studying rare variants in complex diseases as they have deleterious variants that are present at higher frequencies as well as a substantial reduction in rare neutral variation. To explore the potential of the Finnish founder population for studying low-frequency (0.5-5%) v
