Advances in Genetic Polymorphism Research in Rosacea: Mechanisms and Clinical Implications.

Rosacea is a chronic inflammatory skin disease primarily affecting the central region of the face. Typical manifestations include erythema, papules, or pustules on the cheeks, glabella, chin, and nose, with some patients experiencing ocular involvement. The pathogenesis of this disease is influenced by both polygenic inheritance and environmental factors, with abnormalities in innate immunity and neurovascular regulation playing a leading role. Recent genetic studies have identified several key genes, including Human Leukocyte Antigen (HLA), Toll-Like Receptor 2 (TLR2), Interleukin-17 (IL-17),

Blood donor biobank as a resource in personalised biomedical genetic research.

Health questionnaires and donation criteria result in accumulation of highly selected individuals in a blood donor population. To understand better the usefulness of a blood donor-based biobank in personalised disease-associated genetic studies, and for possible personalised blood donation policies, we evaluated the occurrence and distributions of common and rare disease-associated genetic variants in Finnish Blood Service Biobank. We analysed among 31,880 blood donors the occurrence and geographical distribution of (i) 53 rare Finnish-enriched disease-associated variants, (ii) mutations assum

Hypertrophic Cardiomyopathy: Diverse Pathophysiology Revealed by Genetic Research, Toward Future Therapy.

Hypertrophic cardiomyopathy (HCM) is an intractable disease that causes heart failure mainly due to unexplained severe cardiac hypertrophy and diastolic dysfunction. HCM, which occurs in 0.2% of the general population, is the most common cause of sudden cardiac death in young people. HCM has been studied extensively using molecular genetic approaches. Genes encoding cardiac β-myosin heavy chain, cardiac myosin-binding protein C, and troponin complex, which were originally identified as causative genes, were subsequently reported to be frequently implicated in HCM. Indeed, HCM has been consider

CRISPR/Cas9 in soybean research: a measured leap toward genetic refinement.

Soybean [Glycine max (Linn.) Merr] is a globally important source of plant-based protein and edible oil, widely cultivated in India as a major oilseed crop. However, conventional breeding approaches for soybean are laborious, constrained by the genomic complexity of soybean, and face persistent challenges in genetic transformation. Clustered regularly interspaced short palindromic repeats (CRISPR)/Cas-based genome editing has emerged as a precise and efficient tool for rapid trait improvement in soybean. While many existing reviews of soybean have been published, this literature review address

[Research progress on mitochondrial genetic mechanisms underlying the onset of migraine].

Migraine is a common primary headache disorder in clinical practice, and its recurrent nature severely impairs patients' quality of life. Data from the 2021 Global Burden of Disease Study indicate that migraine imposes a heavy burden on patients, their families, and society. In recent years, the association between mitochondrial dysfunction and migraine has attracted increasing attention. Studies have demonstrated that during migraine attacks, skeletal muscle cells exhibit significant mitochondrial morphological changes (such as mitochondrial swelling and cristae disruption), while platelets s

Expanding the methodological repertoire: Integrating multimodal approaches in genetic counseling research.

This paper highlights the importance of methodological diversity in the field of genetic counseling (GC) and demonstrates how alternative multimodal approaches can serve as valuable additions to the GC research toolkit. We use examples from four separate qualitative GC projects, in the United Kingdom and South Africa, to highlight how different multimodal methods such as photo elicitation (PE) and graphic elicitation (GE), body mapping (BM), and conversation analysis (CA) can be incorporated into GC research and how they can produce novel ways for understanding practice, services, and patient

Recent progress in oculopharyngodistal myopathy research from clinical and genetic viewpoints.

Oculopharyngodistal myopathy (OPDM) is a rare muscular disorder characterized by ocular symptoms, pharyngeal symptoms, facial weakness, and distal predominant limb muscle weakness. The cause of the disease was unknown for a long time. Recently, however, it has been reported that expansions of CGG or CCG repeats in LRP12, LOC642361/NUTM2B-AS1, GIPC1, NOTCH2NLC, RILPL1, and ABCD3 are the causes of the disease. Cases sometimes present with neurological symptoms, and the clinical spectrum of diseases caused by expansions of CGG or CCG repeats has been proposed to be called FNOP-spectrum disorder a

Genetic diagnosis in acromegaly and gigantism: From research to clinical practice.

It is usually considered that only 5% of all pituitary neuroendocrine tumours are due to inheritable causes. Since this estimate was reported, however, multiple genetic defects driving syndromic and nonsyndromic somatotrophinomas have been unveiled. This heterogeneous genetic background results in overlapping phenotypes of GH excess. Genetic tests should be part of the approach to patients with acromegaly and gigantism because they can refine the clinical diagnoses, opening the possibility to tailor the clinical conduct to each patient. Even more, genetic testing and clinical screening of at-r

Genetic mouse models in opioid research: current status and future directions.

Synthetic and semi-synthetic opioids are prescribed for the management of severe pain conditions, but their long-term use is often leading to physical dependence and addiction disorders. Understanding the complex neurobiology of the opioid system in preclinical models will be essential for the development of safe and efficacious analgesics. With rising numbers of synthetic opioid users and overdose cases, a better understanding of the neuroanatomical and cellular pathways associated with physical dependence and addiction is expected to guide treatment approaches for opioid use disorders. In th

Research Note: High genetic diversity of infectious bronchitis virus from Mexico.

The avian infectious bronchitis virus (IBV) is a highly mutable coronavirus that causes an acute and highly contagious disease responsible for economic losses to the poultry industry worldwide. Preventing and controlling bronchitis disease is difficulted by the numerous IBV circulating types with limited antigenic cross-protection that hamper the prevention and control by heterologous vaccines. The coding region of the variable spike S1 receptor-attachment domain is used to classify IBV in 7 genotypes (GI-GVII) comprising 35 viral lineages (1-35). Knowledge of the circulating IBV types causing

Past, present, and future of genetic research in borderline personality disorder.

Borderline Personality Disorder (BPD) is a major mental illness with a lifetime prevalence of approximately 1-3%, characterized by a persistent pattern of instability in relationships, mood, impulse regulation, and sense of self. This results in impulsive self-damaging behavior, high suicide rates, and severe functional impairment. BPD has a complex, multifactorial etiology, resulting from an interaction among genetic and environmental substrates, and has moderate to high heritability based on twin and family studies. However, our understanding of the genetic architecture of BPD is very limite

Genetic Analysis of Dent's Disease and Functional Research of CLCN5 Mutations.

Dent's disease is an X-linked inherited renal disease. Patients with Dent's disease often carry mutations in genes encoding the Cl-/H+ exchanger ClC-5 and/or inositol polyphosphate 5-phosphatase (OCRL1). However, the mutations involved and the biochemical effects of these mutations are not fully understood. To characterize genetic changes in Dent's disease patients, in this study, samples from nine Chinese patients were subjected to genetic analysis. Among the nine patients, six were classified as having Dent-1 disease, one had Dent-2 disease, and two could not be classified. Expression of ClC

[Genetic basis of arterial hypertension: new findings in molecular research methods].

Arterial hypertension is thought to derive from an interaction between genetic and environmental factors. Primary hypertension is currently deemed to be under complete polygenic control but highly heterogeneous both from the genetic and the pathophysiologic standpoint. In the last few years a widespread application of molecular medicine techniques to epidemiological genetic studies has led to unprecedented progress in unravelling the genetical basis of human hypertension. The current research strategies along with the most recent results in this field are reviewed.

Perspectives and ethical considerations for return of genetics and genomics research results: a qualitative study of genomics researchers in Uganda.

BACKGROUND: The return of genetics and genomics research results has been a subject of ongoing global debate. Such feedback is ethically desirable to update participants on research findings particularly those deemed clinically significant. Although there is limited literature, debate continues in African on what constitutes appropriate practice regarding the return of results for genetics and genomics research. This study explored perspectives and ethical considerations of Ugandan genomics researchers regarding the return of genetics and genomics research results. METHODS: This was a qualita

Unlocking the potential of genetic research in pulmonary arterial hypertension: Insights from clinicians, researchers, and study team.

Genetic research and testing are increasingly important for understanding and treating pulmonary arterial hypertension. We aimed to explore how attitudes toward genetic research among clinical and research teams impacted the engagement in genetic research and the integration of genetic insights into clinical practice. We conducted 53 semistructured interviews and focus groups with patients, clinicians, and researchers from nine UK Pulmonary Hypertension centers, who had genetic research experience. Transcripts were thematically coded using inductive analysis. In this study, we focus on the res
