Dyslexia is a very common developmental disorder. The aetiology of this complex disorder must in large part still be clarified. Dyslexia segregates in families and the risk for a sibling to become dyslexic is 3.5-fold increased. Different phenotypic dimensions are correlated with dyslexia. These are mainly phonological awareness, phonological decoding, orthographic coding, auditory short-term memory, and rapid naming. The correlated dimensions segregate in families and were found to be heritable. The heritability of word reading lies between 50% and 60%, and that of spelling between 50% and 70

Osteoporosis is a systemic skeletal disease comprising rarefaction of bone structure and loss of bone mass, finally leading to increased fracture risk. As a part of its multifactorial aetiology, twin and family studies have demonstrated an important genetic component of osteoporosis regarding many parameters of bone properties e. g. bone mineral density, with a heredity of 60-80 %. Whole genome screens, linkage analysis and candidate gene research have contributed to our current knowledge about genetic loci in osteoporosis. Genotyping of collagen alpha I, lactose intolerance or estrogen recept

Osteoporosis is a common disease with a strong genetic component. In recent years, some progress has been made in understanding the genetic basis of osteoporosis. Genetic factors contribute to osteoporosis by influencing not only bone mineral density but also bone size, bone quality, and bone turnover. Meta-analysis has been used to define the role of several candidate genes in osteoporosis. Some quantitative trait loci that regulate bone mass identified by linkage studies in humans and experimental animals have been replicated in multiple populations. Genes that cause monogenic bone diseases

Prostate cancer (PC) is the most frequent malignant tumor among men over 50 years old. Its incidence varies according to countries and ethnic groups. Known risk factors are race and positive family history of the disease. Familial aggregation (at least 2 cases in the family) is observed in about 20% of cases and a hereditary form of PC, compatible with a Mendelian inheritance, in 5%. This proportion increases with younger age at diagnosis. Eight putative loci are already identified for hereditary PC but undoubtedly, others will be found in forthcoming studies. The genetic heterogeneity observe

As is becoming increasingly apparent, both atopy and asthma (however they are clinically defined) are a diverse group of related conditions, which are similarly disparate in their origins. Despite this, genetic factors are clearly operational. Speaking to their relatedness, linkages have been found between similar chromosomal sites for both atopy and asthma. Speaking to their divergence, there are also reports of the same phenotypes displaying linkage to different chromosomal areas. The likely explanation for this is that both the atopy and asthma phenotypes are polygenetic, requiring that mul

Many neurodegenerative diseases are exceedingly complex disorders (Fig. 6). In the past decade, we have made tremendous advances in our understanding [figure: see text] of the genetic basis of these disorders. One common characteristic of these disorders is the existence of rare families in which a given disease is inherited as a Mendelian trait. In this article, we have reviewed the genetics of several common neurodegenerative disorders that are associated with cognitive disturbances and for which causative genes have been identified. Further genetic analysis should clarify the roles of known

Osteoporosis is a common multifactorial disorder of reduced bone mass. The disorder in its most common form is generalized, affecting the elderly, both sexes, and all racial groups. Multiple environmental factors are involved in the pathogenesis. Genes also play a major role as reflected by heritability of many components of bone strength. Quantitative phenotypes in bone strength in the normal population do not conform to a monogenetic mode of inheritance. The common form of osteoporosis is generally considered to be a polygenic disorder arising from the interaction of common polymorphic allel

In summary, autism genetics has moved from a time of identification of heritability and determination of risk of "lesser variants" or the "broader phenotype" in relatives to a phase where some cases of autism have a definite basis such as maternally inherited duplications of 15q11-q13, identification of mutations causing AS, Rett syndrome, and FRAXA. The first phase of genome-wide screens has not revealed definitive linkage, but as samples are enlarged and meta-analyses performed, the strongest linkage findings are likely to yield susceptibility variants once fine mapping proceeds. To date, at least 12 types of primary dystonia can be distinguished on a genetic basis. A 3-bp deletion in the DYT1 gene causes early onset, generalized torsion dystonia (TD), and mutations in the GTP cyclohydrolase I and the tyrosine hydroxylase genes result in dopa-responsive dystonia (DYT5). A missense change in the D2 dopamine receptor in one large family (DYT11) has recently been implicated in myoclonus-dystonia. Furthermore, seven other loci for dystonia genes have been mapped to chromosomal regions, including a locus for a mixed dystonia phenotype (DYT6), one form of focal dystonia (DYT

Population genetic studies of vectors are essential for (i) the determination of their taxonomic status and consequently the definition of their vectorial role in the transmission of pathogenic agents; (ii) the evaluation of the species genetic variability and the estimation of their capacities of adaptation to selection pressure; (iii) an estimation of gene flow among populations in order to evaluate their degree of isolation and gene circulation, especially resistance genes. Among the malaria vectors taken as examples on three continents, Africa, South-East Asia and Latin America, the large

Otosclerosis is an early-middle adult life genetic disease affecting bone remodelling in the ear. Current knowledge of otosclerosis as an inherited disease dates to the mid-19th century, and we report here an attempt to understand the genetics of otosclerosis and detect its heterogeneity. The analysis was conducted on 151 otosclerotic families. The results of our study indicate that while heredity plays an important role in the manifestation of the disease a substantial portion of otosclerotic cases could arise due to non-genetic causes.

There are several arguments suggesting that vitiligo is a genetically dependent disease. Certain animals have comparable pigment disturbances (Smyth chickens, C57BL/6J-mi(vit)/mi(vit) mice and B light mice). Genetic studies have demonstrated that certain genes are crucial for the development of vitiligo. Familial cases and cases reported in twins are further arguments. There is a high risk for both children and siblings of a subject with vitiligo. Several HLA studies have been reported. Most show sporadic association between vitiligo and certain HLA groups, but there is no clear and regular as

Molecular studies of the major histocompatibility complex region, its genes, and its products are progressing in a number of different directions, and this progress is becoming of increasing interest not only to immunologists but also to geneticists and clinicians. Here I select only certain aspects of this field, stressing the biological importance of a few recent developments. This review concentrates on two topics of clinical importance and where important recent progress is reported: the "revolution" of HLA genotyping for histocompatibility matching in transplantation and the elucidation o

Genetic resistance to disease and its use in domestic livestock usually ranks last, if at all, amongst preferred disease control measures - usually preceded by measures such as chemotherapy, vector control and vaccination. Thus, interest in genetic resistance is often a reflection of dissatisfaction with other control strategies, and the current emphasis on trypanotolerant cattle in Africa is just such a case. Eighty years of tsetse fly eradication programmes have had little impact on tsetse distribution, although recent research with odour baited targets impregnated with insecticide brings ho

The incidence of genetic abnormalities in our population is rather high. About one out of 13 liveborn children will sooner or later suffer from a fully or partially genetically determined malformation or disease. With the use of numerical models ist has been attempted to predict the dysgenic effects of modern medicine and medical genetics in the next few generations of the Swiss population. Modern medical treatment will result foremost in an increase of multifactorial diseases and malformations. However, the impact on society will probably be smaller than the actual burden of chronic genetic d
