The radioprotectant nano-genistein enhances radiotherapy efficacy of lung tumors in mice.

BACKGROUND: Radiotherapy for non-small cell lung cancer (NSCLC) can be dose-limiting due to treatment-related toxicities. Genistein has been shown to be a robust radioprotective agent in preclinical models. A novel genistein oral nanosuspension formulation (nano-genistein) has demonstrated efficacy in mitigating radiation-induced lung damage in preclinical animal models. However, while those studies have confirmed that nano-genistein can protect normal lung tissue from radiation-induced toxicities, no studies have assessed the effect of nano-genistein on lung tumors. Here, we evaluated the imp

Orobol, 3'-hydroxy-genistein, suppresses the development and regrowth of cutaneous SCC.

Chronic solar ultraviolet exposure is a major risk factor for cutaneous squamous cell carcinoma (cSCC), which is the second most common type of skin cancer. Our previous data showed that total protein and phosphorylation levels of T-LAK cell-originated protein kinase (TOPK) were enhanced in solar-simulated light (SSL)-induced skin carcinogenesis and overexpressed in actinic keratosis (AK) and cSCC human skin tissues compared to those in matched normal skin. Thus, targeting TOPK activity could be a helpful approach for treating cSCC. Our data showed that orobol directly binds to TOPK in an ATP-

Effects of Chronic Dietary Exposure to Phytoestrogen Genistein on Uterine Morphology in Mice.

Genistein is naturally occurring in plants and binds to estrogen receptors. Humans are mainly exposed through diet, but the use of supplements is increasing as genistein is claimed to promote health and alleviate menopausal symptoms. We analyzed diverse uterine features in adult mice chronically fed genistein for different times. The luminal epithelium height was increased in females treated with 500 and 1000 ppm at PND 95, and the width of the outer myometrium was increased in females treated with 1000 ppm at PND 65 compared to that in controls. An increase in proliferation was noted in the i

Protective effects of Genistein on the cognitive deficits induced by chronic sleep deprivation.

Sleep deprivation has been widely reported to cause cognitive dysfunction, and elevation in oxidative stress and inflammation in the body, including the brain, have been suggested as the main factors. Genistein (GE) is an isoflavone widely present in leguminous plants, and it was found to exert a wide spectrum of biological activities, including antioxidant, anti-inflammatory, hepatoprotective, neuroprotective, and antimetastatic effects. In this study, the protective effect of GE on chronic sleep deprivation (CSD)-induced cognitive dysfunction was investigated. The mice were subjected to the

Genistein as Potential Therapeutic Candidate for Menopausal Symptoms and Other Related Diseases.

Plant-derived compounds have recently attracted greater interest in the field of new therapeutic agent development. These compounds have been widely screened for their pharmacological effects. Polyphenols, such as soy-derived isoflavones, also called phytoestrogens, have been extensively studied due to their ability to inhibit carcinogenesis. These compounds are chemically similar to 17β-estradiol, and mimic the binding of estrogens to its receptors, exerting estrogenic effects in target organs. Genistein is an isoflavone derived from soy-rich products and accounts for about 60% of total isofl

Genistein Binding to Copper(II)-Solvent Dependence and Effects on Radical Scavenging.

Genistein, but not daidzein, binds to copper(II) with a 1:2 stoichiometry in ethanol and with a 1:1 stoichiometry in methanol, indicating chelation by the 5-phenol and the 4-keto group of the isoflavonoid as demonstrated by the Jobs method and UV-visible absorption spectroscopy. In ethanol, the stability constants had the value 1.12 × 1011 L²∙mol-2 for the 1:2 complex and in methanol 6.0 × 10⁵ L∙mol-1 for the 1:1 complex at 25 °C. Binding was not detected in water, as confirmed by an upper limit for the 1:1 stability constant of K = 5 mol-1 L as calculated from the difference in solvation free

Equol but not genistein improves early metaphyseal fracture healing in osteoporotic rats.

Healing of predominantly metaphyseal fractures in postmenopausal osteoporosis is delayed and comparatively poor. Hormone replacement therapy could improve fracture healing, but, because of its potential side effects, natural alternatives are more appealing. The aim of this study was to determine if the soy metabolite equol and the native isoflavone genistein, in comparison to 17beta-estradiol, improve metaphyseal fracture healing in ovariectomy-induced osteoporotic bone of the rat. Forty-eight 12-week-old female rats developed severe osteoporosis ten weeks after ovariectomy. After metaphyseal

The role of early life genistein exposures in modifying breast cancer risk.

Review of the existing literature suggests that consumption of soy foods or an exposure to a soy isoflavone genistein during childhood and adolescence in women, and before puberty onset in animals, reduces later mammary cancer risk. In animal studies, an exposure that is limited to the fetal period or adult life does not appear to have the same protective effect. A meta-analysis of human studies indicates a modest reduction in pre- and postmenopausal risk when dietary intakes are assessed during adult life. These findings concur with emerging evidence indicating that timing may be vitally impo

Genistein is neuroprotective in murine models of familial amyotrophic lateral sclerosis and stroke.

Amyotrophic lateral sclerosis (ALS), whether sporadic or familial (FALS), is a progressive, fatal neurodegenerative disorder involving the motor neurons of the cortex, brain stem, and spinal cord. In some studies, the male/female ratio of ALS patients was as high as 2 to 1. In FALS mice, disease onset and mortality were earlier among males than among females. This sexual dimorphism was due to estrogen, as treatment with genistein, a phytoestrogen, eliminated the observed sexual dimorphism in FALS mice. Genistein treatment also protected against oxygen singlet-induced cerebral damage in vivo. H

Dietary genistein supplementation enhances reproductive and bone health in late-laying hens.

Aging in laying hens leads to progressive declines in production performance and egg quality, primarily due to deteriorating ovarian function and bone health. Genistein, a natural isoflavone with anti-inflammatory and antioxidant properties, shows potential as a dietary intervention to alleviate age-related physiological decline. This study evaluated the effects of dietary genistein supplementation on ovarian and skeletal health in aging laying hens. A total of 180 Hy-Line Brown laying hens (60 weeks old) were randomly assigned to either a control group (CON) receiving a basal diet or a genist

Genistein-3'-sodium sulfonate suppresses NLRP3-mediated cell pyroptosis after cerebral ischemia.

Cerebral ischemia-induced pyroptosis contributes to the dissemination of neuroinflammation, and Nod-like receptor protein-3 (NLRP3) inflammasome plays a key role in this process. Previous studies have indicated that Genistein-3'-sodiumsulfonate (GSS) can inhibit neuroinflammation caused by cerebral ischemia, exert cerebroprotective effects, but its specific mechanism has not been comprehensively understood. The aim of this study was to explore the effect of GSS on ischemic stroke-induced cell pyroptosis. SD rats were randomly assigned to Sham group, transient middle cerebral artery occlusion (

Genistein resists castration-resistant prostate cancer by inhibiting the androgen receptor pathway.

Prostate cancer is a prevalent malignancy, often diagnosed at advanced stages and associated with poor prognosis. Genistein, a soybean isoflavone, has demonstrated antitumor effects against castration-resistant prostate cancer in vivo. Our results indicate that genistein effectively downregulates the expression of key enzymes involved in the de novo synthesis pathway, namely AKR1C3, SRD5A2, CYP11A1, and 3βHSD, thus blocking androgen synthesis in CRPC cells. Additionally, genistein inhibits the activation and nuclear translocation of the AR. Collectively, these findings support the potential of

Genistein induces degradation of mutant huntingtin in fibroblasts from Huntington's disease patients.

Mutations in the HTT gene, consisting of expansion of CAG triplets, cause the Huntington's disease (HD), one of the major neurodegenerative disorders. Formation of aggregates of mutant huntingtin (mHTT, the product of the mutant HTT gene) leads to cellular dysfunctions, and subsequent neurodegeneration which manifest clinically as motor abnormalities and cognitive deficits. We recently used immortalized HEK-293 cells expressing the 1st exon of the mutant HTT gene as a cellular model of HD, and showed that the stimulation of autophagy by genistein corrected the mutant phenotype. However, effect

Genistein-induced LKB1-AMPK activation inhibits senescence of VSMC through autophagy induction.

Genistein, the primary isoflavone from soy products, enhances antioxidant enzyme activities and inhibits tyrosine kinase. However, the mechanisms underlying genistein-induced autophagy are not yet completely understood. Autophagy refers to a regulated cellular process for the lysosomal-dependent turnover of organelles and proteins. During starvation or nutrient deficiency, autophagy provides an endogenous mechanism for prolonging survival. Here, we investigated whether genistein exerts autophagic effects through the activation of LKB1-AMPK signaling in VSMCs. Genistein dose- and time-dependent

Genistein and the immune system: experimental evidence, key challenges, and future perspectives.

Genistein (GEN), a soy-derived isoflavonoid and phytoestrogen, exhibits diverse biological activities against allergic and hypersensitivity reactions, autoimmune disorders, infectious diseases, and cancer. Its immunomodulatory properties are crucial to these effects; however, clinical translation remains limited due to the complexity and pronounced context dependence of its immune actions. This review critically summarizes experimental evidence on GEN-mediated immunomodulation across innate and adaptive immune systems, including its effects on cytokine regulation and key molecular signaling pa

A Soy-Derived Isoflavone Genistein Inhibits the Life Cycle of PRRSV In Vitro.

Porcine reproductive and respiratory syndrome virus (PRRSV) has become a global health threat for swine. Because of the limited effectiveness of commercial vaccine strategies and the absence of specific antiviral chemical drugs for PRRS, it is urgent to identify new strategies for preventing and controlling PRRSV infections. Genistein, a soy-derived isoflavone, has shown promise in reducing viremia in PRRSV-infected pigs. However, its antiviral molecular mechanism remains unclear. This study investigates the inhibitory effects and mechanism of genistein on PRRSV infection in vitro. Our results

QbD-driven Formulation Development and Evaluation of Genistein Nanoparticles for Prostate Cancer.

BACKGROUND: Genistein (GEN) shows significant anticancer potential, particularly against prostate cancer. However, its clinical application is limited by poor water solubility, rapid metabolism and excretion, low bioavailability, and lack of targeted delivery to cancer cells, hindering its effectiveness as a chemopreventive or therapeutic agent. OBJECTIVE: In this study, poly-ε-caprolactone (PCL) nanoparticles incorporating polyvinyl alcohol (PVA) as a stabilizer were engineered to encapsulate genistein (GEN) effectively. Utilizing a Quality by Design (QbD) methodology, the development and op

Daidzein and Genistein: Natural Phytoestrogens with Potential Applications in Hormone Replacement Therapy.

Menopause is characterized by a decline in estrogen levels, leading to symptoms such as vasomotor instability, osteoporosis, and increased cardiovascular and cognitive risk. Hormone replacement therapy (HRT) remains the gold standard for managing menopausal symptoms; however, concerns regarding its long-term safety, including elevated risks of cancer and cardiovascular events, have prompted interest in alternative therapies. Phytoestrogens, particularly the isoflavones daidzein and genistein, are plant-derived compounds structurally similar to 17β-estradiol (E2) and capable of binding estrogen

The effect of genistein on IGF-1, PlGF, sFLT-1 and fetoplacental development.

The mechanisms by which genistein, a phytoestrogen, affects fetoplacental development adversely are still poorly understood. It is reported that genistein ingestion modulates thyroid functions, leptin hormone, C-reactive protein, and thyroxin kinase activities. In this study, we evaluated changes in serum and placental insulin-like growth factor-I (IGF-1), placental growth factor (PIGF), and soluble fms-like tyrosine kinase-1 (sFLT-1) in pregnant rats exposed to genistein using ELISA. According to the treatments, Rats were divided into control, 2 mg genistein, and 4 mg genistein groups. Genist

Influence of Genistein on Hepatic Lipid Metabolism in an In Vitro Model of Hepatic Steatosis.

Nonalcoholic fatty liver disease (NAFLD) is among the leading causes of end-stage liver disease. The impaired hepatic lipid metabolism in NAFLD is exhibited by dysregulated PPARα and SREBP-1c signaling pathways, which are central transcription factors associated with lipid degradation and de novo lipogenesis. Despite the growing prevalence of this disease, current pharmacological treatment options are unsatisfactory. Genistein, a soy isoflavone, has beneficial effects on lipid metabolism and may be a candidate for NAFLD treatment. In an in vitro model of hepatic steatosis, primary human hepato
