The first chromosome-level genome assembly of a green lacewing Chrysopa pallens and its implication for biological control.

Many lacewing species (Insecta: Neuroptera) are important predators of pests with great potential in biological control. So far, there is no chromosome-level published genome available for Neuroptera. Here we report a high-quality chromosome-level reference genome for a green lacewing species Chrysopa pallens (Neuroptera: Chrysopidae), which is one of the most important insect natural enemies used in pest biocontrol. The genome was sequenced using a combination of PacBio and Hi-C technologies and assembled into seven chromosomes with a total size of 517.21 Mb, occupying 96.07% of the genome se

Attenuated virulence and genomic reductive evolution in the entomopathogenic bacterial symbiont species, Xenorhabdus poinarii.

Bacteria of the genus Xenorhabdus are symbionts of soil entomopathogenic nematodes of the genus Steinernema. This symbiotic association constitutes an insecticidal complex active against a wide range of insect pests. Unlike other Xenorhabdus species, Xenorhabdus poinarii is avirulent when injected into insects in the absence of its nematode host. We sequenced the genome of the X. poinarii strain G6 and the closely related but virulent X. doucetiae strain FRM16. G6 had a smaller genome (500-700 kb smaller) than virulent Xenorhabdus strains and lacked genes encoding potential virulence factors (

Host-independent metagenomics reveal gut bacteria contribution to Delia antiqua growth by vitamin B6 provision.

Insect guts host a diverse and abundant array of microorganisms. These microbes improve host fitness by extensively involving in a range of crucial physiological processes, which have mainly been revealed by high-throughput sequencing, particularly metagenomics. However, it is almost impossible to make an accurate and complete distinction between the genetic functions of microbial symbionts and insect hosts without host genome data. By comparing metagenomic data from gut germ-free and nonaxenic larvae, we accurately identified the data belonging to the gut microbiome of the onion maggot Delia

Evolutionary Genetics of Insect Mimicry and Masquerade.

As classic examples of adaptive evolution, insect mimicry and masquerade have been studied extensively for more than a century. Both phenomena protect insects from predators and are widely observed in nature. Previous research on phenotypic, genetic, phylogenetic, and behavioral aspects has laid a solid foundation for in-depth investigations. Recent advances in genomic data resources, analytical methodologies, and gene-editing tools have greatly facilitated the elucidation of the genetic and developmental mechanisms underlying insect mimicry and masquerade. This review provides a comprehensive

Cellular remodeling of ovarian follicular epithelial cells transmits an obligate nutritional endosymbiont in a scale insect.

Many insects show complex associations with vertically transmitted endosymbionts. Here, we describe unique cellular remodeling of the oocyte's follicular epithelial cells into endosymbiont-bearing tubular structures in the ensign scale insect Insignorthezia insignis (Hemiptera: Ortheziidae). Vitellogenic oocytes develop a bulge-like distension at the posterior pole. Here, follicular epithelial cells undergo extensive cellular reorganization, remodeling their actin cytoskeleton and plasma membrane to produce cellular protrusions. These tubular protrusions, which are densely packed with the endo

Genetic bases and robustness of the toxicological response to spinosad and lambda-cyhalothrin in Drosophila melanogaster.

Insecticides are valuable and widely used tools for the control of insect pests. However, we know very little about the genes and processes involved in the key steps of the poisoning sequence. Besides, the excessive and indiscriminate use of insecticides is generating mechanisms of resistance in various insect pests. To improve our understanding of insecticide toxicity, we need a powerful model organism to help describe the processes underlying insecticide poisoning and the use of genomic tools to identify and analyze the genetic basis of the toxicological response of the insecticides. In this

Tachinid Endoparasitoid Flies do not Rely on Domesticated Viruses, Unlike Multiple Hymenopteran Lineages.

Viral domestication, the co-option of integrated viral genes for host functions, has been repeatedly documented in endoparasitoid Hymenoptera. Whether this phenomenon extends to other parasitoid insects remains to be assessed. Here, we tested this hypothesis in tachinid flies, the largest group of non-hymenopteran parasitoids. To this end, we investigated patterns of viral endogenization in 52 genomes, including 37 newly sequenced species. Similarly to hymenopteran endoparasitoids, tachinid genomes were found to display numerous endogenous viral elements (EVEs), primarily derived from insect-i

The genome of Phlebotomus chinensis, the primary vector of visceral leishmaniasis in China: insights from chromosome-level assembly and comparative analysis.

BACKGROUND: Phlebotomus chinensis is the primary vector of visceral leishmaniasis (VL) in China. However, the lack of a high-quality genome assembly for this species has limited research on its biology, vector-pathogen interactions, and evolutionary adaptations. To address this critical gap, the first chromosome-level genome assembly of Ph. chinensis was constructed. METHODS: Nanopore long-read sequencing served as the primary method, complemented by Illumina short-read sequencing for base-level error correction and Hi-C mapping for chromosomal anchoring and chromosome-level scaffolding. Geno

The genome of the stable fly, Stomoxys calcitrans, reveals potential mechanisms underlying reproduction, host interactions, and novel targets for pest control.

BACKGROUND: The stable fly, Stomoxys calcitrans, is a major blood-feeding pest of livestock that has near worldwide distribution, causing an annual cost of over $2 billion for control and product loss in the USA alone. Control of these flies has been limited to increased sanitary management practices and insecticide application for suppressing larval stages. Few genetic and molecular resources are available to help in developing novel methods for controlling stable flies. RESULTS: This study examines stable fly biology by utilizing a combination of high-quality genome sequencing and RNA-Seq a

A Chromosome-scale Assembly for the Genome of Southern Corn Rootworm, Diabrotica Undecimpunctata.

Diabrotica undecimpunctata ssp. howardi, the southern corn rootworm or eastern 12-spotted cucumber beetle, is a generalist insect herbivore that causes damage and yield loss to several crops in North America, including maize. Unresolved phylogenetic relationships within and among D. undecimpunctata subspecies are impacting current quarantine policies. We report the chromosome-level haploid genome assembly, icDiaUnde3, constructed using HiFi and Hi-C read data from a single male D. undecimpunctata collected and identified as subspecies howardi based on geographic location and morphology. Agua Salud alphavirus defines a novel lineage of insect-specific alphaviruses discovered in the New World.

The genus Alphavirus harbours mostly insect-transmitted viruses that cause severe disease in humans, livestock and wildlife. Thus far, only three alphaviruses with a host range restricted to insects have been found in mosquitoes from the Old World, namely Eilat virus (EILV), Taï Forest alphavirus (TALV) and Mwinilunga alphavirus (MWAV). In this study, we found a novel alphavirus in one Culex declarator mosquito sampled in Panama. The virus was isolated in C6/36 mosquito cells, and full genome sequencing revealed an 11 468 nt long genome with maximum pairwise nucleotide identity of 62.7 % to Si

Proteomic footprints of a member of Glossinavirus (Hytrosaviridae): an expeditious approach to virus control strategies in tsetse factories.

The Glossinavirus (Glossina pallidipes salivary gland hypertrophy virus (GpSGHV)) is a rod-shaped enveloped insect virus containing a 190,032 bp-long, circular dsDNA genome. The virus is pathogenic for the tsetse fly Glossina pallidipes and has been associated with the collapse of selected mass-reared colonies. Maintenance of productive fly colonies is critical to tsetse and trypanosomiasis eradication in sub-Saharan Africa using the Sterile Insect Technique. Proteomics, an approach to define the expressed protein complement of a genome, was used to further our understanding of the protein com

The highly rearranged mitochondrial genome of the plague thrips, Thrips imaginis (Insecta: Thysanoptera): convergence of two novel gene boundaries and an extraordinary arrangement of rRNA genes.

To help understand the mechanisms of gene rearrangement in the mitochondrial (mt) genomes of hemipteroid insects, we sequenced the mt genome of the plague thrips, Thrips imaginis (Thysanoptera). This genome is circular, 15,407 bp long, and has many unusual features, including (1) rRNA genes inverted and distant from one another, (2) an extra gene for tRNA-Ser, (3) a tRNA-Val lacking a D-arm, (4) two pseudo-tRNA genes, (5) duplicate control regions, and (6) translocations and/or inversions of 24 of the 37 genes. The mechanism of rRNA gene transcription in T. imaginis may be different from that

The complete mitochondrial genome of Reptalus artemisiae (Becker, 1865) (Hemiptera: Cixiidae).

Reptalus artemisiae (Becker, 1865) is a vector of economically important plant pathogens. To date, no complete mitochondrial genome has been available for Pentastirini. We present the first complete mitogenome of R. artemisiae, sequenced using ONT MinION long reads. The circular genome is 15,476 bp long, comprises 13 protein-coding, two rRNA, 22 tRNA genes, and an AT-rich putative noncoding control region. Gene content and organization are consistent with the ancestral insect mitochondrial genome. This dataset provides a crucial reference for phylogenetic studies for both Reptalus and Pentasti

Hemipteran insects experience recurrent and synergistic molecular evolution adapting to diet shift.

BACKGROUND: Hemiptera is an insect order with extremely high physiological and behavioral diversity. Feeding traits have switched and reversed multiple times, but the molecular basis governing this phenotypical change remains unclear. RESULTS: We obtained the high-quality genomes and salivary gland transcriptomes of two essential biocontrol agents, Eocanthecona furcellata and Arma custos (Hemiptera: Heteroptera: Pentatomidae: Asopinae). This subfamily represents a typical insect clade with the reversal of feeding traits from phytophagy to zoophagy. Combined with public data of an additional 3

Complete genome sequence of the entomopathogenic and metabolically versatile soil bacterium Pseudomonas entomophila.

Pseudomonas entomophila is an entomopathogenic bacterium that, upon ingestion, kills Drosophila melanogaster as well as insects from different orders. The complete sequence of the 5.9-Mb genome was determined and compared to the sequenced genomes of four Pseudomonas species. P. entomophila possesses most of the catabolic genes of the closely related strain P. putida KT2440, revealing its metabolically versatile properties and its soil lifestyle. Several features that probably contribute to its entomopathogenic properties were disclosed. Unexpectedly for an animal pathogen, P. entomophila is de

Phylogenomics confirms Palaeoptera monophyly and illuminates the origin of winged insects.

The origin and early radiation of winged insects (Pterygota) represent one of the most transformative events in terrestrial evolution, yet phylogenetic relationships among their basalmost lineages (Ephemeroptera, Odonata and Neoptera) continue to be a central debate within insect systematics. Here, we present the most taxonomically comprehensive phylogenomic analysis to date, leveraging genomic and transcriptomic data from 101 species, including 16 sequenced ephemeropteran genomes. Comparative genomics reveal signatures of ecological specialization in the striped ambrosia beetle Trypodendron lineatum.

BACKGROUND: Beetles (Coleoptera) display exceptional dietary diversity and occupy a wide range of ecological niches, often involving close associations with plants and microbes. Ambrosia beetles (Curculionidae; Scolytinae and Platypodinae) exemplify ecological specialization by cultivating mutualistic fungi within galleries excavated in their host trees' xylem, with the fungi serving as their main food source. The striped ambrosia beetle Trypodendron lineatum is a pest of conifers, relying on its nutritional mutualist Phialophoropsis ferruginea for survival. This fungiculture-based lifestyle p
