Reductive dechlorination performed by organohalide-respiring bacteria (OHRB) enables the complete detoxification of certain emerging groundwater pollutants such as perchloroethene (PCE). Environmental samples from a contaminated site incubated in a lab-scale microcosm (MC) study enable documentation of such reductive dechlorination processes. As compound-specific isotope analysis is used to monitor PCE degradation processes, nucleic acid analysis-like 16S-rDNA analysis-can be used to determine the key OHRB that are present. This study applied both methods to laboratory MCs prepared from enviro

The dechlorination rate in a flow-through porous matrix can be described by the species specific dechlorination rate observed in a liquid batch unless mass transport limitations prevail. This hypothesis was examined by comparing dechlorination rates in liquid batch with that in column experiments at various flow rates (3-9-12 cm day(-1)). Columns were loaded with an inoculated sand and eluted with a medium containing 1mM trichloroethene (TCE) for 247 days. Dechlorination in the column treatments increased with decreasing flow rate, illustrating the effect of the longer residence time. Zeroth o

In situ chemical oxidation (ISCO) followed by a bioremediation step is increasingly being considered as an effective biphasic technology. Information on the impact of chemical oxidants on organohalide respiring bacteria (OHRB), however, is largely lacking. Therefore, we used quantitative PCR (qPCR) to monitor the abundance of OHRB (Dehalococcoides mccartyi, Dehalobacter, Geobacter, and Desulfitobacterium) and reductive dehalogenase genes (rdh; tceA, vcrA, and bvcA) at a field location contaminated with chlorinated solvents prior to and following treatment with sodium persulfate. This study demonstrated the utility in correlating performance and community structure of a trichloroethene (TCE)-dechlorinating microbial consortium; specifically dechlorinators, fermenters, homoacetogens, and methanogens. Two complementary approaches were applied: predicting trends in the microbial community structure based on an electron balance analysis and experimentally assessing the community structure via pyrosequencing and quantitative polymerase chain reaction (qPCR). Fill-and-draw reactors inoculated with the DehaloR^2 consortium were operated at five TCE-pulsing rates between 14 an

Dissolved organic matter (DOM), particularly its thiol moieties, kinetically controls Hg(II) speciation, thereby regulating the pool of bioavailable Hg(II). However, how variations in thiol abundance within DOM influence Hg(II)-DOM reaction kinetics and Hg(II) bioavailability remains unclear. Here, we investigated the Hg(II)-DOM reaction kinetics driven by DOM thiol abundance under dark/light conditions, as well as their effects on microbial Hg(II) uptake and methylation. This was achieved by employing an Escherichia coli-based whole-cell biosensor and a Geobacter sulfurreducens PCA methylatio

The production of methylmercury by some bacteria is a key first step in the accumulation and biomagnification of this toxic substance in aquatic food webs, a major human health concern. By direct measurement of cellular Hg(II) uptake in model iron and sulfate reducing bacteria, we have observed that specific trace metals, such as Zn(II) and Cd(II), inhibit uptake and methylation in these organisms, whereas other metals, such as Ni(II), Co(II), or Fe(II), do not. The inhibition of Hg(II) methylation by Zn(II) was competitive in nature and related to the concentration of inorganically complexed

Cr(VI) contamination is widely recognized as one of the major environmental hazards. To address the problem of remediation of soil Cr(VI) contamination and utilization of waste peanut shells, this study comprehensively investigated the effects of peanut shell-derived biochar loaded with root exudates of hyperaccumulator Leersia hexandra Swartz on Cr(VI) reduction and microbial community succession in soil. This study confirmed that root exudate-loaded peanut shell biochar reduced soil pH while simultaneously increasing DOC, sulfide, and Fe(II) concentrations, thereby facilitating the reduction

The iron redox cycle can enhance anammox in treating low-strength ammonia wastewater. However, maintaining an effective iron redox cycle and suppressing nitrite-oxidizing bacteria in a one-stage partial nitritation and anammox (PN/A) process poses challenges during long-term aeration. We proposed a novel and simple strategy to achieve an efficient iron redox cycle in an iron-mediated anoxic-microaerobic (A/O) process by controlling organic matter (OM) at medium-strength levels (30-110 mg COD/L) in microaerobic granular sludge (MGS)-dominated reactor. The developed A/O process consistently achi

Microbial fuel cells (MFCs) have been extensively studied as toxicity sensors. MFCs have potential as toxicity sensors because when their anodes are exposed to toxic substances or the concentration of the target is changed, the voltage generated by certain microorganisms with active electrical activity attached to the anode changes. However, the underlying reasons behind this phenomenon have not been deeply explored. Therefore, the activity of anodic microorganisms during voltage drops in MFCs under formaldehyde (FA) stress was studied. The composition of the microbial community structure with

Naturally conductive protein nanowires have inspired efforts to engineer electrical conductivity into synthetic fibrous proteins for the development of bioelectronic materials and devices. A comprehensive analysis of charge transport in these systems requires a combination of various measurement methods, instruments and electrode designs. Measurements under direct current (DC) typically focus on charge transport without distinguishing between charged species, requiring alternating current (AC) and electrochemical methods to probe additional phenomena. Here, ionic and electronic charge transpor

Iron reduction and mercury methylation by dissimilatory iron-reducing bacteria (DIRB), Geobacter sulfurreducens and Shewanella oneidensis, were studied, and the relationship of mercury methylation coupled to iron reduction was determined. The ability of both bacteria for reducing iron was tested, and Fe(III) reduction occurred with the highest rate when ferric oxyhydroxide was used as a terminal electron acceptor. G. sulfurreducens had proven to mediate the production of methylmercury (MeHg), and a notable increase of MeHg following the addition of inorganic Hg was observed. When the initial c

Organic thiols (R-SH) are known to react and form complexes with some toxic soft metals such as mercury (Hg) in both biotic and abiotic systems. However, a clear understanding of these interactions is currently limited because quantifying thiols in environmental matrices is difficult due to their low abundance, susceptibility to oxidation, and measurement interference by non-thiol compounds in samples. Here, we report a fluorescence-labeling method using a maleimide containing probe, ThioGlo-1 (TG-1), to determine total thiols directly on bacterial cells and natural organic matter (NOM). We sy

Forty-six candidate phenol/benzoate degrading-iron reducing bacteria were isolated from long-term irrigated tropical paddy soils by enrichment procedures. Pure cultures and some prepared mixed cultures were examined for ferric oxide reduction and phenol/benzoate degradation. All the isolates were iron reducers, but only 56.5% could couple iron reduction to phenol and/or benzoate degradation, as evidenced by depletion of phenol and benzoate after one week incubation. Analysis of degradative capability using Biolog MT plates revealed that most of them could degrade other aromatic compounds such

Microbial production of the neurotoxin methylmercury (MeHg) is a significant health and environmental concern, as it can bioaccumulate and biomagnify in the food web. A chalkophore or a copper-binding compound, termed methanobactin (MB), has been shown to form strong complexes with mercury [as Hg(II)] and also enables some methanotrophs to degrade MeHg. It is unknown, however, if Hg(II) binding with MB can also impede Hg(II) methylation by other microbes. Contrary to expectations, MB produced by the methanotroph Methylosinus trichosporium OB3b (OB3b-MB) enhanced the rate and efficiency of Hg(I

Mercury (Hg) is a pervasive environmental pollutant and poses serious health concerns as inorganic Hg(II) can be converted to the neurotoxin methylmercury (MeHg), which bioaccumulates and biomagnifies in food webs. Phytoplankton, representing the base of aquatic food webs, can take up Hg(II) and influence MeHg production, but currently little is known about how and to what extent phytoplankton may impact Hg(II) methylation by itself or by methylating bacteria it harbors. This study investigated whether some species of phytoplankton could produce MeHg and how the live or dead phytoplankton cell

The release of geogenic arsenic into groundwater, driven by reductive dissolution of Fe(III)/As(V) oxide phases, poses a severe health risk to millions in South and Southeast Asia. However, the microbes and electron donors responsible for the reductive dissolution remain unclear, due to complex a(biotic) interactions in sediments (traditionally used in microcosm incubation studies). In this study, indigenous microbial communities were sampled from arsenic-prone aquifers in Kandal Province, Cambodia, by filtering groundwater through sands coated with Fe(III)/As(V) minerals. This provided a stre

Polarity reversal is one of the effective strategies to rapidly start up denitrifying BESs,but the long-term performances of the denitrifying BESs operated under polarity reversal receive little attention. This study investigated the effects of periodic polarity reversal (PPR) and polarity reversal once only (PRO) on the long-term performances of denitrifying BESs. Repeatable oxidative and reductive currents were observed in the BESs obtained by PPR (PPR-BESs). The peak reductive currents of the PPR-BESs reached 0.95 A/m2, and nitrate was mainly removed by dissimilatory nitrate reduction to am

Bioelectrochemical methanation offers a promising avenue for carbon resource recovery. However, the direct biocathodic conversion of toxic carbon monoxide (CO) into methane, as well as the relevant interfacial extracellular electron transfer pathways and microbial community succession remain largely unexplored. Here, we systematically investigated the CO methanation process within a mixed-culture microbial electrolysis cell. Decreasing the cathodic potential from -0.90 to -1.05 V triggered a significant enhancement in methanogenic activity, boosting the cumulative methane yield from 0.142 to 1

Sustainable recycling strategies are urgently needed to address the rapidly growing accumulation of spent lithium-ion batteries. Current strategies for the first step in recycling, the solubilization of lithium and other metals, are energy intensive and/or generate toxic waste. This study critically evaluated the option of 'microbial electro-extraction' (MEE), a process in which electroactive microbes supply electrons to reduce the nickel, cobalt, and manganese in spent lithium-ion battery powder, releasing the soluble reduced ions of these metals along with soluble Li+.
