A proteomic signature of healthspan.

The focus of aging research has shifted from increasing lifespan to enhancing healthspan to reduce the time spent living with disability. Despite significant efforts to develop biomarkers of aging, few studies have focused on biomarkers of healthspan. We developed a proteomics-based signature of healthspan [healthspan proteomic score (HPS)] using proteomic data from the Olink Explore 3072 assay in the UK Biobank Pharma Proteomics Project (53,018 individuals and 2,920 proteins). A lower HPS was associated with higher mortality risk and several age-related conditions, such as chronic obstructive

Aging and the hypothalamic-pituitary-thyroid axis.

The world's population is increasingly aging, this noted particularly in the Western world where there are ever greater numbers of centenarians and those over 85 years. Given the immense importance of the thyroid gland for optimal health and the fact that morphological and functional changes in the hypothalamic-pituitary-thyroid (HPT) axis take place as a natural adaptation to the aging process, a clear distinction must be made in older individuals between these and the onset of disease. However, this is problematic since, frequently, subtle differences exist between them, making diagnosis a c

Quantitative lipidomics reveals Lipid perturbation of plasma apoptotic extracellular vesicles during physiological aging.

INTRODUCTION: Aging is closely implicated in the pathogenesis of chronic diseases, degenerative conditions, and mortality, making it a core research focus in geroscience. As direct secretory products of apoptotic cells, apoptotic bodies can directly reflect the intensity of apoptotic activity and the degree of systemic and cellular aging. Given their close correlation with senescence progression, apoptotic bodies hold great potential as reliable and sensitive biomarkers for evaluating aging status. However, apoptotic bodies as effective indicators of aging remain to be explored. METHODS: We c

Sex-stratified immune patterns across the canine lifespan and short-term responses to geroprotective interventions.

Canine models are invaluable for translational geroscience, but natural sex-associated immune aging remains under investigated. Here, we characterize the age-associated hematologic and serum cytokine profiles in a controlled cross-sectional cohort of 80 intact laboratory Beagles spanning 1 to 11 years of age. Canine immune aging is heterogeneous and non-linear. While several absolute leukocyte counts declined with age before rising again in geriatric dogs, serum cytokines exhibited distinct, sex-stratified patterns. Although more cytokines varied significantly with age in males than in females

mTOR signaling in aging: from causality to geroprotective interventions and hallmark-level outcomes.

The mechanistic target of rapamycin (mTOR) pathway is an important integrator of processes involved in aging and longevity, coordinating nutrient sensing, metabolic adaptation, and cellular stress responses. This review presents a three-section framework in which mTOR functions as a dynamic signaling hub coordinating multiple biological processes underlying the aging process. Evidence from genetic, experimental, and translational studies supports a causal role for mTOR signaling in lifespan regulation in model organisms, whereas human data remain predominantly associative but biologically cons

Behavioral and neuronal biomarkers of aging and disease: Integration with physical performance and function.

Aging is a heterogeneous process characterized by marked variability in biological, cognitive, and functional trajectories among individuals of the same chronological age. Consequently, there is greater interest in biomarkers capable of identifying early deviations from healthy aging and predicting adverse outcomes. This review provides an updated overview of neuronal and behavioral biomarkers associated with aging and disease and discusses their integration with physical performance and functional outcomes in older adults. Neuronal biomarkers, including neuroimaging, blood-based, and neurovas

[Autoimmunity and ageing : How immune ageing shapes disease phenotype, infection risk and treatment safety in rheumatic diseases].

BACKGROUND: Immune ageing alters tolerance mechanisms, the chronic low-grade inflammatory environment and host infection defence. This is clinically relevant in rheumatology because the number of older patients with inflammatory rheumatic diseases is increasing and age modifies disease phenotype, vaccination responses and treatment safety. OBJECTIVE: To summarize the interfaces between ageing, autoimmunity and chronic inflammation and to derive consequences for prevention and treatment. MATERIAL AND METHODS: Narrative review of recent reviews and key studies on immunosenescence, geroscience,

Immunosenescence and the Geriatric Giants: Molecular Insights into Aging and Healthspan.

Aging is associated with complex immune dysfunction that contributes to the onset and progression of the "geriatric giants", including frailty, sarcopenia, cognitive decline, falls, and incontinence. Central to these conditions is immunosenescence, marked by thymic involution, the loss of naïve T cells, T-cell exhaustion, impaired B-cell class switch recombination, and increased autoreactivity. Concurrently, innate immunity deteriorates due to macrophage, neutrophil, and NK cell dysfunction, while chronic low-grade inflammation-or "inflammaging"-amplifies systemic decline. Key molecular pathwa

Mitochondrial DNA Deletion Mutations: A Molecular Cause of Age-Induced Skeletal Muscle Fiber Dysfunction and Fiber Death Contributing to Sarcopenia.

This chapter describes a molecular basis for age-induced muscle fiber loss involving the mammalian mitochondrial genome (mtDNA). Early studies of human mitochondrial myopathies, which display many phenotypes associated with muscle aging, led to the search for and subsequent discovery of similar genetic and histopathological changes in aging skeletal muscle. A diverse spectrum of mtDNA deletion mutations increase in abundance with age and clonally accumulate to high abundance within individual cells. Deletion accumulation results in a focal loss of electron transport and oxidative phosphorylati

Aging of the eye: Lessons from cataracts and age-related macular degeneration.

Aging is the greatest risk factor for chronic human diseases, including many eye diseases. Geroscience aims to understand the effects of the aging process on these diseases, including the genetic, molecular, and cellular mechanisms that underlie the increased risk of disease over the lifetime. Understanding of the aging eye increases general knowledge of the cellular physiology impacted by aging processes at various biological extremes. Two major diseases, age-related cataract and age-related macular degeneration (AMD) are caused by dysfunction of the lens and retina, respectively. Lens transp

Aging and neuropathic pain: Mitochondria-to-glia cascade, system mechanisms, and therapeutic strategies.

Neuropathic pain (NP) creates a severe pathological condition that primarily affects elderly people because of their accumulated neurobiological changes that make them more susceptible to persistent pain. The aging process leads to multiple mechanisms that combine neurodegeneration with immunosenescence and mitochondrial dysfunction with impaired autophagy and glial priming, and ion channel dysregulation to create a nociceptive environment. This review examines how mitochondrial breakdown and dysfunctional autophagy, and ion channel disturbances with glial cell activation form a interconnected

Beyond risk factors: A capacity framework for cancer survivorship research.

Cancer survivorship research has identified numerous biological, behavioral, psychosocial, health care, and structural factors that influence recovery. However, these factors are typically studied as separate determinants rather than interacting influences. This commentary proposes available survivorship capacity as a unifying framework that explains how these diverse determinants collectively shape recovery and survivorship outcomes. Concepts from geroscience, health care delivery, rehabilitation, occupational therapy, and human factors science were synthesized to develop a conceptual framewo

Aging is not a disease: an evolutionary and comparative biological reappraisal.

The question of whether aging should be classified as a disease has gained prominence in geroscience, fueled by advances in molecular biology and the aspiration to develop interventions that mitigate age-associated functional decline. However, evolutionary models describe aging as an emergent consequence of declining selection gradients and life-history trade-offs rather than as a deviation from species-typical function. Comparative data across taxa reveal substantial heterogeneity in aging trajectories, challenging the assumption of a uniform pathological pattern. At the molecular level, proc

The Preservation of Functional Reserve: A Control Systems Framework for Human Aging.

The dominant clinical manifestation of aging is not mortality but the progressive depletion of functional reserve-the surplus physiological capacity separating independent function from disability. Existing geroscience frameworks describe molecular aging in detail yet give the point-of-care clinician little basis for prioritizing targets. We reframe functional aging as a control systems problem in which aging is modeled as progressive instability within a coupled, bidirectional mitochondrial-epigenetic axis. That axis is called central in two limited senses only: network connectivity and thera

Sirtuin signaling as therapeutic targets of immunosenescence.

INTRODUCTION: Aging is characterized by immunosenescence, encompassing, immune aging and inflammaging. Because immune competence is a critical determinant of lifespan and healthspan, targeting immunosenescence has become a major objective of geroscience. In this context, sirtuins have emerged as compelling therapeutic candidates. AREAS COVERED: This review examines the role of sirtuin signaling in immunosenescence, focusing on its links with nicotinamide adenine dinucleotide (NAD)+ metabolism, oxidative stress, and inflammatory pathways. Pharmacological strategies aimed at enhancing sirtuin a

New Perspectives on Gastric Inflammaging: Integrating Multi-Omics Mechanisms and Gerotherapeutic Strategies in Chronic Gastritis.

Chronic gastritis (CG) is a highly prevalent, age-associated inflammatory disorder of gastric mucosa and a key precursor of gastric cancer in older adults. Beyond Helicobacter pylori infection and environmental insults, accumulating evidence indicates that chronic, low-grade inflammation coupled with aging biology, "gastric inflammaging", plays a central role in driving mucosal degeneration, atrophy, and malignant transformation. Here, we synthesize current mechanistic and multi-omics evidence to conceptualize CG as a tractable model of organ-specific inflammaging. We first summarize how hallm

Rilmenidine extends lifespan and healthspan in Caenorhabditis elegans via a nischarin I1-imidazoline receptor.

Repurposing drugs capable of extending lifespan and health span has a huge untapped potential in translational geroscience. Here, we searched for known compounds that elicit a similar gene expression signature to caloric restriction and identified rilmenidine, an I1-imidazoline receptor agonist and prescription medication for the treatment of hypertension. We then show that treating Caenorhabditis elegans with rilmenidine at young and older ages increases lifespan. We also demonstrate that the stress-resilience, health span, and lifespan benefits of rilmenidine treatment in C. elegans are medi

Assessing health in aging male and female mice: does cardiac function dictate frailty or physical resilience?

As the global population over age 65 is projected to triple by 2050, understanding the physiological mechanisms of aging and the role of sex as a biological variable is critical. Research conducted on old mice demonstrates that age-related cardiac dysfunction is differentially linked to markers of frailty and resilience in a sex-dependent manner. We assessed post-anesthesia recovery time, wire hanging test, and a deficit accumulation-based frailty index as markers of whole-body frailty and resilience, and correlated these variables with cardiac parameters obtained by echocardiography and Doppl

The emerging role of miRNAs in biological aging and age-related diseases.

Ageing is a complex biological process characterised by the accumulation of molecular and cellular damage leading to functional decline and an increased risk of chronic disease and geriatric syndromes. Despite data showing that lifestyle modifications, such as caloric restriction and exercise, can lead to healthy ageing and greatly reduce the incidence of chronic disorders, no medical therapies exist to delay or prevent these conditions effectively. We also lack tools to effectively track the ageing process in a manner that predicts an individual's risk of chronic disease and assess response t
