Gibberellin-mediated PtrLBD39-PtrHDA6-PtrHCT epigenetic path regulates lignin biosynthesis during tension wood formation in Populus trichocarpa.

Tension wood (TW) is a specialized xylem tissue formed in angiosperm trees to counteract mechanical stresses. Increased phytohormones, particularly gibberellins (GAs), and decreased lignin deposition are two well-established features of TW, yet the molecular mechanisms linking GA signaling to lignin repression remain largely unknown. Here we revealed that TW formation in Populus trichocarpa elevates GA accumulation, which activates the expression of PtrLBD39, the most strongly induced transcription factor and a repressor of lignin biosynthesis in TW development. Inhibition of GA biosynthesis a

[TWO SCHOOL-AGED CASES OF FRUIT GIBBERELLIN-REGULATED PROTEIN ALLERGY WHICH NEED INDIVIDUAL REMOVAL STRATEGY IN SCHOOL LUNCH].

Gibberellin-regulated protein (GRP) is a newly discovered allergen in systemic fruit allergies. The kind of fruits which cause allergy is extensive as GRP is universally included in plants. Two children with GRP allergy were reported. Case 1: A 6-year-old boy experienced anaphylaxis while running after school lunch, which included canned peaches. A skin prick test (SPT) and blood examination suggested that he had peach GRP allergy. Six months and three years later, he experienced a similar episode after eating apple and citrus flesh, respectively. Case 2: An 11-year-old boy experienced anaphyl

Proteome analysis provides new insight into major proteins involved in gibberellin-induced fruit setting in triploid loquat (Eriobotrya japonica).

BACKGROUND: Parthenocarpy can be induced by gibberellin (GA) treatment in plants. The fruits of the loquat exhibit many seeds. GA treatment can induce the development of seedless fruit and increase fruit quality during production. However, the molecular mechanism of fruit setting under GA treatment is still unclear. OBJECTIVE: Our aim was to explore GA-induced parthenocarpy in triploid loquat by proteome analysis to identify the differentially expressed proteins. METHODS: A proteome analysis was performed using TMT protein labeling and LC-MS/MS in triploid loquat. RESULTS: A total of 7290 p

Mediation of Flower Induction by Gibberellin and its Inhibitor Paclobutrazol: mRNA and miRNA Integration Comprises Complex Regulatory Cross-Talk in Apple.

Guaranteeing successful flowering is very important in economic plant species, especially apple (Malus domestica Borkh.), which is difficult to induce to flower. However, the gene expression and networks involved in flowering have not been totally characterized. Here, we employed mRNA and microRNA (miRNA) sequencing to understand the different responses to gibberellin- and its inhibitor paclobutrazol- (PAC) mediated flower induction. Significant opposite cytological and morphological changes were observed in treated terminal buds, which led to a reduced flowering rate under gibberellin and an

Overexpression of cotton GhMKK4 enhances disease susceptibility and affects abscisic acid, gibberellin and hydrogen peroxide signalling in transgenic Nicotiana benthamiana.

Mitogen-activated protein kinase (MAPK) cascades are involved in plant development, stress responses and hormonal signal transduction. MAPK kinases (MAPKKs), as the key nodes in these cascades, link MAPKs and MAPKK kinases (MAPKKKs). In this study, GhMKK4, a novel group C MAPKK gene from cotton (Gossypium hirsutum), was isolated and identified. Its expression can be induced by various stresses and signalling molecules. The overexpression of GhMKK4 in Nicotiana benthamiana enhanced its susceptibility to bacterial and fungal pathogens, but had no significant effects on salt or drought tolerance.

MeSRT1 Interacts with MeNF-YC15 and Negatively Mediates Resistance to Cassava Bacterial Blight via Regulation of Gibberellin Metabolism in Manihot esculenta Crantz.

Cassava (Manihot esculenta Crantz) has significant economic value as a vital source of food, feed, and bio-energy. However, the production of cassava is severely restricted by cassava bacterial blight (CBB). Sirtuins (SRTs) are conserved histone deacetylases that play essential roles in the plant energy metabolism and stress response. However, the family of SRTs has not been identified in cassava. In this study, we isolated and identified three MeSRTs in response to CBB infection and found that they were negative regulators of cassava resistance using a transient gene-silencing assay. Further

Genome-wide identification and integrated analysis of gibberellin oxidase genes associated with branch angle and internode elongation in tea plants (Camellia sinensis).

INTRODUCTION: Gibberellin oxidases (GAoxs) are key enzymes involved in maintaining gibberellin (GA) homeostasis and therefore play important roles in regulating plant architecture. However, the GAox genes associated with branch angle and internode elongation in tea plants (Camellia sinensis) remain largely uncharacterized. METHODS: In this study, 33 CsGAox genes were identified in the genome of the tea cultivar `Longjing 43' and classified into four subfamilies. Comparative genomic analysis across eight Camellia genomes was performed, and phenotypic measurements were integrated with endogenou

GhWIP2, a WIP zinc finger protein, suppresses cell expansion in Gerbera hybrida by mediating crosstalk between gibberellin, abscisic acid, and auxin.

Cell expansion is a key determinant for the final size and shape of plant organ, and is regulated by various phytohormones. Zinc finger proteins (ZFPs) consist of a superfamily involved in multiple aspects of organ morphogenesis. However, little is known about WIP-type ZFP function in phytohormone-mediated organ growth. Using reverse genetics, RNA-seq and phytohormone quantification, we elucidated the role of a new WIP-type ZFP from Gerbera hybrida, GhWIP2, in controlling organ growth via regulation of cell expansion. GhWIP2 localizes to the nucleus and acts as a transcriptional repressor. Con

Genetic variation for lettuce seed thermoinhibition is associated with temperature-sensitive expression of abscisic Acid, gibberellin, and ethylene biosynthesis, metabolism, and response genes.

Lettuce (Lactuca sativa 'Salinas') seeds fail to germinate when imbibed at temperatures above 25 degrees C to 30 degrees C (termed thermoinhibition). However, seeds of an accession of Lactuca serriola (UC96US23) do not exhibit thermoinhibition up to 37 degrees C in the light. Comparative genetics, physiology, and gene expression were analyzed in these genotypes to determine the mechanisms governing the regulation of seed germination by temperature. Germination of the two genotypes was differentially sensitive to abscisic acid (ABA) and gibberellin (GA) at elevated temperatures. Quantitative tr

Small Grain and Dwarf 2, encoding an HD-Zip II family transcription factor, regulates plant development by modulating gibberellin biosynthesis in rice.

Homeodomain leucine zipper (HD-Zip) proteins are transcription factors that regulate plant development. Bioactive gibberellin (GA) is a key endogenous hormone that participates in plant growth. However, the relationship between HD-Zip genes and modulation of GA biosynthesis in rice remains elusive. Here, we identified a rice mutant, designated as small grain and dwarf 2 (sgd2), which had reduced height and grain size compared with the wild type. Cytological observations indicated that the defective phenotype was mainly due to decreased cell length. Map-based cloning and complementation tests d

From dormancy to germination: Transcriptome analysis reveals the potential involvement of heat shock proteins in gibberellin-induced seed dormancy release in Solanum torvum.

Solanum torvum, a superior vegetable grafting rootstock and medicinal Solanaceae plant, exhibits strong seed dormancy, which limits its commercial cultivation. Among various strategies explored to improve the germination rate of S. torvum, exogenous application of gibberellin (GA) has been shown to be effective. In this study, a GA concentration of 2.5 mM was established as the optimal for breaking dormancy in S. torvum seeds. Transcriptome analysis of dry, water-soaked, GA-soaked, and GA-induced germinated seeds was conducted to investigate the molecular mechanism of GA-mediated dormancy rele

Engineering a Gibberellin-Switchable Dual-Use Line Based on Ent-Kaurene Oxidase Gene ZmKO1 Enables Two-Line Hybrid Seed Production in Maize.

Male-sterile systems are pivotal for maize hybrid seed production, yet existing systems are constrained by significant limitations. Here, we identified a novel single-gene-controlled recessive genic male sterility (GMS) mutant, defective in filament elongation 1 (def1), which exhibits non-elongated filaments, unopened glumes, and non-exserted anthers, yet undergoes normal microsporogenesis and produces viable pollen. Map-based cloning revealed that def1 is caused by a loss-of-function mutation in the ZmKO1 gene. Previous studies have identified ZmKO1 and ZmKO2 as the two putative KO-encoding g

Integrated metabolomic-transcriptomic profiling of dwarf Camellia reticulata 'Hentiangao' reveals key gibberellin metabolism genes and multi-hormone crosstalk under GA3 treatment.

BACKGROUND: Camellia reticulata 'Hentiangao', a dwarf camellia cultivar, possesses distinctive ornamental traits and high value for urban landscaping. Dwarfism is an important agronomic and ornamental trait, and gibberellin (GA) plays a central role in regulating stem elongation and plant height. However, the molecular mechanisms underlying its dwarf phenotype and the interactions between stem development and endogenous hormone networks remain unclear, limiting the promotion and targeted breeding of this cultivar. We analyzed shoot tips of C. reticulata 'Hentiangao' treated with 800 mg·L-1 GA3

Expression profiling of the gibberellin 2-oxidase gene CsGA2ox2 (Cucumis sativus L.) reveals stage- and tissue-specific transcript accumulation during fruit development.

BACKGROUND: Gibberellins (GAs) regulate fruit set and expansion, and their endogenous levels are controlled in part through catabolic inactivation. GA2ox enzymes convert bioactive GAs into inactive forms, but the expression of GA2ox genes during cucumber fruit development has not been characterised in a tissue-resolved manner. METHODS: Cucumber (Cucumis sativus L.) '9930' fruit were sampled at 0, 7, 14, 21 and 28 days after pollination. The inner fleshy tissue (pulp) and the outer fruit wall (pericarp) were separated, and CsGA2ox2 transcript abundance was quantified by quantitative real-time

Identification of rice Os4BGlu13 as a β-glucosidase which hydrolyzes gibberellin A4 1-O-β-d-glucosyl ester, in addition to tuberonic acid glucoside and salicylic acid derivative glucosides.

Gibberellin 1-O-β-d-glucose ester hydrolysis activity has been detected in rice seedling extracts, but no enzyme responsible for this activity has ever been purified and identified. Therefore, gibberellin A4 glucosyl ester (GA4-GE) β-d-glucosidase activity was purified from ten-day rice seedling stems and leaves. The family 1 glycoside hydrolase Os4BGlu13 was identified in the final purification fraction. The Os4BGlu13 cDNA was amplified from rice seedlings and expressed as an N-terminal thioredoxin-tagged fusion protein in Escherichia coli. The purified recombinant Os4BGlu13 protein (rOs4BGlu

Unraveling the impact of vine growth habit on stem elongation through photosystem redox modulation and a proposed ELIP-GASA22 regulatory module in Glycinesoja.

Plant architecture results from the integration of developmental regulatory networks and environmental signaling pathways. In soybean, the erect growth habit of cultivated soybean (Glycine max) contrasts markedly with the vine growth habit (VGH) of wild soybean (Glycine soja). However, the genetic and transcriptional mechanisms underlying this divergence in gibberellin-mediated stem elongation remain poorly understood. Therefore, this study aims to investigate the genetic, transcriptional, and hormonal basis underlying the differences in growth habit between Williams 82 (erect growth habit) an

Transcriptome and metabolome profiling provide insights into molecular mechanism of pseudostem elongation in banana.

BACKGROUND: Banana plant height is an important trait for horticultural practices and semi-dwarf cultivars show better resistance to damages by wind and rain. However, the molecular mechanisms controlling the pseudostem height remain poorly understood. Herein, we studied the molecular changes in the pseudostem of a semi-dwarf banana mutant Aifen No. 1 (Musa spp. Pisang Awak sub-group ABB) as compared to its wild-type dwarf cultivar using a combined transcriptome and metabolome approach. RESULTS: A total of 127 differentially expressed genes and 48 differentially accumulated metabolites were d

Integration of Transcriptomic and Metabolomic Profiles Provides Insights into the Influence of Nitrogen on Secondary Metabolism in Fusarium sacchari.

Nitrogen availability might play an essential role in plant diseases by enhancing fungal cell growth and influencing the expression of genes required for successful pathogenesis. Nitrogen availability could modulate secondary metabolic pathways as evidenced by the significant differential expression of several core genes involved in mycotoxin biosynthesis and genes encoding polyketide synthase/nonribosomal peptide synthetases, cytochrome P450 and carbohydrate-active enzymes in Fusarium sacchari, grown on different nitrogen sources. A combined analysis was carried out on the transcript and meta

Cloning and functional mechanism of the dwarf gene gba affecting stem elongation and cellulose biosynthesis in jute (Corchorus olitorius).

Plant height (PH) is an important factor affecting bast fiber yield in jute. Here, we report the mechanism of dwarfism in the 'Guangbaai' (gba) of jute. The mutant gba had shorter internode length and cell length compared to the standard cultivar 'TaiZi 4' (TZ4). Exogenous GA3 treatment indicated that gba is a GA-insensitive dwarf mutant. Quantitative trait locus (QTL) analysis of three PH-related traits via a high-density genetic linkage map according to re-seq showed that a total of 25 QTLs were identified, including 13 QTLs for PH, with phenotypic variation explained ranging from 2.42 to 74

Blue or far-red light supplementation induced pre-hardening in the leaves of the Rht12 wheat dwarfing line: hormonal changes and freezing tolerance.

Reduced height (Rht) genes have revolutionised wheat cultivation, but they can compromise freezing tolerance, and only a few alleles are in use. Thus, evaluating the role of other Rht alleles in stress responses is crucial. Far-red supplementation of white light (W+FR) can induce pre-hardening in cereals at 15°C. However, the relevant effect of blue light enrichment (W+B) is poorly described. This study investigates the influence of W+FR or W+B exposure in young winter wheat leaves of a tall (wild-type, rht12) and a dwarf, gibberellin-deficient (near-isogenic line, Rht12) genotype in cv.
