Effects of Ginkgo biloba constituents on fruit-infesting behavior of codling moth (Cydia pomonella) in apples.

Codling moth, Cydia pomonella (L.), is a cosmopolitan pest of apple, potentially causing severe damage to the fruit. Currently used methods of combating this insect do not warrant full success or are harmful to the environment. The use of plant-derived semiochemicals for manipulation with fruit-infesting behavior is one of the new avenues for controlling this pest. Here, we explore the potential of Ginkgo biloba and its synthetic metabolites for preventing apple feeding and infestation by neonate larvae of C. pomonella. Experiments with crude extracts indicated that deterrent constituents of g

Cytochrome P450 enzymes in the biosynthesis of bioactive diterpenoids from traditional chinese medicinal plants.

Cytochrome P450 enzymes play key catalytic roles in the biosynthesis of diterpenoid natural products from traditional Chinese medicinal plants. Operating through a conserved heme-thiolate catalytic cycle and Compound I-driven oxidation, these enzymes mediate most post-cyclization oxidative tailoring of diterpene scaffolds, including hydroxylation, epoxidation, carbon-skeleton rearrangement, and lactone formation. This review surveys the biosynthetic pathways of seven representative diterpenoids, namely tanshinones, paclitaxel, triptolide, andrographolide, ginkgolides, forskolin, and oridonin,

γ-Aminobutyric acid promotes the inhibition of hair growth induced by chronic restraint stress.

Stress plays a critical role in hair loss, although the underlying mechanisms are largely unknown. γ-aminobutyric acid (GABA) has been reported to be associated with stress; however, whether it affects stress-induced hair growth inhibition is unclear. This study aimed to investigate the potential roles and mechanisms of action of GABA in chronic restraint stress (CRS)-induced hair growth inhibition. We performed RNA-seq analysis and found that differentially expressed genes (DEGs) associated with neuroactive ligand-receptor interaction, including genes related to GABA receptors, significantly

Eupalinolide A attenuates bleomycin-induced pulmonary fibrosis by inhibiting lung fibroblast transition.

This work aimed to investigate the effects of eupalinolide A on bleomycin-induced pulmonary fibrosis and the possible underlying mechanism. We found that in vitro, the antifibrotic activity of eupalinolide A was better than that of eupalinolide G and eupalinolide I and that it was also superior to that of ginkgolide A, ginkgolide B, ginkgolide C, and bilobalide. Eupalinolide A significantly suppressed lung fibroblast transition and decreased the production of extracellular matrix (ECM). Mechanistically, eupalinolide A reduced serum response factor (SRF) levels. Knockdown of SRF inhibited lung

A Cross-sectional In Vitro Study on the Synergistic Neuroprotective Effects of Phytochemicals Ferulic Acid and Ginkgolide B against Amyloid Beta-induced Oxidative Stress and Modulation of Multifunctional Enzyme APE1/Ref-1 in Human Neuroblastoma SH-SY5Y Cells.

The Apurinic/Apyrimidinic endonuclease 1 (APE1/Ref-1) is essential for controlling the oxidative DNA base damage via the Base Excision Repair (BER) pathway. While APE1's N-terminal region is important in redox function (named redox effector factor 1, Ref-1) and protein-protein interactions, its C-terminal region is involved in DNA repair activities. In this work, we employed the Aβ25-35 peptide to produce oxidative stress in order to assess the protective effects of two phytochemicals, ferulic acid (FA) and Ginkgolide B (GB), individually and in combination, with a focus on APE1 in different s

Functional identification of two cytochrome P450 enzymes involved in diterpenoid oxidation in Ginkgo biloba.

Ginkgo biloba is a globally renowned medicinal plant. Ginkgo produces structurally unique diterpene metabolites, of which ginkgolides are the most representative bioactive compounds. Although the ginkgo genome contains a large and diverse family of cytochrome P450s (CYPs), only a few CYPs involved in the early steps of ginkgolide biosynthesis have been identified so far. As a result, the contribution of CYP-mediated oxidation to the diversity of ginkgo's diterpene metabolism remains poorly understood. In this study, we used a constructed diterpenoid-producing yeast chassis for rapid functional

High-Fat Diet-Induced Degradation of the Protein Phosphatase 4 Regulatory Subunit 2 Protein Increases Protein Phosphatase 4 Activity in the Aorta and Mediates Endothelial Dysfunction and Vascular Damage.

BACKGROUND: High fat diet (HFD)-induced increases in free fatty acids in the blood increase the risk of cardiovascular diseases by compromising vascular function. We reported that palmitic acid decreases eNOS (endothelial nitric oxide synthase) phosphorylation at serine 633 by reducing the expression of the PP4R2 (protein phosphatase 4 regulatory subunit 2) protein and activating PP4, leading to endothelial dysfunction. This study aimed to elucidate the underlying mechanisms. METHODS: Studies were performed using C57BL/6J male mice fed an HFD or a normal fat diet for 12 weeks. PP4R2 protein e

Computational identification of Ginkgo biloba phytochemicals as dual inhibitors of herpes simplex virus type 1 glycoproteins gB and gD targeting its entry and Alzheimer's disease pathogenesis.

Herpes simplex virus type 1 (HSV-1) causes severe neurological complications such as herpes simplex encephalitis and Alzheimer's disease (AD). Its entry is mediated by glycoproteins gD and gB, which regulate receptor binding and membrane fusion. Targeting these entry glycoproteins represents a promising therapeutic strategy to limit HSV-1 infection and its contribution to AD pathogenesis. Therefore, this study aimed to identify natural dual inhibitors of HSV-1 glycoproteins gB and gD from Ginkgo biloba using an integrated computational approach involving molecular docking, pharmacokinetic, tox
