Cytostatic effects of structurally different ginsenosides on yeast cells with altered sterol biosynthesis and transport.

Triterpene glycosides are a diverse group of plant secondary metabolites, consisting of a sterol-like aglycon and one or several sugar groups. A number of triterpene glycosides show membranolytic activity, and, therefore, are considered to be promising antimicrobial drugs. However, the interrelation between their structure, biological activities, and target membrane lipid composition remains elusive. Here we studied the antifungal effects of four Panax triterpene glycosides (ginsenosides) with sugar moieties at the C-3 (ginsenosides Rg3, Rh2), C-20 (compound K), and both (ginsenoside F2) posit

Ginsenosides Rb1 and Rg1 Suppress Lipopolysaccharide-Induced Inflammatory Gene Expression in Microglia and Astrocytes, Unlike Rb2.

Neuroinflammation, mediated by activated microglia and astrocytes, contributes to the progression of neurodegenerative diseases. Ginsenosides derived from Panax ginseng possess anti-inflammatory properties. However, the comparative efficacy of structurally distinct ginsenosides and their activity across multiple glial cell types remains incompletely understood. In this study, we systematically compared the effects of ginsenosides Rb1 (protopanaxadiol-type), Rb2 (protopanaxadiol-type), and Rg1 (protopanaxatriol-type) in lipopolysaccharide (LPS)-activated BV2 microglia and C8-D1A astrocytes. Development of liquid chromatography/ultraviolet and evaporating light scattering detector platform for simultaneous quantification of ginsenosides.

Ginsenosides, the primary bioactive components in ginseng, offer various health benefits, including anti-inflammatory, anti-osteoporosis, and neuroprotective effects. For consistent product quality and better understanding of these effects, accurate quantification of ginsenosides is essential. Although liquid chromatography-mass spectrometry (LC/MS) can identify up to 200 ginsenosides, it is costly, complex, and requires specialized personnel. In contrast, liquid chromatography coupled with ultraviolet (UV) or evaporative light scattering detectors (ELSD) is more accessible but typically limit

Ginsenosides Regulates Innate Immunity to Affect Immune Microenvironment of AIH Through Hippo-YAP/TAZ Signaling Pathway.

Autoimmune hepatitis (AIH) is characterized by chronic progressive liver inflammatory, but there is still no safe and effective medicine. Therefore, glucocorticoid remains the top choice for AIH treatment. In previous studies, it has been confirmed that ginsenosides (GSS) can produce glucocorticoid-like effects and therapeutic effects on various autoimmune diseases. However, the mechanism of GSS for AIH remains unclear. As an important part of the innate immune system, bone marrow-derived suppressor cells (MDSC) have been identified as an important driver of follow-up acquired immune response

Peripheral anti-inflammatory effects explain the ginsenosides paradox between poor brain distribution and anti-depression efficacy.

BACKGROUND: The effectiveness of ginseng in preventing and treating various central nervous system (CNS) diseases has been widely confirmed. However, ginsenosides, the principal components of ginseng, are characterized by poor accessibility to the brain, and this pharmacokinetic-pharmacological paradox remains poorly explained. Anti-inflammatory approaches are becoming promising therapeutic strategies for depression and other CNS diseases; however, previous studies have focused largely on anti-inflammatory therapies directed at the central nervous system. It is thus of interest to determine wh

Anti-inflammatory and anti-gouty-arthritic effect of free Ginsenoside Rb1 and nano Ginsenoside Rb1 against MSU induced gouty arthritis in experimental animals.

Ginsenoside Rb1 (GsRb1) is the best constituent of ginseng and although it shows clinical efficacy as an antineoplastic, antioxidative and antirheumatic agent, its oral bioavailability is poor due to its limited solubility. In this study, the solubility of GsRb1 was improved by encapsulating it in polymeric nanocapsules (encapsulation efficiency: 99.79%), therefore, improving the oral bioavailability. The encapsulation resulted in stable, homogenous and well-dispersed nano-GsRb1, whose mean particle size and zeta potential were 183.9 nm and +36.9 mV, respectively. A significant improvement was

Rapid analysis of ginsenosides in biological samples using an efficient embedded dispersive solid-phase microextraction method.

A new method for rapid analysis of ginsenosides in biological samples was established using an embedded dispersive solid-phase microextraction (E-DSPE) technique. Lecithin@silica particles were prepared by embedding SiO2 particles with lecithin (mass ratio 4:7) and used as microextraction materials. The optimum extraction conditions of four ginsenosides from biological samples were as followed, 0.20 g of lecithin@silica particles; 10-minute absorption time; the sample of pH 6.0; and elution solvent methanol. The accuracy and matrix effects (ME %) of the method were studied by adding the four g

Estrogen receptor-β of microglia underlies sexual differentiation of neuronal protection via ginsenosides in mice brain.

AIMS: Streptococcus pneumoniae infection in acute bacterial meningitis can lead to widespread brain damage and mortality. Inflammatory responses by immune cells in the brain are thought to determine the degree of brain injury. Yet, the mechanisms underlying host responses to pneumococcal meningitis are largely unknown. To explore host responses as a potential therapeutic target for preventing brain injury after pneumococcal meningitis. METHODS: We evaluated signaling mechanisms that minimize neuronal damage caused by pneumococcal infection; specifically, we assessed pathways related to neuron

Research Progress on the Mechanism of Ginsenoside Rg1 in Inflammatory Bowel Disease.

Inflammatory bowel disease (IBD), a chronic intestinal inflammatory disorder, has witnessed a rising incidence globally. At present, the primary therapeutic approaches for IBD, such as aminosalicylic acid, glucocorticoids, immunosuppressants, and biologics, often entail notable side effects and limited efficacy. Traditional Chinese medicines (TCMs), particularly ginsenosides, have shown promise in the treatment of IBD. Ginsenoside Rg1 (G-Rg1), a characteristic constituent of ginsenosides, exerts a therapeutic impact on IBD. This review primarily elaborates on how G-Rg1 can restore the equilibr

Ginsenoside Rb1/baicalein nanoparticles: a potential nanomedicine for toxoplasmosis therapy.

Toxoplasma gondii (T. gondii), a globally prevalent zoonotic parasite, infects nucleated cells in warm-blooded animals, posing major public health risks. Current treatments (sulfadiazine/pyrimethamine) for the widespread parasite T. gondii have poor cyst penetration and significant side effects. While the natural compound baicalein (BAI, C₁₅H₁₀O₅) shows potent anti-T. gondii activity, its low solubility and high toxicity limit therapeutic use. To overcome BAI's limitations, we engineered ginsenoside Rb1-based nanoparticles (GRb1/BAI NPs) via self-assembly, leveraging GRb1's dual role as a bioc
