INTRODUCTION: Endothelial glycocalyx is a carbohydrate-rich layer lining the luminal side of blood vessels. Its damage was demonstrated in different groups of critically ill patients. Indirect evidence showed that endothelial glycocalyx degradation might be an important factor in pathophysiology of preeclampsia. The aim of our study was to demonstrate endothelial glycocalyx by transmission electron microscopy and to compare its amount in the omentum vessels of pregnant patients with severe preeclampsia and two control groups.
METHODS: Patients with severe preeclampsia who had a cesarean secti

Polytrauma (PT) is frequently associated with hemorrhagic shock (HS), which increases morbidity and mortality. Although various aspects of HS have been addressed in PT patients, the impact of an additional HS is largely unknown regarding the development of multiple organ dysfunction associated with disturbed glycocalyx and barrier function early after trauma. A prospective, longitudinal, mono-centered, observational study enrolling severely injured patients (Injury Severity Score, ISS=38.1±2.6) served for an in-depth analysis of blood (drawn on days 0, 1, 2, 3 and 5) and clinical data (up to 2

BACKGROUND: Glomerular endothelial glycocalyx (eGC) degradation and shedding play key roles in diabetic nephropathy (DN) pathogenesis. Although Panax notoginseng saponins (PNS) have been utilized clinically for diabetes and its complications, the mechanisms by which PNS protects against glomerular eGC degradation, particularly through heparanase-1 (HPSE1)-mediated pathways, remain unclear.
PURPOSE: This study investigated whether PNS protects against glomerular eGC injury in DN by regulating the HPSE1/EGFR/STAT3 signaling axis.
METHODS: In vitro cell models and in vivo animal models of DN we

The endothelial glycocalyx, a glycan-rich layer on the luminal surface of endothelial cells lining blood and lymphatic vessels, plays a crucial role in vascular homeostasis by regulating vascular permeability, immune cell trafficking, and vascular tone. Dysregulated endothelial glycocalyx turnover-whether through altered synthesis, intracellular degradation, or shedding-contributes to endothelial dysfunction in conditions such as sepsis, ischemic events, and chronic inflammatory disorders including diabetes and atherosclerosis. In this review, we examine the structure, function, and turnover o

COVID-19 is a highly infectious respiratory disease caused by a new coronavirus known as SARS-CoV-2. COVID-19 is characterized by progressive respiratory failure resulting from diffuse alveolar damage, inflammatory infiltrates, endotheliitis, and pulmonary and systemic coagulopathy forming obstructive microthrombi with multi-organ dysfunction, indicating that endothelial cells (ECs) play a central role in the pathogenesis of COVID-19. The glycocalyx is defined as a complex gel-like layer of glycosylated lipid-protein mixtures, which surrounds all living cells and acts as a buffer between the c

The endothelial luminal glycocalyx has been largely ignored as a target in vascular pathophysiology even though it occupies a key location. As a model of the inflammatory response, we tested the hypothesis that tumor necrosis factor-alpha (TNF-alpha) can alter the properties of the endothelial apical glycocalyx. In the intact hamster cremaster microcirculation, fluorescein isothiocyanate (FITC)-labeled Dextrans 70, 580, and 2,000 kDa are excluded from a region extending from the endothelial surface almost 0.5 micrometer into the lumen. This exclusion zone defines the boundaries of the glycocal

BACKGROUND: The endothelial surface layer (ESL) regulates vascular permeability to maintain fluid homeostasis. The glycocalyx (GCX), which has a complex and fragile ultrastructure, is an important component of the ESL. Abnormalities of the GCX have been hypothesized to trigger pathological hyperpermeability. Here, we report an integrated in vivo analysis of the morphological and functional properties of the GCX in a vital organ.
METHODS: We examined the behavior of the ESL and GCX, using both electron microscopy (EM) and intravital microscopy (IVM). We also compared morphological changes in t

The intestinal epithelial surface is protected by a multilayered barrier system that includes secreted mucus and a membrane-bound glycocalyx. While gel-forming mucins have been extensively studied, the transmembrane mucins that form the enterocyte glycocalyx remain less well understood. MUC17 has emerged as a dominant and functionally important transmembrane mucin of the small intestinal brush border, where its large, extensively O-glycosylated extracellular domain forms a dense, hydrated barrier between luminal microbes and the epithelial membrane. This review summarizes current knowledge of

Cancer-associated defects in mucin-type O-glycosylation expose truncated glycans on cell-surface glycoproteins and create tumor-associated glycopeptide epitopes. However, the structural principles by which antibodies recognize specific MUC1 glycopeptide sequences remain incompletely defined. Here, we report the molecular basis of MUC1 recognition by 16A, a high-affinity antibody isolated against hypoglycosylated MUC1. Co-crystal structures of 16A Fab bound to site-selectively glycosylated MUC1 peptides revealed that 16A recognizes an STAPPAHG-centered epitope within the MUC1 variable number ta

BACKGROUND: Sepsis damages endothelial cells through glycocalyx degradation, contributing to organ dysfunction. Glycocalyx repair is essential for recovery, but the underlying mechanisms remain unclear. Rhamnan sulfate (RS), a sulfated polysaccharide with anti-inflammatory properties, may support endothelial glycocalyx repair. In this study, we investigated the glycocalyx repair process and the therapeutic potential of RS using a mouse sepsis model.
METHODS: Sepsis was induced in male C57BL/6 mice by cecal ligation and puncture (CLP). Mice were divided into the RS, control (CTL), and sham gro

INTRODUCTION: Neutrophil extracellular traps (NETs) play a critical role in inflammation and coagulation imbalance. Recent studies have demonstrated that activation of gasdermin D (GSDMD) protein and its pore-forming activity are essential drivers of NET generation. This study investigated the association between GSDMD-NETs axis activation and sepsis-induced coagulopathy (SIC), as well as the potential association with glycocalyx damage.
MATERIALS AND METHODS: A prospective cohort of 70 sepsis patients (35 with SIC, 35 non-SIC) admitted to a respiratory intensive care unit was analyzed. This

Burn injuries often lead to severe complications, including acute respiratory distress syndrome (ARDS), driven in part by systemic inflammation and glycocalyx disruption. In this study, we analyzed the sera of 28 patients after burn trauma and utilized single-cell RNA sequencing (scRNA-seq) along with microarray transcriptomic analysis to decipher the impact of burn injury on glycocalyx derangement. We observed the significant upregulation of immune cell-derived degrading enzymes, particularly matrix metalloproteinase-8 (MMP8), which correlated with increased immune cell infiltration and glyco

BACKGROUND: We aimed to assess the longitudinal changes of glycocalyx, a proteoglycan/glycoprotein layer that lines and protects endothelial cells in normotensive pregnancies (NPs); to correlate glycocalyx parameters with kidney injury markers (podocin-positive to nephrin-positive [P/N] ratio); and to explore changes in P/N ratio in preeclampsia.
METHODS: Plasma and urine samples and glycocalyx measurements using side-stream darkfield imaging were collected at each trimester from pregnant women (n=31). Plasma glycocalyx components were measured by ELISA, and urinary P/N-positive extracellular

Oxaliplatin, a platinum-based anticancer drug, is associated with peripheral neuropathy (oxaliplatin-induced peripheral neuropathy, OIPN), which can lead to worsening of quality of life and treatment interruption. The endothelial glycocalyx, a fragile carbohydrate-rich layer covering the luminal surface of endothelial cells, acts as an endothelial gatekeeper and has been suggested to protect nerves, astrocytes, and other cells from toxins and substances released from the capillary vessels. Mechanisms underlying OIPN and the role of the glycocalyx remain unclear. This study aimed to define chan

BACKGROUND: Chronic periodontitis may induce erectile dysfunction (ED), however, the specific mechanism involved is unclear. The endothelial glycocalyx (eGlx) is a structure that can regulate endothelial nitric oxide synthase (eNOS) phosphorylation on the cavity surface of vessels.
AIM: To investigate whether chronic periodontitis leads to ED by affecting the eGlx.
METHODS: Twenty-four 4-week-old male Sprague‒Dawley rats were randomly divided into four groups (n = 6): the control group, chronic periodontitis group, chronic periodontitis + heparin group (subcutaneous heparin 200 U/kg/day, 7 d

Xenotransplantation is a promising approach to reduce organ shortage, while genetic modification of donor pigs has significantly decreased the immunogenic burden of xenotransplants, organ rejection is still a hurdle. Genetically modified pig organs are used in xenotransplantation research, and the first clinical pig-to-human heart transplantation was performed in 2022. However, the impact of genetic modification has not been investigated on a cellular level yet. Endothelial cells (EC) and their sugar-rich surface known as the glycocalyx are the first barrier encountering the recipient's immune

BACKGROUND: Acute pyelonephritis, a risk factor for maternal sepsis, adult respiratory distress syndrome, and preterm labor, is a frequent cause of hospitalization. This condition is characterized by excessive intravascular inflammation and endothelial cell activation and dysfunction. Syndecan-1, a major component of the glycocalyx, is a gel-like layer that covers the luminal surface of healthy endothelial cells, preserving and mediating many endothelial functions. During pregnancy, there is an additional potential source of syndecan-1, the "syncytiotrophoblast glycocalyx," which lines the int

Significantly reduced levels of the anti-inflammatory gaseous transmitter hydrogen sulfide (H2S) are observed in diabetic patients and correlate with microvascular dysfunction. H2S may protect the microvasculature by preventing loss of the endothelial glycocalyx. We tested the hypothesis that H2S could prevent or treat retinal microvascular endothelial dysfunction in diabetes. Bovine retinal endothelial cells (BRECs) were exposed to normal (NG, 5.5 mmol/L) or high glucose (HG, 25 mmol/L) ± the slow-release H2S donor NaGYY4137 in vitro. BACKGROUND: Endothelial glycocalyx (EG) abnormal degradation were widely found in critical illness. However, data of EG degradation in multiple traumas is limited. We performed a study to assess the EG degradation and the correlation between the degradation and organ functions in polytrauma patients.
METHODS: A prospective observational study was conducted to enroll health participants (control group) and polytrauma patients (trauma group) at a University affiliated hospital between Feb 2020 and Oct 2020. Syndecan1 (SDC1) and heparin sulfate (HS) were detected in serum sample of both groups.

High-intensity interval training (HIIT) has been proposed to exert vasculoprotective effects. This study aimed to evaluate whether HIIT affects the microvasculature, including the endothelial glycocalyx barrier, and to identify associated microRNAs (miRNAs). Fifty healthy participants (23.1 ± 3.0 yr) performed a 4-wk 4 × 30-s all-out running HIIT. Sidestream dark-field imaging was performed at baseline and follow-up to detect changes of the sublingual microvasculature including the endothelial glycocalyx. Exercise parameters were determined by continuous running field test and documentation of
