Enhanced glycolysis contributes to the pathogenesis of experimental autoimmune neuritis.

BACKGROUND: With the recognition of the key roles of cellular metabolism in immunity, targeting metabolic pathway becomes a new strategy for autoimmune disease treatment. Guillain-Barré syndrome (GBS) is an acute immune-mediated inflammatory demyelinating disease of the peripheral nervous system, characterized by inflammatory cell infiltration. These inflammatory cells, including activated macrophages, Th1 cells, and Th17 cells, generally undergo metabolic reprogramming and rely mainly on glycolysis to exert functions. This study aimed to explore whether enhanced glycolysis contributed to the

Inhibition of glycolysis in the retina by oxidative stress: prevention by pyruvate.

Intraocular generation of reactive oxygen species (ROS) with consequent oxidative stress has been shown to be a significant factor in the pathogenesis of many vision-impairing diseases such as cataracts and retinal degenerations. Previous studies have shown that pyruvate can inhibit such oxidative stress. This is attributable to its property of scavenging various ROS and consequently inhibiting many of the apparent toxic reactions such as lipid peroxidation and loss of tissue thiols. It is hence expected that ROS will have an adverse effect on tissue metabolism also. The present investigations

Early effects of vitamin A toxicity on hepatic glycolysis in rat.

Vitamin A toxicity, caused by oral administration of 30,000 IU of vitamin A (retinyl palmitate) to young rats (70 to 90 g) once daily for 2 days, increased the levels of lipids, glycogen, and citrate in the liver. Furthermore, hypervitaminosis A decreased the activities of two key hepatic glycolytic enzymes, phosphofructokinase, and pyruvate kinase, without affecting those of hexokinase and glucokinase. It is suggested, therefore, that in addition to the increased activities of key gluconeogenic enzymes, reported earlier, a marked decrease in the activities of phosphofructokinase and pyruvate

Ubiquitination of PFKFB4 by CHIP regulates glycolysis and progression in endometriosis†.

Endometriosis is a common estrogen-dependent gynecological disorder characterized by chronic inflammatory responses and frequently associated with clinical infertility. Recent studies have demonstrated that the pathogenesis of this disease is closely linked to dysregulated post-translational modifications. This study focuses on the regulatory role of the ubiquitin-proteasome system (UPS) in endometriosis and, for the first time, reveals the molecular mechanism by which the E3 ubiquitin ligase STIP1 homology and U-box containing protein 1 (CHIP) specifically binds to and ubiquitinates the glyco

TAB1 regulates glycolysis and activation of macrophages in diabetic nephropathy.

OBJECTIVE AND DESIGN: Macrophages exhibit strong phenotypic plasticity and can mediate renal inflammation by polarizing into an M1 phenotype. They play a pivotal role in diabetic nephropathy (DN). Here, we have investigated the regulatory role of transforming growth factor β-activated kinase 1-binding protein 1 (TAB1) in glycolysis and activation of macrophages during DN. METHODS: TAB1 was inhibited using siRNA in high glucose (HG)-stimulated bone marrow-derived macrophages (BMMs) and lentiviral vector-mediated TAB1 knockdown was used in streptozotocin (STZ)-induced diabetic mice. Western blo

A modelling study of feedforward activation in human erythrocyte glycolysis.

Though feedforward activation (FA) is a little known principle of control in metabolic networks, there is one well-known example; namely, the activation of pyruvate kinase (PK) by fructose-1,6-biphosphate (FBP) in glycolysis. The effects of this activation on the enzyme's kinetics are well characterised, but its possible role in glycolytic control has not been determined, and, experimentally, there is as yet no direct way of modifying the enzyme to remove just the FBP activation without affecting other aspects of the enzyme's kinetics. Given this limitation, we used a detailed numerical simula

Role of fructose 2,6-bisphosphate in the control of heart glycolysis.

The aim of this work was to study whether changes in fructose 2,6-bisphosphate concentration are correlated with variations of the glycolytic flux in the isolated working rat heart. Glycolysis was stimulated to different extents by increasing the concentration of glucose, increasing the workload, or by the addition of insulin. The glycolytic flux was measured by the rate of detritiation of [2-3H]- and [3-3H]glucose. Under all the conditions tested, an increase in fructose 2,6-bisphosphate content was observed. The glucose- or insulin-induced increase in fructose 2,6-bisphosphate content was re

[Intensity of glycolysis and energy metabolism in erythrocytes in experimental hypervitaminosis A].

Administration of 60,000 i.e. of vitamin A into rats within three weeks caused an increase in amount of reticulocytes, in the rate of glucose utilization and in formation of lactic acid by erythrocytes. The activity of glycolytic enzymes was intensified. The activity of hexokinase was increased by 84.6%, activities of aldolase and phosphohexoisomerase were increased by 34%. But in the erythrocytes content of AMP, ADP and ATP was unaltered, probably due to activation of total and Na+, K+-dependent ATPase. The harmful effect of an excess of the vitamin A was manifested in an increased content of

A novel PGK1 activator improves Alzheimer's disease by regulating glycolysis.

Alzheimer’s disease (AD) is a neurodegenerative disorder characterized by the pathological accumulation of amyloid-β (Aβ) plaques and neurofibrillary tangles. Decreased glucose metabolism and ATP levels are well-established early biomarkers of AD. Recent studies indicate that the activation of phosphoglycerate kinase 1 (PGK1), a critical enzyme in the glycolytic pathway, promotes glucose metabolism and ATP production, thus offering potential therapeutic benefits for AD. However, two significant gaps persist: The expression pattern of PGK1 across different brain cell types remains unclear, and

Targeting glycolysis in prostate cancer: Molecular mechanisms and therapeutic advances.

Prostate cancer (PCa) is the second most common cancer among men worldwide and poses a significant threat to male health. A key feature of tumor progression is metabolic reprogramming, which involves the abnormal activation of glycolysis. This metabolic process supports PCa proliferation, metastasis, and drug resistance through rapid energy production, the provision of biosynthetic precursors, and the remodeling of the tumor microenvironment (TME). Key enzymes such as hexokinase 2 (HK2), phosphofructokinase (PFK), pyruvate kinase M2 (PKM2), glucose transporters (GLUTs), and lactate dehydrogena

LRRN3 protects dopaminergic neurons by inhibiting glycolysis in Parkinson's disease.

Leucine-rich repeat neuronal protein 3 (LRRN3) is a multifunctional transmembrane protein with a crucial role in intracellular signal transduction. It is expressed at high levels in neurons. LRRN3 expression has been shown to be associated with Parkinson's disease (PD) and aging. It is involved in regulating cellular energy metabolism. However, the specific mechanism involved remains undetermined. In this study, we investigated whether LRRN3 can regulate the expression of the key glycolytic enzymes HK2 and LDHA as well as lactate levels. We also studied the expression of the apoptosis-related

Demethoxycurcumin suppresses HK2-mediated glycolysis by targeting PTEN/Akt signaling.

Aerobic glycolysis is a hallmark of tumor cells, with the expression of glycolytic enzymes often being upregulated in many cancers, leading to enhanced metabolic activity. Among the key rate-limiting enzymes in this process, Hexokinase 2 (HK2) plays a crucial role in sustaining the biological activities of human cancers. Therefore, HK2 is considered a potential therapeutic target, although effective targeted drugs for oral squamous cell carcinoma (OSCC) treatment are currently lacking. To confirm potent anti-tumor agents that inhibit HK2 expression, we screened a library of 639 natural product

CTRP9 engages AdipoR1 and promotes T cell glycolysis and immunity.

The adiponectin (ADPN) receptor (AdipoR) modulates T-cell responses, but its effects remain controversial since signaling can either promote or inhibit T-cell function. Interaction with the ligand ADPN inhibits T-cell responses, but given the existence of multiple AdipoR ligands, we hypothesize that ligand diversity underlies its differential effect in T-cell immunity. To test this, we use tilapia and mouse models. Tilapia encodes AdipoR1 but lacks ADPN. Instead, an alternative adipokine, CTRP9, engages AdipoR1. We find CTRP9-AdipoR1 interaction triggers Ca2+ influx and activates the CaM-CaMKK

ATF4 promotes renal tubulointerstitial fibrosis through hexokinase II-mediated glycolysis.

BACKGROUND: Renal tubulointerstitial fibrosis is a reliable predictor of progressive chronic kidney disease (CKD). Activating transcription factor 4 (ATF4) has recently emerged as a pivotal player in multiple pathophysiologic processes, particularly the stress response processes. This study aims to explore the role of ATF4 in tubulointerstitial fibrosis from a metabolic perspective. METHODS: A murine model of renal fibrosis was generated via unilateral ureteral obstruction (UUO). Quantitative PCR was employed to assess the expression of inflammation-related genes and fibrotic markers in renal

Translocation of SIRT6 promotes glycolysis reprogramming to exacerbate diabetic angiopathy.

Diabetic angiopathy, a major complication of type 2 diabetes mellitus (T2DM), is driven by vascular dysfunction, metabolic reprogramming, and oxidative stress. NAD+-dependent deacetylase SIRT6, located in the nucleus, is recognized for its role in modulating cardiovascular and metabolic homeostasis through histone deacetylation. However, the functions and mechanisms of accumulation of cytoplasmic SIRT6 in T2DM remain to be elucidated. Herein, a previously unrecognized cytoplasmic role for SIRT6 is identified in promoting pathological glycolysis during diabetic vascular remodeling. Vascular smo
