Streptococcus pneumoniae colonizes human airways, where it acquires sugars from glycosylated mucins using glycoside hydrolases and sugar transport systems. This study identifies widespread nucleotide sequence variation in the promoter of a pneumococcal operon encoding a glycan scavenging system. We identify 78 promoter sequence patterns across 21,155 genomes, with variation clustered within a stretch of adenines, where mutations accumulate via strand slippage during DNA replication. Promoter mutations influence operon transcription, and multiple promoter patterns are co-identified during singl

Egg white powder incorporation into high-protein yogurt may interfere with starter culture performance and acid-induced gel formation. This study prepared egg white powder by ultrasound-heat pretreatment followed by spray drying (UHSD-prepared egg white powder) and evaluated its effects on set-type yogurt. Compared with fresh egg white, the UHSD-prepared egg white powder exhibited a 90.3% reduction in apparent lysozyme activity. Yogurt containing 1% UHSD-prepared egg white powder exhibited improved texture, water-holding capacity, and viscoelastic properties, whereas 4% addition increased pH,

Bacteroides uniformis (Bu) is a promising probiotic, yet its mechanism for utilizing lotus seed resistant starch (LRS) remains unclear. Here, growth analysis, starch structural characterization, transcriptomics, and molecular docking were integrated to elucidate the LRS utilization strategy of Bu. Bu efficiently utilized LRS and released abundant extracellular maltooligosaccharides during fermentation. Structural analyses revealed an exterior-to-interior degradation pattern characterized by the preferential removal of peripheral short chains and progressive surface erosion of starch granules.

A gene of unknown function, Gohir.A02G131900.1, identified in Gossypium hirsutum was studied using computational sequence and structure bioinformatic tools. The encoded protein GhGH5BG-A0A1U8NW40 (UniProt A0A1U8NW40) is predicted to be secreted and localized to the cell wall. Homology and conserved residues indicate it belongs to a plant-specific subgroup of the glycoside hydrolase family 5 and likely has exo-1,3-β-glucosidase activity. This subgroup is unique in containing a fascin-like domain which may have evolved a unique glucan binding site of interest for further research.

Lytic polysaccharide monooxygenases (LPMOs) are mononuclear copper enzymes that catalyse the oxidative cleavage of glycosidic bonds. They are characterised by two histidine residues that coordinate copper in a configuration termed the Cu-histidine brace. Although first identified in bacteria and fungi, LPMOs have since been found in all biological kingdoms. LPMOs are now included in commercial enzyme cocktails used in industrial biorefineries. This has led to increased process yield due to the synergistic action of LPMOs with glycoside hydrolases. However, the introduction of LPMOs makes contr

Glycoside hydrolase family 31 (GH31) contains α-glycoside hydrolases with different substrate specificities involved in various physiological functions. This family has recently been classified into 20 subfamilies using sequence similarity networks. An α-galactosidase from the gut bacterium Bacteroides salyersiae (BsGH31_19, which belongs to GH31 subfamily 19) was reported to have hydrolytic activity against the synthetic substrate p- nitrophenyl α-galactopyranoside, but its natural substrate remained unknown. BsGH31_19 shares low sequence identity (around 20%) with other reported GH31 α-galac

Prunin of desirable bioactivity and bioavailability can be transformed from plant-derived naringin by the key enzyme α-L-rhamnosidase. However, the production was limited by unsatisfactory properties of α-L-rhamnosidase such as thermostability and organic solvent tolerance. In this study, biochemical characteristics, and hydrolysis capacity of a novel α-L-rhamnosidase from Spirochaeta thermophila (St-Rha) were investigated, which was the first characterized α-L-rhamnosidase for Spirochaeta genus. St-Rha showed a higher substrate specificity towards naringin and exhibited excellent thermostabil

Agar is a common component biosynthesized from various marine algae species that is widely applied in various fields including food and pharmaceutical industries. However, the structural composition of agar is highly resisted against chemical and biological hydrolysis. Therefore, tremendous research is exploring various pretreatment strategies to break down the intrinsic chemical structural of agar linkage (i.e. neutral agarose and highly sulfated agaropectin) prior for its industrial potential usage. In this research work, a novel agar degrading bacterium was screened and isolated from agricu

Bacteriophages have been of interest as agents combating undesirable bacteria since their discovery nearly 100 years ago. Currently, intensive research is being conducted into two groups of phage enzymes, which cause damage to bacterial cells. The first group includes lysins responsible for breaking down the cell wall in order to release progeny phages and the second is polysaccharides depolymerases (PDs), which degrade capsular and structural polysaccharides, including exopolysaccharides (EPS) - a dominant bacterial biofilm component. PDs can be attached to a phage tail or present as a free f

Aliphatic 1,2-alkanolamines inhibited beta-glucanase from Candida utilis. The highest inhibitory effect was observed with 1-dodecylamino-3-chloro-2-propanol (Ki = 1.0 mumol/L) and it was much higher than that of D-glucono-delta-lactone. Simple 1,2-alkanolamines, like 2-aminoethanol or 2-methylaminoethanol, did not exhibit any significant inhibitory effect.

This study aimed to quantify verbascoside (VEB), perform molecular docking studies of VEB with the α-glucosidase (GL) of Bacillus stearothermophilus, and evaluate the inhibition of the enzyme by L. dulcis preparations. The substrate concentration and presence of reduced glutathione were evaluated for their effect on the in vitro inhibition of the GL enzyme. Assays were also performed in the presence and absence of simulated gastric fluid. The antidiabetic fractions 2 and 3 were the most inhibited GL, but their activity were significantly decreased in the presence of gastric fluid. Chromatograp

Glycosyl hydrolases (GHs) are carbohydrate-active enzymes that hydrolyze a specific β-glycosidic bond in glycoconjugate substrates; β-glucosidases degrade glucosylceramide, a ubiquitous glycosphingolipid. GHs are grouped into structurally similar families that themselves can be grouped into clans. GH1, GH5, and GH30 glycosidases belong to clan A hydrolases with a catalytic (β/α)8 TIM barrel domain, whereas GH116 belongs to clan O with a catalytic (α/α)6 domain. In humans, GH abnormalities underlie metabolic diseases. The lysosomal enzyme glucocerebrosidase (family GH30), deficient in Gaucher d

Glucocerebrosidase (GCase), which is encoded by the GBA1 gene, has lysosomal glycoside hydrolase activity that hydrolyzes glucosylceramide. Defects in GCase lead to the accumulation of glucosylceramide, which causes the development of the lysosomal storage disease known as Gaucher's disease. Loss-of-function mutations in the GBA1 gene are the most important genetic risk factor for synucleinopathies, such as Parkinson's disease and dementia with Lewy bodies. Recent studies on PD genes associated with lysosomal function suggest that GCase activity is decreased in cell models of PD and in neurons

Bletillae Rhizoma (Baiji), the dried tuber of Bletilla striata (Thunb.) Reichb.f. (BS), a traditional medicinal plant widely used in Chinese medicine. While polysaccharides from BS tuber (BSP) have attracted attention for their broad industrial applications, their detailed structural features and biological functions remain limited. In this study, a neutral polysaccharide (BSPN, 178 kDa), constituting about 80% of BSP, was extracted and purified. Structural analysis elucidated BSPN as a glucomannan (mannose: glucose = 4: 1), featuring a backbone of →4)-β-D-Manp-(1 → and →4)-β-D-Glcp-(1 → linka

Dietary carrageenan (CGN) is a widely used food additive, yet its microbial metabolism in the human gut and health implications remain unclear. This study elucidated a syntrophic pathway wherein Bacteroides thetaiotaomicron (B. theta) degrades CGN via specific sulfatases and glycoside hydrolases, releasing inorganic sulfate. This sulfate fuels the growth and metabolic activity of the sulfate-reducing bacterium Desulfovibrio desulfuricans (D. desulfuricans). In vitro co-culture experiments demonstrated that this cross-feeding interaction significantly enhanced hydrogen sulfide (H2S) production.

The in situ relevance of biosynthetic gene clusters (BGCs) remains poorly understood. We apply meta-omics to characterize BGC diversity and activity along a peatland redox gradient. From seven metagenomes, we recover 9,694 BGCs, spanning diverse taxa, most lacking close relatives in reference databases, indicating extensive novelty. Only 9-27% of this potential is expressed in situ, with Acidobacteriota, despite moderate repertoires, accounting for over half of all BGC transcription. Talented producers with up to 24 clusters are largely silent, and expression is inversely related to BGCs per g
