Gray and white matter trajectories in patients with bipolar disorder.

OBJECTIVES: Findings on brain structural abnormalities in patients with bipolar disorder (BP) are inconsistent and little is known about age-related evolution of these changes. We employed a cross-sectional, case-control study to compare structural age-related brain trajectories in patients with BP and healthy control subjects (HC) over a period of approximately 50 years. The primary aim was to understand whether white (WM) and gray matter (GM) abnormalities are present from the beginning of the illness and how they change over time. METHODS: Seventy-eight patients with BP and 78 HC matched f

Accelerated Gray and White Matter Deterioration With Age in Schizophrenia.

OBJECTIVE: Although brain changes in schizophrenia have been proposed to mirror those found with advancing age, the trajectory of gray matter and white matter changes during the disease course remains unclear. The authors sought to measure whether these changes in individuals with schizophrenia remain stable, are accelerated, or are diminished with age. METHOD: Gray matter volume and fractional anisotropy were mapped in 326 individuals diagnosed with schizophrenia or schizoaffective disorder and in 197 healthy comparison subjects aged 20-65 years. Polynomial regression was used to model the i

Gray matter abnormalities in paranoid schizophrenia and their clinical correlations.

Numerous imaging studies have shown structural brain abnormalities in schizophrenia. Recently, voxel-based morphometry enabled whole brain analysis beyond the regions of interest (ROI). Regional gray matter concentrations of magnetic resonance (MR) images from 35 patients with paranoid schizophrenia were compared with those from 35 age- and sex-matched controls, and their clinical correlations were explored using voxel-based morphometry. Gray matter concentrations in the patients were significantly reduced in the left insular and dorsolateral prefrontal regions, and bilaterally in the medial f

Ventrolateral periaqueductal gray matter and the control of tonic immobility.

Tonic immobility is an inborn defensive behavior characterized by a temporary state of profound and reversible motor inhibition elicited by some forms of physical restraint. The periaqueductal gray matter (PAG) contains neural circuits involved in descending pain modulation, as well as in the modulation of TI. We have reported previously that the cholinergic stimulation of the ventrolateral PAG increases the duration of TI in guinea pigs. In the present study, we attempted to characterize further the modulation of TI by pharmacological alteration of the neurochemistry of the ventrolateral PAG

On the Correlations of Gray Matter with Schizotypy in Mentally Healthy Subjects.

We analyzed the relationships between morphometric characteristics of brain gray matter and schizotypy. Mentally healthy subjects (n=164, age 18-35 years) completed Russian version of SPQ-74 test and underwent high-field 3T MRI. Cortical thickness in the right frontal pole (determined with FreeSurfer 6.0.0) positively correlated with negative schizotypy factor. The revealed features can reflect the protective mechanisms (resilience) against the development of mental disorders and also can be a result of individual ontogenesis trajectories manifested in deceleration of the decrease of the corte

White matter and cortical gray matter microstructural alterations in migraine: a NODDI and DTI analysis.

BACKGROUND: The pathophysiological mechanism of migraine remains elusive, thereby impeding the effective treatment of the disease. Current neuroimaging research focuses on changes in brain functional connectivity, functional networks, and macrostructural alterations, which reflect abnormal neuronal function during the disease process. The plasticity changes in neuronal structures and neurotransmitter system dysregulations potentially play a crucial role in migraine onset and chronicity of migraine. This study utilizes multimodal neuroimaging techniques to investigate the microstructural and ne

Widespread reductions in white matter-gray matter functional connectivity in long-term fibromyalgia syndrome patients.

Long-term fibromyalgia syndrome (FMS) patients experience chronic pain, emotional disturbances, and poor treatment responses, yet the neural mechanisms underlying disease chronicity remain unclear. This study aimed to investigate the white matter-gray matter functional connectivity (WM-GM FC) in long-term FMS patients to explore changes in brain network communication associated with chronicity and their relationship with clinical symptoms. A total of 52 FMS patients (30 long-term, 22 short-term) and 49 healthy controls (HCs) underwent resting-state functional magnetic resonance imaging (fMRI).

Fronto-Parietal gray matter and white matter efficiency differentially predict intelligence in males and females.

While there are minimal sex differences in overall intelligence, males, on average, have larger total brain volume and corresponding regional brain volumes compared to females, measures that are consistently related to intelligence. Limited research has examined which other brain characteristics may differentially contribute to intelligence in females to facilitate equal performance on intelligence measures. Recent reports of sex differences in the neural characteristics of the brain further highlight the need to differentiate how the structural neural characteristics relate to intellectual ab

Periaqueductal gray matter echogenicity: An irreversible marker of chronic migraine?

AimAlthough periaqueductal grey matter (PAG), assessed by transcranial sonography, has been proposed as a biomarker of migraine chronification, its longitudinal evolution remains unknown. This study aimed to evaluate whether changes in clinical diagnosis-from chronic migraine to episodic migraine (CM-EM) or from episodic migraine to chronic migraine (EM-CM)-were associated with modifications in PAG echogenicity.MethodsIn a prospective longitudinal cohort study we reanalysed a cohort of 115 participants (65 patients with migraine and 50 controls) previously assessed in a baseline study. Transcriptomic signatures of gray matter volume alterations in hepatic encephalopathy.

It has been revealed that brain gray matter volume (GMV) abnormalities are present in patients with hepatic encephalopathy (HE). However, the GMV alterations in HE that have been uncovered are inconsistent, and their correlation with gene expression profiles is still unknown. We conducted a quantitative neuroimaging meta-analysis on seven studies, involving 142 HE individuals and 193 HCs, to identify consistent patterns of GMV alterations in HE. We then utilized the Allen Human Brain Atlas database to investigate transcriptome-neuroimaging spatial correlations, exploring gene expression profil

Increased gray matter within temporal cortical networks in sexual sadism.

Sexual sadism is an extreme, pathological manifestation of sadistic traits. While the psychological and forensic characteristics of sexual sadism have been extensively studied, the underlying neurocircuitry is far from being understood. In this study, n = 232 incarcerated men completed structural MRI scans, n = 116 of whom had been convicted of sexual offenses. Sexual offenders were assessed for the presence and severity of sadism using the Sexual Sadism Scale (SeSaS). Multivariate (source-based morphometry (SBM)) and univariate (voxel-based morphometry (VBM)) techniques were used to identify

Altered Gray Matter Myelin in Type IIb Focal Cortical Dysplasia.

BACKGROUND AND OBJECTIVES: Myelin is altered in several neurologic disorders. Published data demonstrate reduced white matter myelin content and lower oligodendrocyte cell number in postsurgical brain specimens obtained from patients with focal cortical dysplasia (FCD) and temporal lobe epilepsy; a pathogenic role of dysfunctional myelin in focal epilepsies has been proposed. Based on this evidence, our study aims to investigate the myelination status in the gray matter in postsurgical brain specimens from patients with FCDIIb. METHODS: We collected specimens from patients with a histopatholo

Longitudinal Gray Matter Trajectories in Pediatric Mild Traumatic Brain Injury.

BACKGROUND AND OBJECTIVES: This prospective, longitudinal cohort study examined trajectories of brain gray matter macrostructure after pediatric mild traumatic brain injury (mTBI). METHODS: Children aged 8-16.99 years with mTBI or mild orthopedic injury (OI) were recruited from 5 pediatric emergency departments. Reliable change between preinjury and 1 month postinjury symptom ratings was used to classify mTBI with or without persistent symptoms. Children completed postacute (2-33 days) and/or chronic (3 or 6 months) postinjury T1-weighted MRI, from which macrostructural metrics were derived u

Ventrolateral periaqueductal gray matter integrative system of defense and antinociception.

Defensive responses are neurophysiological processes crucial for survival during threatening situations. Defensive immobility is a common adaptive response, in rodents, elaborated by ventrolateral periaqueductal gray matter (vlPAG) when threat is unavoidable. It is associated with somatosensory and autonomic reactions such as alteration in the sensation of pain and rate of respiration. In this study, defensive immobility was assessed by chemical stimulation of vlPAG with different doses of NMDA (0.1, 0.3, and 0.6 nmol). After elicitation of defensive immobility, antinociceptive and respiratory

Gray matter changes in asymptomatic C9orf72 and GRN mutation carriers.

Frontotemporal dementia (FTD) is a neurodegenerative disease with a strong genetic basis. Understanding the structural brain changes during pre-symptomatic stages may allow for earlier diagnosis of patients suffering from FTD; therefore, we investigated asymptomatic members of FTD families with mutations in C9orf72 and granulin (GRN) genes. Clinically asymptomatic subjects from families with C9orf72 mutation (15 mutation carriers, C9orf72+; and 23 non-carriers, C9orf72-) and GRN mutations (9 mutation carriers, GRN+; and 15 non-carriers, GRN-) underwent structural neuroimaging (MRI). Visuospatial short-term memory and dorsal visual gray matter volume.

Visual short-term memory (VSTM) is an important cognitive capacity that varies across the healthy adult population and is affected in several neurodevelopmental disorders. It has been suggested that neuroanatomy places limits on this capacity through a map architecture that creates competition for cortical space. This suggestion has been supported by the finding that primary visual (V1) gray matter volume (GMV) is positively associated with VSTM capacity. However, evidence from neurodevelopmental disorders suggests that the dorsal visual stream is more broadly vulnerable and atypical volumes o
