Circulating growth-and-differentiation factor-15 in early life: relation to prenatal and postnatal growth and adiposity measurements.

BACKGROUND: Growth-and-differentiation-factor-15 (GDF15) is a regulator of energy homeostasis. To determine the relationship between circulating GDF15 and parameters of metabolic health, we assessed longitudinally GDF15 concentrations in infants born either appropriate- (AGA) or small-for-gestational-age (SGA), the latter population known to be at risk for metabolic alterations, particularly after a rapid postnatal catch-up in weight. METHODS: The study cohort consisted of 103 infants (70 AGA and 33 SGA). Assessments included body length, weight, and ponderal index (PI); fasting glucose, insu

Growth Differentiation Factor-15 in Acute Coronary Syndromes: Prognostic Value and Barriers to Clinical Implementation.

High-sensitivity cardiac troponin has made the diagnosis of myocardial infarction (MI) faster and more precise, but it does not measure the broader biological vulnerability that often determines the outcome after an acute coronary syndrome (ACS). Growth differentiation factor-15 (GDF-15) is induced by ischemic stress, inflammation, oxidative injury, renal dysfunction, metabolic disease, and ageing. This biology explains its appeal in ACS, but also its diagnostic limitation: GDF-15 is not cardiac-specific and should not be used as an alternative to electrocardiography and high-sensitivity tropo

Association of growth differentiation factor-15 with muscle wasting and anemia in chronic kidney disease.

BACKGROUND: Cachexia, characterized by muscle wasting and anemia, is a major complication of chronic kidney disease (CKD). Growth differentiation factor-15 (GDF-15), a stress-responsive cytokine implicated in cancer cachexia, is markedly elevated in CKD and linked to disease progression. However, the role of GDF-15 in CKD-related cachexia remains unclear. METHODS: We conducted a cross-sectional study of 268 patients with stage 3-5 CKD not yet on dialysis. Plasma GDF-15 concentrations were measured using immunoassays. Lean body mass was assessed by multifrequency bioimpedance spectroscopy with

Relationship between growth differentiation factor 15 and functional prognosis and severity in traumatic brain injury.

Circulating growth differentiation factor 15 (GDF-15) increases in response to inflammation and tissue damage. Its association with functional prognosis in cerebral infarction and subarachnoid hemorrhage is established; however, its role in traumatic brain injury (TBI) and its relationship with Sequential Organ Failure Assessment (SOFA) score, an indicator of systemic organ damage in TBI, remains unclear. This study aimed to explore the correlation between GDF-15 and functional outcomes at discharge in patients with TBI and stroke, including its association with SOFA scores in TBI. Patients wi

Diagnostic and prognostic values of circulating growth differentiation factor-15 and osteopontin in uterine sarcoma.

Uterine sarcoma is an aggressive malignancy that is difficult to distinguish from benign leiomyomas pre-operatively. This study aimed to identify circulating biomarkers to improve differential diagnosis and prognostication. The serum and tissue levels of growth differentiation factor-15 (GDF15), progranulin (PGN), and osteopontin (OPN) were measured in patients with uterine sarcoma (n = 38) and leiomyoma (n = 67). The levels were correlated with diagnosis and patient survival. The serum and tissue levels of GDF15 and OPN were significantly higher in uterine sarcomas than in leiomyomas (p < 0.0

Growth differentiation factor 15 (GDF15) levels are associated with malnutrition in acutely admitted older adults.

BACKGROUND AND AIMS: The aging process is often accompanied by high risk of malnutrition and elevated levels of growth differentiation factor 15 (GDF15). GDF15 is an increasingly recognized biomarker for regulation of metabolism, but few studies have investigated the connection between GDF15 and malnutrition in older age and how it relates to other features of aging such as decreased appetite and physical function. Therefore, we investigated the associations between GDF15 levels and nutritional status, appetite, and physical function in acutely admitted older adults. METHODS: Plasma GDF15 lev

Metformin and growth differentiation factor 15 (GDF15) in type 2 diabetes mellitus: A hidden treasure.

Type 2 diabetes mellitus (T2DM) is a chronic endocrine disorder due to the reduction of insulin sensitivity and relative deficiency of insulin secretion. Growth differentiation factor 15 (GDF15) belongs to the transforming growth factor beta (TGF-β) superfamily and was initially identified as macrophage inhibitory cytokine-1 (MIC-1). GDF15 is considered a cytokine with an anti-inflammatory effect and increases insulin sensitivity, reduces body weight, and improves clinical outcomes in diabetic patients. GDF15 acts through stimulation of glial-derived neurotrophic factor (GDNF) family receptor

Growth Differentiation Factor-15 as a Predictor of Idiopathic Membranous Nephropathy Progression: A Retrospective Study.

Idiopathic membranous nephropathy (IMN) is a major cause of nephrotic syndrome. No biomarker to predict the long-term prognosis of IMN is currently available. Growth differentiation factor-15 (GDF-15) is a member of the transforming growth factor-β superfamily and has been associated with chronic inflammatory disease. It has the potential to be a useful prognostic marker in patients with renal diseases, such as diabetic nephropathy and IgA nephropathy. This study examined whether GDF-15 is associated with the clinical parameters in IMN and showed that GDF-15 can predict IMN disease progression

Histamine receptor 2-mediated growth-differentiation factor-15 expression is involved in histamine-induced melanogenesis.

Vitiligo is a progressive depigmenting disorder. Histamine has been shown to induce melanogenesis via histamine receptor 2, suggesting the possibility of histamine as a repigmenting agent for the treatment of vitiligo. However, the role and signaling mechanism of histamine are still unclear in melanogenesis, especially in relation to growth-differentiation factor-15, which is a protein belonging to transforming growth factor beta and found to be overexpressed in metastatic or malignant melanoma. We found that histamine induces growth-differentiation factor-15 in melanoma cell lines such as SK-

Growth differentiation factor (GDF)-15 blocks norepinephrine-induced myocardial hypertrophy via a novel pathway involving inhibition of epidermal growth factor receptor transactivation.

Accumulating evidence suggests that growth differentiation factor 15 (GDF-15) is associated with the severity and prognosis of various cardiovascular diseases. However, the effect of GDF-15 on the regulation of cardiac remodeling is still poorly understood. In this present study, we demonstrate that GDF-15 blocks norepinephrine (NE)-induced myocardial hypertrophy through a novel pathway involving inhibition of EGFR transactivation. Both in vivo and in vitro assay indicate that NE was able to stimulate the synthesis of GDF-15. The up-regulation of GDF-15 feedback inhibits NE-induced myocardial
