Bacteria associated with ovine gut parasites Trichuris ovis and Haemonchus contortus.

An associated microbiome of any host helps it in different metabolic processes ranging from the decomposition of food to the maturation of gametes. Organisms with a parasitic mode of life, though present at nutritious sites inside their host, maintain their own microbiome. Nevertheless, the comprehensive characterization and functionality of microbiome in parasitic organisms remain understudied. We selected two nematode parasites of Kashmir Merino sheep viz;Haemonchus contortus and Trichuris ovis based on their higher prevalence, difference in mode of nutrition, habitation site and effect on h

Immunization with thiol-binding proteins from Haemonchus contortus adult worms partially protects goats against infection during prepatency.

To contribute of the knowledge of the immune mechanisms underlying the response to the immunization of goats with thiol-binding proteins fractions (PBS-TSBP) from Haemonchus contortus (H. contortus) adult worms, this study analyzed the degree of protection and the immune responses developed against the parasite after vaccination with this antigenic complex during the time-elapsing between challenge with L3 of the parasite and the development of adult worms, evidenced by the appearance of first faecal eggs (prepatent period or prepatency). Goat kids immunized with PBS-TBSP generated an immune r

The global diversity of Haemonchus contortus is shaped by human intervention and climate.

Haemonchus contortus is a haematophagous parasitic nematode of veterinary interest. We have performed a survey of its genome-wide diversity using single-worm whole genome sequencing of 223 individuals sampled from 19 isolates spanning five continents. We find an African origin for the species, together with evidence for parasites spreading during the transatlantic slave trade and colonisation of Australia. Strong selective sweeps surrounding the β-tubulin locus, a target of benzimidazole anthelmintic drug, are identified in independent populations. These sweeps are further supported by signals

Pathology of Haemonchus contortus in New World camelids in the southeastern United States: a retrospective review.

Most small ruminant farms in tropical climates are plagued by Haemonchus contortus, a hematophagous, abomasal parasite. Heavy burdens of this parasite can cause anemia, hypoproteinemia, weight loss, and mortality in susceptible animals. Haemonchus contortus is becoming a major health concern in New World camelids as well, namely llamas (Llama glama) and alpacas (Vicugna pacos), yet little research has been conducted regarding its prevalence or pathology in these species. Herein, we present a retrospective review of llamas and alpacas that were admitted to The University of Georgia Veterinary T

Genetic diversity and population structure of Haemonchus contortus isolates from sheep across Xinjiang, China.

Haemonchus contortus, a highly pathogenic gastrointestinal nematode, significantly impacts small ruminant production, causing substantial economic losses in sheep and goat farming. This study examined the genetic diversity and population structure of 171 H. contortus isolates collected from the abomasa of sheep slaughtered across 8 distinct regions in Xinjiang, China. Using sequence analysis, phylogenetic reconstruction and population genetic analyses of the mitochondrial nad4 gene, we identified 163 haplotypes, with haplotype diversity ranging from 0.995 to 1.000 and nucleotide diversity from

EFFECT OF FOUR DIFFERENT PRESERVATIVES ON AUTOFLUORENCE AND PEANUT AGGLUTIN LECTIN BINDING CHARACTERISTICS OF HAEMONCHUS EGGS.

Haemonchus contortus is the most economically important trichostrongyle nematode infecting various ruminants. Fecal egg counts are used to measure the intensity of trichostrongyle infections, but egg morphology alone can't differentiate Haemonchus eggs from the other less pathogenic trichostrongyle genera also infecting ruminants. The lectin, peanut agglutinin (PNA), binds selectively to Haemonchus eggs and has become a useful tool for diagnosing Haemonchus infections during fecal egg counts. Unfortunately, this binding is not stable when eggs are stored either within fecal samples or when iso

Implantation of a vaccine platform for extended antigen release (VPEAR) induces long-term immunity against Haemonchus contortus in sheep.

The nematode Haemonchus contortus causes severe anemia in sheep and goats. Drug-resistant isolates are common, prompting a need for parasite control measures beyond chemotherapeutics. Vaccination is one promising approach for mitigation of clinical signs associated with haemonchosis. One challenge for H. contortus vaccine efforts is the need to administer repeated boosting doses at regular intervals. In this study, we evaluated a vaccine platform for extended antigen release (VPEAR) designed to initiate and maintain long-term immunity following a single immunization event in sheep. We compared

Ethiopian isolates of nematophagus fungi have effectively killed Haemonchus contortus infective larvae in vitro.

INTRODUCTION: The effectiveness of anthelmintics in controlling gastrointestinal parasitism in livestock is increasingly threatened by the emergence of drug resistance. To address this issue, biological control methods have been proposed as potential alternatives. This research aimed to evaluate the efficacy of nematophagous fungi isolated from soil samples against Haemonchus contortus infective larvae. MATERIAL AND METHODS: Soil samples were collected from Ambo and Hawasa, representing the central and southern regions of Ethiopia, respectively. The fungi were cultured on 2% water agar using

Threshold levels of artificial infection with Haemonchus contortus impacting lamb physiology and production.

This study examined the effects of levels of artificial Haemonchus contortus infection on lamb behaviour and physiology. Forty 6-month-old Romney-cross wethers were assigned to one of four treatments (n = 10/treatment): 8000 L3 larvae (High; H), 4000 L3 larvae (Medium; M), 1000 L3 larvae (Low; L), or water (Control; C). Lambs were monitored 1 week pre- and 6 weeks post-treatment. Weekly measurements included faecal egg counts, liveweight, blood counts, and cytokine concentrations (e.g., IL-6). Behaviour was continuously monitored using cameras and accelerometers. Faecal egg counts increased in

El Niño and La Niña differentially drive transmission dynamics of the small ruminant parasite Haemonchus contortus across agroecological zones in Southern Africa.

The El Niño La Niña Southern Oscillation (ENSO) exerts a significant influence on agroecological environments and plays a crucial role in influencing crop productivity, yet its impact on animal diseases has been largely overlooked, despite the evident impact of climate on disease transmission. This study aimed to evaluate the impact of ENSO on the transmission potential (Q0) of Haemonchus contortus (H. contortus) across agroecological zones (AEZs) in southern Africa across the region's typical rainy season (November-April). Key results indicate that stronger El Niño events can significantly de

Chromosome-contiguous genome for the Haecon-5 strain of Haemonchus contortus reveals marked genetic variability and enables the discovery of essential gene candidates.

Millions of livestock animals worldwide are infected with the haematophagous barber's pole worm, Haemonchus contortus, the aetiological agent of haemonchosis. Despite the major significance of this parasite worldwide and its widespread resistance to current treatments, the lack of a high-quality genome for the well-defined strain of this parasite from Australia, called Haecon-5, has constrained research in a number of areas including host-parasite interactions, drug discovery and population genetics. To enable research in these areas, we report here a chromosome-contiguous genome (∼280 Mb) for

Ramming the parasites: Evaluation of quarantine procedures against Haemonchus contortus at sheep markets in Sweden.

In Sweden, it is recommended to treat rams at sheep markets with ivermectin and then keep them in quarantine to minimise the risk of transmission of Haemonchus contortus between farms. The aim of this study was to evaluate the effectiveness of this practise. Accordingly the gastrointestinal parasite status of 50 rams purchased at two sheep markets in central Sweden in autumn 2019 was investigated using faecal samples collected on the day of sale (test 1), 12 ± 3 days after ivermectin injection (test 2, in quarantine) and a few months later before inclusion in the new flocks (test 3). We used b

Ivermectin induces oxidative stress and mitochondrial damage in Haemonchus contortus.

Haemonchus contortus poses a severe threat to livestock, leading to substantial economic losses. The extensive use of ivermectin (IVM), an effective anthelmintic drug, has resulted in the development of resistant strains, reducing the drug's effectiveness and making disease management more challenging. Hence, understanding the underlying mechanisms of IVM resistance is crucial. IVM has been shown to induce apoptosis and oxidative stress imbalance in cancer cells, thereby inhibiting tumor growth; however, whether there is a similar impact on H. contortus has not been well-documented. In this st

Population replacement of benzimidazole-resistant Haemonchus contortus with susceptible strains: evidence of changes in the resistance status.

The spread of anthelmintic resistance (AR) in nematode populations threatens the viability of sheep production systems worldwide, and warrants the adoption of sensitive, practical, and standardized tests to detect AR. The aim of this study was to characterize the replacement of an Haemonchus contortus population resistant to benzimidazoles (BZDs) by a susceptible one, by means of both phenotypic and genotypic techniques. Phenotypic methods to assess BZD resistance included in vivo tests, such as the fecal egg count reduction test (FECRT), and in vitro tests, such as the egg hatch assay (EHA).

Vaccines against blood-feeding nematodes of humans and livestock.

This paper summarises the progress towards vaccine development against the major blood-feeding nematodes of man and livestock, the hookworms and Haemonchus contortus, respectively. The impact of the diseases and the drivers for vaccine development are summarized as well as the anticipated impact of the host immune response on vaccine design. The performance requirements are discussed and progress towards these objectives using defined larval and adult antigens, many of these being shared between species. Specific examples include the Ancylostoma secreted proteins and homologues in Haemonchus a
