BACKGROUND: The small ruminant parasite Haemonchus contortus is the most widely used parasitic nematode in drug discovery, vaccine development and anthelmintic resistance research. Its remarkable propensity to develop resistance threatens the viability of the sheep industry in many regions of the world and provides a cautionary example of the effect of mass drug administration to control parasitic nematodes. Its phylogenetic position makes it particularly well placed for comparison with the free-living nematode Caenorhabditis elegans and the most economically important parasites of livestock a

This study evaluated the effects of homeopathic treatment on control of Haemonchus contortus infection in sheep. Twenty lambs were randomized to three treatments: treated with the homeopathic medicines, Ferrum phosphoricum, Arsenicum album and Calcarea carbonica; treated with a conventional antihelminthic, doramectin, and an untreated control group. Fecal and blood samples were taken from each animal on days 18, 38 and 68 after start of treatment. A significant reduction in number of H. contortus larvae (p<0.01) was observed for animals in the homeopathic treatment group compared to the contro

Fourteen nematode parasite-naïve kids were given a mixed larval inoculum (approximately 17,250 infective larvae) via stomach tube. Twenty-two days after infection, the kids were randomly assigned to control and treatment groups of seven animals each. Mean eggs per gram for control and treatment groups were 4304 and 5315, respectively. Kids in the treatment group were given ivermectin (0.2 mg kg-1) administered by subcutaneous injection. At necropsy, the mean numbers of Haemonchus contortus and Trichostrongylus colubriformis for control and treatment groups were 2259 and 0, and 3033 and 773, re

The Th17 immune response is a newly identified response that may play an important role in resistance to Haemonchus contortus (H. contortus) infection. Recombinant protein Hcftt-2 (rHcftt-2) has been shown to impart a degree of immunoprotection in infected goats and is capable of inducing a Th17 immune response. To identify the peptides primarily responsible for inducing the Th17 response, four peptides (Ftt-1, Ftt-2, Ftt-3, and Ftt-4) derived from rHcftt-2 were amplified and expressed. Our studies established that peptide Ftt-1 induces goat PBMCs to secrete IL‑17 in vitro by activating STAT3/

Benzimidazole resistance in Haemonchus contortus represents a widespread challenge in small ruminant production worldwide. Benzimidazoles remain one of the most commonly used drug classes, due to their broad spectrum of activity, affordability, and ease of administration. However, extensive and often inappropriate use of these compounds has led to the rapid emergence and global spread of resistance, particularly in H. contortus, a highly pathogenic and genetically diverse gastrointestinal nematode. Resistance to benzimidazoles is primarily associated with point mutations in the isotype-1 β-tub

BACKGROUND: The gastrointestinal nematode, Haemonchus contortus (Barber's pole worm), is a member of the Trichostrongilidae family with a major ovine parasite that resides in the abomasum of sheep, goats, and other ruminants. The spicule lengths provided the quickest and easiest character for separating most populations of H. contortus and Haemonchus placei.
AIM: Detection and diagnosis of H. contortus in slaughtered small ruminants from Basrah city using PCR technique with specific genes as targets: ITS2 gene (HcI) of ribosomal DNA and (Nad4) gene of mitochondrial DNA. Then, the gene sequenc

RNA interference (RNAi) on parasitic nematodes has been described as a valuable tool for screening putative targets that could be used as novel drug and/or vaccine candidates. This study aimed to set up a pipeline to identify potential targets using RNAi for vaccine/anti-parasite therapy development against Haemonchus contortus, a blood-feeding abomasal nematode parasite. The available H. contortus sequence data was mined for targets, which were tested for essentiality using RNAi electroporation assays. A total of 56 genes were identified and tested for knockdown using electroporation of first

Haemonchus contortus is a blood-sucking gastrointestinal nematode that infects all ruminants and causes significant economic losses in production. Characterizing the genetic variability of H. contortus populations is crucial for understanding patterns of disease transmission and developing effective control strategies against haemonchosis. This study aimed to identify the genetic variability of H. contortus isolates in small ruminants from slaughterhouses in Bangladesh. During January to December 2015, 400 abomasa samples were collected and 186 were found to be positive for Haemonchus. A 321-b

Anthelmintic resistance in grazing livestock systems has been spreading worldwide in prevalence and severity. Therefore, alternative measures including the use of herbal anthelmintic is considered as one of the successful approaches for the control of anthelmintic resistance. In the present report, we describe the chemical constituents of Myrtus communis essential oil, its in vitro anthelmintic effect against the most pathogenic gastrointestinal parasite of sheep; Haemonchus contortus and its in vivo anthelmintic potential using an in vivo gastrointestinal parasite model of rodents; i.e. Helig

Haemonchus contortus is a parasitic nematode of ruminants which causes significant losses to many farmers worldwide. Since the drugs currently in use for the treatment of haemonchosis are losing their effectiveness due to the drug-resistance of this nematode, a new or repurposed drug is highly needed. As the antipsychotic drug sertraline (SRT) has been shown to be effective against the parasitic nematodes Trichuris muris, Ancylostoma caninum and Schistosoma mansoni, the aim of the present study was to evaluate the possible effect of SRT on H. contortus. The potential hepatotoxicity of SRT was

AIM: Haemonchus contortus is the most pathogenic nematode infesting the digestive tract of goats and sheep worldwide leading to a tremendous loss in a variety of routes. Economic losses due to haemonchosis in subtropic and tropic areas are usually caused by poor weight gain, minimized growth, loss of production, and mortality. The prevalence of haemonchosis in Indonesia is 89.4% in goat, and annual loss achieved 1 million US dollars. This study evaluated in vitro effects of Gigantochloa apus crude aqueous extract as an anthelmintic on H. contortus morphology and morphometry.

Haemonchus contortus is one of the most important parasite nematodes of small ruminants around the world and causes great economic losses in livestock production. Control of gastrointestinal nematode infections, like haemonchosis, relies mainly on anthelmintic drugs, but its excessive and inappropriate use has caused serious drug resistance issues in many countries, including Uruguay, where sheep production occupies an important place in the country's economy. Benzimidazole (BZ) anthelmintics have been used for decades to treat sheep against H. contortus infection and resistance to this anthel

ETHNOPHARMACOLOGY RELEVANCE: Acacia cochliacantha is a small tree whose foliage is traditionally used in Mexico for treatment of kidney pain, gastrointestinal illnesses and to kill intestinal parasites. In recent decades, the study of vegetal extracts has offered other possible alternatives for the control of Haemonchus contortus. Considering that this nematode affects dramatically the health and productivity of small ruminants, the aim of this study was to identify the anthelmintic compounds from A. cochliacantha hydro-alcoholic extract (HA-E) through an ovicidal test.

Acetylcholine receptors (AChRs) are pentameric ligand-gated ion channels involved in the neurotransmission of both vertebrates and invertebrates. A number of anthelmintic compounds like levamisole and pyrantel target the AChRs of nematodes producing spastic paralysis of the worms. The muscle AChRs of nematode parasites fall into three pharmacological classes that are preferentially activated by the cholinergic agonists levamisole (L-type), nicotine (N-type) and bephenium (B-type), respectively. Despite a number of studies of the B-type AChR in parasitic species, this receptor remains to be cha
