Dimorphic frataxin and its gene regulation by sex steroids in hamsters.

The role of frataxin (FXN) has been studied extensively in Friedreich ataxia patients, however, the molecular bases underlining the sex steroid-dependent gene expression profiles of FXN in adult tissues are unknown. I describe the molecular characterization of hamster FXN by examining the sexually dimorphic expression and its regulation by sex steroids. Sequence analysis of FXN cDNA showed 630 bp-long ORF encoding 209 amino acids. qPCR analysis revealed that FXN is detected in a wide range of tissues, with the highest expression in the heart, liver, and epididymis, and the weakest expression i

The interaction of ascorbic acid with hemoglobin: Relevance to hemin release and lipid peroxidation.

The interactions between L-ascorbic acid (i.e., Vitamin C, VC) and bovine hemoglobin (Hb) were comprehensively analyzed using fluorescence, circular dichroism, UV-vis absorption spectra and molecular docking method, to elucidate the structural mechanisms through which VC altered Hb redox states and stabilities. VC interacted with the central cavities of Hb to form Hb-VC complex via static quenching mechanism involving nonradiative energy transfer, with van der Waals forces and hydrogen bonds playing predominant roles in the binding processes. VC was surrounded by residues such as α1-Thr137, α1

Lucina pectinata oxyhemoglobin (II-III) heterodimer pH susceptibility.

Lucina pectinata live in high concentrations of hydrogen sulfide (H2S) and contains one hemoglobin, Hemoglobin I (HbI), transporting H2S and two hemoglobins, Hemoglobin II (HbII) and Hemoglobin (HbIII), transferring dioxygen to symbionts. HbII and HbIII contain B10 tyrosine (Tyr) and E7 glutamine (Gln) in the heme pocket generating an efficient hydrogen bonding network with the (HbII-HbIII)-O2 species, leading to very low ligand dissociation rates. The results indicate that the oxy-hemeprotein is susceptible to pH from 4 to 9, at acidic conditions, and as a function of the potassium ferricyani

The hematin-dihydroartemisinin adduct mobilizes a potent mechanism to suppress β-hematin crystallization.

Malaria remains a significant public health challenge in equatorial regions of the world, largely owing to the parasite's emerging resistance to the recently introduced drugs of the artemisinin (ART) family. In the human body, most ART-derivative drugs are metabolized to dihydroartemisinin (DHA), which, in the parasite, after activation by heme, can form a hematin-dihydroartemisinin adduct (H-DHA). Here we test whether and how H-DHA inhibits hematin crystallization, the main constituent of the heme detoxification pathway of malaria parasites. We find that H-DHA is a poor inhibitor of classical

Mechanism of Peroxynitrite Interaction with Ferric M. tuberculosis Nitrobindin: A Computational Study.

Nitrobindins (Nbs) are all-β-barrel heme proteins present along the evolutionary ladder. They display a highly solvent-exposed ferric heme group with the iron atom being coordinated by the proximal His residue and a water molecule at the distal position. Ferric nitrobindins (Nb(III)) play a role in the conversion of toxic peroxynitrite (ONOO-) to harmless nitrate, with the value of the second-order rate constant being similar to those of most heme proteins. The value of the second-order rate constant of Nbs increases as the pH decreases; this suggests that Nb(III) preferentially reacts with pe

Evaluation of a point-of-care haemozoin assay (Gazelle device) for rapid detection of Plasmodium knowlesi malaria.

Plasmodium knowlesi is the major cause of zoonotic malaria in Southeast Asia. Rapid and accurate diagnosis enables effective clinical management. A novel malaria diagnostic tool, Gazelle (Hemex Health, USA) detects haemozoin, a by-product of haem metabolism found in all Plasmodium infections. A pilot phase refined the Gazelle haemozoin identification algorithm, with the algorithm then tested against reference PCR in a larger cohort of patients with P. knowlesi mono-infections and febrile malaria-negative controls. Limit-of-detection analysis was conducted on a subset of P. knowlesi samples ser

A second mechanism employed by artemisinins to suppress Plasmodium falciparum hinges on inhibition of hematin crystallization.

Malaria is a pervasive disease that affects millions of lives each year in equatorial regions of the world. During the erythrocytic phase of the parasite life cycle, Plasmodium falciparum invades red blood cells, where it catabolizes hemoglobin and sequesters the released toxic heme as innocuous hemozoin crystals. Artemisinin (ART)-class drugs are activated in vivo by newly released heme, which creates a carbon-centered radical that markedly reduces parasite density. Radical damage to parasite lipids and proteins is perceived to be ARTs' dominant mechanism of action. By contrast, quinoline-cla

Comprehensive Insight into the Protein-Surface Biomolecular Interactions on a Smart Material: Complex Formation between Poly(N-vinyl Caprolactam) and Heme Protein.

Proteins are naturally occurring biopolymers that exhibit a wide range of functional applications. Meticulous knowledge about biomolecular interactions between polymeric biomaterials and body fluids or proteins is essential for designing biospecific surfaces and understanding protein-polymer interactions beyond existing limitations. In this regard, we studied the comparative effect of heme proteins such as cytochrome c, myoglobin, and hemoglobin on the phase behavior of poly(N-vinyl caprolactam) (PVCL) aqueous solution and demonstrated various biomolecular interactions in the polymer-protein c

Prediction Model for Antimalarial Activities of Hemozoin Inhibitors by Using Physicochemical Properties.

The rapid spread of strains of malaria parasites that are resistant to several drugs has threatened global malaria control. Hence, the aim of this study was to predict the antimalarial activity of chemical compounds that possess anti-hemozoin-formation activity as a new means of antimalarial drug discovery. After the initial in vitro anti-hemozoin-formation high-throughput screening (HTS) of 9,600 compounds, a total of 224 hit compounds were identified as hemozoin inhibitors. These 224 compounds were tested for in vitro erythrocytic antimalarial activity at 10 μM by using chloroquine-mefloquin

Oxygen Activation and Radical Transformations in Heme Proteins and Metalloporphyrins.

As a result of the adaptation of life to an aerobic environment, nature has evolved a panoply of metalloproteins for oxidative metabolism and protection against reactive oxygen species. Despite the diverse structures and functions of these proteins, they share common mechanistic grounds. An open-shell transition metal like iron or copper is employed to interact with O2 and its derived intermediates such as hydrogen peroxide to afford a variety of metal-oxygen intermediates. These reactive intermediates, including metal-superoxo, -(hydro)peroxo, and high-valent metal-oxo species, are the basis

Comprehensive characterization of the major nemoglobin from the giant kidney worm Dioctophyme renale.

Globins are widely distributed heme proteins involved in oxygen transport and redox-related processes. Their diversity and functional roles remain poorly understood in nematodes. Dioctophyme renale, the giant kidney worm, is a Clade I nematode that infects mammals, including humans, yet no globin from this group has been experimentally characterized. Here we report the first biochemical and biophysical characterization of a Clade I nemoglobin, P17, an abundant soluble globin isolated from pseudocoelomic fluid of D. renale and responsible for its characteristic red pigmentation. Structural mode

Heme limitation induces LHR2, an essential gene for Leishmania pathogenesis.

Leishmania spp. are intracellular parasites that cause leishmaniasis, a devastating disease with no effective treatment. These parasites are heme auxotrophs and must scavenge this essential cofactor from the host. Transcriptomic analysis of Leishmania major promastigotes cultured in the presence or absence of heme revealed numerous differentially expressed genes. Among those of unknown function, LHR2 (Leishmania Heme Response-2) was the most upregulated gene in response to heme limitation. LHR2 encodes a mitochondrial hemoprotein that likely protects this organelle from elevated levels of reac

Clearing of hemozoin crystals in malaria parasites enables whole-cell STED microscopy.

Malaria is a devastating mosquito-borne parasitic disease that manifests when Plasmodium parasites replicate within red blood cells. During the development within the red blood cell, the parasite digests hemoglobin and crystalizes the otherwise toxic heme. The resulting hemozoin crystals limit imaging by STED nanoscopy owing to their high light-absorbing capacity, which leads to immediate cell destruction upon contact with the laser. Here, we establish CUBIC-P-based clearing of hemozoin crystals, enabling whole-cell STED nanoscopy of parasites within red blood cells. Hemozoin-cleared infected

Erythroid spectrin binding modulates peroxidase and catalase activity of heme proteins.

Hemoglobin oxidation due to oxidative stress and disease conditions leads to the generation of ROS (reactive oxygen species) and membrane attachment of hemoglobin in-vivo, where its redox activity leads to peroxidative damage of membrane lipids and proteins. Spectrin, the major component of the red blood cell (RBC) membrane skeleton, is known to interact with hemoglobin and, here this interaction is shown to increase hemoglobin peroxidase activity in the presence of reducing substrate ABTS (2', 2'-Azino-Bis-3-Ethylbenzothiazoline-6-Sulfonic Acid). It is also shown that in the absence of reduci

Heme as key regulator of major mammalian cellular functions: molecular, cellular, and pharmacological aspects.

Heme (iron protoporphyrin IX) exists as prosthetic group in several hemoproteins, which include respiration cytochromes, gas sensors, P450 enzymes (CYPs), catalases, peroxidases, nitric oxide synthases (NOS), guanyl cyclases, and even transcriptional factors. Hemin (the oxidized form of iron protoporphyrin IX) on the other hand is an essential regulator of gene expression and growth promoter of hematopoietic progenitor cells. This review is focused on the major developments occurred in this field of heme biosynthesis and catabolism and their implications in our understanding the pathogenesis o

Spectral demonstration of semihemoglobin formation during CN-hemin incorporation into human apohemoglobins.

The incorporation of CN-hemin into three human adult apohemoglobin species (apohemoglobin, alpha-apohemoglobin, and apohemoglobin modified at its beta93 sulfhydryl with p-hydroxymercuribenzoate) has been monitored at micromolar concentrations in 0.05 M potassium phosphate buffer, pH 7.0, at 10 degrees C. In all cases, Soret spectral blue shifts accompanied CN-protohemoglobin but not CN-deuterohemoglobin formation. This finding in conjunction with isofocusing studies provided evidence of a CN-protosemi-alpha-hemoglobin intermediate, the formation of which appeared to be a direct consequence of

Nitric oxide and lung surfactant.

Inhalation of nitric oxide (NO) is an experimental treatment for severe pulmonary hypertension. Being rapidly metabolized by hemoglobin, inhaled NO causes selective vasodilation in the pulmonary vascular bed. In addition to the vascular smooth muscle, other pulmonary structures are exposed to inhaled NO, resulting in suppression of NO synthesis in a variety of pulmonary cells and in potential toxicity. NO is a free radical that interacts with a number of proteins, particularly metalloproteins. Together with superoxide radical, it rapidly forms highly toxic peroxynitrite. Peroxynitrite is invol

[Does nitric oxide stress exist?].

Ten years ago, the term "oxidative stress" (sigma -O2) was created to define oxidative damage inflicted to the organism. This definition brings together processes involving reactive oxygen species production and action such as free radical production during univalent reduction of oxygen within mitochondria, activation of NADPH-dependent oxidase system on the membrane surface of neutrophils, flavoprotein-catalyzed redox cycling of xenobiotics and exposure to chemical and physical agents in the environment. Since the discovery of the nitric oxide biosynthetic pathway, the deleterious effects of

Neuroglobin reduces stress-induced oxidative damage and amyloid burden in a three-dimensional human neuroblastoma cell culture.

Neuroglobin (Ngb) is a nerve hemeprotein that acts as a neuroprotectant against multiple brain injuries, including oxidative stress and β-amyloid toxicity. Besides inhibiting apoptosis and scavenging ROS/RNS, Ngb overexpression promotes autophagy, a highly conserved catabolic process essential for maintaining cellular homeostasis under both basal and stress conditions. Although Ngb upregulation has proven effective in animal models of neurological diseases, its effects have not yet been studied in three-dimensional (3D) cell cultures, which better mimic in vivo tissue architecture than monolay

Leghemoglobin is nitrated in functional legume nodules in a tyrosine residue within the heme cavity by a nitrite/peroxide-dependent mechanism.

Protein tyrosine (Tyr) nitration is a post-translational modification yielding 3-nitrotyrosine (NO2 -Tyr). Formation of NO2 -Tyr is generally considered as a marker of nitro-oxidative stress and is involved in some human pathophysiological disorders, but has been poorly studied in plants. Leghemoglobin (Lb) is an abundant hemeprotein of legume nodules that plays an essential role as an O2 transporter. Liquid chromatography coupled to tandem mass spectrometry was used for a targeted search and quantification of NO2 -Tyr in Lb. For all Lbs examined, Tyr30, located in the distal heme pocket, is t
