Comparative metabolism of benzo[a]pyrene and drugs in human liver.

The oxidative metabolism of antipyrine, hexobarbital, coumarin, zoxazolamine, 7-ethoxycoumarin, and the chemical carcinogen benzo[a]pyrene (BP) was studied in 32 adult human livers obtained at autopsy. When enzyme activity for one substrate was plotted against enzyme activity for a second substrate for each of the 32 livers, statistically significant correlations were found between the rates of metabolism of BP and the rates of metabolism of each of the other five drug substrates. The degree of correlation was dependent upon the substrate pair that was studied. Highly significant statistical c

[The effect of epiphysectomy on the dynamics of circadian mobility in rats].

In the paper were studied the rats' individual circadian locomotor activity rhythm patterns and their day and night time sensitivity to hexobarbital sodium and diazepam. Pinealectomy without altering the rhythm patterns significantly increased their amplitude and enhanced the hypnotic effect of hexobarbital both in the daytime and at night and that of diazepam only in the daytime. The animal rhythm patterns and the circadian variations of their sensitivity to the hypnotic drugs were suggested to depend on the individual variations of the pineal activity.

Inhibition of hepatic drug-metabolizing enzymes by arachidonic acid.

1. The effects of arachidonic acid on hepatic drug-metabolizing enzymes was investigated in male ICR-Swiss mice. 2. A single administration of arachidonic acid, 100 mg/kg i.p., doubled the hexobarbital sleeping time. Arachidonic acid in vitro gave a type I binding spectrum with hepatic microsomes; it inhibited the metabolism of hexobarbital and of ethylmorphine, two type I binding drugs, but not that of aniline, a type II binding drug; the inhibition of hexobarbital metabolism by arachidonic acid was competitive. 3. Repeated administration of arachidonic acid up to a total dose of 1000 mg/kg i

Effect of repeated exposure to aniline, nitrobenzene, and benzene on liver microsomal metabolism in the rat.

Exposure of rats to aniline at daily doses of 50 mg/kg of body weight over a month stimulated the microsomal metabolism as manifested by (1) acceleration of p-hydroxylation of anilin and N-demethylation of aminopyrine in 9-000 times g postmitochondrial supernatant of the liver, (2) shortening the sleeping time after hexobarbital, and (3) reduction of the antipyretic effect of phenacetin. In the rats exposed to nitrobenzene in a similar manner to aniline, nitroreduction of nitrobenzene and p-hydroxylation of aniline remained unaffected; the antipyretic effect of phenacetin was decreased, wherea

A comparative study of the effect of some steroid hormones on the response of rats to other drugs.

The effect of a series of steroid hormones on the pentetrazol convulsing action, hexobarbital narcotic action and hepatic drug metabolizing enzyme activities was determined in rats. All steroid compounds used antagonized the pentetrazol effect: the most potent was cortisone and the least potent testosterone. Glucocorticoids and androgens shortened the hexobarbital sleeping time and increased the hepatic drug metabolizing enzyme activity. Estradiol exhibited the opposite effect, whereas progesterone and desoxycorticosterone did not affect these two parameters.

Effect of carbon monoxide inhalation exposure in mice on drug metabolism in vivo.

Experiments were undertaken to evaluate the action of carbon monoxide (CO) on the mixed-function oxidase (MFO) system in vivo. Mice were exposed to 500 ppm CO for 8 h per day in an inhalation chamber under dynamic airflow conditions. Hexobarbital (150 mg/kg, i.p.), zoxazolamine (150 mg/kg, i.p.) or ethanol (2 mg/g, i.p.) was given to each group of mice during CO exposure and disappearance of the drug from blood or brain was determined while CO exposure continued. The experiments were repeated with different groups of animals which were exposed to CO for 3 or 5 days. Hexobarbital and ethanol me

Changes in rat liver monooxygenase activities after administration of atenolol, nifedipine and diltiazem alone and in combination.

The effects of the Ca2+ antagonists nifedipine (NF) and diltiazem (DL) and of the cardioselective beta 1-adrenergic blocking agent atenolol (AT) on the hexobarbital (HB) sleeping time and on the activity of some liver drug-metabolizing enzyme systems in male Wistar rats were studied. Two hours after single oral administration, atenolol (150 mg/kg) did not change hexobarbital sleeping time, while nifedipine (50 mg/kg) and diltiazem (30 mg/kg) prolonged it by 171.2 and 99.6%, respectively. Coadministration of atenolol with diltiazem or with nifedipine significantly prolonged hexobarbital sleep b

Effect of cannabidiol on cytochrome P-450 isozymes.

Cannabidiol (CBD) has been shown to inhibit mouse hepatic mixed-function oxidations of several drugs after acute treatment, whereas repetitive treatment resulted in the restoration of drug-metabolizing capabilities. We have found that acute CBD treatment modestly decreased cytochrome P-450 content but markedly decreased hexobarbital hydroxylase, erythromycin N-demethylase, and 6 beta-testosterone hydroxylase activities. Repetitive CBD treatment, on the other hand, resulted in the restoration of cytochrome P-450 content as well as hexobarbital hydroxylase and erythromycin N-demethylase activiti

Activity of some mixed function oxidases in prenatally carbon disulphide (CS2) treated rats.

Prenatal exposure of albino rats to carbon disulphide (CS2) at 10 and 0.03 mg/m3 led to inhibition and retardation of the development of mixed function oxidase system as shown by the prolongation of hexobarbital sleeping time in the early postnatal period. The rate of the effect was dose-dependent. The speed of recovery of the normal function of the system depended upon the rate of its initial inhibition. Hexobarbital sleeping time in the early postnatal period is suggested as a sensitive indicator of the effect of prenatal CS2 exposure.

Induction of hypersensitivity of brain by hydralazine treatment in rats.

Pretreatment with hydralazine caused prolongation of hypnosis induced by hexobarbital (80 mg/kg, i.p.) or intracerebroventricular injection of phenobarbital (1 mg/rat). Under the same condition, brain concentration of hexobarbital was lower than that of no hydralazine treatment. In addition, no appreciable change in liver microsomal drug metabolizing enzyme activities of rats after hydralazine treatment was observed. These findings strongly suggested that hydralazine increases the sensitivity of brain to barbiturates, which leads to prolongation of sleeping time without inhibition of drug meta

Effect of manganese on the hepatic microsomal mixed function oxidase enzyme system in the rat.

Experiments were conducted to examine the effect of manganese on the hepatic mixed function oxidase system in the rat. Acute treatment with manganese chloride (1-10 mg Mn/kg, ip) produced a significant prolongation of hexobarbital hypnosis in male rats on Days 2 and 3 following metal administration. The threshold dose of manganese to produce this alteration in response was 5 mg Mn/kg and the altered response returned to control values by Day 5. The prolonged hexobarbital hypnosis resulted from Mn inhibition of the hepatic microsomal mixed function oxidase system, the activity of which was asse

Effect of cimetidine or ranitidine pretreatment on hepatic mixed function oxidase activity in the rat.

This study compared the effect of single equimolar oral doses of cimetidine (100 mg/kg) or ranitidine (139 mg/kg) on rat hepatic mixed function oxidases. Cimetidine significantly (p less than 0.05) increased hexobarbital sleeping time and prolonged aminopyrine and theophylline elimination. In contrast, ranitidine did not significantly affect hexobarbital sleeping time and theophylline elimination but significantly (p less than 0.025) increased aminopyrine elimination. Aminopyrine N-demethylase activity in vitro was significantly (p less than 0.05) inhibited by cimetidine pretreatment but signi

Loss of substrate binding capacity of the hepatic microsomal cytochrome P-450 in Fasciola hepatica infected rats: toxicological implications.

Experimental fascioliasis in the rat is responsible for a dramatic decrease in the drug-metabolizing ability of the hepatic monooxygenase system. The present investigation, through a spectroscopic study of hexobarbital interaction with microsomal cytochrome P-450 and in vitro and in vivo studies of hexobarbital metabolism in the rat, demonstrates that this decrease is due to an alteration in the structure of the hemoprotein (loss of substrate binding capacity of cytochrome P-450 followed by denaturation). These results might be responsible for a decreased safety margins for those flukicidal ag

The effects of atropine on the tolerance and the convulsions seen after withdrawal from forced barbital drinking in the rat.

Male rats were forced to drink a barbital solution as their only drinking fluid for 33 weeks. During the last part of the treatment the average dose of barbital was around 200 mg/kg/day. In the abstinence period after barbital treatment, tolerance was recorded with a hexobarbital anaesthesia threshold utilizing an EEG criterium. Convulsions were recorded in jiggle cages. On days 3 and 28 of the abstinence period, i.p. pretreatment with atropine (2, 4, and 8 mg/kg) was given 1.5 h before threshold determinations. The barbital treatment induced a clear tolerance to the hexobarbital, which still

Expression of a human liver cytochrome P-450 protein with tolbutamide hydroxylase activity in Saccharomyces cerevisiae.

The human liver cytochrome P-450 (P-450) proteins responsible for catalyzing the oxidation of mephenytoin, tolbutamide, and hexobarbital are encoded by a multigene family (CYP2C). Although several cDNA clones and proteins related to this "P-450MP" family have been isolated, assignment of specific catalytic activities remains uncertain. Sulfaphenazole was found to inhibit tolbutamide hydroxylation to a greater extent than mephenytoin or hexobarbital hydroxylation. The inhibition by sulfaphenazole was competitive for tolbutamide and hexobarbital hydroxylation but with much different Ki values (5

Differences due to species, sex and pregnancy in microsomal induction by diphenylhydantoin in rabbits and rats.

Diphenylhydantoin sodium (DPH), 100 mg/kg/day for 3 days markedly induced aminopyrine, hexobarbital and aniline in vitro biotransformation, and increased P-450 level in microsomes and liver microsomal protein content in female and male rabbits. However, in pregnant rabbits, only aniline biotransformation was stimulated by DPH. In female nonpregnant rats, DPH also significantly stimulated in vitro hepatic biotransformation of aminopyrine, hexobarbital and aniline, and increased P-450 level in microsomes and liver to body weight ratio. In male rats, however, none of the pathways was stimulated s

[Effects of various anesthesia procedures used in forceps delivery on the cardiopulmonary adaptation of the newborn infant. 2. Modification of pulmonary adaptation].

Mature newborns reveal the same pulmonal adaption to extrauterine conditions after forceps delivery in general anesthesia for the mother beginning with propanidid or hexobarbital as newborns after spontaneous delivery. Intervalls of apnoe, being physiological during neonatal adaption up to the fourth life-hour, occurred in all three tested groups in the same frequency. Genuine apnoe in combination with following bradycardia and cyanosis couldn't be observed. Moreover during the whole examination time all mean values of respiration frequency were physiological. Therefore we believe that there i

Drug metabolism in rats with arthritis induced by 6-sulfanilamidoindazole (6-SAI).

Hexobarbital sleeping time was prolonged and ethylmorphine N-demethylation was inhibited after a single dosage or seven administration of 6-SAI to old rats. These effects were independent of the development of arthritis. Changes in cytochrome P-450 concentration after 6-SAI treatment were insignificant and thus not responsible for the decrease in drug metabolism. In vitro 6-SAI inhibited ethylmorphine N-demethylation; the inhibition was of a mixed type. 6-SAI bound to cytochrome P-450 and induced a type II spectrum. The magnitude of hexobarbital-induced type I spectral changes was diminished b
