Fusarium graminearum, the causal agent of Fusarium head blight, is a common pathogen on small grain cereals worldwide and produces various trichothecenes [deoxynivalenol (DON) is predominant] during infection. A previous study has revealed that DON production is positively correlated with the occurrence of carbendazim (MBC) resistance. Here, we identified and characterized two putative genes encoding hexokinase in F. graminearum (FgHXK1 and FgHXK2), which is a rate-limiting enzyme in DON biosynthesis. The expression level of hexokinase genes and the production of pyruvate, which is the precurs

The frequencies of glucose-6-phosphate dehydrogenase (G-6-PD), pyruvate kinase (PK) and hexokinase (HK) deficiency were determined in different regions of Saudi Arabia. G-6-PD deficiency was found to range from 0.045 to 0.220 for the male and 0.020 to 0.125 for the female population. The highest frequencies were found to exist in the regions which are endemic to malarial parasite and have high frequencies of sickle cell and thalassaemia genes. Partial deficiencies of PK and HK were encountered in each region, however, no case of complete deficiency of these enzymes was identified. Further inve

Hyperglycemia is common in patients with bladder cancer and has been implicated in disease progression, yet the molecular link between a high-glucose milieu and tumor aggressiveness remains poorly defined. Here we identify a noncanonical, nuclear role of hexokinase 2 (HK2) that couples systemic hyperglycemia to MYC-driven glycolysis and stemness in bladder cancer. High glucose promotes nuclear translocation of HK2, where HK2 directly binds the central region of MYC to form a functional transcriptional complex. This HK2-MYC complex occupies the promoters of key glycolytic genes, including HK2 a

BACKGROUND AND OBJECTIVE: The aim of this study was to clinically and molecularly characterize the largest cohort of patients with HK1 associated retinal disease. Hexokinase 1 (HK1) variants have been reported to cause retinitis pigmentosa (RP), but a clearly defined genotype-phenotype correlation has not been well established.
PATIENTS AND METHODS: A retrospective, consecutive, single-center case series was performed on patients with molecularly confirmed HK1-associated disease. Patients between 2016 and 2024 were included.
RESULTS: Sixty eyes from 30 patients were assessed. Thirty patients

B lymphocyte activation triggers metabolic reprogramming essential for B cell differentiation and mounting a healthy immune response. Here, we investigate the regulation and function of glucose-phosphorylating enzyme hexokinase 2 (HK2) in B cells. We report that both activation-dependent expression and mitochondrial localization of HK2 are regulated by the phosphatidylinositol 3-kinase (PI3K) signaling pathway. B cell-specific deletion of HK2 in mice caused mild perturbations in B cell development. HK2-deficient B cells show impaired functional responses in vitro and adapt to become less depen

Defective pericyte-endothelial cell interaction in tumors leads to a chaotic, poorly organized and dysfunctional vasculature. However, the underlying mechanism behind this is poorly studied. Herein, we develop a method that combines magnetic beads and flow cytometry cell sorting to isolate pericytes from tumors and normal adjacent tissues from patients with non-small cell lung cancer (NSCLC) and hepatocellular carcinoma (HCC). Pericytes from tumors show defective blood vessel supporting functions when comparing to those obtained from normal tissues. Mechanistically, combined proteomics and met

Chronic myeloid leukemia (CML) is a myeloproliferative disease which uniquely expresses a constitutively active tyrosine kinase, BCR/ABL. As a specific inhibitor of the BCR-ABL tyrosine kinase, imatinib becomes the first choice for the treatment of CML due to its high efficacy and low toxicity. However, the development of imatinib resistance limits the long-term treatment benefits of it in CML patients. In the present study, we aimed to investigate the roles of miR-202 in the regulation of imatinib sensitivity in CML cell lines and the possible mechanisms involved in this process. We found miR

In this study we compare the specific activities and isoenzyme patterns of five enzymes--phosphoglucose isomerase, phosphoglucomutase, hexokinase, lactate dehydrogenase, and alkaline phosphatase--in term placenta with the analogous enzymes in a clone of choriocarcinoma cells grown in culture. Phosphoglucose isomerase, phosphoglucomutase, and lactate dehydrogenase specific activities of the choriocarcinoma did not differ by more than two or three times from the mean activities of the comparable enzymes in placenta; the specific activity of hexokinase in the choriocarcinoma amounted to 14 per ce

The heart is the most energy-demanding organ in the body and has a dynamically adaptable metabolic state. Under normal conditions, the adult heart primarily relies on mitochondrial oxidative phosphorylation for ATP generation, whereas glycolysis supplies a minor portion of the energy. However, under pathological conditions, glycolysis is increased, and mitochondrial oxidative phosphorylation is reduced. Hexokinase 2 (HK2) is the first rate-limiting enzyme in the glycolytic pathway in cardiomyocytes. Accumulating evidence in recent years has indicated that HK2 confers protection to the heart un

Alveolar echinococcosis (AE), caused by Echinococcus multilocularis infection, leads to destructive hepatic lesions and severe hepatic fibrosis. Lactylation, a post-translational modification, has been linked to fibrotic progression in hepatocytes. However, the role of E. multilocularis infection in hepatic lactylation remains unclear. This study aims to elucidate the relationship between hepatic fibrosis and lactylation induced by E. multilocularis infection and fill this knowledge gap. The results showed that lactate concentrations in the livers and sera of AE mice were significantly elevate

BACKGROUND: Facial infiltrating lipomatosis (FIL) is a rare congenital disorder characterized by adipose hyperplasia and osseous overgrowth, driven by somatic PIK3CA mutations. While PIK3CA-induced metabolic reprogramming elevates lactate levels, the role of histone lactylation in FIL pathogenesis remains unclear.
METHODS: Adipose stem and progenitor cells (ASPCs) from FIL adipose tissue were isolated. Glycolysis inhibitors (2-DG, oxamate), lactate supplementation, and siRNA-mediated knockdown were used to modulate lactylation. CUT&Tag sequencing, Western blot, qPCR, ChIP-qPCR and functional

NLRP3 inflammasome activation is a highly regulated process for controlling secretion of the potent inflammatory cytokines IL-1β and IL-18 that are essential during bacterial infection, sterile inflammation, and disease, including colitis, diabetes, Alzheimer's disease, and atherosclerosis. Diverse stimuli activate the NLRP3 inflammasome, and unifying upstream signals has been challenging to identify. Here, we report that a common upstream step in NLRP3 inflammasome activation is the dissociation of the glycolytic enzyme hexokinase 2 from the voltage-dependent anion channel (VDAC) in the outer

With technological advancements, radiotherapy (RT) has become an effective non-surgical treatment for hepatocellular carcinoma (HCC), comprehensively improving the local control rate of patients with HCC. However, some patients with HCC still experience radio-resistance, cancer recurrence, and distant metastasis following RT. Our previous study has revealed that hexokinase 2 (HK2), a potent oncogene, was overexpressed in radio-resistant HCC cell lines; however, its role in HCC radio-resistance remains elusive. Here, we confirmed the upregulation of HK2 in HCC tissue, which is related to unfavo

BACKGROUND: Inflammation plays a pivotal role in the pathogenesis and treatment of sepsis-associated lung injury (S-ALI). Although hexokinase 2 (HK2) is known to be involved in regulating inflammation, metabolism, and mitochondrial homeostasis, its mechanism in S-ALI remains unclear.
METHODS: In the cecal ligation and puncture (CLP)-induced S-ALI mouse model, haploinsufficiency of HK2 or pharmacological inhibition of HK2 by Lonidamine (LND) was administered. The effects of gene silencing or pharmacological inhibition of HK2 on pulmonary inflammation, lipid metabolism and mitochondrial damage

Neuropathic pain, caused by nerve damage, lacks effective treatments. Astrocytes drive chronic neuroinflammation in the dorsal horn, but directly targeting them is challenging due to their heterogeneity and beneficial roles. The regulation of deleterious astrocyte subset emergence in pain is also unknown. Here, we identify distinct astrocyte clusters and elucidate mechanisms by which metabolic regulation of neuroinflammatory astrocyte subsets during pain pathogenesis. We show that the astrocyte specifier Sox9 transcriptionally regulates hexokinase 1 (Hk1), the enzyme that catalyzes the rate-li

INTRODUCTION: Antibodies to hexokinase 1 (HK1) and kelch-like 12 (KLHL12) have been identified as potential biomarkers in primary biliary cholangitis (PBC), and this study assesses changes of these antibodies over time and if they are associated with clinical outcomes.
METHODS: Two hundred fifty-four PBC patients (93.3% female, 51 ± 12.3 years old) were tested for anti-HK1 and anti-KLHL12, antimitochondrial (AMA), anti-gp210, and anti-sp100 antibodies. One hundred sixty-nine patients were tested twice and 49 three times within 4.2 (0.8-10.0) years. Biochemistry and clinical features at diagno

The glycolytic enzyme hexokinase (HK2), which is aberrantly expressed in various types of tumours, is associated with metastasis. However, its role in the progression and metastasis of tongue squamous cell carcinoma (TSCC) remains unclear. The results of our study showed that HK2 expression is often deregulated in TSCC patients. Increased HK2 expression was associated with tumour stage, clinical stage, lymph node metastasis, but not pathological grade, and reduced overall survival. Microarray and western blotting analyses revealed increases in HK2 expression in TSCC cells with higher metastati

The intracellular human bacterial pathogen Chlamydia trachomatis pursues effective strategies to protect infected cells against death-inducing stimuli. Here, we show that Chlamydia trachomatis infection evokes 3-phosphoinositide-dependent protein kinase-1 (PDPK1) signaling to ensure the completion of its developmental cycle, further leading to the phosphorylation and stabilization of MYC. Using biochemical approaches and imaging we demonstrate that Chlamydia-induced PDPK1-MYC signaling induces host hexokinase II (HKII), which becomes enriched and translocated to the mitochondria. Strikingly, p
