Antidiabetic Properties of Histamine H1 Receptor Antagonists: A Systematic Review.

Allergies and diabetes mellitus are highly prevalent, frequently co-occurring diseases. Antihistamines, the first-line treatment for allergies, block H1 receptors but also exhibit anti-inflammatory and antioxidant properties that may improve glucose metabolism. This systematic review examined the evidence for the potential antidiabetic effects of commonly used first- and second-generation antihistamines. A comprehensive search of the PubMed database (1978-May 2026) yielded 17 eligible in vivo, in vitro, clinical, and in silico studies. Most reports indicated that antihistamines improve glucose

Effect of histamine H2-receptor antagonists on vitamin B12 absorption.

OBJECTIVE: To discuss the potential of histamine H2-receptor antagonists (H2RAs) to cause malabsorption of vitamin B12 (cyanocobalamin). DATA SOURCES: Pertinent literature was identified via a MEDLINE search. Journals and references cited in published articles also were used as data sources. STUDY SELECTION: Studies evaluating the effect of H2RAs on vitamin B12 absorption were reviewed. DATA SYNTHESIS: H2RAs decrease acid secretion by the gastric parietal cells. Gastric acid and pepsin produced by these cells are required for the cleavage of vitamin B12 from dietary sources. Intrinsic facto

Some behavioral effects of histamine H1 antagonists in squirrel monkeys.

Graded dose of the histamine H1 antagonists tripelennamine (0.1-3.0 mg/kg), promethazine (0.1-3.0 mg/kg), diphenhydramine (0.3-10.0 mg/kg) and chlorpheniramine (0.3-1.7 mg/kg) increased rates of nonsuppressed responding under a second-order schedule of food presentation to a maximum beyond which responding was increased less or decreased. In contrast, the H2 antagonist, cimetidine, had no effect or decreased responding at the highest doses studied (56.0-100.0 mg/kg). Intermediate doses of tripelennamine, diphenhydramine and promethazine also increased rates of food-maintained responding that w

Histamine H2-receptor antagonists in the treatment of duodenal ulcers.

A double-blind trial of histamine H2-receptor antagonists and placebo was carried out in 46 patients with endoscopically proven duodenal ulceration. H2-receptor antagonists (metiamide and cimetidine) produced a significantly greater degree of completely healed ulcers and fewer complete failures than placebo. The results of the trial indicate that H2-receptor antagonists promote the healing of ulcers. The action of these compounds would appear to be related to the inhibition of acid secretion. Neither the duration of the illness, rate of recurrence of the ulcers of level of gastric acid secreti

Constitutive activity of the histamine H(1) receptor reveals inverse agonism of histamine H(1) receptor antagonists.

Transient expression of the wild-type human histamine H(1) receptor in SV40-immortalised African green monkey kidney cells resulted in an agonist-independent elevation of the basal levels of the second messenger inositoltrisphospate. Several histamine H(1) receptor antagonists, including the therapeutically used anti-allergics cetirizine, loratadine and epinastine reduced this constitutive histamine H(1) receptor activity. Inverse agonism, i.e., stabilisation of an inactive conformation of the human histamine H(1) receptor, may therefore be a key component of the anti-allergic mechanism of act

Are Histamine H3 Antagonists the Definitive Treatment for Acute Methamphetamine Intoxication?

BACKGROUND: Methamphetamine (METH) is classified as a Schedule II stimulant drug under the United Nations Convention on Psychotropic Substances of 1971. METH and other amphetamine analogues (AMPHs) are powerful addictive drugs. Treatments are needed to treat the symptoms of METH addiction, chronic METH use, and acute METH overdose. No effective treatment for METH abuse has been established because alterations of brain functions under the excessive intake of abused drug intake are largely irreversible due in part to brain damage that occurs in the course of chronic METH use. OBJECTIVE: Modulat

Intravenous histamine H2-receptor antagonists and parenteral nutrition: patterns of use.

Patterns of use of i.v. histamine h2-receptor antagonists (H2RAs) and parenteral nutrient solutions in patients receiving both given separately or as admixtures were studied. Consecutive adult patients at a university teaching hospital were placed in an "admixed group," consisting of all those who received an i.v. H2RA as a parenteral nutrient solution additive, or a "nonadmixed group," consisting of all those who received an i.v. H2RA and a parenteral nutrient solution as separate infusions. Data were collected for many variables, including interruptions in therapy, route and method of admini

Structural characteristics of histamine H2 receptor antagonists that scavenge hypochlorous acid.

Gastric and duodenal ulcers are characterized by hypersecretion of gastric acid and pepsin, inflammation of the mucosa and an influx of neutrophils. These cells can produce reactive oxygen species after stimulation. Particularly hydroxyl radicals and hypochlorous acid (HOCl) can be cytotoxic and damage the gastroduodenal mucosa. It has been shown that histamine H2 receptor antagonists such as cimetidine, ranitidine and famotidine are good hydroxyl radical scavengers. This study was undertaken to investigate whether these agents were able to scavenge the cytotoxic HOCl, and if so, which part of

Inhibition of morphine antinociception by centrally administered histamine H2 receptor antagonists.

The actions of zolantidine dimaleate and five other histamine H2 receptor antagonists, given into the lateral ventricle of rats, were assessed on nociceptive responses in the presence and absence of systemically administered morphine. On the tail flick response, zolantidine induced a time- and dose-dependent inhibition of morphine antinociception, with no effect on responses in the absence of morphine. Zolantidine and another H2 receptor antagonist, tiotidine, also inhibited morphine responses in the hot plate test. Four other H2 receptor antagonists of varying structure, brain-penetrating abi

Long-term treatment of systemic mastocytosis with histamine H2 receptor antagonists.

A patient with systemic mastocytosis, accompanied by gastric hypersecretion and duodenal ulcer, was treated with metiamide followed by daily cimetidine for 44 months. Treatment with cimetidine resulted in healing of the ulcer, without recurrence and marked amelioration of the cutaneous symptoms of mastocytosis. Complete suppression of basal gastric hypersecretion was documented after 33 months of treatment and cimetidine and Vitamin B12 absorption remained normal. Cimetidine reduced the patient's cutaneous response to intradermal histamine without affecting leucocyte histamine release. No cime

Effect of histamine-H2-receptor antagonists on the exocrine pancreatic secretion.

Pancreatic exocrine secretion has been studied in 10 patients with documented duodenal ulcer before and after 4 weeks of cimetidine treatment (1 g/die). This chronic 'short term' cimetidine administration did not result in any substantial modification of exo-pancreatic response stimulated by Secretin and Caerulein. From this point of view the drug could be employed in the treatment of pancreatic insufficiency in order to obtain a better utilization of pancreatic enzyme supplement. It is, however, questionable if such a 'long-term' therapy may be responsible for the appearance or the relapse of

The pharmacology of burimamide and metiamide, two histamine H2-receptor antagonists.

Burimamide and metiamide are two histamine H2-receptor antagonists. Evidence is presented that indicates the competitive nature and the specificity of the antagonism. Metiamide is about ten times more potent than burimamide and is also more effective than burimamide when given orally. Both compounds inhibit gastric secretion and the evidence is consistent with this inhibition being due to competitive antagonism of H2 receptors in the gastric mucosa. Burimamide, unlike metiamide, causes release of catecholamines even at dose levels that are just sufficient to produce H2-receptor antagonism. Bur

Histamine H2-receptor antagonists and gastric acid secretion -- a progress report.

Histamine H2-receptor antagonists, including burimamide, metiamide and cimetidine, are effective antagonists of histamine-stimulated acid secretion from mammalian, avian or reptilian gastric mucosa. Acid secretion stimulated by gastrin or pentagastrin is also inhibited by these drugs, but there is disagreement about the effects of these drugs on acid secretion resulting from activation of acetylcholine receptors. Based on the pharmacological evidence possibilities of treatment by these drugs were discussed in cases with excessive stimulation of acid secretion due to high blood levels of histam
