Nitric oxide influences the release of histamine and glutamate in the rat hypothalamus.

To investigate the influence of nitric oxide (NO) on the release of histamine and glutamate, the anterior hypothalamus of anaesthetized rats was superfused through a push-pull cannula either with artificial cerebrospinal fluid (CSF) or with various drugs dissolved in CSF. Hypothalamic superfusion with the NO-donating compounds linsidomine (200 mumol/l) or diethylamine-NO (DEANO, 100 mumol/l) led to a pronounced and sustained decrease in the histamine release rate, whereas the release rate of glutamate was enhanced. Superfusion with the inhibitor of NO synthase L-NG-nitro-L-arginine methyl este

Terfenadine, an H1 antihistamine, inhibits histamine release in vivo in the human.

A double-blind, placebo-controlled clinical trial was performed on 12 subjects with allergic rhinitis to evaluate the effect of 1-wk pretreatment with terfenadine (60 mg twice daily or 300 mg twice daily) on the response to sequential nasal challenges with allergen and histamine. The response to allergen challenge was monitored by counting sneezes and by quantifying the levels of histamine, kinins, TAME-esterase activity, and albumin in recovered nasal lavages. Following histamine challenges, sneezing and the levels of albumin were recorded. Terfenadine significantly reduced sneezing and the l

The inhibitory effect of ketotifen on in vitro histamine release in allergic patients.

Since the discovery of ketotifen as a protective drug of hypersensitivity reactions against an antigen, there have been numerous works that have demonstrated this action both in food and inhalant allergies. In this work we studied the inhibitory capacity of this substance in vitro on the membrane of the basophils of individuals sensitized with positive histamine release test in whole blood against the allergen in question. This was compared with the effect obtained with other drugs (ASA, betamethasone, clemastine, DSCG, oxatomide). Forty-five patients were studied: 30 pollen allergies and 15 w

Development of a new in vivo anaphylactic histamine release assay in rats.

A new method has been developed to detect antigen-induced histamine release in the blood of allergic rats. This in vivo model of systemic anaphylaxis has been utilized to evaluate drugs for histamine release inhibition activity. Compounds were administered by various routes at appropriate times prior to or concomitant with antigen challenge. One minute after challenge the rats were bled into tubes containing heparin and aminoguanidine. Histamine concentrations were determined by a radioenzyme technique after separation of the plasma. Experiments demonstrated that: (a) rats passively sensitized

Induction of histamine release and desensitization in human leukocytes. Effect of anaphylatoxin.

Hog anaphylatoxin (AT) in concentrations from 0.5 to 5 mug/ml gives a dose-dependent histamine release from human leukocytes. Concentration of 100 mug/ml AT give the same high histamine release as 5 mug/ml. This is in contrast to the histamine release obtained with anti-IgE or allergen, which give low histamine release with high doses. The histamine release obtained with AT is completed in 20 sec and the reaction is temperature- and calcium-dependent. Treatment of cells with AT in the presence or absence of calcium makes them insensitive to another challenge with AT. Such treated cells are ful

[Studies on the degranulation and histamine release of purified rat peritoneal mast cells. The inhibitory effect of histamine and other chemicals].

We know that the mast cell or basophil degranulation and the release of chemical mediators such as histamine may play an important role in inducing immediate type allergic reactions. In this experiment, employing purified rat peritoneal mast cells (RPMC) the degranulation pattern of RPMC and percent release of histamine from RPMC by pharmacologic (compound 48/80, kallikrein or peptidoglycan) or allergic (IgE-anti IgE complex) stimuli were examined. The inhibitory effects of catecholamine (isoproterenol), an adenylate cyclase stimulator, methylxanthine (IBMX: 3-isobutyl-1-methyl-xanthine), a ph

Acute central administration of immepip, a histamine H3 receptor agonist, suppresses hypothalamic histamine release and elicits feeding behavior in rats.

Histamine suppresses feeding behavior via histamine H1 receptors in the hypothalamus. This study was performed to examine whether the acute reduction of histamine release in the hypothalamus caused by immepip, a histamine H3 agonist, modulates the feeding behavior of rats. Rats had a catheter implanted in the third cerebral ventricle (i3v) and were given central injections of phosphate-buffered-saline or immepip (100-300 pmol/rat). Following the i3v administration of immepip, the rats developed dose-dependent hypokinesia within 10 min of administration. Next to hypokinesia, the rats showed sig

Influence of histamine release on postoperative vomiting (POV) following gynaecological laparoscopic surgery.

OBJECTIVE: The mechanisms leading to the high incidence of postoperative vomiting (POV) after gynaecological laparoscopic surgery are still unknown. The effectiveness of POV-prophylaxis using H1 + H2-receptor antagonists has been demonstrated, suggesting a role for histamine in the pathogenesis of POV. However, histamine levels were not measured in these studies. The aim of this study was to investigate the incidence of plasma histamine release and its association with POV after gynaecological laparoscopic surgery. MATERIAL OR SUBJECTS: Twenty-two female patients, aged 20-56 y, classified ASA

Ketotifen reduces sneezing but not histamine release following nasal challenge with antigen.

We evaluated the effect of pre-treatment with 1 and 2 mg b.i.d. of ketotifen on the early response to nasal challenge in a double-blind cross-over trial. Weekly nasal challenges were performed in 10 allergic subjects after 1 hr and 1, 2, 3 and 4 weeks of ketotifen administration. The response to nasal challenge was monitored by counting the number of sneezes, the assessment of subjective symptoms, and by measuring the levels of histamine and TAME-esterase activity in recovered nasal lavages. The number of sneezes diminished significantly after a single dose of drug with both the 1 and 2 mg dos

Effect of calmidazolium (R24571) on histamine release from isolated rat mast cells.

The effects of selected calmodulin-antagonists, i.e. calmidazolium (R24571), trifluoperazine, cis- and transflupenthixol, chlorpromazine, and imipramine, on rat mast cells and on mast cell histamine release were investigated. The drugs induced histamine release, apparently by cytotoxic effects, with a rank order of potency in accordance with their lipid solubility and with maximal release at calmidazolium (5 mumol/l), trifluoperazine (40 mumol/l), cis- and trans-flupenthixol (50 mumol/l), chlorpromazine (100 mumol/l), and imipramine (500 mumol/l). Somatostatin-induced histamine release in mast cell incubates from thymusaplastic nude rats.

Both cyclic and linear somatostatin-14 are potent histamine secretagogues in rat mast cell incubates. Linear somatostatin-14 is more effective than cyclic somatostatin-14 in a concentration range up to 10(-6) mol/l, in normal Sprague-Dawley rats, and even more responsive in sensitized hooded Lister rats. This additional mast cell stimulation capacity of linear somatostatin-14 depends on circulating IgE levels. No differences are observed in athymic rat mast cell incubates between the linear and the cyclic isomer of somatostatin-14. A direct unspecific interaction of linear somatostatin with ce

Neurotensin stimulates histamine release from the isolated, spontaneously beating heart of rats.

Neurotensin (NT) evoked a transient, dose-dependent histamine release (ED50 170 ng ml-1) from the rat perfused heart. Histamine release by NT occurred within seconds and lasted less than 2 min. The histamine releasing effect of NT was followed by a dose-dependent increase of the perfusion pressure and a slight tachycardia. The histamine releasing effect of NT was completely abolished in hearts derived from rats pretreated for 3 days with high doses of compound 48/80. The coronary vasoconstrictor effect of NT was increased in hearts derived from compound 48/80-pretreated rats. The mast cell inh

H2 antihistamines augment antigen-induced histamine release from human basophils in vitro.

H2 antihistamines, including cimetidine, burimamide, metiamide, and tiotidine, consistently augmented antigen-induced histamine release from human basophils in vitro when control histamine release was less than 20% of total. This effect was specific to the H2-receptor blocking activity of these drugs: equivalent degrees of receptor blockade by four different H2 antihistamines resulted in equipotent enhancement; H1-receptor antagonists did not alter histamine release; and aminoguanidine and amodiaquine, agents that inhibit histamine metabolism but do not block H2 receptors, did not enhance hist

Dysfunctional inhibitory muscarinic receptors mediate enhanced histamine release in isolated human bronchi.

In human airways mucosal mast cells are under the control of inhibitory muscarinic receptors. The described experiments tested, whether the inhibitory potency of two muscarinic receptor agonists (oxotremorine, acetylcholine) becomes impaired in advanced chronic obstructive pulmonary disease (COPD). Isolated human bronchi obtained from 26 patients with lung cancer were separated into two groups. Group 1 patients suffered from moderate COPD (mean FEV1 56%; range 34-71%; mean pack years of cigarette smoking 50, range 20-96; one non-smoker). Group 2 patients had no or only a mild form of COPD; mea

Physiologic responses and histamine release after nasal antigen challenge. Effect of atropine.

We enrolled nine allergic subjects in a double blind, placebo-controlled study to examine the effect of premedication with 0.6 mg of atropine on nasal antigen challenge. The challenge consisted of unilaterally stimulating the nasal septum with diluent followed by three increasing doses of antigen and recording responses bilaterally. Sneezes, symptoms, and nasal airway resistance (NAR) were recorded. Secretions were collected using preweighed filter paper discs and histamine was measured. Antigen challenge with the subjects on placebo led to significant dose-dependent increases in sneezes, symp

Helicobacter pylori potentiates histamine release from serosal rat mast cells in vitro.

Helicobacter pylori seems to be involved in the etiology of peptic ulcer and chronic gastritis. Histamine is fundamental in gastric secretion modulation, and some features of H. pylori-associated gastritis (edema, vasodilatation, inflammatory cell infiltration) are typical of the histamine-mediated response. This in vitro study has been undertaken as a preliminary step, in order to find a possible link between H. pylori and histamine release. H. pylori isolated from gastric biopsies has been tested as whole washed bacterium, whole formalin-killed bacterium, and crude cell wall preparation with
