[Host-adaptive mechanism of H5N1 avian influenza virus hemagglutininn].

The H5N1 subtype is a highly pathogenic avian influenza virus currently circulating in birds in parts of Asia and northeast Africa, which has caused fatal human infections since 1997. Continuous circulation of the virus in endemic areas has allowed genetically diverse viruses to emerge, increasing the risk of H5N1 human infection. Although human infections with H5N1 have to date been limited, experimental evidence of the aerosol transmission of mutated viruses in a mammalian infection model has revealed the pandemic potential of H5N1 virus. One of the most important viral factors for host-adap

Ecological fitness and strategies of adaptation of Bartonella species to their hosts and vectors.

Bartonella spp. are facultative intracellular bacteria that cause characteristic hostrestricted hemotropic infections in mammals and are typically transmitted by blood-sucking arthropods. In the mammalian reservoir, these bacteria initially infect a yet unrecognized primary niche, which seeds organisms into the blood stream leading to the establishment of a long-lasting intra-erythrocytic bacteremia as the hall-mark of infection. Bacterial type IV secretion systems, which are supra-molecular transporters ancestrally related to bacterial conjugation systems, represent crucial pathogenicity fact

Comparative pangenomics unveils distinct host adaptation trends and conserved biosynthetic potential in microbiome Clostridia.

To thrive in diverse ecological niches, bacteria adopt various lifestyles that range from living freely in the soil to forming close associations with human and animal hosts. However, the impact of these adaptation processes on their genomes and metabolisms remains largely unexplored beyond the genus level. Investigating these evolutionary dynamics at higher taxonomic levels can enhance our understanding of the relationship between host adaptation and functional capabilities. Here, we examine the evolutionary trajectories and metabolic capabilities of the Clostridia class, which displays a wid

SARS-CoV-2 intra-host recombination promotes epistatic spike interactions and temperature-dependent adaptation.

Prolonged infections in immunocompromized individuals are associated with accumulation of unexpected combinations of mutations, as exemplified by the Omicron variant. Here, we show that intra-host evolution during a chronic SARS-CoV-2 infection in the pre-Omicron era was associated with mutations that modulate antibody evasion, cell entry efficiency, spike processing, and fusogenicity. Single spike N-terminal mutations S13I and W64G exerted deleterious impacts on S1/S2 cleavage, syncytia formation, and cell entry but combined through an inferred recombination event with P330S to restore functi

Proteome Remodelling in Candida auris During Early Host Adaptation In Vitro and In Vivo.

Candida auris is an emerging fungal pathogen that is posing a serious global health threat due to its high transmissibility and multidrug resistance profile. Despite recent molecular advances in scrutinizing this enigmatic microbe, much of our understanding regarding its pathomechanisms remains unelucidated. Since microbial pathogenesis is modulated by a dynamic interplay between the host and the pathogen, dissecting such host-pathogen interactions involving C. auris can shed novel insights into its pathogenic cascade. As such, to characterize the virulence repertoire of C. auris, this study a

Impact of Homologous Recombination on Core Genome Evolution and Host Adaptation of Pectobacterium parmentieri.

Homologous recombination is a major force mechanism driving bacterial evolution, host adaptability, and acquisition of novel virulence traits. Pectobacterium parmentieri is a plant bacterial pathogen distributed worldwide, primarily affecting potatoes, by causing soft rot and blackleg diseases. The goal of this investigation was to understand the impact of homologous recombination on the genomic evolution of P. parmentieri. Analysis of P. parmentieri genomes using Roary revealed a dynamic pan-genome with 3,742 core genes and over 55% accessory genome variability. Bayesian population structure

Five centuries of genome evolution and multi-host adaptation of Campylobacter jejuni in Brazil.

Consumption of raw, undercooked or contaminated animal food products is a frequent cause of Campylobacter jejuni infection. Brazil is the world's third largest producer and a major exporter of chicken meat, yet population-level genomic investigations of C. jejuni in the country remain scarce. Analysis of 221 C. jejuni genomes from Brazil shows that the overall core and accessory genomic features of C. jejuni are influenced by the identity of the human or animal source. Of the 60 sequence types detected, ST353 is the most prevalent and consists of samples from chicken and human sources. Notably

The role of influenza-A virus and coronavirus viral glycoprotein cleavage in host adaptation.

While receptor binding is well recognized as a factor in influenza-A virus (IAV) and coronavirus (CoV) host adaptation, the role of viral glycoprotein cleavage has not been studied in detail so far. Interestingly, recent studies suggest that host species may differ in their protease repertoire available for cleavage. Furthermore, it was shown for certain bat-derived CoVs that proteolytic activation provides a critical barrier to infect human cells. Understanding the role of glycoprotein cleavage in different species and how IAV and CoVs adapt to a new protease repertoire may allow evaluating t

mSphere of Influence: the Importance of Metabolism for Pathogen Adaptation to Host-Imposed Stresses.

Jorge Amich studies several aspects of sulfur and nitrogen metabolism in Aspergillus fumigatus, with the ultimate aim of identifying targets for the development of novel antifungals. In this mSphere of Influence article, he reflects on how "Sub-Telomere Directed Gene Expression during Initiation of Invasive Aspergillosis" (A. McDonagh, N. D. Fedorova, J. Crabtree, Y. Yu, S. Kim, et al., PLoS Pathog 4:e1000154, 2008, https://doi.org/10.1371/journal.ppat.1000154) impacted his thinking about in vivo metabolism and how to investigate it.

Clinical S. aureus Isolates Vary in Their Virulence to Promote Adaptation to the Host.

Staphylococcus aureus colonizes epithelial surfaces, but it can also cause severe infections. The aim of this work was to investigate whether bacterial virulence correlates with defined types of tissue infections. For this, we collected 10⁻12 clinical S. aureus strains each from nasal colonization, and from patients with endoprosthesis infection, hematogenous osteomyelitis, and sepsis. All strains were characterized by genotypic analysis, and by the expression of virulence factors. The host⁻pathogen interaction was studied through several functional assays in osteoblast cultures. Additionally,

Virulence determinants involved in differential host niche adaptation of Neisseria meningitidis and Neisseria gonorrhoeae.

Neisseria meningitidis and Neisseria gonorrhoeae are the only pathogenic species of the genus Neisseria. Although these two species are closely related, they specialized on survival in completely different environments within the human host-the nasopharynx in the case of N. meningitidis versus the urogenital tract in the case of N. gonorrhoeae. The genetic background of these differences has not yet been determined. Here, we present a comparison of all characterized transcriptional regulators in these species, delineating analogous functions and disclosing differential functional developments

Host immunosuppression and antifungal pressure cooperatively shape fungal adaptation and pulmonary microenvironment in cryptococcosis.

Cryptococcus neoformans remains a major opportunistic pathogen causing life-threatening pulmonary and systemic infections in immunocompromised individuals. Effective disease control is challenged by limited therapeutic options and by the pathogen's remarkable ability to adapt under immune and pharmacological pressures. Corticosteroids, while indispensable for managing inflammatory and autoimmune conditions, can disrupt immune homeostasis and worsen fungal infections. To investigate how corticosteroid-induced immunosuppression interacts with antifungal therapy, we employed a murine model of pul

Cracking bacterial persisters: from molecular mechanisms and host adaptation to novel methodologies and countermeasures.

Persisters represent phenotypic tolerance characterized by a MIC value within the susceptible range, along with reduced metabolic activity marked by ATP depletion, a feature likely implicated in persistent and recurrent infections. Although prior research has addressed certain issues associated with persisters, the latest research advances spanning from bacterial physiology to host-pathogen interactions are lacking. Therefore, this review aims to elucidate the definition, distinction, formation mechanisms, host-pathogen adaptation, novel methodologies, and eradication strategies related to bac

The Dual Role of RsiP in Regulating Virulence and Host Adaptation in Bacillus anthracis.

Bacillus anthracis displays susceptibility to penicillin despite harboring a β-lactamase gene, a phenotype governed by the anti-sigma factor RsiP. While RsiP represses σP-dependent β-lactamase expression, its broader roles in physiology and virulence remain unclear. This study aimed to define the global regulatory functions of RsiP beyond antibiotic resistance. Deletion of rsiP significantly upregulated the nprR gene, which is an important quorum-sensing (QS) system regulator and enhanced protease secretion. The ΔrsiP mutant caused higher mortality in cellular and Galleria mellonella models an

Adaptation to an amoeba host drives selection of virulence-associated traits in Vibrio cholerae.

Predation by heterotrophic protists drives the emergence of adaptive traits in bacteria, and often these traits lead to altered interactions with hosts and persistence in the environment. Here we studied adaptation of the cholera pathogen, Vibrio cholerae during long-term co-incubation with the protist host, Acanthamoeba castellanii. We determined phenotypic and genotypic changes associated with long-term intra-amoebal host adaptation and how this impacts pathogen survival and fitness. We showed that adaptation to the amoeba host leads to temporal changes in multiple phenotypic traits in V. ch

Host Cell Tropism and Adaptation of Blood-Stage Malaria Parasites: Challenges for Malaria Elimination.

Plasmodium falciparum and Plasmodium vivax account for most of the mortality and morbidity associated with malaria in humans. Research and control efforts have focused on infections caused by P. falciparum and P. vivax, but have neglected other malaria parasite species that infect humans. Additionally, many related malaria parasite species infect nonhuman primates (NHPs), and have the potential for transmission to humans. For malaria elimination, the varied and specific challenges of all of these Plasmodium species will need to be considered. Recent advances in molecular genetics and genomics

Gene duplications circumvent trade-offs in enzyme function: Insect adaptation to toxic host plants.

Herbivorous insects and their adaptations against plant toxins provide striking opportunities to investigate the genetic basis of traits involved in coevolutionary interactions. Target site insensitivity to cardenolides has evolved convergently across six orders of insects, involving identical substitutions in the Na,K-ATPase gene and repeated convergent gene duplications. The large milkweed bug, Oncopeltus fasciatus, has three copies of the Na,K-ATPase α-subunit gene that bear differing numbers of amino acid substitutions in the binding pocket for cardenolides. To analyze the effect of these

Genomic and phenotypic heterogeneity of Klebsiella aerogenes in a single host: plasmid loss and tissue-specific virulence gene expression during host adaptation.

Klebsiella aerogenes is an opportunistic pathogen increasingly associated with healthcare-associated infections and severe respiratory complications. However, it's in vivo adaptation during host colonization remains poorly understood. We characterized three clonal K. aerogenes ST93 isolates sequentially recovered on days 4, 14, and 19 of hospitalization from a patient with fatal viral pneumonia and concurrent abdominal sepsis. While initially identified generically as Klebsiella spp. by automated clinical systems, Whole-Genome Sequencing and Average Nucleotide Identity confirmed their identity

Structural Receptors and Host Adaptations Affecting the Bacteriophage Targeting of Vibrio Pathogens.

The widespread exposure of multidrug-resistant bacterial strains due to excessive use of antibiotics has accelerated the bacteriophage applications as highly specific antibacterial agents competent to target resistant pathogenic Vibrio through receptor-mediated adsorption and lytic infection. Vibrio pathogens are responsible for substantial economic losses, environmental instability, and foodborne infections in humans through contaminated seafood and water. However, the molecular determinants remain incompletely understood, commanding the Vibrio-phage host specificity, host range, and resistan

Molecular evolution and adaptations of Legionella pneumophila from amoebae hosts to macrophages.

Legionella pneumophila is an environmental bacterium that emerged from a prolonged co-evolution and adaptation to free-living amoebae as the natural hosts. Within these protozoan hosts, L. pneumophila evolved to evade amoebae predation and remodel their vacuoles into endoplasmic reticulum (ER)-derived vacuoles that evade phagosomal-lysosomal fusion. The L. pneumophila-amoebae co-evolution fortuitously has facilitated infection of human alveolar macrophages, resulting in pneumonia known as Legionnaires' Disease. Intracellular replication and host manipulation are orchestrated by the Dot/Icm Typ
