Plasmodium infection and dysbiosis: A new paradigm in the host-parasite interaction.

The mucosal immune system contributes for the largest component of the tissue immune system due to its massive surface area and constant exposure to the microbiota. The gut microbiota comprises a complex micro-ecosystem in the intestine and plays a major role in regulating innate and adaptive immunity. Several studies revealed that infectious diseases involve bidirectional interactions in the gut microenvironment, including changes in the gut microbiota composition. During Plasmodium infection, an increase of pro-inflammatory cells in the lamina propria and a shift in the composition of the gu

MicroRNAs in the Host-Apicomplexan Parasites Interactions: A Review of Immunopathological Aspects.

MicroRNAs (miRNAs), a class of small non-coding regulatory RNAs, have been detected in a variety of organisms ranging from ancient unicellular eukaryotes to mammals. They have been associated with numerous molecular mechanisms involving developmental, physiological and pathological changes of cells and tissues. Despite the fact that miRNA-silencing mechanisms appear to be absent in some Apicomplexan species, an increasing number of studies have reported a role for miRNAs in host-parasite interactions. Host miRNA expression can change following parasite infection and the consequences can lead,

Bovine cryptosporidiosis: impact, host-parasite interaction and control strategies.

Gastrointestinal disease caused by the apicomplexan parasite Cryptosporidium parvum is one of the most important diseases of young ruminant livestock, particularly neonatal calves. Infected animals may suffer from profuse watery diarrhoea, dehydration and in severe cases death can occur. At present, effective therapeutic and preventative measures are not available and a better understanding of the host-pathogen interactions is required. Cryptosporidium parvum is also an important zoonotic pathogen causing severe disease in people, with young children being particularly vulnerable. Our knowledg

An Overview of Trypanosoma brucei Infections: An Intense Host-Parasite Interaction.

Trypanosoma brucei rhodesiense and T. brucei gambiense, the causative agents of Human African Trypanosomiasis, are transmitted by tsetse flies. Within the vector, the parasite undergoes through transformations that prepares it to infect the human host. Sequentially these developmental stages are the replicative procyclic (in which the parasite surface is covered by procyclins) and trypo-epimastigote forms, as well as the non-replicative, infective, metacyclic form that develops in the vector salivary glands. As a pre-adaptation to their life in humans, metacyclic parasites begin to express and

Parasite transmission in social interacting hosts: monogenean epidemics in guppies.

BACKGROUND: Infection incidence increases with the average number of contacts between susceptible and infected individuals. Contact rates are normally assumed to increase linearly with host density. However, social species seek out each other at low density and saturate their contact rates at high densities. Although predicting epidemic behaviour requires knowing how contact rates scale with host density, few empirical studies have investigated the effect of host density. Also, most theory assumes each host has an equal probability of transmitting parasites, even though individual parasite loa

Leishmania cell surface prohibitin: role in host-parasite interaction.

Proteins selectively upregulated in infective parasitic forms could be critical for disease pathogenesis. A mammalian prohibitin orthologue is upregulated in infective metacyclic promastigotes of Leishmania donovani, a parasite that causes visceral leishmaniasis. Leishmania donovani prohibitin shares 41% similarity with mammalian prohibitin and 95-100% within the genus. Prohibitin is concentrated at the surface of the flagellar and the aflagellar pole, the aflagellar pole being a region through which host-parasite interactions occur. Prohibitin is attached to the membrane through a GPI anchor.

Helicobacter pylori and gastroduodenal disease: pathogenesis and host-parasite interaction.

Helicobacter pylori has been shown to be the cause of chronic active gastritis and the evidence that it is involved in the development of peptic ulcer disease and gastric cancer is compelling. Narrow host range, tissue specificity, and chronic inflammation are hallmarks of infection. The study of virulence determinants has just begun but it seems likely that urease, adhesins, cytotoxins, and mediators of inflammation will prove to be important.

Can predators stabilize host-parasite interactions? Changes in aquatic predator identity alter amphibian responses and parasite abundance across life stages.

The role of parasites can change depending on the food web community. Predators, for instance, can amplify or dilute parasite effects on their hosts. Likewise, exposure to parasites or predators at one life stage can have long-term consequences on individual performance and survival, which can influence population and disease dynamics. To understand how predators affect amphibian parasite infections across life stages, we manipulated exposure of northern leopard frog (Rana pipiens) tadpoles to three predators (crayfish [Orconectes rusticus], bluegill [Lepomis macrochirus], or mosquitofish [Gam

A review on the interactions between dendritic cells, filarial parasite and parasite-derived molecules in regulating the host immune responses.

Lymphatic filariasis (LF) is the second leading cause of parasitic disabilities that affects millions of people in India and several other tropical countries. The complexity of this disease is endorsed by various immunopathological consequences such as lymphangitis, lymphadenitis and elephantiasis. The immune evasion strategies that a filarial parasite usually follows are chiefly initiated with the communication between the invaded parasites and parasite-derived molecules, with the Toll-like receptors (TLRs) present on the surface of the antigen-presenting cells (APCs). Classically, the filari

The Trichinella Super-Pangenome Reveals the Evolution of Encapsulation and Predicted Host-Parasite Protein Interactions.

The muscle capsule of Trichinella is a critical structure that impedes immune attacks and drug penetration, yet the molecular mechanisms underlying its formation remain poorly understood. Using a high-quality super-pangenome comprising 12 Trichinella species, we compared extensive genomic variations between encapsulating and non-encapsulating lineages. Pangenomic analysis revealed an open-genome architecture dominated by dispensable genes, with nonencapsulated lineages exhibiting a high load of structural variation (SV). These findings suggest that the capsule represents an evolutionary innova

Host-parasite interactions of the sardine, Sardina pilchardus (Walbaum, 1792) from the Atlanto-Iberian Waters.

The European sardine, Sardina pilchardus (Walbaum, 1792) is one of the most abundant small pelagic clupeids and holds significant commercial value. Adult specimens were obtained along the Western and Southern Iberia, covering three ICES sub-divisions: 9aCN-Northerwestern Iberia, 9aCS-Southern Iberia and 9aS-South Iberia. A parasitological survey was carried out to characterize sardine parasite fauna and to assess its potential as a proxy for the spatial distribution of free-living parasite biodiversity and sardine trophic interactions, based on parasite richness and diversity. Parasites were a

Analysis of the role of Dirofilaria repens macrophage migration inhibitory factors in host-parasite interactions.

INTRODUCTION: Dirofilaria repens is a zoonotic parasitic filarial nematode that infects carnivores and occasionally humans. Knowledge of the host-parasite molecular interactions enabling the parasite's avoidance of the host immune response in subcutaneous dirofilariasis remains limited. Parasitic orthologues of host macrophage migration inhibitory factor (MIF) are molecules potentially involved in this process. MATERIAL AND METHODS: Complementary DNA encoding two D. repens MIF orthologues (rDre-MIF-1 and rDre-MIF-2) was cloned into a pET-28a expression vector. The recombinant proteins were pr

Roles of Glycoconjugates and Mucinases from Carcinogenic Liver Fluke, Opisthorchis viverrini, in Host: Parasite Interactions.

Glycoconjugates, glycoproteins, glycolipids, proteoglycans, and glycoconjugate-modifying enzymes play important roles in host-parasite interactions. In this chapter, we focus on the current knowledge of structures and biological roles of glycoconjugates and M60-like metallopeptidases in survival and pathogenesis of the carcinogenic liver fluke, Opisthorchis viverrini. Understanding this chapter could lead to the development of novel molecular tools for detecting, preventing, and treating parasitic O. viverrini infection.

Multi-omics data elucidate parasite-host-microbiota interactions and resistance to Haemonchus contortus in sheep.

BACKGROUND: The integration of molecular data from hosts, parasites, and microbiota can enhance our understanding of the complex biological interactions underlying the resistance of hosts to parasites. Haemonchus contortus, the predominant sheep gastrointestinal parasite species in the tropics, causes significant production and economic losses, which are further compounded by the diminishing efficiency of chemical control owing to anthelmintic resistance. Knowledge of how the host responds to infection and how the parasite, in combination with microbiota, modulates host immunity can guide sele

Host-parasite interactions in infections due to Entamoeba histolytica: A tale of known and unknown.

Entamoeba histolytica (E. histolytica) is an enteric microaerophilic protozoan parasite responsible for millions of cases worldwide. Majority of the infections due to E. histolytica remain asymptomatic; however, it can cause an array of symptoms ranging from devastating dysentery, colitis, and abscesses in different vital organs. The interactions between the E. histolytica and its host are a multifaceted chain of events rather than merely destruction and invasion. There are manifold decisive steps for the establishment of infections by E. histolytica which includes degradation of mucosal layer

Effects of different roadway deicing salts on host-parasite interactions: The importance of salt type.

The application of roadway deicing salts is increasing the salinity of freshwater systems. Increased salinization from salts, such as NaCl, CaCl2 and MgCl2, can have direct, negative impacts on freshwater organisms at concentrations found in nature. Yet, our understanding of how these salts can indirectly impact freshwater organisms by altering important ecological interactions, such as those between hosts and their parasites, is limited. Using a larval amphibian and infectious free-living helminth (i.e. trematode) model, we examined whether exposure to environmentally relevant concentrations

Synergistic parasite-pathogen interactions mediated by host immunity can drive the collapse of honeybee colonies.

The health of the honeybee and, indirectly, global crop production are threatened by several biotic and abiotic factors, which play a poorly defined role in the induction of widespread colony losses. Recent descriptive studies suggest that colony losses are often related to the interaction between pathogens and other stress factors, including parasites. Through an integrated analysis of the population and molecular changes associated with the collapse of honeybee colonies infested by the parasitic mite Varroa destructor, we show that this parasite can de-stabilise the within-host dynamics of D
